A Study on the Amino Acid Components Soil Humus Composition (토양부식산(土壤腐植酸)의 형태별(形態別) Amino 산(酸) 함량(含量)에 관(關)한 연구(硏究))

  • Contents and distribution of amino acids in the humic acid and fulvic acid fractions of different types ($R_p$, B, A, P) were investigated. Extracted humic and fulvic acids were purified and analyzed. The results are summarized as the following: (1) Composition of Humus The total humus ($H_T$), amount of humic acid (a), amount of fulvic acid (b), and ${\Delta}logK$ all decrease in the order of $R_p$ > B > A > P type. The same trend was observed in the total nitrogen and carbon. (2) Contents and composition of amino acids in humic acids. 1) The total amounts of amino acids in the humic acid fraction of different types were in the following order for soils under coniferous forest trees: $R_p$ > B > A > P type, but for soils under deciduous forest trees the order was P > A > $R_p$ > B type. There were positive correlationships between total amino acids and total carbon and ${\Delta}logK$ for humic acids from soils under coniferous forest trees, but a negative correlationship was existed. between total amino acids and C/N ratios. No significant correlation was found for samples taken from soils under deciduous forest trees. 2) The ratios of one group of amino acids to the others were compared. The ratios of acidic amino acids were in the order of P > $R_p$ > B > A type. those of neutral amino acids followed the order of $R_p$ > B > A > P type and those of the basic amino acids were in the order of B > A >$R_p$ > P type for soils under coniferous forest trees. Contents of total amino acids were in the order of the neutral > the acidic > the basic amino acids. For the soils under deciduous forest trees the order of the ratio was different. Acidic amino acids followed the order of A > P > B > $R_p$ type, neutral ones followed the order of P > $R_p$ > A > B type, and the basic amino acids did the order of $$P{\geq_-}$$ A > B $$\geq_-$$ $-R_p$ type where the difference was very small. 3) In general aspartic aicd, glycine and glutamic acid were the major components in all samples. Histidine, tyrosine and methionine belonged to the group contained in a small amount. (3) Contents and composition of amino acids in fulvic acids. 1) The total amounts of amino acids of different types of fulvic acids were in the order of $R_p$ > B > P > A type regardless of origin of samples. There were positive correlationships observed between the toal amino acids and total carbon and ${\Delta}logK$ for soils under coniferous forest trees. For soils under deciduous forest trees, positive correlationships were observed among total amino aicds, total nitrogen, total humus ($H_T$), total humic aicd (a), and ${\Delta}logK$, but a negative correlationship existed between total amino acids and C/N ratio. 2) Thr ratio among acidic amino acids, neutral amino acids and basic amino acids of different types were $R_p$ > B > P > A type. In this respect there was no difference between the two soils. 3) In general glycine, aspartic acid, and alanine were the major constituents in all samples of different types, while tyrosine and methionine were contained in a small amount. Virtually no amount of arginine was measured.

Functional Properties of Soybean Curd Whey Concentrate by Nanofiltration and Effects on Rheological Properties of Wheat Flour Dough (나노여과에 의한 순물 농축액의 기능적 특성 및 밀가루 반죽의 리올로지 성질에 미치는 영향)

  • This study was designed to investigate the feasibility of utilizing concentrates of sunmul(soybean curd whey), the waste by-product of soybean curd processing, as functional food ingredients. Sunmul was concentrated by nanofiltration fo11owing ultrafiltration and then freeze-dried. The oil adsorption capacity of the nanofiltraion(NF) powder(97.33g/100g) was similar to that of sunmul powder(94.17g/100g), but was lower than that of ISP(isolated soy protein). However, the water holding capacity of NF powder could not be determined because the NF powder completely dissolved in water. The protein solubilities of sunmul powder and ISP in distilled H$_{2}$O, 0.1M and 0.5M NaCl were lowest at pH 4.0 and increased at more acidic or alkaline conditions. However, the protein solubility of NF powder was at its minimum at pH 6.0 and increased at more acidic or alkaline conditions. Emulsifying activity indexes of NF powder in 4% and 6% solution were minimal at pH 4.0 and 6.0, respectively, which were 3 to 8 times lower than that of sunmul powder. The emulsion stability of 4% sunmul solution was lowest at pH 4.0, but that of NF powder was highest at pH 5.0 and decreased at more acidic or alkaline conditions at all concentrations of solution. The total free amino acid contents of protein in sunmul, and NF power were 99.07 and 2,110.10mg%, respectively, and NF powder exhibited especially high threonine content. Rapid viscosity analysis of dough with 1 to 5% added NF powder demonstrated that all of the peak and final viscosities decreased with increasing NF powder concentration compared to the control.

