Anaerobic Acid Tolerance Response in Salmonella typhimurium (Salmonella typhimurium의 혐기적 산내성도 평가)

  • Salmonella typhimurium can encounter a wide variety of environments during its life cycle. In nature, S. typhimurium can experience and survive dramatic acid stresses that occur in diverse ecological niches ranging from pond water to phagolysosomes. These survival mechanism is aquired by the Acid Tolerance Response(ATR) in Salmonella. The ATR of S. typhimurium is a complex inducible phenomenon in which exposures to slight or moderate low pH will produce a stress response capable of protecting the organism against more severe acid challenges. ATR in Salmonella has two different systems that are called RpoS dependent and independent. We found that ATR in anaerobic was showed RpoS independent because rpoS$\Omega$AP had ATR as S. typhimurium UK1. Using the P22 MudJ(Km, lacZ) operon fusion technique and a lethal selection procedure combining low pH(pH4.5) and sodium acetate(10mM, pH4.5), we isolated LF487 aatA::MudJ which showed acid sensitive in anaerobic condition. aatA locus was determined at 12 min on Salmonella Genetic Map. The survival rate of aatA mutant was showed significantly diminished at pH4.3 than virulent wild type Salmonella in anaerobic condition(5% $CO_2$, 5% H$_2$, 90% $N_2$). Therefore isolated gene was confirmed important gene for anaerobic ATR system.

Dry Enteric Coating Process of Lactic Acid Bacteria by Hybridization System (Hybridization system을 이용한 유산균의 장용성 건식 피복)

  • Surface-modified powders were produced by hybridization system using core freeze-dried lactic acid bacteria (Lactobacillus acidophilus ATCC 43121) and enteric coating materials. Scanning electron microscopy showed that the surface of freeze-dried lactic acid bacteria changed to smooth round shape during surface reforming process, although no significant physical damages affecting the activity of the lactic acid bacteria were observed based on viability and salt-tolerance tests. Signigicant difference was not found in acid tolerance test probably due to the inherent acid tolerance of L. acidophilus ATCC 43121. Significantly improved heat tolerance was obtained by surface modification process. Among the tested coating materials, Sureteric showed a higher surface- reforming ability than Eudragit S100 and L100-55. Core : coating ratio agent of 9 : 1 (w/w) with rotor speed of 15,000 rpm for 3 min were determined to be optimum conditions for the process.

Physiological Characteristics Related to Cold Injury in Rice (수도 냉해에 관련된 생리적 특성 고찰)

  • Cold stress influence plant growth through a wide range of growth characters. Adverse effects of low temperature to plant growth come from results of colligative and complex physiological responses to cold stress. To evaluate more exactly cold tolerance of crop plant, it is needed to observe physiological changes induced by cold stress and to analyze relationships between intraspecific variations in physiological factors related to cold tolerance and the extent of cold tolerance in the field. Therefore, the composition and unsaturation ratio of fatty acids in phospholipid, a constituent of membrane, the transition-temperature in respiratory activity of mitochodria, the chlorophyll fluorescence as a factor related to photosynthesis were investigated in rice plant and data on these factors were compared with the degree of cold tolerance obtained in the field experiment. Also, effects of hardening and Mn++ treatment were evaluated as a method to reduce chilling injuries. The unsaturation ratio of fatty acids, whether rice plants were grown in a natural condition or under the chilling stress, was higher in the cold- tolerant varieties and was significantly correlated with the degree of cold tolerance (1-9) observed in the field experiment. And it was also increased by chilling treatment or hardening treatment, due to a reduction in palmitic acid content and an increase in linolenic acid content. The transition-temperature of respiratory activity of mitochodria isolated from etiolated rice seedlings ($25^{\circ}C$, two week-grown in the dark), was correlated with the degree of cold tolerance in the field, cold -tolerant varieties showing a lower transition-temperature. It was not influenced by growth stages. The intensity of chlorophyll fluorescence was highly correlated with the degree of cold tolerance, cold-tolerant varieties having a higher fluorescence intensity. By foliar application of Mn, the transition-temperature of respiratory activity was lowered as much as 0-2$^{\circ}C$ in all tested varieties. Soil application of Mn induced more significant effect in cold-susceptible varieties with a possibility of reducing chilling injuries. On the whole, there were high correlationships among the degree of cold tolerance, the unsaturation ratio of fatty acids in phospholipid, the transition- temperature of respiratory activity and chlorophyll fluorescence except for a few varieties. The transition- temperature of respiratory activity appeared to be negatively correlated with the unsaturation ratio of fatty acids. and the chlorophyll fluorescence to be positively correlated with the unsaturation ratio. This implies that these physical and physiological factors were very closely related to cold tolerance and can be used as an effective index of the evaluation of cold tolerance of crop plant. But other factors as well as three factors discussed above are needed to be considered colligatively and altogether with a systematic analysis for the more exact evaluation of cold tolerance. in rice cultivars. in rice cultivars.

