  • The plant of Pedicularis resupinata var. oppositifolia(Scrophulariaceae) is described to be used for treatment of rheumatoid arthritis, malignant abscess (tumor), urolith and diuretics in oriental medicinal books. This study is concerned with some general pharmacological activities. It was observed that the methanol extract of the aerial part of this plant showed some effects on acute toxicity, carrageenin induced edema in rat paw, bile juice secretion and antibacterial effect. From this result it was obtained that the methanol extract revealed the antiinflammatory activity and bile juice secretion increasing effect. Thus, it was fractionated and buthanol fraction has obvious inhibition effect on edema and bile juice secretion increasing effect. The total extract showed low acute toxicity i.e., the minimum lethal dose was more than 8000mg/kg in oral administration in mice.

Study on Inhalation Toxicity of 1-Hexene in SD Rats (SD Rats를 이용 1-hexene의 흡입독성 연구)

  • The purpose of this study was to investigate the acute (4 hours) and repeated-dose (6 hours a day, 5 days a week, 4 weeks) toxic effects of 1-hexene on Sprague-Dawley (SD) rats which were treated by inhalation. The results were as follows; 1. The median lethal concentration(LC$_{50}$) was estimated 52,694 ppm (confidence limit 95%; 49,494~55,447 ppm) in acute inhalation. Abnormal clinical signs related to the 1-Hexene were not observed with the acute inhalation dose. Cross findings of necropsy revealed on evidence of specific toxicity related to the 1-hexene. II. By repeated inhalation exposure the body weight of male were more or less reduced by the dose of 2,500 ppm and 5,000 ppm compared with control group. However there were no significant variation hematology and blood biochemistry for the exposed rats compared with the control rats. Abnormal clinical signs and gross findings of necropsy related to the 1-hexene were not shown. In conclusion when we exposed 1-hexene to SD rats for 4 weeks, 5 days per week, 6 hours per day, the Lowest observed effect level (LOEL) was over 2,500 ppm and Non observed effect level (NOEL) was below 500 ppm.

Single and Four-week Intravenous Toxicity Studies of DA-3585, a Recombinant Human Erythropoietin, in Rats (재조합 사람 적혈구 조혈인자 DA-3585의 랫드에 대한 단회 및 4주반복 정맥투여 독성시험)

  • DA-3585 is a recombinant human erythropoietin produced by Dong-A pharmaceutical Co. Ltd. using recombinant DNA technique. Recently, recombinant human erythropoietin (rHu-EPO) has been used to treat various types of anemia. In this study, we examined acute and subacute toxicity of DA-3585 in rats. DA-3585 was intravenously administered to rats at dose levels of 0, 6,250, 12,500 and 25,000 lU/kg for single dose toxicity study and at dose levels of 0,100,500 and 2,500IU/kg daily for 4 week-repeated dose toxicity study. In the single dose toxicity study, there were no death, clinical signs and changes in body weight gain related to the treatment. Necropsy revealed no evidence of toxicity related to DA-3585, In the repeated dose toxicity study, all the rats survived throughout the study. There were no treatment-related changes in clinical signs, food and water intake, and body weight. Hematological examination showed increases in the number of erythrocytes, hemoglobin concentration, hematocrit value and mean corpuscular volume, and decrease in the number of platelet in 500 and 2,500 lU/kg dosed groups. Extramedullary hematopoiesis in the spleen and erythroid hyperplasia in the bone marrow were noted as treatment-related histological changes. Toxicologically significant changes were not observed in blood biochemistry, urinalysis, organ weights and in any other examinations. The treatment-related changes observed in this study were hematological or histological changes associated with pharmacological effects of DA-3585. On the basis of the results of this study, LD5n value of DA-3585 was above 25,000 lU/kg and the no-observed-adverse-effect-level was estimated to be 100 lU/kg.

