Changes of haemolymph proteins in Pieris rapae L. during the cuticle formation and hardening process (배추흰나비의 큐티클 形成과 硬化에 따른 혈림프 단백질의 變化)

  • Changes and possible origin of haemolymph proteins during the cuticle formation and hardening are determined by means of acrylamide gel electrophoresis and immunodiffusion. The results by acrylamide gel electrophoresis showed at least 19 protein bands in the haemolymph and 13 fractions in the fat body with relatively constant pattern during the period of cuticle formation and hardening. Both haemolymph and fat body proteins are generally characterized by the presence of three to four heavy stained bands and several thin bands near the top region of the gel. At least over five haemolymph proteins are constantly present during this period. Immunodiffusion tests show that of total eight to nine pupal haemolymph proteins two proteins were already detected in the fat body before pupation and other two proteins were also found in the fat body immediately after pupation, suggesting fat body as possible source of these two haemolymph proteins.

A Study on the Antimicrobial Finishing of Artificial Suede by Allylamine Copolymers (Allylamine계 항균제를 이용한 인조스웨드 직물의 항균코팅에 관한 연구)

  • This study was carried out to develope antimicrobial artificial suede by coating with water soluble polyurethane resin and the copolymer of N,N'-dialkyl-N,N'-dialkyl ammonium chloride (DADAAC) and acrylamide as a antimicrobial additve. The copolymer of DADAAC and acrylamide was synthesized by free radical initiation and intra-intermolecular propagation, and the prepared copolymers had sufficient compatibility with water soluble polyurethane resin. The MIC values of the prepared copolymers and antimicrobial characteristics of the artificial suede coated by polyurethane were evaluated. With the increase in the proportion of DADAAC, which is antimicrobially active part in the DADAAC/acrylamide copolymers, the MIC value becomes lower. The MIC value of DADAAC-AA (1 : 1) copolymer is below 30 ppm against S. aureus, and below 90 ppm against K pneumoniae. The artificial suede coated by water soluble polyurethane resin with 1.0% owl concentration of DADAAC/acrylamide copolymer has good antimicrobial fastness as to show colony reduction of above 90% and 80% against S. aureus and K. pneumoniae respectively in the shake flask test after 10 times of washing, and above 95% and 85% after 10 times of dry-cleaning. The elastic recovery of coated suede fabric is not affected up to 1.0% owf concentration of DADAAC-AA copolymer in the polyurethane coating.

  • Polypropylene(PP) films were treated with plasma glow discharge to produce peroxy radicals on the surfaces. The peroxy radicals formed on the PP film surfaces were subsequently used for the graft polymerization of acrylic acid and acrylamide in an aqueous solution by heating, respectively. Introduction of acrylic acid and acrylamide on the PP film could be confirmed by the observation of carbonyl and primary amine absorptions based on carboxylic acid and amide, respectively. And introduction of functional group could be confirmed by weight analysis and ESCA. The water contact angle(90$^{\circ}$) of PP film was constant, irrespective of elapsed time, while plasma-treated and functional monomer-grafted PP films were slowly increased with elapsed time, showing the rearrangement of surface polar groups in air condition. The water contact angle$(90^\circ)$ of PP film was decreased by the plasma treatment$(56^\circ)$ and further decreased by the grafting of acrylic acid$(34^\circ)$ and acrylamide$(37^\circ)$, indicating increased hydrophilicity of the modified surfaces. The water contact angle of plasma-treated PP film increased a little as time elapsing. The half-life periods of surface voltage on acrylic acid-(31sec) and acrylamide-grafted PP(42sec) were significantly decreased when compared to those on PP(950sec) and plasma-treated PP film(241sec). In the experiments using acid, basic and disperse dyes, absorbance and $\Delta{E}$ values of functional monomer-grafted PP films were significantly increased than that of oxygen plasma-treated one.

  • Styrene-acrylamide co-polymer was immobilized on porous partially sub-$2{\mu}m$ silica monolith particles and inner surface of fused silica capillary ($50{\mu}m$ ID and 28 cm length) to result in ${\mu}LC$ and CEC stationary phases, respectively, for separation of anomeric D-glucose derivatives. Reversed addition-fragmentation transfer (RAFT) polymerization was incorporated to induce surface polymerization. Acrylamide was employed to incorporate amide-functionality in the stationary phase. The resultant ${\mu}LC$ and CEC stationary phases were able to separate isomers of D-glucose derivatives with high selectivity and efficiency. The mobile phase of 75/25 (v/v) acetonitrile (ACN)/water with 0.1% TFA, was used for HPLC with a packed column (1 mm ID, 300 mm length). The effects of pH and ACN composition on anomeric separation of D-glucose in CEC have been examined. A mobile phase of 85/15 (v/v) ACN/30 mM sodium acetate pH 6.7 was found the optimized mobile phase for CEC. The CEC stationary phase also gave good separation of other saccharides such as maltotriose and Dextran 1500 (MW~1500) with good separation efficiency (number of theoretical plates ~300,000/m).