Bile and Acid Tolerance of Lactic Acid Bacteria Isolated from Dadih and Their Antimutagenicity against Mutagenic Heated Tauco

  • Antimutagenicity of milk cultured with lactic acid bacteria isolated from dadih on the mutagenicity of heated salty and sweet tauco was examined using streptomycin dependent (SD) 510 strain of Salmonella typhimurium TA 98 as a tester culture. Cultured milk samples exhibited widely antimutagenic activity against mutagenic heated salty and sweet tauco. Lc. lactis subsp. lactis R-22, Lc. lactis subsp. casei R-35, Lc. lactis subsp. casei R-52 and E. faecalis subsp. liquefaciens R-55 exhibited no inhibitory effect on the mutagenic heated salty tauco. Mutagenicity of heated sweet tauco was inhibited by cultured milks stronger than that of heated salty tauco. Milk cultured with Lc. lactis subsp. cremoris R-48, Leuc. mesentroides R-51 and Lc. lactis subsp. casei R-68 showed high inhibition against the mutagenicity of both heated salty and sweet taucos. Antimutagenic activity of the cultured milks against mutagenic heated tauco was attributed to the bacterial cells. Among the three strains which showed high antimutagenicity, only Leuc. mesentroides R-51 was tolerant to both acid and bile; so this strain can be used as probiotic in preventing the occurrence of mutagenesis caused by mutagenic heated food like tauco.

Probiotic Characteristics of Lactobacillus acidophilus KY1909 Isolated from Korean Breast-Fed Infant (한국인 유아 분변에서 분리한 Lactobacillus acidophilus KY1909의 프로바이오틱 특성)

  • The purpose of this study was to isolate lactic acid bacteria that produced L(+) lactic acid from infant feces. Thirteen colonies were isolated with a MRS-plate containing 0.5% $CaCO_3$ to determine their ability to produce lactic acid. Based on their lactic acid production, 10 strains of Lactobacillus were identified to assess the ratio of lactate isomer using HPLC. A strain producing L-lactic acid was identified as Lactobacillus acidophilus, using API carbohydrate fermentation patterns and physiological tests, and named KY1909. The strain exhibited good acid tolerance in an artificial gastric juice as well as high bile resistance in MRS containing 0.5% bile acids. L. acidophilus KY1909 produced D(-) and L(+) lactic acid at a ratio of 6 : 94; whereas commercial strains of Lactobacillus acidophilus produced D(-) and L(+) lactic acid at a ratio of 1 : 1. These results demonstrate the L. acidophilus KY1909 can be utilized in fermented milk products and dietary supplements as a probiotic culture.

Bile Salts Degradation and Cholesterol Assimilation Ability of Pediococcus pentosaceus MLK67 Isolated from Mustard Leaf Kimchi (갓김치에서 분리된 Pediococcus pentosaceus MLK67의 담즙산 분해능 및 콜레스테롤 동화능)

