Cytomegalovirus (CMV)-associated esophageal ulcer is rare in immunocompetent infants. The presence of inclusion bodies and immunohistochemical staining for CMV in biopsy specimens obtained during esophagogastroduodenoscopy (EGD) indicate that such ulcers occur because of CMV infection. A 7-week-old female infant who experienced frequent vomiting and feeding intolerance was diagnosed with a massive CMV-associated ulcer in the distal esophagus. The ulcer improved after conservative treatment using proton-pump inhibitors; however, ganciclovir was not administered. In a follow-up EGD biopsy specimen, no CMV inclusion bodies were present, and immunohistochemical staining results for this virus were negative. The presence of CMV inclusion bodies indicates active viral replication. If persistent inclusion bodies or positive immunohistochemical staining for CMV is observed in follow-up biopsy specimens, ganciclovir may be used to treat CMV-associated esophageal ulcers.
A fungal strain able to produce filter paper activity (FPase) was isolated from soil by testing the ability to hydrolyze using filter paper. The isolated strain was identified as an Aspergilus sp. judging from its morphological and microscopical characteristics. The cellulase-xylanase system of Aspergillus sp. 8-17 was detected in situ after gel electrophoresis in the presence of SDS and showed that each protein pattern had a distinct polypeptide composition. ${\beta}$-1,4-Glucanase, cellobiohydrolase, and xylanase activity profiles differ from protein patterns. The Aspergillus sp. 8-17 hydrolytic enzymes responsible for the hydrolysis of ${\beta}$-glucan, MUC, and xylan have multiple active forms.
Objectives : Schisandrae Fructus (SF) has traditionally been used to balance level of body fluid and to strengthen kidney function. It has been reported that the SF extract has antioxidant, hepatoprotective, neuroprotective and anticancer effects. This study investigated an antiproliferative effect of SF extract on PC-3 human prostate cancer cells and analyzed active ingredients of SF extract qualitatively and quantitatively. Methods : We examined the antiproliferative effect of SF extract with MTT assay, DAPI staining and annexin-V/7-AAD double staining. The active ingredients of SF extract were identified by using HPTLC and HPLC/DAD system. Results : SF-chloroform fraction inhibited growth of PC-3 cells and changed the morphology of nucleus in a dose dependent manner. A dose-dependent apoptotic cell death was also measured by flow cytometry analysis. It was analyzed that SF-chloroform fraction contained more schizandrin than other fractions by using HPTLC and HPLC/DAD system. Conclusions : These results suggest that SF extract and schizandrin may be a potential chemotherapeutic agent for the control of PC-3 human prostate cancer cells.
This study was performed to understand the interaction between Ophiostomataceae and basidiomycetes fungi during cultures, and whether the basidiomycetes fungi inhibit the growth and decolorize dark pigments of blue staining fungi. The conjoint cultivation was studied on 2% malt extract agar. The ability of basidial cultures to decolorize dark pigments of ophiostomatoid fungi was the main characteristics estimated during this study. More than half of basidial cultures were characterized by deadlock interaction with blue staining fungi. In the dual cultures, where basidial partners were presented by Agaricus bisporus(64), Laetiporus sulphureus(L01/89), Trametes versicolor(09) and unknown fungus(02), antagonism was found at the phase of primary contact of colonies. Replacement interaction resulted usually in decreasing dark colour of substrate was observed for 11 basidial cultures that were belonging mainly to white-rot fungi. Among them Abortiporus biennis(123), Antrodiella hoehnelii(S28/91), Bjerkandera fumosa (137), and Gleophyllum odoratum(124) were characterized by the absence of deadlock-phase: they began to grow over dark colonies of their partners just after primary contact. Basidiomycetes did not affect strongly the pigments of Ceratocystis spp. and Leptographium sibirica isolates, but completely decolorized colonies of Ophiostoma ips and to a smaller degree Ophiostoma minus. Antrodiella hoehnelii(S28/91), Bjerkandera fumosa(137), Gleophyllum odoratum(124) and Trametes versicolor(B18/91) cultures were found to be the most active in decreasing dark color of blue staining fungi colonies. The cultures were recommended for further development as agents of biopulping of wood chips and bio-control of blue stain in woods.
Object : Warming acupuncture (WA) has been used in Oriental Medicine for the treatment of physical disabilities caused by ligament damage. Here, the effects of WA on injured ligament tissues were investigated using the rat model. Methods : The rats were induced injury on the right hind ankle, and 4 weeks later, WA was given onto the acupoint GB4O(Quixu) of the injury area on a weekly basis for 6 weeks. Main outcome was measured by levels of Erk1/2. Hoechst nuclear staining and collagen staining in the ligament tissue. Result : Levels of active form of Erk1/2 kinase were increased in the injured ligament with WA compared with the control ligament induced injury only, and this change correlated with cell number increases in the ligament by WA. Type III, but not type I, collagen mRNA and protein levels were elevated in the injured ligament treated with WA. Moreover, histological staining showed increased re-organization of collagen fibers in the ligament by WA. Conclusions : The present data suggest that WA performance to the injured ligament may facilitate the healing process via increasing cellular activity.
