The anaerobic bacterium P. productus was known to produce acetate from CO, $CO_2$ and $H_2$. In this research the production of acetate at high concentration from carbon monoxide was studied. In this research the effect of $CO_2$, $H_2$, and CODH co-factor on cell growth and acetate production was studied. Also the toxicity of acetate on cell growth was examined. Acetate formation in lab scale fermenter was tested and specific acetate production rate of $0.43\;g/L \;{\cdot}\;hr\;{\cdot}\;O.D.$ was obtained and the concentration of the acetate obtained in a fermenter higher than 40g/L.
Objectives: The aim of the study was to evaluate the anti-asthmatic effect of alismatis rhizoma and alisol acetate B combination therapy in a murine asthma model. Methods: C57BL/6 mice were sensitized to and challenged with a mixture of ragweed, dust mite, and aspergillus to induce an asthma animal model. Alismatis rhizoma extract and alisol acetate B combination therapy was co-administered only in the experimental group. To evaluate the anti-asthmatic effect of the combination therapy, inflammatory cell counts in bronchoalveolar lavage (BAL) fluid were determined, and tissue was examined histologically with hematoxylin and eosin (H & E) and periodic acid-Schiff (PAS) stains, by enzyme-linked immunosorbent assay (ELISA) of IgE, IL-4, and IL-5, and with reverse transcription polymerase chain reaction (RT-PCR) of IL-5, IL-33, MUC5AC. Results: Alismatis rhizoma and alisol acetate B combination therapy reduced the number of inflammatory cells, alleviated histologic features, and down-regulated all the investigated asthma mediators, IgE, IL-4, IL-5, IL-33, and MUC5AC. Conclusions: According to the above results, alismatis rhizoma and alisol acetate B combination therapy may have therapeutic potential for asthma.
Objectives: This study was undertaken to investigate the action mechanism of Holotrichia (HT) at parameter related to dementia. Methods: HT was tested for the effects on acetylchonine esterase and monoamine oxidase activities, lipid peroxidation, antioxidation in brain of lead acetate-treated rats. Lead acetate were treated firstly into samples for 10 days, and then lead acetate and HT were set with them for 20 days. Results: The level of lipid peroxide, acetylcholine esterase and monoamine oxidase activities, enzyme activities and ratio of type conversion of xanthine oxidase increased in lead acetate-treated rats were decreased as highly as normal group by HT. Superoxide dismutase, catalase and glutathione peroxidase activities, the level of acetylcholine decreased in lead acetate-treated rats were increased as lowly as normal group by HT. Conclusions : These results suggest that HT might have an effect on treatment of dementia according to decreasing the activities of acetylcholine esterase, monoamine oxidase and level of lipid peroxide in brain.
Bleeding after cardiopulmonary bypass remains a cause for concern, requiring reexploration of the chest in approximately 3 percent of patients who have had operations on the heart. We examined the possibility that this problem might be alleviated by desmopressin acetate [DDAVP], synthetic vasopressin analogue that lacks vasoconstrictor activity. In a prospective, randomized trial, we studied the effect of intraoperative desmopressin acetate in 20 patients [the treated group 10 patients and the control group 10 patients] undergoing various cardiac operations requiring cardiopulmonary bypass. The result showed that the early postoperative [during first 24hrs] and mean postoperative blood loss [first 3 days] of the treated group were significantly reduced than the control group[447\ulcorner199ml in the treated group versus 746\ulcorner199ml in the treated group versus 746\ulcorner295 ml in the control group, p=0.014; mean\ulcornerstandard deviation and 675\ulcorner276 ml in the treated group versus 1006\ulcorner303 ml in control group, p=0.019]. The mean red-cell transfusion in first 3days were reduced in the treated group than the control group [3.3\ulcorner1.7 units vs 4.9\ulcorner1.7units, P=0.051]. There were no untoward side effects of desmopressin acetate. We conclude that the administration of desmopressin acetate can be recommended to reduce blood loss and blood conservation in patients undergoing cardiac operations.
Leptin has a major role in the regulation of food intake and energy homeostasis. In addition, leptin participates in many physiological functions including regulation of lipid metabolism. Bovine recombinant leptin protein was produced in E. coli cells in order to understand function of leptin in the regulation of lipid metabolism. The leptin expression vector was constructed in pGEX-4T-3 vector and transformed into E. coli BL21 cells. Expression of the GST-leptin fusion protein was induced with IPTG. The fusion protein was purified using glutathione sepharose 4B batch method, and the recombinant leptin was eluted after thrombin protease digestion. The effect of leptin on glucose transport was examined in the differentiated adipocytes of 3T3-L1 cells. Leptin had no effect on basal and insulin-stimulated glucose transport in 3T3-L1 cells (p>0.05). Effect of recombinant leptin on glucose and acetate transport was examined in adipose tissues of Korean cattle (Hanwoo). Insulin stimulated glucose transport in both intramuscular and subcutaneous adipose tissues (p<0.05), but leptin did not affect glucose transport in both adipose tissues (p>0.05). Insulin stimulated acetate transport in bovine adipose tissues (p<0.05), but leptin did not affect acetate transport (p>0.05). Northern and RT-PCR analyses showed that mRNA levels of uncoupling protein-2 were increased by leptin treatment in 3T3-L1 cells without statistical difference (p>0.05). In conclusion, bovine recombinant leptin did not affect glucose and acetate transport in both 3T3-L1 adipocytes and bovine adipose tissues, while it stimulates UCP-2 mRNA expression in 3T3-L1 cells.
