Inhibitory Effects of Flavonoids Isolated from the Leaves of Stewartia koreana on Nitric-oxide Production in LPS-stimulated RAW 264.7 Cells (노각나무 잎에서 분리된 플라보노이드에 의한 대식세포에서 산화질소 생성 억제효과)

  • Five phenolic compounds were isolated from the ethyl acetate fraction of leaves from Stewartia koreana, and their nitric-oxide (NO) inhibitory activities were measured to identify the major active constituents responsible for the efficacy of the extract against inflammatory reactions. These five compounds were quercetin (1), quercitrin (2), hyperin (3), quercetin-3-O-(6"-O-galloyl)-${\beta}$-D-galactopyranoside (4), and kaempferol 3-O-[2",6"-di-O-(trans-p-coumaroyl)]-${\beta}$-D-glucopyranoside (5). Among the separated compounds in the EtOAc fraction, compounds 4 and 5 were isolated for the first time, and no study has yet reported their anti-inflammatory effects. The compounds were identified by spectroscopic analysis, and the isolated compounds showed significant NO inhibitory effects in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Compound 5 showed the most potent inhibitory effect (63.35% inhibition) against LPS-induced NO production compared to that of compound 1 (17.17%), compound 2 (5.0%), compound 3 (3.92%), and compound 4 (6.32%) at $10{\mu}g/ml$ concentration. NO production was inhibited by suppressing the protein expression of inducible nitric-oxide synthase in LPS-stimulated RAW 264.7 macrophages. These results indicate that kaempferol 3-O-[2",6"-di-O-(trans-p-coumaroyl)]-${\beta}$-D-glucopyranoside might be the major active compound responsible for the anti-inflammatory effects of S. koreana.

Anti Inflammatory Activity of Viburnum dilatatum Thunb. Extract as Cosmetic Ingredient (화장품 소재로서 가막살나무 추출물의 항염증 효능)

  • The crude ethanol extracts and their solvent-partitioned fractions derived from the leaf and twig of Viburnum dilatatum Thunb. were investigated for their 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging efficacy. The results showed that the butanol-soluble fraction ($SC_{50}\;=\;110.30\;{\mu}g/mL$) exhibited higher anti-oxidant activity than the crude ethanol extract ($SC_{50}\;=\;117.03\;{\mu}g/mL$) in the DPPH assay model. Then, the effects of the same extract samples on the production of nitric oxide were examined in LPS-stimulated RAW264.7 cells. Although the hexane and methylene chloride-soluble fraction showed a weak anti-oxidant activity, they exhibited potent inhibitory activity of NO production above 50 % at a concentration of $10\;{\mu}g/mL$. The hexane-soluble fraction also showed the inhibitory effect on mRNA expression of pro-inflammatory mediators such an TNF-$\alpha$, IL-$1{\beta}$, IL-6, iNOS and COX-2 in LPS-stimulated RAW264.7. These results suggest that the solvent extracts of Viburnum dilatatum Thunb. could be used as an anti-irritation ingredient.

SOME PHYSIOLOGICAL STUDIES ON THE UTILIZATION OF ORGANIC SUBSTRATES BY EUGLENA GRACILIS VAR. BACILLA 10616 IN LIGHT AND IN DARKNESS ("유-그레나"의 명암배양에 따르는 유기질의 이용과 호흡 및 생장에 대하여)

