Prospective Study for Korean Red Ginseng Extract as an Immune Modulator following a Curative Surgery in Patients with Advanced Colon Cancer

  • In this paper, we present evidence that the Korean red ginseng extract shows the immunomodulatory activities during postoperative chemotherapy after curative surgery in patients with advanced colon cancer. We measured the circulating interleukin-2 (IL-2), interleukin-8 (IL-8)and interleukin-10 (IL-10) as a immune modulator to evaluate the effect of Korean red ginseng. The mean preoperative value of IL-2 was similar in the non-RG group and the RG group (5.72 pg/ml versus 6.87 pg/ml, p>0.05). The mean value of IL-2 was compared with IL-2 from healthy control group, there was no significant difference (14.89 pg/ml versus 14.22 pg/ml, p>0.05). The mean preoperative value of IL-8 was higher in the non-RG group comparing with the RG group (30.92 pg/ml versus 36.25 pg/ml, p < 0.05). At postoperative 3 month, the mean values of IL-8 from non-RG and RG group down to 24.56 pg/ml and 21.46 pg/ml respectively. The IL-8 of RG group at 3 month showed no difference with that of HC group(21.46 pg/ml versus 16.31 pg/ml, p>0.05). The preoperative mean value of IL-10 of non-RG, RG and HC group was 11.56 pg/ml, 10.8 pg/ml, and 3.68 pg/ml respectively. At postoperative 3 month, the mean values of IL-10 from non-RG and RG group down to 8.45 pg/ml and 5.04 pg/ml respectively. In spite of decreasing IL-10 levels of both cancer Patients group with time, there was still significant difference with that of HC group (non-RG versus HC group, p=0.00, RC versus HC group, p=0.04). The results of this study suggest that the red ginseng extract may have some immunomodulatory properties associated with IL-2, IL-8 and IL-10 activity in patients with advanced colorectal cancer during postoperative chemotherapy. We think to need the further studies and a larger sample size to fully evaluate the antitumor effect of ginseng and need to establish this mechanism of action as well as identify the active components associated with antitumor activity and immunomodulation in patients with advanced colorectal cancer.

Effect of phosphorus application on appearance of algal water bloom and rice yield in rice-barley double cropping system

  • Algal communities are important to maintain the aquatic ecosystems function. Algae have short life cycles, they respond quickly to environmental change and their diversity and density can indicate and the quality of their habitat. The bloom forms before the rice seedings have emerged, it may present a physical barrier that prevents the seedlings from penetrating the floodwater. Wind may also move the algal bloom, pushing the young plants beneath the surface. Another harmful action develops when the water dries up and the algae form a layer at the bottom of the field. The layer envelops the seedlings, which are not yet deeply rooted, and drag them to the surface when the water is let in again. Soil utilization pattern can be the mail facter affecting soil physico-chemical properties, especially in soil phosphorus (P). Solid content of the algae culture solution increased with the increase in the nitrogen rather the phosphors concentration. Phosphoric acid was treated with conventional treatments (100-0%, before transplanting time-tillering stage), 50-50%, 0-100%, and un-treated. The herbicide was treated on the 7 DAT (day after transplanting). Green algae samples were collected 20 DAT. Total phosphoric acid was the highest at 0.06 in 50-50% treatment in 20 DAT. The amount of green algae was about twice (9.8 mg/20ml) that of un-treated. Total number of green algae was 54 species(Green algae 35 species, Euglena 9 species, Stone wheel 10 species). Among the phosphoric acid treatment methods, the number of occurrences of green algae were the highest with 39 species in 0-100%, followed by 50-50%, 28 species, conventional treatments, 22 species, non-treatment, 18 species, respectively. Rice Yield was not significantly different by phosphoric treatment time, but slightly higher than un-treated. The maximal algal biomass was observed about 2weeks or 1 month after transplanting; the subsequent decrease of the biomass was related to the consumption by grazers and to a deficient light under the rice canopy. Maximal algal growth was observed just before tillering. To estimate the suitable method of phosphorus application in puddled-soil drill seeding of rice, available phosphorus appearance of algal water bloom, and rice yield were investigated in paddy soil of rice-barley double cropping system.