A Review on the Classification of Skin Toxicity Hazards Due to Skin Contact with Chemical Substances (화학물질 피부접촉에 의한 피부독성 유해성 분류에 관한 고찰)

  • Objectives: In this study, we analyze statistics on industrial accidents caused by chemical skin contact and provide skin toxicity hazard information on the related domestic system and circulation volumes. Methods and Results: We analyzed occupational fatalities and skin diseases caused by chemical leaks and contact from 2007 to 2016(10 years) and surveyed data on occupational skin diseases using the 2014 work environment survey data. The NIOSH Skin Notation Profiles for 57 chemical substances, which are provided to prevent occupational skin diseases, were searched and hazard information on skin contact with chemical substances was classified. In order to identify skin toxicity information among domestically distributed and legally regulated substances and to investigate skin-toxic substances, MSDS basic data on 19,740 chemical substances provided on the homepage of Korea Occupational Safety & Health Agency were searched. Acute toxicity(dermal) category 1-4 substances totaled 1,020, and the number of chemical substances classified as category 1 and 2 substances were 135 and 137, respectively. In the chemical substances prescribed by the Ministry of Employment and Labor, 173 substances were classified into acute toxicity(dermal) categories 1-4, 58 of which correspond to category 1 or 2. Conclusions: Within the present range of industrial accidents, the proportion of skin diseases due to contact with chemicals is not high. However, there is always a risk of occupational skin diseases due to increasing chemicals and due to the use of new chemicals. It is hoped that this information will be used by workplace safety and health officials and health and safety experts to prevent acute toxity(dermal) due to chemical skin contact.

TOXICOLOGICAL STUDIES ON RAW AND PROCESSED (PREBREWED) ACONITI TUBERS; ACUTE, SUBACUTE TOXICITY STUDIES AND ASSAY OF ACONITINE ALKALOIDS (生附子와 修治附子에 관한 毒性연구 : 급성 및 아급성 독성과 Aconitine 알칼로이드 함량분석)

  • Aconiti Tuber is the root of Aconitum sp (Ranunclaceae) which has been considered as one of the most important medicinal plant having cordiotonic, diuretic and analgesic effect. On the other hand, it has been known that Aconiti Tuber contained toxic agent, aconitine alkaloids so that only processed Aconiti Tubers have been used as herbal drug traditionally. For the safety evaluation of processed Aconiti Tuber, quantitative determination of aconitine and acute, subacute toxicity test were performed on 5 commercial processed Aconiti Tubers. Arapid and precise method using HPLC has been developed for the separation and determination of aconitine. Samples were extracted with hydrochloric acid (pH3) and hot water decoction. In case of d-HCL extracts, the contents of aconitine were from 0.08 mg/g to trace. But in case of hot water decoction extracts, the contents of aconitine were not detected. For the investigation of Aconiti Tuber toxicity in rats, hot water decoction samples and methanol extracts were tested. 1) Acute toxicity test Hot water decoction sample and methanol extracts from Aconiti Tuber did not show any toxic effects in rats by an oral administration. $LD_50values of 2 extracts were above 10.0 g/kg. 2) Subacute toxicity study In the repeated administration study, hot water decoction samples were given orally to Sprague-Dawlay rats for 2 week at daily doses of 5.0 g/kg. The results are as follows; No toxic manifestation, body weight changes and lethality were observed during wxperimental period. There were no significant changes in serum enzyme activities such as GOT, GPT, LDH, ALP between treated and control groups. However CPK values were decreased in the Subuja-treated group. (P<0.01). In addition, no gross and microscopic changes were noted in Aconiti Tuber-treated groups.

