  • S. fradiae exhibited the highest acetanilide p-hydroxylation activity among the Streptomyces spp. screened. Studies with inhibitors (metyrapone, 2. 6-dichloroindophenol, $\alpha,\alpha'$-dipyridyl, o-phenanthroline) and an absorption peak after CO treatment suggested that S. fradiae hydroxylase activity was due to cytochrome p-450. This hydroxylase activity was increased to ten times in the cell extract containing 0.5 mM sodium azide. Furthermore, the sedimentary activity in $105,000\times{g}$ centrifugal forces and solubilization of the activity with Triton-X 100 implied that this enzyme was membrane bound monooxygenase. pH Optimum of the enzyme was 6.5 in membrane bound state.

  • This study investigated characteristics of daily life activity experiences and their relations to stress in early adolescence. A sample of 134 8th graders carried electronic watches programmed to signal 7 times a day for a week at random times and reported their daily life activity experiences when signaled. Boys spent 33% of their waking hours in schoolwork, 46% in leisure, and 21% in maintenance activities. Girls spent 32% in schoolwork, 43% in leisure, and the other 25% in maintenance. Both boys and girls experienced more positive affect, cognitive, and motivation during leisure, whereas more negative psychological states during schoolwork. The major stress was schoolwork-related. The stress was significantly related to objective and subjective experiences of daily life activity.

  • The time-kill curves of LB20304, a novel fluoroquinolone that has potent antibacterial activity against gram-positve and gram-negative bacteria, were calculated at the concentrations of 1/4-, 1/2-, 1-, 2- and 4-times the MIC against Staphylococcus aureus 77, Escherichia coli 3739E, Pseudomonas aeruginosa 1912E. The bactericidal activity of LB20304 for these strains was very rapid and comparable to that of ciprofloxacin. LB20304 produced a decrease in the $log_10$ CFU per milliliter of${\geq}$3 within 2 h at 4-times the MIC for all strains and consitently prevented regrowth of bacteria after 24 h of incubation. The MBCs (Minimal Bactericidal Concentration) of LB20304 against test organisms were equal to or at most four-times higher than the MICs.

  • The anaerobic digester with sludge from sewage treatment plant was operated in the laboratory for two year to investigate the enumeration and activity of methanogenic microorganisms. In this experimental study, the effects of HRT on the degradation characteristics of organic materials and on the number of methanogenic bacteria produced were investigated. By making the media with the repeated wxperiment, the number and activity of methanogenic bacteria were measured. The increase of the removal rate of organic acid in the digester was oberved at HRT of 2 days. The total number of methane forming bacteria estimated by the MPN method showed 2.3 $\times$ $ at HRT of 3 days, 7$\times$$ of 5 days and 7.9$\times$$ $/ml of 10 days. The optimum incubation time for measuring the number of methanogenic bacteria was found as more than four weeks. The PMA revealed 161ml CH$/l day at HRT of 10 days and the PUA 290mg COD/l day. At the incubation time 4.3 days, the maximum value of CH$ *59.1%) was found. At this time, $ was found as 15.3% and $ 25.6%.

  • This study was carried out to investigate the effects of nucleic acid related compounds and metal ions on activities of cytidine deaminase from Bacillus subtilis ED 213. The purified cytidine deaminase was weakly inhibited by 1mM GMP, IMP and ATP, but not affected by other nucleic acid related compounds such as CMP and UDP. The apparent Km values for cytidine, deoxycytidine, 5 methylcy tidine, fluorodeoxycytidine, and 5 bromocytidine were calculated to be 6.6$\times$10-4M, 6.0$\times$10-4M, 0.9$\times$10-4M, 0.8$\times$10-4M, and 2.0$\times$10-3M, respectively. The cytidine deaminase was completely inhibited by 1mM Hg2+, and mildly inhibited over 40% by metal ions such as Na+ and Fe2+. However the enzyme activity was activated more than 40% by 1mM Mg2+.

  • Effects of root, stem and leaf extract of sancho (Zanthoxylum schinifolium) on the inhibition of lipid peroxidation in the hepatic microsome of rat, DPPH radical scavenging activity and activated partial thromboplastin times (APTT) were examined in vitro. The highest inhibition of hepatic microsomal lipid peroxidation was observed by ethyl acetate fraction than that of methylene chloride fraction of the root and stem extracts. The high inhibition of lipid peroxidation was determined in the leaf, the root and the stem in order. The DPPH radical scavenging activity of ethyl acetate fraction was higher than that of n-butanol fraction and it was similar to the root and the steam extract. It was similar to the inhibition of hepatic microsomal lipid peroxidation. The DPPH radical scavenging activity was the highest in 2.500mg/mL of ethyl acetate fraction and it was 4.4 fold higher than that of $\alpha-tocopherol$, as an antioxidant standard. The DPPH radical scavenging activity was dependent on the extract concentration in the range of 0.125-5.000 mg/mL. The throm-boplastin times were higher than that of n-butanol fraction and it was similar to the root and the steam extracts. The leaf extract showed the highest antithrombogenic effect followed by the stem and then the root extract. The activated partial thromboplastin times were ependent on the extract concentration in the range of 0.100-2.000 mg/mL. Consequently, the effects of antioxidative, DPPH radical scavenging activity and antithrombogenic of Z. schinifolium was observed due to the inhibition of lipid peroxidation and the DPPH radical scavenging activity by methylene chloride, n-butanol and ethyl acetate fraction of the leaf extract. • The antimicrobial and glucosyltransferase(GTase) inhibition activity were searched for 30 species of various folk drugs and by products of food industry. Among them, two species, gallnut and red grape husk water extracts, were selected for the powerful antimicrobial and GTase inhibition activity. The polyphenol fractions of gallnut and red grape husk were showed very greater antimicrobial activity on both Gram(+) and (-), B. subtilis and E. coli. The minimum antimicrobial activity of gallnut polyphenol fraction were 1.0mg for B. subtilis and 3,0mg for E. coli. Red grape husk was 2.0mg for B. subtilis and 3.0mg for E coli. The polyphenol fractions of gall nut and red grape husk were showed powerful GTase inhibition activity. The concentrations of these fractions for 80% inhibition of GTase activity were 1.08$\times$10$\^$-3/mg/㎖ and 1.08$\times$10$\^$-2/mg/㎖, respectively. The most abundant compound in these fraction seems to be polyphenol derivatives. From these results, we think that the gallnut and the red grape husk polyphenol fraction had more antimicrobial and anti-plaque activities than artificial synthetic preservatives as an economic point of view.

  • This study evaluated the effects of heat treatment on antioxidant activity of Rehmannia radix Libosch (RRL). RRL was heated at various temperatures ($110-150^{\circ}C$) for various times (1-5 hr), and the total polyphenol, flavonoid content, and antioxidant activity were investigated. With increased heating temperature and exposure time, total content of polyphenol, flavonoid, as well as antioxidant activity increased. The highest total polyphenol and flavonoid contents were 21.65 and 3.56 mg/g, respectively, these values were occurred after heating for 3 hr at $150^{\circ}C$ (RRL was 5.09 and 0.83 mg/g, respectively). The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was highest value of 83.46% after heating for 3hr at $150^{\circ}C$. The 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) cation radical scavenging activity was highest value of 20.43mg ascorbic acid (AA) eq/g after heating for 2 hr at $150^{\circ}C$. There were highly significant differences in the total polyphenol, flavonoid content, and antioxidant activity among heating temperatures and times (p<0.001), with heating temperature having the greater effect.