  • An analytical diffusion model for flood routing with backwater effects and lateral flows is developed. The basic diffusion equation is linearized about an average depth of (H + h), and is solved using the boundary conditons which take into account the effects of backwater and lateral flows. Scouring phenomenon around pier which affects on the support function of pier and the stabilization if river bed is a complex problem depending on flow properties and river bed state as well as pier geometry. therefore, there is no uniting theory at present which would enable the designer to estimate, with confidence, the depth of scour at bridge piers. The various methods used in erosion control are collectively called upstream engineering, HEC-RAS Model, underwater blasting. They consist of reforestation, check-dam construction, planting of burned-over areas, contour plowing and regulation of crop and grazing practices. Also included are measures for proper treatment of high embankments and cuts and stabilization of streambanks by planting or by revetment construction. One phase of reforestation that may be applied near a reservoir is planting of vegetation screens. Such screens, planted on the flats adjacent to the normal stream channel at the head of a reservoir, reduce the velocity of silt-laden storm inflows that inundate these areas. This stilling action causes extensive deposition to occur before the silt reaches the main cavity of the reservoir.

Tumorigenic Effects of Endocrine-Disrupting Chemicals are Alleviated by Licorice (Glycyrrhiza glabra) Root Extract through Suppression of AhR Expression in Mammalian Cells

  • Endocrine-disrupting chemicals (EDCs) have been reported to interfere with estrogen signaling. Exposure to these chemicals decreases the immune response and causes a wide range of diseases in animals and humans. Recently, many studies showed that licorice (Glycyrrhiza glabra) root extract (LRE) commonly called "gamcho" in Korea exhibits antioxidative, chemoprotective, and detoxifying properties. This study aimed to investigate the mechanism of action of LRE and to determine if and how LRE can alleviate the toxicity of EDCs. LRE was prepared by vacuum evaporation and freeze-drying after homogenization of licorice root powder that was soaked in 80% ethanol for 72 h. We used 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) as an EDC, which is known to induce tumors or cancers; MCF-7 breast cancer cells were used as a tumorigenic model. These were treated with TCDD and various concentrations of LRE (0, 50, 100, 200, $400{\mu}g/mL$) for 24, 48, and 72 h. As a result, TCDD stimulated MCF-7 cell proliferation, but LRE significantly inhibited TCDD-induced MCF-7 cell proliferation in a dose- and time-dependent manner. Expression of TCDD toxicity-related genes, i.e., aryl hydrocarbon receptor (AhR), AhR nuclear translocator, and cytochrome P450 1A1, were subsequently down-regulated by LRE in a dose-dependent manner. Analysis of cell cycle distribution after treatment of MCF-7 cells with TCDD and various concentrations of LRE showed that LRE inhibited the proliferation of MCF-7 cells via G2/M phase arrest. Reverse transcription-polymerase chain reaction and Western blot analyses also revealed that LRE dose-dependently increased the expression of the tumor suppressor genes p53 and p27 and down-regulated the expression of cell cycle-related genes. These data suggest that LRE can mitigate the tumorigenic effects of TCDD in breast cancer cells by suppression of AhR expression and cell cycle arrest. Thus, LRE can be used as a potential toxicity-alleviating agent against EDC-mediated disease.

Effect of Cooking Methods and Reheating on Texture of Sausage (시판소시지의 조리방법 및 재가열이 조직특성에 미치는 영향)

  • The Purpose of this study was to evaluate the effect of cooking methods and reheating on texture characteristics of sausage. Sausage samples were cooked(microwaving, gas-broiling and frying), reheated on day 3 and 6 after refrigeration and put to evaluation of sensory and texture characteristics. The results of sensory evaluation showed that hardness, salty taste, rancid flavor and after-taste were increased but wetness and chewiness were decreased by reheating after refrigeration. Overall acceptability was the highest when the samples were gas broiled and reheated on day 3 after refrigeration. As for the estimation of texture changes by rheometer, hardness, cohesiveness, elasticity, gum-miness and chewiness tended to increase by reheating after refrigeration. Hardness, cohesiveness, gumminess and chewiness were the highest when the samples were gas-broiled and reheated on day 3 after refrigeration.