We examined the availability of the lacZ gene (${\beta}$-galactosidase gene) as a reporter of foreign gene transfer in the cysts of Artemia franciscana (A. franciscana) to conduct a risk assessment of living genetically modified organisms (LMOs) in the marine ecosystem. The LacZ gene was transferred to decapsulated cysts by particle bombardment, and its insertion and expression were assessed by means of polymerase chain reaction (PCR) and X-gal staining. X-gal staining indicated lacZ expression in all A. franciscana examined (including the control group), which exhibited not only negative but also positive PCR amplification. Endogenous ${\beta}$-galactosidase is highly active in the whole body of A. franciscana during all stages of the life cycle. Thus, the lacZ gene is unsuitable as a reporter for foreign gene transfer in A. franciscana cysts, because it is difficult to discriminate between exogenous and endogenous ${\beta}$-galactosidase activity.
Goniothalamin is an active compound extracted from Goniothalamus griffithii, a local plant found in northern Thailand. Goniothalamin inhibits cancer cell growth but is also toxic to normal cells. The aims of this study were to identify the cytotoxic effect of goniothalamin and the mechanism of cell death in human HL-60 and U937 cells. Cytotoxicity was determined by MTT assay and cell cycle profiles were demonstrated by staining with propidium iodide (PI) and flow cytometry. Apoptosis was confirmed by staining with annexin V-FITC/propidium iodide (PI) and flow cytometry. Reduction of mitochondrial transmembrane potential was determined by staining with dihexyloxacarbocyanine iodide and flow cytometry and expression of Smac, caspase-8 and -9 was demonstrated by Western blotting. Goniothalamin inhibited growth of HL-60 and U937 cell lines. An increase of SubG1 phase was found in their cell cycle profiles, indicating apoptosis as the mode of cell death. Apoptosis was confirmed by the flip-flop of phosphatidylserine using annexin V-FITC/PI assay in HL60 and U937 cells in a dose response manner. Furthermore, reduction of mitochondrial transmembrane potential was found in both cell types while expression of caspase-8, -9 and Smac/Diablo was increased in HL-60 cells. Taken together, our results indicate that goniothalamin-treated human leukemic cells undergo apoptosis via intrinsic and extrinsic pathways.
Horizontal starch gel electrophoresis, follow by enzyrne-specific staining, separate and visualize several legume esterase isozyme. Using extracts prepared from cotyledon, radicle and plumule of legume seedlings germinated 5 days. The results were as follows. 1. The number and staining intensity of esterase isozyme bands varies depending on the plant species. tissues and developmental stage. 2. Bands in the cotyledon of field bean seedling expressed 4 and 1 in radicle. 3. In soybean cultivars, cotyledon of IIwangkum-kong had 3 bands and 1 band in the examined tissues of Paldal-kong and Jangkyung-kong seedling. 4. The cotyledon and radicle of french bean seedling had 3 bands, respectively. 5. The highest esterase isozyme activity appears to be expressed in the cotyledon and radicle of french bean, as indicated by intensity of stain, with the Paldal-kong particulary active.
Streptomyces blastmyceticus strain 12-6 was isolated from a forest soil sample of Cheonan area on the basis of strong antifungal activities against plant pathogenic fungi. Butanol extracts of the cultural filtrates were active against C. acutatum, C. coccodes, C. gloeosporioides, F. oxysporum, and T. roseum. Active fractions were prepared by thin layer chromatography using silica gel plate; 12-6-2 ($R_f$ 0.36), 12-6-3 ($R_f$ 0.44). Scanning electron microscopy showed that the active fractions caused a change in surface texture of fungal spores from smooth surface to wrinkled surface. The lethal effect on the spores of the active fractions varied from 56% to 100%. It was shown that the spores of C. acutatum were more sensitive to the antifungal fractions than the spores of F. oxysporum. Fluorescence staining using TOTO-1 indicated that the antifungal fractions could make the spores more sensitive to the fluorescence dye. Thus, it was suggested that antifungal agents prepared in this study exhibited the antifungal activity by damaging the plasma membrane of both fungal spores and hyphae. Identification of antifungal agents in the active fraction using GC-MS analysis revealed the presence of cyclo-(Leu-Pro) and 9-octadecenamide as major components that have already been known as antifungal substances.
MTT (3-(4, 5-dimethyl thiazol-2-yl)-2, 5-diphenyl tetrazolium bromide) reduction assay is a method that validates the viability of an active cell. Dehydrogenase in mitochondria converts yellow colored insoluble tetrazolium salt to purple colored water-soluble formazan. Sperm also have mitochondria in the midpiece, therefore sperm viability could be evaluated by MTT reduction assay. Several studies have already demonstrated the capability of application of the MTT reduction assay to sperm of several species in Hepes-BSA buffer. Because most liquid semen was diluted in extender like BTS (Beltsville Thawing Solution), Modena or Androhep when it is used or transferred, semen needed another dilution in Hepes-BSA buffer to assess sperm viability. In this study, we evaluated boar sperm viability especially in BTS extended semen and compared the efficiency of this test with eosin-nigrosin staining. We used the fresh BTS extended semen from a local A.I center. Semen sample was diluted to $3.0{\times}10^7$ sperms/ml in BTS. The rates of formazan production were measured in 96-well microtiter plates immediately and 1h after incubation at $17^{\circ}C$ using a spectrophotometer at wave length 560 nm. Simultaneously, split samples of the same semen were tested, using eosin-nigrosin staining to compare the efficiency of the MTT assay of sperm viability in BTS. The correlation between the results of these tests was calculated using Student-t test and ANOVA. The results revealed a strong correlation between the results of MTT reduction rate and the results that were simultaneously determined by eosin-nigrosin staining at 1 h. In conclusion, the MTT reduction test was an effective and simple method to validate sperm viability and it could be used as a simple tool to evaluate sperm viability in the local A.I center and laboratory.
