High-Resolution Microarrays for Mapping Promoter Binding sites and Copy Number Variation in the Human Genome

  • NimbleGen has developed strategies to use its high-density oligonucleotide microarray platform (385,000 probes per array) to map both promoter binding sites and copy number variation at very high-resolution in the human genome. Here we describe a genome-wide map of active promoters determined by experimentally locating the sites of transcription imitation complex binding throughout the human genome using microarrays combined with chromatin immunoprecipitation. This map defines 10,567 active promoters corresponding to 6,763 known genes and at least 1,196 un-annotated transcriptional units. Microarray-based comparative genomic hybridisation (CGH) is animportant research tool for investigating chromosomal aberrations frequently associated with complex diseases such as cancer, neuropsychiatric disorders, and congenital developmental disorders. NimbleGen array CGH is an ultra-high resolution (0.5-50 Kb) oligo array platform that can be used to detect amplifications and deletions and map the associated breakpoints on the whole-genome level or with custom fine-tiling arrays. For whole-genome array CGH, probes are tiled through genic and intergenic regions with a median probe spacing of 6 Kb, which provides a comprehensive, unbiased analysis of the genome.

  • Due to the aging of economically active population in the world, the average age of workers in the domestic construction industry is also steadily increasing. This study analyzes the aging of construction workers in small-size construction sites in Busan, where the aging of the economically active population is the most severe in Korea. As a result of the study on the aging of construction workers in six construction sites with a construction cost of less than 500 million won in Busan, it was found that 65% of the workers were over 50 years of age. In addition, several experts interviews and survey revealed that 'securing welfare facilities for construction workers' and 'diminishing prejudice about the construction industry" were the most urgent and effective methods to overcome the aging problems. The results of this study can be served as basic data to solve the aging problems in the industry.

Influence of defective sites in Pt/C catalysts on the anode of direct methanol fuel cell and their role in CO poisoning: a first-principles study

  • Carbon-supported Pt catalyst systems containing defect adsorption sites on the anode of direct methanol fuel cells were investigated, to elucidate the mechanisms of H2 dissociation and carbon monoxide (CO) poisoning. Density functional theory calculations were carried out to determine the effect of defect sites located neighboring to or distant from the Pt catalyst on H2 and CO adsorption properties, based on electronic properties such as adsorption energy and electronic band gap. Interestingly, the presence of neighboring defect sites led to a reduction of H2 dissociation and CO poisoning due to atomic Pt filling the defect sites. At distant sites, H2 dissociation was active on Pt, but CO filled the defect sites to form carbon π-π bonds, thus enhancing the oxidation of the carbon surface. It should be noted that defect sites can cause CO poisoning, thereby deactivating the anode gradually.

Detection of Active Intra-Abdominal Bleeding from Malignant Tumors in Two Dogs Using Contrast-Enhanced Ultrasonography

  • Contrast-enhanced ultrasonography (CEUS) has been applied to evaluate parenchymal organs in human and veterinary medicine. However, to our knowledge, there is no report on the identification of active bleeding and the bleeding site in veterinary clinical patients. Herein, we describe the use of CEUS in two cases of abdominal bleeding caused by ruptured lesions with malignant abdominal tumors. One dog had a splenic hemangiosarcoma, which had metastasized to the liver; the other dog had hepatic cell carcinomas in the left hepatic lobe, which were lobectomized, and another nodule was identified in the right hepatic lobe. Immediately after the rupture of these oncogenic lesions was suspected, CEUS was performed to identify the bleeding sites. The active bleeding sites were confirmed by hyperechoic pooling signs in the arterial phase, and extravasation could be observed within the defects showing hypoechoic perfusions in the delayed phase of the CEUS. Microbubbles were also observed in the ascites; thus, CEUS could detect the presence of hemorrhage and accurately identify the bleeding site. Collectively, the study findings suggest the usefulness of CEUS in emergent situations as it enables rapid and noninvasive evaluation of bleeding points in case of active bleeding in dogs.

Recent Developments of Metal-N-C Catalysts Toward Oxygen Reduction Reaction for Anion Exchange Membrane Fuel Cell: A Review

  • Metal-N-C (MNC) catalysts have been anticipated as promising candidates for oxygen reduction reaction (ORR) to achieve low-cost polymer electrolyte membrane fuel cells. The structure of the M-Nx moiety enabled a high catalytic activity that was not observed in previously reported transition metal nanoparticle-based catalysts. Despite progress in non-precious metal catalysts, the low density of active sites of MNCs, which resulted in lower single-cell performance than Pt/C, needs to be resolved for practical application. This review focused on the recent studies and methodologies aimed to overcome these limitations and develop an inexpensive catalyst with excellent activity and durability in an alkaline environment. It included the possibility of non-precious metals as active materials for ORR catalysts, starting from Co phthalocyanine as ORR catalyst and the development of methodologies (e.g., metal-coordinated N-containing polymers, metal-organic frameworks) to form active sites, M-Nx moieties. Thereafter, the motivation, procedures, and progress of the latest research on the design of catalyst morphology for improved mass transport ability and active site engineering that allowed the promoted ORR kinetics were discussed.

