A study on the Equilibrium sorption of Silk fibroin by Reactive dye. (견에 대한 반응성 염료의 평형론적 연구)

  • The equilibrium sorptions of C.I. Reactive Blue 19 and C.I. Reactive Blue 19 and C.I. Acid Blue 138 on Silk fibroin were investigated in the range of 50$^{\circ}C$, 70$^{\circ}C$, 90$^{\circ}C$ and to the pH range from 2.0 to 10.5. The results obtained are summarized as follows: 1) The amount of sorption of reactive dye was increased with the decrease of pH in dyeing solution and temperature. The amount of fixation showed the maximum value to pH 8.5 and 70$^{\circ}C$. 2) In acidic region, the sorption behavior of acid dye was similar to that of reactive dye, and Langmuir adsorption constant was increased with the decrease of pH. 3) Langmuir constant of both dyes was decreased with the increase of temperature, while standard affinity was increased. 4) The reaction of both dyes was exothermic and the values of $\Delta$S$^{\circ}$ were positive. 5) It was found that the sorption behavior of dyes against Silk fibroin could be described as Langmuir adsorption and Nernst distribution in lower pH region.

Biological Activities of a Korean Traditional Prescription, Nogyongdaebotang (전통 한약탕제인 녹용대보탕의 생리활성 효과)

  • This study was performed to examine in vitro biological activities such as antioxidative, nitrite scavenging effect, tyrosinase inhibitory effect and antithrombotic activity of a Korean traditional prescription, Nogyong-daebotang, composed of oriental medical herbs and antler, nourishing the blood, helthiness, strengthening of vital power and promotion of growth. The concentration of total phenolic compounds of the prescription sample was 151.3$\pm$2.6 mg%. The electron donating abilities (EDA) by reduction of 2.2'-diphenyl-1-picrylhydrazyl (DPPH) was 80.9%, inhibition rate of lipid peroxidation in thiocyanate method used linoleic acid was 88.1%. Nitrite scavenging effects of the sample were more than 70% at acidic pH, but it was pH dependent, showing the highest at pH 1.2 and the lowest at pH 6.0. Inhibition rate against tyrosinase of the sample was above 80%. Platelet aggregation induced by ADP (adenosine-5'-diphosphate) was inhibited up to 30% and the inhibitory effect was dependent on concentration of the sample.

pH-mediated Regulation of Pacemaker Activity in Cultured Interstitial Cells of Cajal

  • Interstitial cells of Cajal (ICCs) are pacemakers in gastrointestinal tracts, regulating rhythmicity by activating nonselective cation channels (NSCCs). In the present study, we investigated the general characteristics and pH-mediated regulation of pacemaker activity in cultured interstitial cells of Cajal. Under voltage clamp mode and at the holding potential of -60 mV, the I-V relationships and difference current showed that there was no reversal potential and voltage-independent inward current. Also, when the holding potentials were changed from +20 mV to -80 mV with intervals of 20 mV, there was little difference in inward current. In pacemaker activity, the resting membrane potential (RMP) was depolarized (In pH 5.5, $23{\pm}1.5$ mV depolarized) and the amplitude was decreased by a decrease of the extracellular pH. However, in case of increase of extracellular pH, the RMP was slightly hyperpolarized and the amplitude was decreased a little. The melastatin type transient receptor potential (TRPM) channel 7 has been suggested to be required for intestinal pacemaking activity. TRPM7 produced large outward currents and small inward currents by voltage ramps, ranging from +100 to -100 mV from a holding potential of -60 mV. The inward current of TRPM7 was dramatically increased by a decrease in the extracellular pH. At pH 4.0, the average inward current amplitude measured at -100 mV was increased by about 7 fold, compared with the current amplitude at pH 7.4. Changes in the outward current (measured at +100 mV) were much smaller than those of the inward current. These results indicate that the resting membrane potential of pacemaking activity might be depolarized by external acidic pH through TRPM7 that is required for intestinal pacemaking activity.

