An In vitro Enzymatic Digestion Method for Estimation of the Acrylamide Contents of Foods

  • In this study, the acrylamide contents of foods were estimated via liquid chromatography (LC)/mass spectrometry (MS)/MS after the food matrix constituents had been degraded with digestive enzymes (i.e., pepsin and pancreatin) and extracted with water. The quantities of acrylamide released from samples of cereal, potato chips, peanuts, and coffee were $62{\pm}5.1,\;970,\;106{\pm}20$, and 890 ppb, respectively. No acrylamide was detected in samples of soybean curd (tofu), fish cake, and ham. Compared to the amounts of acrylamide detected after extraction with water only, we noted no significant differences in the soybean curd, fish cake, potato chip, ham, and coffee samples. However, the quantities of acrylamide released from the cereal and peanut samples were approximately 2-fold larger following pretreatment with the digestive enzymes. This study presents a new in vitro enzymatic digestion method which allows for a more accurate estimation of the acrylamide contents of foods.

The Effect of Acrylamide on the Ultrastructures of Nervous System of the Mouse (생쥐 신경계의 미세구조에 미치는 Acrylamide의 영향)

  • The effed of acrylamide on the nervous system has heen morphologically studied using light and electron microscopes. The light micrographs on central and pedpheral nervous tissues of mouse treated with acrylamide monomer showed total vacuolation of spinal cord, cell degradation containing neuron and neuroglia, and distal nerve fiber degeneration. The electron micrographs showed ultrastrudural changes. Abnormal mitochondria in neuron, splitting of myelin sheath in lumbar ventral root nerve, partial disintergration of myelin sheath and axoplasmic degeneration in sciatic nerve, and overafl polyneuropathies in nervous system were observed. These results suggest that acrylarnide intoxicated mouse shows distal behavioral neuropathy as an earlist clinical sign, but the initial effect of acrylamide on the nervous system seems to appear at nearly the same time in both central and peripheral nervous systems.

  • The effects of acrylonitrile and acrylamide on the enzyme action of nitrile hydratase of Brevibacterium sp. CH1 and CH2 strains used for the biotransformations of nitriles were studied. The excessive substrate (acrylonitrile) and product (acrylamide) inhibited the enzyme activity competitively. In comparison with 0.2 mol/l of CH1 strain, the substrate inhibition of CH2 strain began to appear only at a high acrylonitrile concentration of 0.91 mol/l. In a packed bed reactor, dispersed plug flow model was proposed and this model was proved to be valid by the experiment. Also acrylamide productivity decreased sharply when acrylamide concentration in the substrate solution exceeded 20% (wt/v).

Isolation of a Pseudomonas aeruginosa Strain Capable of Degrading Acrylamide

  • A new strain of Pseudomonas aeruginosa growing in a rice field contaminated with herbicide and effluents of a factory manufacturing explosives was isolated. This isolate showed excellent growth in unusually high concentration of acrylamide (60 mM). It utilized acrylamide as the sole source of carbon and nitrogen for growth. Other amides such as acetamide, butyramide, isobutyramide, and methacrylamide were also utilized for the growth by this isolate. Acrylamide was degraded into acrylic acid and ammonia by the enzyme amidase. More than $65\%$ of added acrylamide (40 mM) was converted into acrylic acid after 40 h of growth of the culture. Amidase activity was inducible, the highest activity being observed with isobutyramide ($12.5{\mu}M$ ammonia/mg protein/min). These results demonstrate that this bacterium can degrade a variety of amides.

  • Photografting reaction onto styrene-butadiene rubber (SBR) as a function of monomer concentration, irradiation time and carbon black content has been studied using ultraviolet (UV). Acrylamide and benzophenone are used as monomer and photoinitiator, respectively. FT-IR ATR and static contact angle analysis using distilled water are used to measure the graft ratio of acrylamide onto SBR surface. Graft ratio of acrylamide increases with acrylamide concentration and irradiation time and contact angle tends to decrease with increasing graft ratio. It is observed that graft ratio increases with carbon black content.