  • The objective of this study was to evaluate the acid and bile tolerance, bile salt hydrolase (BSH) activity, and cholesterol assimilation ability of lactic acid bacteria isolated from mustard leaf kimchi. MLK11, MLK22, MLK27, MLK41, and MLK67 were relatively acid- and bile-tolerant strains, with more than $10^5$ CFU/ml after incubation in simulated gastric juice and intestinal fluid, while MLK53 was the most sensitive strain to acid and bile. Strains MLK22 and MLK67 deconjugated the highest level of sodium glycocholate with more than 3.5 mM of cholic acid released, while deconjugation was lowest by strains MLK13 and MLK41 which released only 1.35 mM and 1.16 mM, respectively. Specially, strains MLK22 and MLK67 showed higher deconjugation of sodium glycocholate compared to sodium taurocholate and conjugated bile mixture. Although strains MLK22 and MLK67 exhibited maximal BSH activity at the stationary phase, MLK22 had somewhat higher total BSH activity compared to MLK67 towards both sodium glycocholate and sodium taurocholate. Meanwhile, cholesterol removal varied among tested strains (p<0.05) and ranged from 5.22 to 39.16 ${\mu}g$/ml. Especially, MLK67 strain assimilated the highest level of cholesterol in media supplemented with 0.3% oxgall, cholic acid, and taurocholic acid (p<0.05). According to physiological and biological characteristics, pattern of carbohydrate fermentation, and 16S rDNA sequence, strain MLK67 that may be considered as probiotic strain due to acid and bile tolerance and cholesterol-lowering effects was identified as Pediococcus pentosaceus MLK67.

Enhancement of Ethanol Tolerance of Lactose Assimilating Yeast Strain by Protoplast Fusion

  • In order to construct a yeast strain having high ethanol tolerance together with good lactose fermentation ability, the protoplast fusion using Saccharomyces cerevisiae STV 89 and Kluyveromyces fragilis CBS 397 was carried out. Auxotrophic mutants of K. fragilis were obtained as a selection marker by treatment of ethylmethane sulfonate. The best mutant for protoplast fusion was selected based on the capabilities of ${\beta}-galactosidase$ production and lactose fermentation. The protoplast fusion using polyethylene glycol and calcium chloride solution led to the fusion frequence of $3{\times}10^{-6}$ and a number of fusants were obtained. Among these fusants, a fusant F-3-19 showed the best results in terms of ethanol tolerance, ${\beta}-galactosidase$ activity and lactose fermentation. The performance of lactose fermentation and ethanol tolerance by this fusant were better than those of K. fragilis. Study on the ethanol tolerance having relation to fatty acid composition and intracellular ethanol concentration revealed that the fusant F-3-19 had a higher unsaturated fatty acids content and accumulated less amount of intracellular ethanol compared with a parent of K. fragilis.

Tolerance of the Mutants of Pyricularia oryzae against Mercuric Chloride (자외선조사에 의해 유도된 도열병균의 승홍에 대한 내성에 관하여)

  • Tolerance against mercuric chloride in mutants of Pyricularia oryzae Cavara induced by ultraviolet(UV) irradiation has been investigated. The tolerant isolates obtained using ultraviolet(UV) irradiation were maintained a high level of tolerance even after 15 times transfer to the chemical free media. Each isolate of mutants UM-1, UM-2, and UM-3 on the successive monoconidial culture has genetically homogeneous for tolerance. The tolerant isolates sporulated less and showed a higher percentage of germination of conidia on media without the chemical than the parental isolate. The xotins released from the parental and the tolerant isolates have been identified as piricularin and $\alpha$-picolinic acid by paper-chromatography. The tolerant isolates produced more piricularin and its virulence on seedlings of rice varieties were higher than the parental isolates. Both piricularin production and virulance on rice were highest in the UM-2 isolate.

Selection and Characterization of Tomato Plants for Osmotic Stress Tolerance Derived from a Gamma Ray Irradiation (감마선 돌연변이원에 의한 Osmotic 스트레스 저항성 토마토 계통 선발 및 특성)

  • The present study has been performed to select the osmotic tolerant lines using polyethylene glycol (PEG 6000) through an in vitro and in vivo mutagensis with a gamma-ray. During the screening, we selected three mutant lines that seemed to confer elevated osmotic tolerance in high concentrations of PEG 6000. Fruits of these mutants (Os-HK101, Os-HK102 and Os-HK103) were increased to sugar concentration, L-glutamine acid, vitamin C content and lycopine content than those of the wild type. Also the chlorophyll contents were few decreased more in the three mutant lines than the WT plants. Our results suggest that the Os-HK101 is characterized as osmotic stress tolerance considering the sugar concentration and lycopine content. It is expected that the result of this study can be used for breeding more competitive species with respect to contents in sugar or functional chemicals from the selected osmotic resistant lines.
