중성자방사선에 피폭된 생쥐에 대한 인삼제제의 효과에 관하여 (The Effects of Korean Ginseng Components for the Mouse Irradiated 1 by Neutron(Besource))

  • When mice irradiated by neutron (Be) are fed with ginseng concentrate, ginseng powder, and adaptagen of which the major ingredient is ginseng alkaloid to neutron (Be source) irradiated mouse, the following results are obtained. 1. The 50% lethal dose (LD50) for the neutron irradiation were 4 days at 600 rad, 7 days at 500 rad, 16 days at 400 rad, 33 days at 375 rad, and 55 days at 350 rad. In thistest, the standard amollntofirradiation was set at 375 rad/8 min. 2. Some spots appeared in the tail of the neutron-irradiated mouse because of blood congestion, and some had its tip tails cut. But the group administered with adaptagen did not show any of these symptoms. 3. The neutron irradiated mouse showed darkening the color of their lung-chloasmas while none of the adaptagen group had this symptom. 4. The lung tissue of the neutron irradiated mouse showed an increase of the karyolysis and cytoplasmic vacuole. 5. When both neutron irradiation and the ginseng sllbstances were given to the mouse at the same day, the 50% lethal days were increased to 29-33 days for the group administered with ginseng extract. 67 days for the group given with the ginseng powder. and 80 days for the groilp arith the adaptagen. 6. The survival rate of those fed with adaptagen for 33 days before the neutron-irradiation was 100%, while the 50% lethal daysofthe group fed with ginsengextract were 39 days and that of the group fed with ginseng powder were 69 days. 7. The serum valued of ${\gamma}$-globulin, IgG, and albumin were returned to normal condition in the group fed with adaptagen for 33 days before the neutron-irradiation. But those of the group which were given the irradiation and the ginseng substances at the same day did not show such a recovery.

X-방사선($^{60}Co$)에 조사된 새앙쥐의 상해에 대한 "아답태겐"$^{\textregistered}$의 효과에 관한 연구 (A Study on the Effect of "ADAPTAGEN"$^{\textregistered}$ Korean Ginseng Components, for the Injured Mouse by X-ray($^{60}Co$) Irradiation)

  • The results ok feeding experiments to the mice with ginseng extract, ginseng Powder, and ADAPTAGEN, for 30 days before X-ray irradiation and for 40 days after the X-ray irradiation at 750 rads were as follow: 1. The 50% lethals days (LD50, ) by the X-ray irradiation were 9 days at 1, 000 rads. 10 days at 900 rads, 11 days at 800 rads, 14 days at 760 rads, and 19 darts at 750 rads. Therefore, the standard radiation dose was set at 750 radb/8 min. 2. The 80% of the control group mice exposed to the X-ray radiation without ginseng feeding died in periods ranging from 14 to 24 days and the 20~30% of the ginseng extract and ginseng powder feeding groups died. But the 100% of the mice fed with ADAPTAGEN survived. 3. Testicles of the control group became smaller in weight than the nomad group by 26.5 to 29.0% and those of the ginseng extract and ginseng powder feeding group reduced by 44.6 to 60.4%. However, testicles of the ADAPTiIGEN feeding group increased in size by 77.4% to 87.1% and in weight by 61%, showing a recovery phenomenon approarhing to those of the ordinary mice. The ADAPTAGEN feeding group mice were also as active in color as the ordinary ones. 4. An electron micrograph(X8, 000X2.2) of the liver cells of the mice which had been 40 days after X-ray irradiation showed as follows; The control group appeared that is physiological action stopped due to the frequent occurrence of morphological change of the nucleus and diffusion of chromosome, reduction in microspores and expansion of microsomts, and endoplasmic change of mitochondria. The liver cells or the ADAPTAGEN feeding group were in a state similar to those of the ordinary mice restoring to normalcy In contrast, the liver cells of the ginseng extract and ginseng powder feeding groups were still far from being normal. 5. A serological analysis showed that the control group sharply decreased in albumin, Y-g1obu1in, and IgG so far as to cause dystrophy and to weaken antibody resistance but that ginseng extract and ginseng powder feeding groups, though in a little more restoring state than the control group, were still far from the normal group. The ADAPTAGEN feeding group restored to a state as comparable to the normal group in the contents of albumin ${\gamma}$-globulin, IgG and serum protein. In order words, it is noteworthy that ADAPTAGEN feeding was effective in revitalizing the destroyed cells of a living body and that it has the function of normalizing antibody components.

