Preparation of Red Ginseng Extract Rich in Acidic Polysaccharide from Red Tail Ginseng Marc Produced After Extraction with 70% Ethyl Alcohol (홍미삼 알콜 추출박을 이용한 산성다당체 다량 함유 홍삼 엑기스 제조)

  • In this study, we investigated the appropriate conditions to extract acidic polysaccharide and to prepare red ginseng extract being rich in acidic polysaccharide from red tail ginseng marc produced after manufacturing alcoholic extract from red tail ginseng. Amount of acidic polysaccharide in red tail ginseng marc was about 11%. The best condition for the extraction of acidic polysaccharide from the marc was using of 3~5 mg of $\alpha$-amylase/g residue/15 ml of distilled water, and the amount of acidic polysaccharide in water extract of the residue treated with $\alpha$-amylase was about 27%. So, it is possible to manufacture red ginseng extract being rich in acidic polysaccharide using water extract of red tail ginseng alcoholic residue as extraction solvent. From the above results, we suggest that red tail ginseng residue produced by manufacturing alcoholic extract of red tail ginseng has high potencies in the utilization of waste material.

  • This study was carried out to investigate the optimum conditions for extraction of soluble acidic polysaccharides from ginseng marc. Method of carbazole-sulfuric acid was applied to determine the amount of acidic polysaccharides in ginseng marc. The amounts of soluble acidic polysaccharides in water extract of ginseng marc were increased with increasing extraction temperature. The contents of acidic polysaccharides were not significantly different despite the extraction time increasing from 0.5 hours to 6 hours. To estimate the rehydration rate of the freeze dried polysaccharide, the extracted acidic polysaccharide fraction powder was determined the amount of soluble acidic polysaccharides by carbazole-sulfuric acid method again. The rehydration rate of acidic polysaccharides from water-extract of red ginseng marc at room temperature was 100%. On the other hand, the rehydration rate of acidic polysaccharide of red ginseng marc at boiling temperature was about 50%. The rehydration rate of acidic polysaccharides from water-extract of white ginseng marc at room temperature was 50%. The rehydration rate of acidic polysaccharide of red ginseng marc at boiling temperature was about 40%. The rate of soluble acidic polysaccharide of Red Ginseng is higher than that of White Ginseng. We can find out the maximum extraction method of soluble acidic polysaccharide from ginseng marc.

  • Rice endosperm, the portion that remains after milling, is the part of the rice seed that is primarily consumed as a source of nutrients. There have been many studies on polysaccharides, such as hemicellulose, cellulose, and pectins, derived from the cell walls of various plant groups. It has been reported that the acidic polysaccharide fractions, which contain water-soluble pectins that have been shown to have pharmacological effects in vivo and in vitro, have common chemical structures that include galacturonic acid polymers, rhamnose, arabinose, and galactose. However, few studies have been conducted on the acidic polysaccharides contained in the endosperm of rice. In this study, we quantitatively compared the differences in the acidic polysaccharide contents from samples from two of the main varieties of rice consumed as staple foods, japonica and indica, using a colorimetric method. Rice samples were collected from 39 different regions in Korea, China, Thailand and Vietnam. Acidic polysaccharide fractions were obtained by precipitation of the alcohol-insoluble residue (AIR) and enzyme treatment of each sample. The total amount of carbohydrates and uronic acid in each acidic polysaccharide fraction were measured using the phenol-sulfuric acid method and the carbazole-sulfuric acid method, respectively. The differences in the total polysaccharide contents in the acidic polysaccharide fractions were not statistically significant (p = 0.07), but the uronic acid contents were significantly different between the two groups (p = 0.04).

  • The method for colorimetric determination of acidic polysaccharide from Panax ginseng was investigated. It is possible to apply the method of carbazole-sulfuric acid to determination of pectin, and also to measure the amount of pectin in the mixture of various high molemlu compounds such as starch. cellulose and gum, etc. When the method of carbazole-sulfuric acid was applied to determine the amount of acidic polysaccharide, optical density at 525 nm increased linearly with an increase in the concentration of pure acidic polysaccharide. Effective extraction temperature with water for the determination of the amount of ginseng acidic polysaccharide (GAP) was $80{\circ}C$. In order to separate or concentrate GAP it was appropriate to precipitate the extract only once with 80% ethyl alcohol. GAP was very stable at $100{\circ}C$ for 4 hrs in aqueous solution and between pH values of 5.0~ 12.0.

  • Toxohormone-L is a lipolytlc factor, found in ascites fluid of sarcoma 180-bearing mice and of patients with hepatoma. A substance that inhibited the lipolytic action of Toxohormone-L was isolated from Korean red ginseng powder. This substance had a pectin-like a-1,4-polygalacturonan backbone with some acetoxyl groups, and so was an acidic polysaccharide. Acidic polysaccharide was found to inhibit significantly toxohormone-L-induced lipolysis at its concentration of 10$\mu\textrm{g}$/ml.