  • This study was conducted to investigate the effect of pH on the formation of acrylamide and acrylate from glucose and fructose reacting with amino acid enantiomers by the Maillard reaction. The acrylamide content was increased with increasing pH, except for Fru/D-Asn system. Both acrylamide and acrylate contents were higher in the glucose-based system compared to the fructose-based system at pH 10.0. However, according to amino acid enantiomers, only the acrylamide content showed a difference in the fructose-based system. In addition, the acrylate content was increased with increasing pH except in the Glc/L-Asn system. Acrylate formation was observed specifically at pH 4.0 for both the Glc/D-Asn and Fru/D-Asn systems.

  • Acrylamide, N-alkyl substituted acrylamides and N,N-diethylacrylamide were reacted with aryl bromides in the presence of triethylamine and palladium acetate-triorthotolyl phosphine catalyst to form the various substituted aryl conjugated enamides. These reactions proceeded selectively and (E)-isomer of aryl conjugated enamides was obtained. N-alkyl substituted acrylamides were more reactive than acrylamide or N,N-diethylacrylamide and gave high yields of vinylated products. Aryl bromides with electron withdrawing group showed good reactivity but aryl bromides with electron donating group showed poor reactivity or no reactivity for acrylamide or N,N-diethylacrylamide.

Simple Analytical Method for Acrylamide in the Workplace Air Adsorbed by Charcoal Tube (활성탄관을 사용하여 포집한 작업환경 공기중 아크릴아마이드의 분석법)

  • For the ambient monitoring of acrylamide, the adequate condition of sampling and analysis was checked. The adequate adsorbents and desorption solvents were tested. The combination of charcoal tube as a adsorbent and acetone as a desorption solvent showed 87% desorption efficiency. Flame ionization detector was used to detect acrylamide. The detection limit was 0.814 mg acrylamide in 1 L acetone. It is the equivalent concentration of 0.0203 mg acrylamide in $1m^3$ air if the volume of air collected was 40L. The permissible exposure level (PEL) of acrylamide in the workplace air recommended by Occupational Safety and Health Administration (OSHA, USA) is 0.3 mg acrylamide in $1m^3$ air. So, it is very simple and economic analytical method for acrylamide to be set in the industrial hygiene laboratories.

  • Non-aggregated nanoscale $\alpha$-Al$_2$O$_3$ powders were prepared successfully by polyacrylamine (PAA) gel method. The method was very simple and polymer network inhibited the aggregate of $\alpha$-Al$_2$O$_3$ powders. In this investigation, nanoparticles of $\alpha$-Al$_2$O$_3$ with a diameter of about 8-15 nm were fabricated by calcining the gel precusors with various concentrations of aluminum sulfate, acrylamide and N,N'-methylene-bis-acrylamide (BIS) in air at 110$0^{\circ}C$ for 2 h. The molar ratio of aluminum sulfate to acrylamide did not have any influence on the size of particles. On the other hand, as the molar ratio of BIS to acrylamide increased, the size of nanoparticles decreased.

  • N-n-Propylacrylamide has been synthesized from acrylamide and n-propyl bromide. N -n Propylacrylamide was copolymerized with acrylamide at $60^{\circ}C$ in tetrahydrofuran using ${\alpha},{\alpha}'$-azobisisbutyronitrile as initiator. The synthesized monomer and copolymers have been identified by NMR and FT-lR spectrophotometer. The swelling properties of the crosslinked homopolymers were investigated at different temperatures. Three types of hydration layer around the back-bone structure of gels were determined. The thermal properties of copolymers were also measured by differential scanning calorimeter and thermogravimetry. As the amounts of N-n-propylacrylamide are increased, the enthalpic changes associated with endothermic transition and glass transition of the copolymers are decreased. As the amount of N-n-propylacrylamide is increased, the thermal stability is increased. The activation energies of thermal decomposition and dehydration for the poly (acrylamide-co-N -n-propylacrylamide) have been evaluated by Freeman and Carroll's method. As the amounts of N-n- propylacrylamide are increased, the activation energies of thermal decomposition and dehydration are increased.