  • Morphine produces wide-spread immunesuppression, such as lymphokine production, phagocytic activity, natural killer cell activity, cell proliferation, and cell-mediated immunity. On the other hand, one of the representative pharmacological act ions of Panax ginseng is immune enhancement. In this study, we investigated the effects of ginseng total sponin (GTS), and Adaptagen (trade name of ginseng product) on morphine-induced immunesuppression. Morphine hydrochloride (10 mg/kg, sc), GTS (400 mg/kg, oral), Adaptagen (400 mg/kg, oral ) were administered to mice for 14 consecutive days.

  • 본 연구에 사용한 인삼 성분 중 Adaptagen(ADT), Ginseng Extract(GE) Ginseng Total Saponin(GTS)는 자발 운동을 각각 억제하였다. GE alc GTS는 Morphine 만성 투여에 의한 내성 및 의존성 형성을 억제하였으며, morphine에 의한 자발운동과 연속 투여에 의한 자발운동의 증강작용을 억제하였다. 또한 morphine의 만성 투여로 인한 Dopamine 수용체 초과민성을 억제하였다.

  • The effects of ginseng prepation, Adaptagen$\^$R/ (AD), on the central dopaminergic nervous system in the learning-impaired rats were studied. The learning impaired rats were rendered by the intracerebroventricular infusion of ethylcholine aziridium (AF64A), 3 nmol/each side. Three days after the infusion of AF64A, AD were orally intubated daily for five days, 200 mg/kg. The control groups were intubated with distilled water. Twenty four hours after the last intubation, The changes in the specific bindings of dopamine receptors, the concentrations of dopamine (DA) and metabolites, The activities of tyrosine hydrosylase (TH) and monoamine oxidase (MAO) were analyzed using receptor radiography, HPLC-ECD and the methods in enzyme-assays, respectively.

방사선 세포 손상에 대한 인삼 Alkaloid 분획의 보호 효과에 관한 실험적 연구 (An Experimental Study of Radioprotective Effect of Ginseng Alkaloid Fraction on Cellular Damage)

  • 본 연구에서는 인삼의 알카로이드 분획 (alkaloid fraction)을 주요성분으로 함유하는 Adaptagen의 방사선 방어제로서의 효과를 구명(究明)하기 위하여, 방사선조사에 의한 마우스 공장 소낭선 세포 손상 및 회복을 측정하고, 마우스 비장임파구 미세핵 측정법을 시험관 내 및 생체에서 실시함으로써 본 제재의 방사선 방어효과를 평가하였으며, 실험결과 얻어진 결론은 다음과 같다. 1. 마우스 공장소낭선세포의 방사선 손상은 대조군에 비해 alkaloid 분획 투여시와 수용성 분획 투여시 공히 감소하였으나, alkaloid 분획 투여시 더욱 현저하였다. 2. 마우스 공장소낭선세포 손상의 회복 및 증식은 대조군에 비해 alkaloid 분획 투여시와 수용성 분획 투여시 공히 현저히 증가하였다. 3. 시험관내 시험에서 방사선에 의한 임파구 미세핵 형성도는 쌍핵세포의 형성율이 낮아 통계학적 의의는 없으나 약제 투여 군에서 빈도가 낮은 경향을 보였으며 alkaloid 분획 투여시 더욱 현저하였다. 4. 생체내 시험에서 방사선에 의한 임파구 미세핵 형성빈도는 대조군에 비해 alkaloid 분획 주여시와 수용성 분획 투여시 공히 감소하였으나 alkaloid 분획 투여시 더욱 현저하였다. 이상의 결과에서 인삼의 alkaloid 및 수용성 분획이 방사선에 의한 염색체 손상을 억제하고 손상된 세포의 회복 (repair) 및 증식 (regeneration)을 촉진하여 방사선 방어효과를 나타냄을 알 수 있었으며, 이는 독성이 비교적 없는 자연산생물 (natural products)로서 방사선방어제로 임상에 직접 사용 할 수 있다.