Background: To date, the anti-wrinkle efficacy of phellodendri cortex has not been defined. In this study, we investigated the nitric oxide (NO) production and elastase inhibitory activities of 80% methanol extract of Phellodendri cortex and its ethyl acetate fraction. Methods and Results: We prepared 80% methanol extract, and its fractions from phellodendri cortex. The treatment of RAW 264.7 cell with $25{\mu}g/m{\ell}$ 80% methanol extract and ethyl acetate fraction resulted in no toxicity. We conducted assays of nitric oxide (NO) production and elastase inhibition. In the NO production assay, the ethyl acetate fraction showed an inhibitory effect approximately 17 times stronger than the 80% methanol extract. In elastase inhibitory assay, the ethyl acetate fraction also showed a stronger effect than the 80% methanol extract. In order to standardize the extract and fraction, we used TLC to separate the extract and observed the plate under UV light. We confirmed that the known pharmacological ingredients berberine, and palmatine in the 80% methanol extract and the ethyl acetate fraction. Conclusions: These results indicated that phellodendri cortex extract and its ethyl acetate fraction produced strong inhibitory effect on elastase and NO production.
비파 부위별 추출물의 아질산염 소거능 및 항돌연변이 효과를 조사한 결과는 다음과 같다. 추출용매 중 활성이 높게 나타난 메탄올 추출물을 비극성∼극성 용매별로 계통분획한 에칠아세테이트 분획물에서 잎은 80%, 씨를 제거한 과실 81%, 과피 73% 및 종자 63%로 아질산염 소거 효과가 가장 높게 나타났다. 메탄올 추출물의 분획하여 Salmonella typhimurium TA 100에 대한 항돌연변이 효과를 측정한 결과, 잎과 씨를 제거한 과실의 에칠아세테이트 분획물에서 각각 53.1%, 48.3%로 높게 나타났다. 에틸아세테이트 분획물을 농도별로 첨가하여 항돌연변이 효과를 확인한 결과, 잎과 씨를 제거한 과실의 경우 에틸아세테이트 분획물 2.5 mg과 5.0 mg 첨가시 각각 65.5%, 71.2%와 63.4%, 69.2%로 분획물의 농도가 높을수록 항돌연변이 효과가 높게 나타났다.
Propolis를 몇 가지 용매로 추출하여 분획물을 얻고 이를 palm oil과 lard에 단독으로 혹은 synergist와 함께 첨가하여 항산화 효과를 Rancimat을 이용하여 측정하였다. 용매별 추출,수율은 68.1%(75% ethanol), 75.5%(99% ethanol), 67.4%(methanol), 86.7%(chloroform), 72.6%(ethyl acetate), 65.6%(butanol)였다. Methanol 추출물이 항산화 효과가 가장 우수 하였고 palm oil 보다는 lard에서 더욱 효과적이었다. Ethanol(75%) 추출물을 chloroform, ethyl acetate, butanol 및 water 순으로 분획 하였을 때 ethyl acetate 분획물은 조추출물보다 항산화 효과가 상승하였다. 또 ethyl acetate fraction 600 ppm에 synergist로 ascorbic acid(200ppm)을 palm oil에 첨가 했을 때와 ${\dalta}-toocopherol(200ppm)$을 lard에 첨가했을 때 높은 항산화 상승 효과를 보였다.
본 연구에서는 식중독 유발세균에 대한 항균활성이 우수한 천연 항균성 물질을 검색하기 위해 예로부터 민간과 한방에서 널리 이용되어 온 청피를 각종 유기용매로 추출하여 식중독 유발세균에 대한 항균활성을 조사해 보았다. 청피를 petroleum ether, chloroform, ethly acetate, methanol를 이용하여 실온에서 각각 용매별로 계통 분획하고, 열수추출물을 얻은 후, 9종의 식중독 유발세균 (Staphylococcus-aureus, Bacillus cereus, Salmonella enteritidis, Shigella flexneri, Ercherichia coli. Salmonella typhimurium, Shigella dysenteriae, Pseudomonas aruginoia, Shigella sonnei) 에 대하여 항균효과를 조사하였다. 청피 추출물의 농도별 항균 활성 검색에서는 청피의 ethyl acetate 추출물이 가장 큰 항균 효과를 보였으며 Staphylococcus auren와 Pseudomonas aeruginosa가 가장 민감하게 반응하는 균주였다. 청피의 ethyl acetate 추출물과 백화사설초의 methanol 추출물을 혼합하여 항균력을 측정해 본 결과 두 추출물을 섞어 첨가했을 경우가 단독으로 사용했을 시 보다 상승효과를 나타내었다. 또한 청피의 ethyl acetate 추출물이 식중독 유발세균의 성장에 미치는 효과를 검정하기 위해 Bacillus cereus 및 Shigella sonnei의 배양액에 청피의 ethyl acetate 추출물온 각각 5,000 ppm 농도로 첨가했을 시, Bacillus cereus의 생육이 24시간 이상까지 억제됨을 관찰할 수 있었고, Shigella sonnei의 생육도 12시간까지 지연시킬 수 있었다.