  • This study was devised to observe the inhibitory effects of ethanol treatment precipitate (crude acidic polysaccharide) from Korean red ginseng on a lipolytic action of Toxohormone-L which has been known as lipolytic and anorexigenic factors. Toxohormone-L was obtained by partial purification of the ascites fluid from mice which had been inoculated with sarcoma-180. The yields of crude acidic polysaccharide from Korean red ginseng was 63.5%. In vitro, at the concentration of 500rg /ml, the inhibition rate of lipolysis by the crude acidic polysaccharide of Korean red ginseng was 38.8% and the total inhibitory activity per gram of ginseng material was 4,928 nit. In vivo, the red ginseng polysaccharide(40mg/ml in saline soon.) 16ul/g of body weight was injected to the sarcoma-180 bearing mice once In 3 days until death. The effects against the extension of life span was little but body weight gain of sarcoma-180 bearing mice decreased significantly by administration of Korean red ginseng polysaccharide compared to those of the control group.

  • Effect of an acidic polysaccharide fraction In Korean white and red ginseng on lipolytic action of Toxohormone-L was studied. Crude acidic polysaccharide fraction was extracted from main and lateral root of Korean white and red ginseng separately and purified several times. Inhibitory effect of crude polysaccharide fraction was determined by unit (1 unit is loft inhibition rate per Is sample). Yields of purified crude polysaccharide fraction of main and lateral root of red ginseng were 2.9 and 2.2 times higher than those of white ginseng, respectively. Inhibitory effects of main root of white and red ginseng, 11.hen final reaction concentrations of sample were 50, 100, 200, 500 $\mu$g/ml, were 37.2가 and 23.7% higher than those of lateral root of white and red ginseng. Inhibitory effect of main root of red ginseng was 2.3 times higher than that of white ginseng.

The contents of starch in water-extract of red ginseng marc were increased with increasing extraction temperature. The starch amounts in water extract of red ginseng marc extracted for 48 hours were increased. The yields of polysaccharide precipitated from water-extract of red ginseng marc were increased with increasing extraction temperature. The hydration rate of acidic polysaccharides and starch from water-extract of red ginseng marc were decreased with increasing extraction temperature. The contents of starch were not significantly different despite of the extraction time increasing from 6 hours to 48 hours at $8^{\circ}C$. However, the rehydration rate of acidic polysaccharide for 48 hours were decreased at $8^{\circ}C$. The rehydration rate of acidic polysaccharide and starch extracted from 6 hours to 24 hours at $25^{\circ}C$ were not significantly different, but those extracted for 48 hours were increased. From the above results, we suggest that by altering the extraction conditions in red ginseng marc it is possible to develop optimum conditions for extraction that modulate the proportions of acidic polysaccharide and starch.

Preparation of Ginseng Concentrate with High Content of Acidic Polysaccharide from White Tail Ginseng Marc (백삼 알코올 추출박을 이용한 산성다당체 다량 함유 백삼 농축액 제조)

  • Preparation of ginseng concentrate with high content of acidic polysaccharide from white tail ginseng marc that was obtained after preparation of white tail ginseng extract. As a result of extraction of white tail ginseng under various concentrations of ethanol (0∼90%), both amount of acidic polysaccharide and extraction yield decreased by increasing the ethanol concentration. However, acidic polysaccharide extracted by water from white tail ginseng marc was increased in accordance with the increase of ethanol concentration. The optimal condition for the extraction of acidic polysaccharide from the marc was treatment of $\alpha$-amylase in 390∼650 unit/g residue/15 mL of distilled water for 5 min at 4$0^{\circ}C$. The amount of acidic polysaccharide in water extract of the marc was increased from 8.3% to 10.5% by the treatment of $\alpha$-amylase. A new ginseng extract mixture was manufactured by mixing 50% ethanol extract of white tail ginseng and water extract of alcoholic residue in the ratio of 8:2 (w/w). Crude saponin content and acidic polysaccharide content were 10.5% and 17%, respectively. The mixture had a same crude saponin content and twice acidic polysaccharide content comparing to 50% ethanol extract of white tail ginseng. It suggests that preparation of new ginseng concentrate with high content of acidic polysaccharide from white tail ginseng marc has high potencies in the utilization of waste material.

Study on Atitumor and Immunomodulating Acivities of Polysaccharide Fractions from Panax ginseng : Comparison of effects of neutral and acidic polysaccharide fraction

  • The crude polysaccharide from Panax ginseng prepared by hot water extration and precipiation with ethanol was further fractionated into neutral and acidic fractions by DEAE- cellulose ion exchange chromatography. The chemical compositions were 85.0% carbohydrorate and 15.0% protein for the neutral fraction, and 28.4% carbohydrate, 10.0% protein and 29.0% uronic acid for the acidic fraction. The acidic fraction was more effective in increasing of the ratio of spleen to body weight, the number of antibody secreting cells to SRBC and phagocytic activity of reticuloendothelial system, as well as antitumor activity against the solid form of sarcoma 180 in ICR mice than the neutral fraction. All polysaccharide fractions were mitogenic to cultured spleen cells of C57BL/6 mice. However, FA was different from FN in the co-mitogenicities with lectin mitogens. Both crude and acidic fractions potentiated remarkably the mitogenic activity of PHA-P or LPS in dose-dependent manner but neutral fraction enhanced only that of LPS. Three polysaccharide fractions had no effect on that of Con A. These results suggest that the acidic fraction may stimulate B and Td cells as well as macrophages while the neutral fraction may simulate only B cells and macropages.
