Subchronic Inhalation Toxicity of Trichloroacetonitrile on the Sprague Dawley Rats

  • Trichloroacetonitrile is used as an intermediate in insecticides, pesticides, and dyes. In Korea alone, over 10 tons are used annually. Its oral and dermal toxicity is classified as category 3 according to the globally harmonized system of classification and labelling of chemicals, and it is designated a toxic substance by the Ministry of Environment in Korea. There are no available inhalation toxicity data on trichloroacetonitrile. Thus, the present study performed inhalation tests to provide data for hazard and risk assessments. Sprague-Dawley rats were exposed to trichloroacetonitrile at concentrations of 4, 16, or 64 ppm for 6 hour per day 5 days per week for 13 weeks in a repeated study. As a result, salivation, shortness of breath, and wheezing were observed, and their body weights decreased significantly (p < 0.05) in the 16 and 64 ppm groups. All the rats in 64 ppm group were dead or moribund within 4 weeks of the exposure. Some significant changes were observed in blood hematology and serum biochemistry (e.g., prothrombin time, ratio of albumin and globulin, blood urea nitrogen, and triglycerides), but the values were within normal physiological ranges. The major target organs of trichloroacetonitrile were the nasal cavity, trachea, and lungs. The rats exposed to 16 ppm showed moderate histopathological changes in the transitional epithelium and olfactory epithelium of the nasal cavity. Nasal-associated lymphoid tissue (NALT) and respiratory epithelium were also changed. Respiratory lesions were common in the dead rats that had been exposed to the 64 ppm concentration. The dead animals also showed loss of cilia in the trachea, pneumonitis in the lung, and epithelial hyperplasia in the bronchi and bronchioles. In conclusion, the no-observed-adverse-effect level (NOAEL) was estimated to be 4 ppm. The main target organs of trichloroacetonitrile were the nasal cavity, trachea, and lungs.

  • Environmental toxicities of priority water pollutants were evaluated by two selected bioassays, Lettuce seed germination/elongation test and Microtox acute toxicity test. Toxic chemicals (heavy metals, polycyclic aromatic hydrocarbons, and phenolic compounds) inhibited the germination rate and root elongation of Lettuce seed, as well as the bioluminescence of Microtox bacteria. When test biota were exposed to target chemicals, the sensitivity of Lettuce bioassay was relatively lower than that of Microtox bioassay. However, Lettuce bioassay may be a good candidate for prescreening the environmental toxicities of priority water pollutants, since the testing method with Lettuce seed was relatively easier and more economic than with Microtox bacteria. Toxicity tests were conducted to compare the validity and sensitivity of both bioassays for sediment from a small stream passed through urban area as well as leachate from a municipal solid waste landfill. From experimental results, we found that Lettuce test and Microtox test are compensated each other as a battery of bioassay for evaluating the environmental toxicities of field samples obtained from a small stream contaminated by pollutants.

  • A ginseng preparation (GP) consisting of ginseng ex., lycium fructus ex., four kinds of vitamines and caffein was evaluated for acute toxicity and general pharmacology. Average lethal doses $(LD_{50})$ of GP in male mice were 2,988mg/kg (i.p.) and more than 3g/kg (p.o.). In dosage of 300 and 900mg/kg (p.o.) showed no analgesic activity in both tests of the writhing method induced by acetic acid and of tail pressure method and no effect on the pentetrazole-induced convulsion. However, it appeared to have a hypothermic action only in dose of 900mg/kg. The duration of hypnosis induced by hexobarbital sodium in mice was shortened by GP, being probably due to caffein. GP Produced no marked contraction of isolated ileum and uterus in high concentrations of up to $1{\pm}10^{-3}g/ml$. These results suggested that GP did not show any considerable central nervous depressant activity and exhibited very weak actue toxicity in mice.

  • To examine safety of Ganoderma lucidum, it was extracted with hot water (Fraction A). After the extract was dialyzed and freeze-dried, a polysaccharide fraction (Fraction B) was obtained and examined for acute and subacute toxicity. In the acute toxicity tests of Fr. A and Fr. B on mice, both agents did not show any serious and lethal effects. The results showed that 50% lethal doses were higher than 5,000 mg/kg. The experiments of oral administration of Fr. A (5,000 mg/kg) to mice for 30 days showed that there were no changes in body weight, hematological features and organ weight.