  • The adsorption and ion exchange properties of tin phosphate were studied in an aqueous solution of KCl, varing the pH and metal ion concentration in the solution. The data were explained on the basis of chemical equilibrium. Tin phosphate behaved as an acidic ion exchanger and had an adsorption selectivity toward the bivalent transition metal ions in the following order : Cu+2 > Co+2 > Ni+2. As in the case of a weekly acidic exchanger, the change in hydration of metal ions played the dominant role in determining the selectivity of tin phosphate. In all cases the extent of adsorption increased with an increase in temperature and concentration. The apperance of irregular kinks in the titration curves justified the presence of several exchangeable adsorption sites with different pKa values.

Studies on the Marginal leaf chlorosis of Ginseng Plant (1) The Effect of Excess Manganese uptake on the occurrence of marginal leaf chlorosis (인삼의 엽록형 황증에 관한 연구 제1보 Mn의 과잉흡수가 엽록형 황증발생에 미치는 영향)

  • This study was conducted to determine the cause of the occurence of marginal leaf chlorosis in ginseng plants (Panax ginseng C.A. Meyer), and to determine its emersion in fields (practically) and in pots (experimentally). The following results were obtained. In the Present investigation, ginseng plants raised in acidic soil containing a high a moue t of Mn showed marginal leaf chlorosis. Henre it Ivas suggested that the shoot growth and root weights became grad gractually lower. The leaves having marginal leaf chlorosis contained low amounts of N, P,. Ca, Mg, and Na and the Fe/Mn ratios were low. There was a corresponding increase in Mn uptake. It was founrl that in soils where marginal leaf chlorisis occured the pH urar brlolv 4.2 to 4.9 and the Ca, Mg and Na content was decreased thus effectively increasing the available manganese in the soil. The Mn/Fe ratios in the yellow leaf margins of ginseng Plants affected by the Mn toxicity was over 2.0 compared to the general Mn/Fe ratio of 0.50 for healthily leaves, stems and roots. Typically when ginseng plants grow fields having soil with a pH below about 5.0, there tenor to be an uptake of excess Mn. When ginseng plants are grown in a nutrient sand culture solution It with an increased Mn concentration, they accumulate large amounts of Mn in the roots and in the shoots. In both casts marginal leaf chlorosis appeared in the emersions. In the Present investigation, ginseng plants raised in acidic soil and containing a high amount of Mn showed marginal leaf chlorosis.

Geochemical Behavior of Metals in the Contaminated Paddy Soils around Siheung and Deokeum Mines through Laboratory Microcosm Experiments (실내 microcosm실험에 의한 시흥광산 및 덕음광산 주변 오염 논토양내 중금속의 지구화학적 거동 연구)

  • Seasonal variations in vertical distributions of metals were investigated in the contaminated paddy soils around Siheung Cu-Pb-Zn and Deokeum Au-Ag mines. Geochemical behavior of metals was also evaluated with respect to redox changes during the cultivation of rice. Two microcosms simulating the rice-growing paddy field were set up in the laboratory. The raw paddy soils from two sites showed differences in mineralogy, metal concentrations and gecochemical parameters, and it is suggested that high proportions of exchangeable fractions in metals may give high dissolution rates at Deokeum. In both microcosms of Siheung and Deokeum, redox differences between surface and subsurface of paddy soils were maintained during the flooded period of 18 weeks. Siheung soil had neutral to alkaline pH conditions, while strongly acidic conditions and high Eh values were found at the surface soil of Deokeum. The concentrations of dissolved Fe and Mn were higher in the subsurface pore waters than in interface and upper waters from both microcosms, indicating reductive dissolution under reducing conditions. On the contrary, dissolved Pb and Zn had high concentrations at the surface under oxidizing conditions. From the Siheung microcosm, release of dissolved metals into upper waters was decreased. presumably by the trap effect of Fe- and Mn-rich layers at the interface. However, in the Deokeum microcosm, significant amounts of Pb and Zn were released into upper water despite the relatively lower contents in raw paddy soil, and seasonal variations in the chemical fractionation of metals were observed between flooded and drained conditions. Under acidic conditions, rice may uptake high amounts of metals from the surface of paddy soils during the flooded periods, and increases of exchangeable phases may also increase the bioavailability of heavy metals in the drained conditions.

Increase of Functional Saponin by Acidic Treatemnt and Temperature of Red Ginseng Extract (홍삼엑기스의 산(pH) 및 온도처리에 의한 기능성 사포닌 함량증대)

  • To increase the contents of functional ginsenosides by conversion, especially ginsenoside-$Rg_3$ and $Rh_2$, the extracts of red ginseng were treated with high temperature and citric acid or apricot extract. When the extracts were subject to $120^{\circ}C$ for 2 hours, the content of ginsenoside-$Rg_3$ was increased 2 times than in control. When the extracts were subject to $120^{\circ}C$ and acidic conditions adjusted with citric acid, the ginsenoside-$Rg_3$, was detected 2.8 times, but other ginsenoside were decreased heavily to 65%. When the extract were treated with for 12 hours at $80^{\circ}C$, the content of ginsenoside-$Rg_3$ was increased to 3.3 times, Also, when the red ginseng extracts were treated with apricot extract, the ginsenoside-$Rg_3$ was detected to 4 times than in control, but other ginsenoside were decreased lightly to 35%, not same as at the $120^{\circ}C$ treatment.