  • In order to characterize the catalytically active sites on carbon black, acetylene chemisorption had been examined recently at 773 and 873 K by using a pulse technique. As the inject ion was repeated at 773 K, the adsorbed amount gradually decreased and eventually the adsorption did not occur any more. At 873 K a constant amount of $C_2H_2$ was consumed repeatedly after several injections. Good linear relationships were obtained between the methane decomposition rate at 1123 or 1173 K and the cumulative acetylene adsorption at 773 K or the constant acetylene consumption at 873 K. Reasonable models for the associative acetylene chemisorption at 773 K and the constant acetylene consumption at 873 K on the armchair face at the edges of graphene layers were proposed. The constant consumpt ion may be explained by the "$C_2H_2$-addition-hydrogen- abstract ion (CAHA)" mechanism.

  • Lipomyces starkeyi KSM 22 elaborates an enzyme that has both dextranase and amylase activities in a single protein of 100 kDa. Competition studies, using different amounts of dextran and starch as substrates, gave a competition plot consistent with the hypothesis that the hydrolysis of dextran and starch occurs at two independent active sites, each specific for starch and dextran, respectively.

심해 극한 환경에서의 중형저서동물 군집 (Meiobenthic Communities in Extreme Deep-sea Environment)

  • The spatial patterns of meiobenthic communities in deep-sea sediment were examined. Sediment samples for analyzing of meiobenthic community structure were collected using a remote operated vehicle (ROV), multiple corer TV grab at 20 stations at five sites. In all, 15 meiofauna groups were recorded. Nematodes were the most abundant taxon. Benthic foraminiferans, harpacticoid copepods, polychaetes, and crustacean naupii were also dominant groups at all sites. The total meiofauna density at the study sites varied from 49 to 419 ind./$10cm^2$. The maximum density was recorded at a site located in Challenger Deep in the Mariana trench where simple benthic foraminifera with organic walls flourish. These distinctive taxa seem to be characteristic of the deepest ocean depths. Active hydrothermal sediments contain up to 150 harpacticoid copepods per $10cm^2$ of sediment. In a inactive ridge sediments, devoid of macrofaunal organisms:, the abundance of harpacticoid copepods never exceeded 15 ind./$10cm^2$. Multivariate analysis (multidimensional scaling) revealed significant differences in community structure among the three regions; near an active hydrothermal vent, in the deepest ocean depths and at typical deep-sea bed sites.

EPR Studies of the Active Sites of Carbon Monoxide Dehydrogenase from Clostridium thermoaceticum

  • The active sites of the nickel and iron-containing enzyme, carbon monoxide dehydrogenase (CODH) from clostridium thermoaceticum were investigated using Electron Paramagnetic Resonance (EPR) technique. CODH exhibits several spectral features called NiFeC, $g_{ave}=1.82$, $g_{ave}=1.86$. FCII signals which are originated from different clusters in this enzyme. CODH is know to catalyze two different kinds of reactions - acetyl-CoA synthesis and CO oxidation. The acetyl-CoA synthesis activity can be followed by monitoring CO/acetyl-CoA exchange. The addition of 1,10-phenanthroline (phen) to CODH selectively destroyed the CO/acetyl-CoA exchange activity and eliminated the NiFeC signal completely. CO oxidation activity and other EPR signals were unaffected. Such behavior demonstrates that CODH has two distinct active sites and that the NiFe complex is only responsible for the CO/acctyl-CoA exchange activity. Phen caused the removal of only 30% of Ni in the NiFe complex ($0.3Ni/{\alpha}{\beta}$) as shown by the quantitative metal analysis. The phen-treated CODH could be reactivated fully by incubation In $Ni^{2+}$ solution. Radioactive $^{63}Ni^{2+}$ was used to quantitate the amount of the $Ni^{2+}$ incorporated into phen-treated enzyme and showed that the amount was the same as the removed by the phen treatment. i.e. $0.3Ni/{\alpha}{\beta}$. This indicates that only 30% of NiFe complexes are labile and responsible for the CO/acctyl-CoA exchange activity, the other 70% are non-labile and have no exchange activity. This is the first clear evidence that the NiFe complex is heterogencous and labile and non-labile Ni sites arc interacting differently with substrates and chelating agents like phen.

Electrical Characteristics of Solution Processed DAL TFT with Various Mol concentration of Front channel

  • In order to investigate the effect of front channel in DAL (dual active layer) TFT (thin film transistor), we successfully fabricated DAL TFT composed of ITZO and IGZO as active layer using the solution process. In this structure, ITZO and IGZO active layer were used as front and back channel, respectively. The front channel was changed from 0.05 to 0.2 M at fixed 0.3 M IGZO of back channel. When the mol concentration of front channel was increased, the threshold voltage (VTH) was increased from 2.0 to -11.9 V and off current also was increased from 10-12 to 10-11. This phenomenon is due to increasing the carrier concentration by increasing the volume of the front channel. The saturation mobility of DAL TFT with 0.05, 0.1, and 0.2 M ITZO were 0.45, 4.3, and $0.65cm2/V{\cdot}s$. Even though 0.2 M ITZO has higher carrier concentration than 0.05 and 0.1 M ITZO, the 0.1 M ITZO/0.3 M IGZO DAL TFT has the highest saturation mobility. This is due to channel defect such as pores and pin-holes. These defect sites were created during deposition process by solvent evaporation. Due to these defect sites, the 0.1 M ITZO/0.3 M IGZO DAL TFT shows the higher saturation mobility than that of DAL TFT with front channel of 0.2 M ITZO.