  • Tetramine toxicity due to sea snail ingestion is generally mild and has a good prognosis. Tetramine toxicity acts on the acetylcholine receptor, affecting the neuromuscular junction and autonomic nervous system. A 78-year-old female patient visited the emergency room with vomiting and dyspnea after eating sea snails. At the time of admission, the vital signs recorded were 140/80 mmHg-105/min-24/min-36.5℃, and 90% oxygen saturation. Arterial blood test revealed hypercapnia (pCO2 58.2 mmHg) and respiratory acidosis (pH 7.213, HCO3- 22.5 mmol/L), whereas other blood tests showed no specific findings. Due to decreased consciousness and hypoxia, endotracheal intubation and mechanical ventilation were administered to the patient. Successful weaning was accomplished after 12 hrs, and the patient was discharged without any further complications. Although tetramine toxicity rarely results in acute respiratory failure due to paralysis of the respiratory muscle, caution is required whilst treating the patient.

Effects of Size, Impurities, and Citrate Capping on the Toxicity of Manufactured Silver Nano-particles to Larval Zebrafish (Danio rerio)

  • Objectives: This study was conducted to identify factors determining the toxicity of manufactured silver nano-particles (AgNPs) on aquatic organisms. Methods: For this purpose, we prepared several AgNPs with varied characteristics, including hydrodynamic size (nano-$^{ABC}Ag^{Cit}\;vs$-sized-$^{ABC}Ag^{Cit}$), impurities ($^{ABC}Ag$ stock vs $^{ABC}Ag$), and citrate capping ($^{ABC}Ag^{Cit}$), using a commercially available manufactured AgNP ($^{ABC}Ag$ stock). Acute tests were conducted using larval zebrafish (Danio rerioI). In addition, in order to determine the ecotoxicological potentials of various capping agents, toxicity tests were conducted with microbes, waterfleas, and fish for eight different capping agents that are used for NPs. Results: The toxicity of AgNPs in terms of 96 h fish $LC_{50}$ increased in the following order: $^{ABC}Ag$ stock < $^{ABC}Ag=^{ABC}Ag^{Cit}=nano-^{ABC}Ag^{Cit}$ < ${\mu}$-sized-$^{ABC}Ag^{Cit}$ < $AgNO_3$. After removing impurities by dialysis, 96 h $LC_{50}$ value decreased significantly from $126.6{\mu}g/L$ (95% confidence intervals [CI]: 107.0-146.2) ($^{ABC}Ag$ stock) to $78.6{\mu}g/L$ (CI: 72.7-84.8) ($^{ABC}Ag$). For ${\mu}$-sized-$^{ABC}Ag^{Cit}$ (ranging between 3.9 and 40.6 nm) and $^{ABC}Ag^{Cit}$ (40.6 nm and $9.1{\mu}m$), the 96 h $LC_{50}$ of the former ($43.9{\mu}g/L$, CI: 36.0-51.7) was approximately two-fold lower than that of the latter ($87.0{\mu}g/L$, CI: 73.5-100.3). Conclusions: In this study, we found that for acute lethality, the contribution of impurities and particle size was significant, but that of citrate was negligible.

Toxicity Test of butachlor to Medaka (뷰타클로르의 송사리에 대한 독성시험)

  • In order to evaluate the toxic effects of butachlor, a herbicide widely used for control of weeds in paddy field, on medaka (Oryzias latipes), acute toxicity tests for five developmental stages and early life stage toxicity test of were conducted. As the results of acute toxicity test, $96h-LC_{50}s$ for 1 day, 1 week, 2 weeks, 2 months and 5 months after hatching of O. latipes were 0.68, 0.52, 0.38, 1.09 and $0.45\;mg\;L^{-1}$, respectively. This indicated that the most sensitive stage was 2 weeks after hatching. The early life stage toxicity test showed that no statistically significant hatching period and hatching success of embryo was observed at all concentrations of butaclor. However, 0.05 and $0.1\;mg\;L^{-1}$ of butachlor showed statistically significant post hatching survival with p<0.1. Abnormalities of larva were 2.1, 2.3 and 10% at 0.025, 0.05 and $0.1\;mg\;L^{-1}$ of concentration, respectively. They showed abnormal vertebral axis, craniofacial alteration and retarded yolk-sac resorption. The total length and weight were decreased depending on butachlor concentration the end of test. Weight of larva was showed more sensitive toxic indicator than total length. The toxicological responses of O. latipes to butachlor expressed as LOEC(lowest observed effect concentration), NOEC(no observed effect concentration) and MATC(maximum acceptable toxicant concentration) values were 0.025, 0.013 and $0.018\;mg\;L^{-1}$, respectively.