A Study on the Acute Toxicity of Leejung-tang(Lizhong-tang) and Fermented Leejung-tang(Lizhong-tang) Extract in ICR Mice (ICR 마우스를 이용한 이중탕 및 발효이중탕의 급성독성 연구)

  • Objectives : This study was carried out to investigate the acute toxicity and safety of $Leejung$-$tang$($Lizhong$-$tang$) and fermented $Leejung$-$tang$($Lizhong$-$tang$) extract. Methods : To evaluate their acute toxicity and safety, 0(control group), 1250, 2500 and 5000 mg/kg of $Leejung$-$tang$($Lizhong$-$tang$) and fermented $Leejung$-$tang$($Lizhong$-$tang$) extracts were orally administered to 35 male and 35 female ICR mice. After a single administration, we observed survival rates, general toxicity, changes of body weight, and autopsy. Results : Compared with the control group, we could not find any toxic alteration in any of treated groups (1250, 2500 and 5000 mg/kg). Conclusions : $LD_{50}$ of $Leejung$-$tang$($Lizhong$-$tang$) and fermented $Leejung$-$tang$($Lizhong$-$tang$) were over 5000 mg/kg and it is very safe to ICR mice.

Acute Toxicity Study on Socheongryong-Tang and Fermented Socheongryong-Tang extracts in Mice (소청룡탕 및 발효 소청룡탕 추출물의 단회투여 독성에 관한 연구)

  • Objectives : Socheongryong-Tang (小靑龍湯, SCRT) has been widely used to treat respiratory disease. In this study, we investigate the acute toxicity and safety of fermented Socheongryong-Tang extract. Methods : To evaluate the acute toxicity and safety, 0 (control group), 1250, 2500 and 5000 mg/kg of Socheongryong-Tang and fermented Socheongryong-Tang extracts were orally administered to male and female ICR mice. After single administration, we observed survival rates, general toxicity, changes of body weight for the 14 days and autopsy. Results : Compared with the control group, we could not find any toxic alteration in all treated group (1250, 2500 and 5000 mg/kg). Conclusions : $LD_{50}$ of Socheongryong-Tang and fermented Socheongryong-Tang extracts might be over 5000 mg/kg and it is safe to ICR mice.

Correlation and Sensitivity of Acute Toxicity of Pesticides on the Common Carp(C. carpio) and Killifish(O. latipes) (농약에 대한 잉어(C. carpio)와 송사리(O. latipes)의 급성독성 상관성 및 감수성)

  • The recommended species in fish acute toxicity test for pesticide registration in korea are common carp (Cyprinus carpio), killifish(Oryzias latipes) and loach(Misgurnus anguillicaudatus). But most of fish toxicity was evaluated using common carp. The common carp has several problems such as difficulty of room breeding, difference of sensitivity to breeding condition, limited experimental period, etc. For that reason, it is necessary to reexamine of test species. The present study investigated the sensitivity and correlation of $LC_{50}$ values that determined using the acute toxicity test with common carp and killifish. And this study is a realistic approach to evaluated appropriate species for fish acute toxicity test of pesticide. As a result, correlation coefficient(R) of the commom carp and killifish was high correlation of 0.8480 and the $LC_{50}$ values was not significant in the interspecies sensitivity. When the $LC_{50}$ values of common carp and killifish was compared with fish toxicity guideline in Korea, fish toxicity class has fluctuated in 18 pesticides with not very special tendency. Therefore, it is necessary to review of killifish has more advantage to standardization of fish culture and in the side of correlation and sensitivity of acute toxicity.

Assessment of Contact and Oral Toxicity of Four Neonicotinoid Insecticides to Bumblebees (Bombus terrestris) (네오니코티노이드계 4종 농약의 서양뒤영벌 급성 접촉 및 섭식 독성평가)