Effect of Soy Isoflavones on the Expression of $TGF-{\beta}1$ and Its Receptors in Cultured Human Breast Cancer Cell Lines

  • The two major isoflavones in soy, genistein and daidzein, are well known to prevent hormone-dependent cancers by their anti estrogenic activity. The exact molecular mechanisms for the protective action are, however, not provided yet. It has been reported that genistein and daidzein have a potential anticancer activity through their antiproliferative effect in many hormone-dependent cancer cell lines. Transforming growth $factor-\beta1(TGF-\beta1)$ has also been found to have cell growth inhibitory effect, especially in mammary epithelial cells. This knowledge led to a hypothetical mechanism that the soy isoflavones-induced growth inhibitory effect can be derived from the regulation of $TGF-\beta1$ and $TGF-\beta$ receptors. In order to test this hypothesis, the effects of the soy isoflavones at various concentrations and periods on the expression of $TGF-\beta1$and $TGF-\beta$ receptors were investigated by using Northern blot analysis in human breast carcinoma epithelial cell lines, an estrogen receptor positive cell line (MCF-7) and an estrogen receptor negative cell line (MDA-MB-231). As a result, only genistein has shown a profound dose-dependent effect on $TGF-\beta1$ expression in the $ER^+$ cell line within the range of doses tested, and the expression levels are correspondent to their inhibitory activities of cell growth. Moreover, daidzein showed down-regulated $TGF-\beta1$ expression at a low dose, the cell growth proliferation was promoted at the same condition. Therefore, antiproliferative activity of the soy isoflavones can be mediated by $TGF-\beta1$ expression, and the effects are mainly, if not all, occurred by ER dependent pathway. The expression of $TGF-\beta$ receptors was induced at a lower dose than the one for $TGF-{\beta}1$ induction regardless of the presence of ER, and the expression patterns are similar to those of the cell growth inhibition. These results indicated that the regulation of $TGF-\beta$ receptor expression as well, prior to $TGF-\beta1$ expression, may be involved in the antiproliferative activity of soy isoflavones. Little or no expression of $TGF-\beta$ receptors was found in the MCF-7 and MDA-MB-231 cells, suggesting refractory properties of the cells to growth inhibitory effect of the $TGF-\beta$. The soy isoflavones can seemingly restore the sensitivity of growth inhibitory responses to $TGF-\beta1$ by re-inducing $TGF-\beta$ receptors expression. In conclusions, our findings presented in this study show that the antitumorigenic activity of the soy isoflavones could be mediated by not only $TGF-\beta1$induction but $TGF-\beta$ receptor restoration. Thus, soy isoflavones could be good model molecules to develop new nonsteroidal antiestrogenic chemopreventive agents, associated with, regulation of $TGF-\beta$ and its receptors.

  • The sand table training is one of the most effective training method in military operations which can accomplish missions such as simulation and rehearsal without limitations related to time, space, money and so on. Previous sand table training has many problems like that the sand table cannot represent real field condition because of its physical properties. So, it is hard to be preserved and impossible to include much of information into them. In this paper, we make an approach based on Augmented Reality(AR) to solve these problems and propose an efficient military training briefing tool with virtual sand table environment described as actual battle field Virtual Tactical Map(VTM) can realize a virtual military training with simple action like moving marker or tangible interface by hand. Real-time state information of VTM gives us more organic intelligence for entire situation. Tangible AR interface provides users with a contents authoring tool that is natural, intuitive and easy to deal with as interaction between user in real world and system that augmented real world with virtual object. VTM is a newly designed military training briefing tools. A military training content can be reproduced and it is possible that user uses this content later. Thus, it shows us potential possibilities of AR applications on military leaning field.