Changes in Quality of Soybean Sprouts Washed with Electrolyzed Water during Storage (전기분해수 처리에 따른 콩나물의 저장 중 품질변화)

  • The storage quality of soybean sprouts washed with various electrolyzed waters was investigated. The washing solutions consisted of tap water (TW), sodium hypochlorite electrolyzed water (SHEW), and slightly acidic electrolyzed water (SAEW). The number of bacteria on the soybean sprouts after 5 min of exposure to TW, SHEW, and SAEW resulted in >0.5, 2.0, and 2.0 log CFU/g reductions, respectively. At both $4^{\circ}C$ and $25^{\circ}C$, the number of bacteria, weight loss ratio, and b value rapidly increased, and pH and L value rapidly decreased in soybean sprouts washed with TW. Whereas, the number of bacteria, pH, weight loss ratio, and color slowly changed in the soybean sprouts washed with SHEW and SAEW. Consequently, these results indicate that washing with electrolyzed water is an effective means of maintaining the quality and enhancing the shelf-life of soybean sprouts; both SHEW and SAEW reduced bacterial growth without affecting the other properties of soybean sprouts during storage.

Analysis of the Plasma Proteins from Bovine and Porcine Blood and Their Emulsifying Activity (소 및 돼지 혈액에서의 혈장단백질 분석 및 이들의 유화능)

  • Plasma proteins were obtained from bloods of slaughtered bovine and porcine and analyzed by Fast Protein Liquid Chromatography (FPLC). Serum albumin content decreased in the following order: Porcine Plasma Protein (PPP)> Bovine Plasma Protein (BPP)> Whey Protein Concentrate (WPC). Protein contents of BPP, PPP, and WPC determined by Kjeldahl method were 85.79%, 82.30%, and 84.38%, respectively. Compared to WPC, plasma proteins had higher emulsifying activity index (EAI) below 2% protein concentration and slightly lower EAI above 4% protein concentration. Plasma proteins had higher EAI in the acidic pH range and more dependence on NaCl than WPC. Also, EAI of plasma proteins with NaCl was higher in the acidic range than that of WPC. These results indicated that plasma protein can be utilized as a raw material for emulsifier.

Weather Patterns and Acid Rain at Kimhae Area (김해지역의 산성강우와 기압유형)

  • This study was carried out to investigate the characteristics of acidity in the precipitation and weather patterns that were influenced it at Kimhae area from March, 1992 to June, 1994. The range of pH value in the precipitation at Kimhae is 3.45 to 6.80 and the average is pH 4.62, and the major anion components associated with acidity in the precipitation are $Cl^-, SO_4^{-2}, NO_3^-$. These distributions are to be expected the influence of industrialization such as, urbanization and construction of industrial complex at Kimhae area and the long range trasporting of air pollutants from China. The weather patterns governing the acid rain at Kimhae were classified broadly into four types(Cyclone(type I-a, type I-b), Migratory Anticyclone(type II), Tropical Cyclone(type III), Siberia High(type IV) and weather pattern which had the most occurrence frequency of acid rain was type I-a and the average pH value of precipitation in this pattern was 4.45, and we are found that the source area of air mass which was accompanied with high acidic precipitation in Kimhae was the central China include with Peking through the analysis of surface weather maps, 850 hPa wind fields, and the streamline analyses.

Novel Cationic Microbial Polyglucosamine Biopolymer from New Enterobacter sp. BL-2 and Its Bioflocculation Efficacy

  • A new bacterium BL-2 excreting a novel cationic polyglucosamine biopolymer was isolated from the spoiled leaves of Chinese cabbage and identified as Enterobacter sp. BL-2. The isolated Enterobacter sp. BL-2 was cultivated in pH-stat fed-batch culture using acetic acid as the feeding stock at pH 8.0, resulting in 17.11 g/l of cells and 1.53 g/l of an extracellular biopolymer after 72 h. The excreted biopolymer was purified by a three-step procedure, involving ethanol precipitation and deproteinizations, to a nearly homogeneous state, and its molecular weight was found to be 106 kDa. It was composed of glucosamine, rhamnose, and galactose at a molar ratio of 86.4:1.6:1.0, respectively, indicating a rarely found novel high-glucosamine-containing biopolymer. The FT-IR and $^{13}C-NMR$ spectra of the novel cationic polyglucosamine biopolymer PGB-l revealed a close identity with chitosan from crab shell. It can effectively flocculate various suspended solids, including kaolin clay, $Ca(OH)_2,\;Al_{2}O_3$, active carbon, microbial cells, and acidic dyes.