  • BACKGROUND: Bumblebees have been shown to be very effective pollinators for most greenhouse tomatoes. Neonicotinoid insecticides are one of the most widely used pesticides in tomato crops in Korea. METHODS AND RESULTS: This study was carried out to investigate the toxicity of four neonicotinoid insecticides (clothianidin, dinotefuran, imidacloprid and thiamethoxam) to bumblebees based on the OECD guidelines (No.246, 247). The 48 hr LD50 (㎍ a.i. /bumblebee) values in the acute contact toxicity tests were determined as follows: clothianidin, 0.467; dinotefuran, 3.741; imidacloprid, 3.967; and thiamethoxam, 0.747. The 48 hr LD50 values in the acute oral toxicity tests were determined as follows: clothianidin, 0.005; dinotefuran, 0.056; imidacloprid, 0.325; and thiamethoxam, 0.018. The acute contact and oral toxicity of the test insecticides to bumblebees from most to least toxic was clothianidin > thiamethoxam > dinotefuran > imidacloprid. CONCLUSION: This study provided the basic toxicological data of neonicotinoid insecticides for bumblebees. In the near future, acute toxicity and mixture toxicity of other pesticides to bumblebees could be determined using this method.

The Study on Acute and Subacute Toxicity and Sarcoma-180 Anti-cancer Effects of Triglii Semen Herbal-acupuncture (파두약침(巴豆藥鍼)의 급성(急性) 아급성(亞急性) 독성실험(毒性實驗) 및 Sarcoma-180 항암효과(抗癌效果)에 관(關)한 실험적(實驗的) 연구(硏究))

  • Objective: The purpose of this study was to investigate the acute and subacute toxicity and sarcoma- 180 anti-cancer effects of Herbal acupuncture with Triglii Semen in mice and rats. Method: Balble mice were injected intraperitoneally with Triglii semen Herbal acupuncture for $LD_{50}$ and acute toxicity test. Sprague Dawley rats were injected intraperitoneally with Triglii semen Herbal acupuncture for subacute toxicity test. The Triglii semen Herbal acupuncture was injected on Chung-wan(CV12) of mice with S-180 cancer cell line. Results: 1. In acute toxicity test, the $LD_{50}$ value was $7.49{\times}10^3$ml, 0.30ml/kg.2. The body weights of mice treated with Triglii semen Herbal acupuncture increased during the acute toxicity test. 3. In acute toxicity test of serum biochenrical values of mice, total protein was decreased in treatment groups I, 2 and 3, albunrin was decreased in treatment groups 2 and 3 compared to the control group. GOT was increased in treatment group I and Alk. Phosphatase was increased in treatment groups 1,2 and 3 compared to the normal group(p<0.05). 4.ln subacute toxicity test, severe tissue injury was found in lung and liver. 5. In subacute toxicity test, the body weight was decreased in treatment groups I and 2 compared to the normal group and the weight of liver. lung and kidney were increased in treatment groups 1, 2 and 3 compared to the normal group.(p<0.05) 6. In subacute toxicity test, RBC, HGB and HCT were decreased in treatment groups 1 and 2 compared to the normal group. MCV was increased in treatment group1 compared to the normal group, MCH was increased in treatment groups 1 and 2 compared to the control group in complete blood count test.(p<0.05) 7. In subacute toxicity test, total protein was decreased in treatment groups 1 and 2 compared to the nonnal group, BUN was increased in treatment groups 1 and 2 compared to the nonnal group, creatinine and uric acid were decreased in treatment groups 1 and 2 compared to the normal group, glucose was increased in treatment group 2 compared to the nonnal group, triglycelide was decreased in treatment groups I and 2 compared to the normal group, total cholesterol was increased in treatment groups 1 and 2 compared to the control group. GOT was decreased in treatment group 2 compared to the normal and control group, AIk. Phosphatase was increased in treatment group 1 compared to the normal and control group.(p<0.05) 8. Median survival time was 17days in treatment group 2 for S- 180 cancer cell treated with Triglii semen Herbal acupuncture. 9. Natural killer cell activity was insignificant for S-180 cell treated with Triglii semen Herbal acupuncture.(p<0.05) 10. lnterieukin-2 productivity was decreased for S-180 cell treated with Triglii semen Herbal acupuncture compared to the normal and control group.(p<0.05) Conclusion: According to the results, we can conclude Herbal-acupuncture with Triglii semen caused toxicity, and caused no effects in S-180 cancer cell.