The Development of the Hantan River Basin, Korea and the Age of the Sediment on the top of the Chongok Basalt (한탄강유역의 발달과정과 전곡현무암 위의 퇴적물의 연대)

  • The development of the Hantan river basin can be divided into three stages. The first stage include the ancient Hantan channel system prior to the Chongokni basalt which yield dates of about 0.6 mya from the K/Ar dating method. During this period the Baekuyri formation was formed. The Baekuyri formation is widely observed under the Chongokni basalt along the current river system. The second stage is the period when stream channels stayed on the top of the basalt plateau. Aggradation and deggradation were continued by meandering and braiding channel systems until major stream channel was formed. The currently remaining deposit on the top of the basalt was formed by lacustrine and fluvial systems in this period. During this period Pleistocene hominid was present on edge of water and flood plain and left Paleolithic material. This period was begun at the time of the final basalt flow dated about 300,000 BP. The third stage is designed for the time when the Hantan river channel was dropped down to a level from which the channel could not influence the top of the basalt any more No more deposit could be formed but erosion by surface water has been continued on the top of the basalt since then. The dropping of the Hantan river channel was probably not very long after the final flow of the basalt. Because of frost action and heavy concentrated precipitation in the basin area along with blocky and clumnar joint structure of the basalt, erosional process of the basalt is believed to have been carried out within a relatively short time. The lowering of the Hantan river channel was probably completed in a cycle of major fluctuation of world cimate. Also, the redclay on the top of the basalt is believed to have been formed during a warm period around 200,000 BP, which accords with the climatic change suggested above fair1y well. The Paleolithic materials in tile deposits fell accordingly into approximately same time period.

  • The damping characteristics of a cylindrical structure containing oil and bearing balls is investigated for external bending forces. The experimental data obtained through the use of bearing balls with viscous oil in a column is given and analyzed. The viscous action of the oil and inertia effects of the balls on the inside of column create a drag force. The drag force dampens the vibration of the column. This study aims to search for an optimum combination of oil and balls which would produce maximum damping. Machining oils of various viscosities along with ball bearings of various sizes place inside cantilevered aluminium tubes of various diameters to create a rig on which the damping properties of the oil and balls can be studied. The contileved tubes are studied in both horizontal and vertical positions in order to gauge the effect of gravity on the system. The actions of the ball in the column and damping characteristics are investigated according to the dimensionless terms. The Buckingham theorem is used to reduce the variables and to predict the damping of an oil ball column. Though the damping ratio remains fairly constant in the horizontal position of column, the damping ratio begins to increase as the ratio of the number of balls and column length rise above 0.28 in the vertical position of oil ball column. The ratio of the ball diameter to column diameter influences the damping ratio with an optimum diameter ratio. Slenderness ratio and gravity effects on the damping ratio ane investigated.

Induction of Apoptosis by Pachymic Acid in T24 Human Bladder Cancer Cells (T24 인체방광암 세포에서 pachymic acid에 의한 apoptosis 유발)

  • Pachymic acid (PA) is a lanostane-type triterpenoid derived from the Poria cocos mushroom. Several beneficial biological features of PA provide medicine with a wide variety of valuable effects, such as anticancer and anti-inflammatory activity; it also has antioxidant effects against oxidative stress. Nonetheless, the biological properties and mechanisms that produce this anti-cancer action of PA remain largely undetermined. In this study, we investigated the pro-apoptotic effects of PA in T24 human bladder cancer cells. It was found that PA could inhibit the cell growth of T24 cells in a dose-dependent manner, which was associated with the induction of apoptotic cell death, as evidenced by the formation of apoptotic bodies and chromatin condensation and accumulation of cells in the sub-G1 phase. The induction of apoptotic cell death by PA was connected with an up-regulation of pro-apoptotic Bax and Bad protein expression and down-regulation of anti-apoptotic Bcl-2 and Bcl-xL proteins, and inhibition of apoptosis family proteins. In addition, apoptosis-inducing concentrations of PA induced the activation of caspase-9, an initiator caspase of the mitochondrial-mediated intrinsic pathway, and caspase-3, accompanied by proteolytic degradation of poly (ADP-ribose)-polymerase. PA also induced apoptosis via a death receptor-mediated extrinsic pathway by caspase-8 activation, resulting in the truncation of Bid and suggesting the existence of cross-talk between the extrinsic and intrinsic pathways. Taken together, the present results suggest that PA may be a potential chemotherapeutic agent for the control of human bladder cancer cells.