A Study on the Applicability of Acrylic Water Leak Repair Materials used to Repair Cracks in Conduits and Underground Structures (관거 및 지하구조물 균열 보수에 사용되는 아크릴 누수 보수재의 적용성에 대한 연구)

  • Various injection materials, such as asphalt-based injection materials, urethane-based injection materials, cement- based injection materials, and acrylic-based injection materials, are used for the repair of aged conduits and underground structures with cracks. In this study, research was conducted on an environmentally friendly acrylic- based leak repair material that exhibits good curing properties even in humid conditions and stability in temperature fluctuations. To compare the performance of the improved acrylic leak repair material with the existing acrylate injection material, experiments were conducted using KS standard methods, including underwater length change rate tests, underwater leakage resistance tests, and chemical performance tests. The comparative experiments revealed that the improved acrylic leak repair material showed no changes in shrinkage due to humidity, temperature variations, or chemical reactions compared to the existing acrylate injection material. In the underwater resistance test, the improved acrylic leak repair material did not show any leakage. Additionally, to assess the environmental impact of the improved acrylic leak repair material, acute fish toxicity tests and acute oral toxicity tests were conducted, and the results showed no mortality and no specific concerns with the test specimens. The experimental results led to the conclusion that the improved acrylic leak repair material is considered to be superior in performance, environmentally safe, and harmless to the human body. Based on various experimental results, it is inferred that the improved acrylic leak repair material is suitable for use as a repair material for cracks in manholes and underground structures compared to the existing acrylate repair material. This study aims to propose valuable data for future technological development by evaluating the applicability of acrylic leak repair materials.

Venlafaxine-Induced Acute Toxic Hepatitis (Venlafaxine에 의한 급성 독성 간염 1예)

  • Venlafaxine is among the most widely prescribed antidepressants. It is extensively metabolized to O-desmethylvenlafaxine via cytochrome P450 (CYP) 2D6. We report a case of acute toxic hepatitis resulting from venlafaxine in a 54-year-old woman with pain disorder. During venlafaxine treatment, laboratory tests revealed elevated liver enzymes with a maximum of 169 IU/L for aspartate transaminase (AST) and 166 IU/L for alanine transaminase (ALT). AST and ALT levels returned to normal after 6 days of discontinuation of venlafaxine. The patient was finally diagnosed with acute toxic hepatitis through liver biopsy. This case indicates the importance that clinicians should be aware of the hepatotoxicity of venlafaxine in practice.

인삼, 한국 및 중국산 산양삼의 간 보호 효능에 관한 실험적 연구

  • Objectives : This study was aimed at investigating liver protection mechanism of Cultivated Ginseng and Cultivated Wild Ginseng of Korean and Chinese by inducing liver toxicity through $CCl_4$ and t-BHP in mice and evaluated serological findings. Methods : Experiment groups was categorized into untreated normal group, treated control group, and orally administered Cultivated Ginseng and Cultivated Wild Ginseng of Korean and Chinese experimental groups. At the termination of experiment, gross examination of the liver as well as Total bilirubin, AST, and ALT contents in the serum were evaluated. Results : 1. In the $CCl_4$ induced acute hepatotoxicity test, total bilirubin, AST and ALT didn't show significant differences between the control and experimental groups. 2. In the t-BHP induced acute hepatotoxicity test, total bilirubin, AST and ALT didn't show significant differences between the control and experimental groups. Conclusion : Taken together, Cultivated Ginseng and Cultivated Wild Ginseng of Korean and Chinese cannot be effectively used for recovering the liver functions in acute hepatotoxicity tests using $CCl_4$ and t-BHP. Further researches, for example treated long period, must be tried to verify the efficacies.