  • We have studied the polymerization kinetics of acrylamide in aqueous solution with potassium persulfate as an initiator by using quantitative real-time in situ IR spectroscopy and monitoring the profiles of peaks in the range 1900-850 cm$\^$-1/. The conversion of acrylamide was calculated from the disappearance of the peak at 988 cm$\^$-1/, which is the out-of-plane bending mode of the=C-H unit, normalized to the C=O stretching peak at 1675 cm$\^$-1/, as an internal standard. For reaction temperatures in the range 40-65$^{\circ}C$ and initiator and monomer concentrations of 0.9-2.6 mmol/L and 0.5-1.1 mol/L, respectively, we deduced that the rate of monomer consumption follows the relation R$\_$p/=k[K$_2$S$_2$O$\_$8/]$\^$0.5/ [Μ]$\^$1.35${\pm}$0.10/. In addition, we obtained activation parameters from an evaluation of the kinetic data.

  • A bacterial strain of Brevibacterium sp. CHI was isolated from soil and used to produce an enzyme (nitrile hydratase) necessary for carrying out the bioconversion of acrylonitrile to acrylamide. Various immobilization methods and enzyme stability were investigated. The nitrile hydratase showed the maximum stability at pH 7 for the free cells. EDTA and phenyl methyl sulfonyl fluoride were selected as the protease inhibitor and the enzyme stability was evaluated by changing inhibitor concentration. Acrylamide beads were the best carriers among four carriers we tested in terms of stability and physicoehemical strength. The storage stability of the immobilized cells decreased with increasing acrylamide concentration of the gel phase at 4$^{\circ}C$, and was very low at acrylarnide concentration above 25%.

  • Polystyrene foam (styrofoam) was treated with low-temperature oxygen plasma by means of immobilization and grafting techniques in order to modify its hydrophobic surface property to hydrophilic one using hydrophilic monomers of acrylic acid and acrylamide, and its surface chemical structure, morphology, and hydrophilicity were examined by ESCA, field emission scanning electron microscope (FESEM), and contactangle meter. The experimental evidences, such as the increases of O/C and N/C ratios in ESCA spectrum, thin film deposition, decrease in contact-angle, strongly suggested that the plasma treatments were useful methods for the preparation of hydrophilic surface. Contact angle diminished drastically from $84^{\circ}$ to $18{\sim}19^{\circ}$. Acrylamide, compared to acrylic acid, appeared to play a decisive role, and to be more powerful agent for improving its surface hydrophilicity.

  • Food products selected based on their annual sales and international acrylamide research data were analyzed for quantitation of acrylamide. Samples including raw food, substitute meal, snack, drink, and sauce products were analyzed by LC/MS/MS methods adopted by PDA. Upon comparison, concentrations of acrylamide in these products were similar to those analyzed in other countries.

Development of Isotope Dilution-Liquid Chromatography/Tandem Mass Spectrometry as a Candidate Reference Method for the Determination of Acrylamide in Potato Chips

  • An isotope dilution-liquid chromatography/tandem mass spectrometric method was developed as a candidate reference method for the accurate determination of acrylamide in potato chips, starch-rich foodstuff cooked at high temperature. Sample was spiked with 13C3-acrylamide and then extracted with water. The extract was further cleaned up with an Oasis HLB solid-phase extraction (SPE) cartridge and an Oasis mixed-phase cation exchange (MCX) SPE cartridge. The extract was analyzed by using LC/ESI/Tandem MS in positive ion mode. LC with a medium reversed-phase (C4) column was optimized to obtain adequate chromatographic retention and separation of acrylamide. MS was operated to selectively monitor [M+H]+ ions of the analyte and its isotope analogue at m/z 72 and m/z 75, respectively. Sample was also analyzed by the LC/MS with selectively monitoring the collisionally induced dissociation channels of m/z 72 → m/z 55 and m/z 75 → 58. Compared to the LC/MS chromatograms, the LC/MS/MS chromatograms showed substantially reduced background chemical noises coming from solvent clusters formed during ESI spray processes and interferences from sample matrix. Repeatability and reproducibility studies showed that the LC/MS/MS method is a reliable and reproducible method which can provide a typical method precision of 1.0% while the LC/MS results are influenced by chemical interferences.

The Stabilization of Liquid Crystal Emulsions by Acrylamide Copolymers (Acrylamide Copolymers에 의한 Liquid Crystal Emulsions의 안정성에 관한 연구)

  • There are several methods to fabricate Polymer Dispersed Liquid Crystal(PDLC) films. One of them, so-called Nematic Curvilinear Aligned Phase(NCAP) film, is based on emulsion technology. To produce NCAP systems various water soluble polymers, such as partially hydrolyzed polyvinylalcohol(PVA) and polyvinyl pyrrolidone(PVP), which can form stable emulsion of liquid crystal(LC) without any stabilizers were used. In this work, we studied the dependence of emulsion stability on nature and composition of copolymers composed of water-soluble and water-insoluble moiety. We found that interfacial surface tension depends on the composition of comonomer, the copolymer concentration in the water, and the nature of hydrophobic chain. The Acrylamide -styrene(AA-ST) copolymer showed the lowest interfacial surface tension among the tested copolymers at the same concentration. Since the interfacial surface tension decreases with increasing the compatibility of copolymer with LC phase the AA-ST copolymer has the best compatibility with LC molecules. It is believed that molecules adsorbing easily on the surface of LC droplets allows the LC emulsion system to be more stable.