Effect of immersion into solutions at various pH on the color stability of composite resins with different shades

  • Objectives: This study examined the color changes of a resin composite with different shades upon exposure to water with different pH. Materials and Methods: Nanohybrid resin composites (Filtek Z350XT, 3M ESPE) with four different shades (A2, A3, B1, and B2) were immersed in water with three different pH (pH 3, 6, and 9) for 14 day. The CIE $L^*a^*b^*$ color coordinates of the specimens were evaluated before and after immersion in the solutions. The color difference (${\Delta}E^*$) and the translucency parameter (TP) were calculated using the color coordinates. Results: ${\Delta}E^*$ ranged from 0.33 to 1.58, and the values were affected significantly by the pH. The specimens immersed in a pH 6 solution showed the highest ${\Delta}E^*$ values (0.87 - 1.58). The specimens with a B1 shade showed the lowest ${\Delta}E^*$ change compared to the other shades. TP ranged from 7.01 to 9.46 depending on the pH and resin shade. The TP difference between before and after immersion in the pH solutions was less than 1.0. Conclusions: The resulting change of color of the tested specimens did not appear to be clinically problematic because the color difference was < 1.6 in the acidic, neutral, and alkaline solutions regardless of the resin shade, i.e., the color change was imperceptible.

Mobility of silver nanoparticles (AgNPs) and oxidative degradation of endocrine disrupting chemicals by saturated column experiments (포화컬럼실험에서 산화공정을 적용한 내분비계 장애물질의 제거 및 은나노물질의 거동 연구)

  • We applied column experiments to investigate the environmental fate and transport of silver nanoparticles(AgNPs) in fully saturated conditions of porous media. These column experiments were performed to emphasize oxidation method with $H_2O_2$ concentration and acidic conditions. The mobility of AgNPs was decreased with the increasing ionic strength that the surface charge of AgNPs(zeta potential) was neutralized with the presence of positive ions of $Na^+$. Additionally, it was also affected due to that not only more increased aggregated size of AgNPs and surface charge of quartz sand. The decreased breakthrough curves(BTCs) of bisphenol-A(BPA) and $17{\alpha}$-ethynylestradiol(EE2) were removed approximately 35.3 and 40%. This is due to that endocrine disrupting chemicals(EDCs) were removed with the release of $OH{\cdot}$ radicals by the fenton-like mechanisms from acidic and fenton-like reagent presenting. This results considered that higher input AgNPs with acidic conditions is proved to realistic in-situ oxidation method. Overall, it should be emphasized that a set of column experiments employed with adjusting pH and $H_2O_2$ concentration in proved to be effective method having potential ability of in-situ degradation for removing organic contaminants such as BPA and EE2.

  • Ultrastructural changes in Z-line, M-line and myofilaments of bovine psoas muscle produced by leukocyte lysosomal enzymes in vitro at different pH values (pH 7.0 and 4.0), temperatures (37 and $4^{\circ}C$) and time intervals (12, 24 hours at $37^{\circ}C$ and 36, 168 hours at $4^{\circ}C$) were studied by transmission electron microscope. Muscle incubated with leukocyte lysosomal enzymes at pH 7.0 produced distinguishable degradation of Z-line, M-Line and H-zone at both temperatures but at acidic pH (pH 4.0), Z-line were very stable and myofilaments were severely disintegrated.

