Antitumor Effect of Mugwort (Artemisia princeps Pampan.) in vivo

  • This study was devised to observe the antitumor activity of mugwort (Artemisia princeps Pampan.) against ICR mice inoculated with sarcoma-180 cells. The antitumor compounds were partially purified from petroleum ether extract of mugwort by silicic acid column chromatography. The active fraction used in in vivo test was obtained under the elution with acetone in silicic acid column chromatography. When the acetone fraction was intraperitoneally injected to the mice which had been subcutaneously inoculated on the left groin with sarcoma-180, the growth rate of tumor (sarcoma-180 mass) was inhibited by 30%. In case the acetone fraction was injected to the mice which had been inoculated intraperitoneally with sarcoma-180, the average life span was prolonged by 20%. After the injection of the active fraction, the spleen index and ${\gamma}-globulin$ ratio (%) were increased significantly (p<0.05). The administration of acetone fraction did not cause any abnormality in the body and the homeostasis of mice. Those observations suggest that the acetone fraction of mugwort extract has an antitumor effect in vivo.

Antioxidative Activity of Extrcts from Fruit of Curdrania tricuspidata (꾸지뽕나무 열매 추출물의 항산화 활성)

  • The comparative activities of acetone, ethanol, and aqueous fractions extracted from fruit powder of Cudrania tricuspidata by different temperature were tested by in vitro experimental models; peroxidation of linoleic acid and autooxidation of rat hepatic and renal microsomes by using thiobarbituric acid (TBA) for assay of free malondialdehyde production, and scavenging activities of free radicals by DPPH (α, α'-diphenyl-β-picrylhydrazyl). In DPPH method, acetone fraction extracted at 30℃ showed the highest free radical scavenging activities and acetone fractions extracted at 30℃ and 60℃ and ethanol fraction extracted at 30℃ showed stronger than BHT (butylated hydroxitoluene) although used ten-fold lower concentrations. In thiocyanate method used linoleic acid an inhibitory effects of all fractions showed higher than control treatment. TBA method used linoleic acid showed the highest antioxidative activity in acetone fraction extracted at 30℃ and 60℃. an inhibition activity against lipid peroxidation in hepatic microsomes of rats showed the highest at acetone faction extracted at acetone fraction among extracted fractions was shown to be the most potent antioxidative properties and this action was more potent in fractions extracted at 30℃ than those extracted at 60℃.

  • Adsorption experiments of binary mixed gases composed of acetone/methylethylketone (MEK), MEK/benzene, MEK/toluene, and benzene/toluene were carried out on activated carbon fixed-bed. The variations of equilibrium adsorption capacity according to type and fraction of binary gas were investigated. In case of binary gases composed of acetone/MEK and benzene/toluene, equilibrium adsorption capacities of MEK and toluene were increased according to the increase of fraction of MEK and toluene, but equilibrium adsorption capacities of acetone and benzene were decreased. In case of binary gases composed of MEK/benzene and MEK/toluene, equilibrium adsorption capacities of benzene and toluene were increased according to the increase of fraction of benzene and toluene, but equilibrium adsorption capacities of MEK was decreased.

Antimicrobial Effect of Caridina denticulata denticulata (새뱅이 (Caridina denticulata denticulata) 추출물의 항균효과)

  • After extracting Caridina denticulata denticulata, the raw material of salt-fermented Toha, with the use of water, ether, acetone and methanol in order, we had the antimicrobial activity test for nine strains of bacteria which are harmful to the human body and food. It is as follows; solvent extract yield of the powder of vacuum freeze dried Caridina denticulata denticulata was 52% of water fraction, 7.4% of ether fraction, 1.0% of acetone fraction and 0.8% of methanol fraction. The result of solvent extract antimicrobial search of Caridina denticulata denticulata said that with the sample of 20$\mu\textrm{g}$/disk, ether fraction resulted in clear one of 8~12mm, acetone fraction 8~12mm, methanol fraction 7~9mm and water fraction didn't have antimicrobial effect. The minimal inhibitory concentration of solvent extract of Caridina denticulata denticulata per area was that ether fraction was 250~500$\mu\textrm{g}$/$m\ell$, acetone fraction 125~500$\mu\textrm{g}$/$m\ell$ and methanol fraction 250~500$\mu\textrm{g}$/$m\ell$. The minimal bactericidal concentration of solvent extract per area of Caridina denticulata denticulata showed that ether fraction was 2, 500~10, 000$\mu\textrm{g}$/$m\ell$, acetone fraction 1, 250~10, 000$\mu\textrm{g}$/$m\ell$ and methanol fraction 5, 000~10, 000$\mu\textrm{g}$/$m\ell$.

Comparative Study on the Cytotoxic Activities of Red Ginseng of Korea and China (한국 및 중국 홍삼의 암세포 증식억제 효과 비교연구(III))

  • A study was performed to compare the anticancer effects of Korean and Chinese red ginseng roots. The whole crude extracts or chloroform, methanol and acetone fractions of the crude extracts were added in the culture medium of three cancer cell lines, a mouse leukemia cell line ($P_{388}$), a human colon carcinoma cell line (HT-29) and a human rectal carcinoma cell line (HRT-18), to screen the growth inhibition effects. The results are summarized as follows : 1. Crude extracts of both Korean and Chinese red ginseng roots inhibited the proliferation of all the three cancer cell lines tested in a dose dependent manner. However, the growth inhibition effects of Korean red ginseng extracts were significantly greater than that of Chinese red ginseng. 2. An acetone fraction showed the greatest antiproliferative effects among the 11'hole crude extracts, chloroform, methanol and acetone fractions of the crude extracts. 3. These results suggest that the active antiproliferative components of the crude extracts are present mostly in the acetone fraction.