Improved Homologous Expression of the Acidic Lipase from Aspergillus niger

  • In this study, the acidic lipase from Aspergillus niger (ANL) was homologously expressed in A. niger. The expression of ANL was significantly improved by the expression of the native ANL with the introns, the addition of the Kozak sequence and the optimization of the signal sequences. When the cDNA sequence of ANL fused with the glaA signal was expressed under the gpdA promoter in A. niger, no lipase activity could be detected. We then tried to improve the expression by using the full-length ANL gene containing three introns, and the lipase activity in the supernatant reached 75.80 U/ml, probably as a result of a more stable mRNA structure. The expression was further improved to 100.60 U/ml by introducing a Kozak sequence around the start codon due to a higher translation efficiency. Finally, the effects of three signal sequences including the cbhI signal, the ANL signal and the glaA signal on the lipase expression were evaluated. The transformant with the cbhI signal showed the highest lipase activity (314.67 U/ml), which was 1.90-fold and 3.13-fold higher than those with the ANL signal and the glaA signal, respectively. The acidic lipase was characterized and its highest activity was detected at pH 3.0 and a temperature of 45℃. These results provided promising strategies for the production of the acidic lipase from A. niger.

  • To determine the response of soybean cultivars to nitrogen in relation to acidity, nodulation, content of allantoin nitrogen and major chemical compositions of soybean plants were investigated with two cultivars under two levels of soil pH and four levels of nitrogen fertilization in a field and nutri-culture experiments. Nodulation and contents of allantoin nitrogen, total nitrogen, potassium, calcium, magnesium and manganese of soybean plants decreased under acidic condition, but the degree of decrease was smaller in the variety Jangbaegkong compared to the variety Danyeobkong. On the other hand, nodulation, content of allantoin nitrogen and potassium decreased with increased nitrogen fertilization, particulary under pH 7 condition. Nodulation was positively correlated with the content of allantoin nitrogen of soybean plants. The content of total nitrogen of soybean plants increased with increased nitrogen fertilization, and this tendency was remarkable under acidic condition and in the variety Jangbaegkong.

Factors Affecting the Extraction of Protein from Antarctic Krill (남빙양산(南氷洋産) 크릴단백질(蛋白質)의 추출조건(抽出條件))

  • To recover proteins from antarctic krill(Euphausia superba) for the use of food material, some factors affecting the extraction of protein were investigated. The protein solubility profile showed a minimum solubility level(33.2-38.8%) within the range of pH4.0-4.5 and very high solubility levels as 56.8% at pH2.0 and 80.7% at pH11.0. The extraction yield increased as the solvent-to-krill ratios increase in which a ratio of 5:1(volume of solvent/weight of krill) was found to be preferable from the point of handling convenience and extraction yield. The extraction temperatures did not seem to be important variables on extraction of protein. The extraction of krill protein occurred fairly rapidly with little further extraction of protein after 30 minutes. The extraction of protein was slightly decreased at both acidic(pH2.0) and alkaline(pH11.0) conditions with the increasing concentration of sodium chloride. The extractibility of krill protein at strong alkaline condition(pH11.0) was higher than at strong acidic condition(pH2.0) under the same concentration range as 1-6% of sodium chloride. In phosphate treatments, the extraction of protein was slightly influenced by presence of sodium chloride as the concentration range of 3-4% in the aqueous solvent by which maximize the extraction yield as over 80%.

Enhancement of Interfacial Adhesion of Epoxy/Red Mud Nanocomposites Produced by Acidic Surface Treatment on Red Mud (Red Mud의 산처리에 의한 에폭시/Red Mud 나노복합재료의 계면 결합력 향상)

  • In this work, red mud (RM) was chemically modified by 0.1, 1, and 5 M H3PO4 solution to prepare epoxy/RM nanocomposites. The effect of chemical treatment on pH, acid-base values, specific surface area, and porosity of RM surface was analyzed. To estimate the mechanical interfacial properties of epoxy/RM nanocomposites, the critical stress intensity factor (K$_{IC}$) was measured. From the experimental results, it was clearly revealed that the porosity, specific surface area, and acid values of RM surface were developed as the increase of the treatment concentration due to the increase of acidic functional group, including hydroxyl group on RM surface. The mechanical interfacial properties of epoxy/treated-RM nanocomposites were higher than those of epoxy/RM as-received due to an improvement of interfacial bonding between basic matrix and RM surface.