Studies on Antioxidative Substances of Ganoderma lucidum (영지의 항산화성 물질에 관한 연구)

  • To study antioxidative activities of Ganoderma lucidum, its extracts were fractionated by various organic solvents with different polarity the extracts were purified by thin chromatography, silicagel column chromatography and preparative liquid chromatography. In antioxidative activity tests using thiocyanate method, TBA method and weighing method, fraction 5 from the hexane extract and fraction II from the methanol extract showed antioxidative activity. When the antioxidative activities were expressed as TBA value using a homogeneous liver extracte of rats, the relative antioxidative activities of fraction 5 and fraction II were increased by 13.0% and 54.6%, respectively.

  • To investigate the antitumor activity of mugwort (Artemisia princeps Pampan), petroleum ether extract of mugwork was partially purified by a silica gel chromatography. Among several fractions, the fraction which was obtained under the elution with acetone, showed potent cytotoxicity against mouse leukemia cell line(Ll210), human colon cancer cell line (HCT-48) and human hepatoma cell line (Hep G2) , but was less effective with normal cell line(mouse embryo cell). Acetone fraction appeared to be glycolipid by Benedict test and the major fatty acids of the lipid were C16 ; 0 , C 18: 3by GC/MS analysis.

Cytotoxic Effects and Components of Lipid Fractions from Soybean Products on Cancer Cell Lines (대두식품 지질추출물의 세포독성 및 지질성분분석)

  • The cytotoxic effects of lipid extracts from soybean products were studied using K562 human leukemia cell, Yac1 mouse leukemia cell and S 180 mouse sarcoma cell. Total lipids from soybean powder, soybean curd residue and doenjang were extracted with chloroform/methanol (2 : 1) and water saturated butanol, consecutively, and fractionated into acetone supernatants (AS fraction) and acetone precipitates (AP fraction) by adding excess acetone. AS fraction of doenjang lipids showed the strongest cytotoxic effects on K562, Yac1 and S180 cancer cells, whereas each lipid fraction of soybean curd residue also showed relatively weak cytotoxic effects on cancer cells but soybean powder did not. AS and AP fractions of doenjang contained more free fatty acids than those of soybean curd residue and soybean. And when lipid fractions were digested with 0.4 N KOH/methanol, doenjang lipid fractions showed to contain some alkali-stable substances which showed positive reaction with ninhydrin solution on silica TLC separation.

Studies of Antipyretic Component of the Earthworm (지렁이(지룡)의 해열성분에 관한 연구)

  • In order to confirm the exact antipyretic component in the earthworm, etherial extract of American earthworm(Red Worm) was fractionated into five fractions by using silica gel column chromatography and thin layer chromatography. The fraction including free fatty acids was found to possess artipyretic response and standard arachidonic acid showed marked antipyretic response on typhoid vaccinated rabbits. Arachidonic acid was identified from the free fatty acid fraction of the earthworm by using gas liquid chromatography. Thus it was considered that the antipyretic activity of the free fatty acid may be due to the presence of arachidonic acid. Lipid-free earthworm powder was extracted with phosphate buffer (pH, 8.0, 0.1M) and all the proteins was salted out by ammonium sulfate. The crude precipitate was dialyzed and the impure proteins were eliminated at pH 5.4 and 4.6. The remaining protein solution was fractionated with various concentrations of acetone. The acetone fractions were identified by using S.D.S. polyacrylamide gel electrophoresis and disc gel electrophoresis. The precipitate at 85% acetone concentration and the remaining proteins in the supernatant did not exhibit the antipyretic activity.

Antioxidant Effect of the Fraction Isolated from Pyrus ussuriensis Leaves (산돌배나무(Pyrus ussuriensis) 잎 분획물의 항산화 효과)

  • This study was conducted to confirm the application as ingredients of cosmetics through an examination of the function for anti-oxidant activity of the fraction isolated from Pyrus ussuriensis leaves. The dried leaf of Pyrus ussuriensis were extracted with acetone-$H_2O$ (6:4, v/v), concentrated and fractionated with the upper layer of acetone on a separatory funnel. Each fraction was freeze dried, then a portion of acetone soluble powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol as eluents and also used the MIC-gel using a series of aqueous methanol as developing solvent. The isolated compounds were identified by silica-gel TLC. The concentration of total phenolic compound of Pyrus ussuriensis acetate soluble fraction was high, 914 mg/g. The results obtained from the analyses of the anti-oxidanat effects of Pyrus ussuriensis acetate fraction can be summarized as follows: In the result of DPPH scavenging radical activity, Pyrus ussuriensis acetate soluble fraction showed more than 80% at 100 ppm. SOD-like activity of one of Pyrus ussuriensis acetate soluble fractions was 77% at 1000 ppm. Xanthine oxidase inhibition of Pyrus ussuriensis acetate soluble fraction was 38% at 100 ppm. From these results, we confirmed that acetate fraction of Pyrus ussuriensis has a great potential as a natural ingredients with a natural antioxidant and antimicrobial source.