  • 1) The comparative studies of the quantitative measurement of growth characteristics and utilization of substrates by Euglena gracilis var. bacilla 10616 in the light and in darkness have been carried out. Eodogenous respiration, effect of respiratory inhibitors and responses to the added substrates for the exogenous respiration are also investigated. 2) All cultures are grown in the open air under the continuous illumination of fluorescent light of 3500 lux at room termperature, the growth rate of the culture in the basal medium added 0.5% lactate is found to be the highest. The growth rate decreases successively for the cultures of 0.5% sucinate, 0.5% Na-acetate, 0.5% malate, and control. There is no growth in the basal meidum added 0.5% butyrate and 0.5% hydroquinone. The similar results are obtained for the mentioned cultures in the darkness. However, the growth rate in basal medium added 0.5% glucose and 0.5% sucrose does seem to increase in the darkness unlike the illumination. 3) The endogenous rate of respiration for the organism cultured photosynthetically is about 12.94ul 02/mg/hr, in basal medium and the respiratory quotient is about 0.84. The rate is decreased by starvations to 6.5ul 02/mg/hr, about to a half, but the respiratory quotient does net change. 4) The oxygen consomption during initial 2 hours in suspending solution ranging from pH 4.5 to pH 9.3 is highest at pH 4.5 in which the algae had grown, at pH 5.5 and at pH 6.9. 5) Endogenous respiration of the cells is strongly inhibited by 0.1M of potassium cyanide, malomic acid, sodium fluoride and iodo-acetic acid. It is also strongly inhibited by 0.01M of potassium cyanide. 6) The respiratory response to added substrates for the exogenous respiration in the organism is coincided with the rate in the basal medium added the substrate in light and in darkness, whether the cells are fed or starved. 7) According to the results of this study, there seems to be the flexibility of the interconversion between photosynthesis and chemosynthesis, heterotropic mode of metabolism, in Euglena gracilis var. bacillaris, and that this organism utilizes the lactate most. It also may be suggested that the enayme systems linked in the each steps of Embden-Myerhof-Parnas path way and TCA cycle seem to exist in this organism.

Effect of Tumor Necrosis Factor-${\alpha}$(TNF) on the Expression of Oncogenes in ME-180 Human Cervical Carcinoma Cells (종양괴사인자(TNF)가 ME-180 사람 경부 암종세포에서 종양 발생 유전자의 발현에 미치는 영향)

  • Tumor necrosis factor-${alpha}$ (TNF) induced a cytotoxic response in ME-180 cervical carcinoma cells in vitro. This cytotoxic response was accompanied by a temporal series of mitogenic stimuli : increased c-fos, c-jun and jun-B expression. Depletion of protein kinase C (PKC) by exposure of ME-180 cells to 100ng/ml phorbol myristate acetate (PMA) for 24hours almost completely abolished TNF-mediated increase in these signals, indicating that a PKC-dependent pathway is involved in TNF-mediated increases in the expression of c-fos, c-jun and jun-B. Characteristics of TNF receptors after exposure to 100ng/ml PMA or 24hours were not altered, suggesting that diminished induction of these oncogenes by TNF after PMA treatment is not due to any changes at the receptor level. To examine whether a PKC-dependent pathway is involved in TNF-mediated cytotoxicity in ME-180 cells, cytotoxicity was measured after depletion of PKC. No apparent changes in cytototoxicity after PKC depletion suggest that a PKC-dependent pathway is not involved in TNF-mediated cytotoxicity. Furthermore, results from cytotoxicity tests after exposure to staurosporine (PKC inhibitor) did not show any changes in the TNF-mediated cytotoxicity, confirming that a PKC-dependent pathway is not involved in this process. These data indicate that 1) TNF induces expression of c-fos, c-jun and jun-B oncogenes via a PKC-dependent pathway and 2) PKC-dependent expression of these three oncogenes by TNF may not be involved in TNF-mediated cytotoxicity in ME-180 cells.

  • Cell-cell adhesion managed by various adhesion molecules is known to be one of important phenomena found in numerous immunological responses or diseases such as immunostimulation, rheumatoid arthritis and allergic diseases. In this study, we examined the regulatory role of ginsenosides (G)-Rb1, reported to display immunostimulatory and anticancer effects, on cell adhesion, the up-regulation of surface adhesion molecules and morphological changes using monocytic U937 and macrophage-like RAW264.7 cells. G-Rb1 significantly up-regulated U937 cell-cell adhesion mediated by both CD29 and CD43. It also enhanced U937 cell-fibronectin adhesion, while CD29 blocking antibody P5D2 strongly suppressed it. In agreement, this compound also significantly increased the surface level of CD29 as well as CD43. Furthermore, this compound differentially modulated CD82 up-regulation and morphological changes triggered by lipopolysaccharide (LPS) and phorbol-12-myristate-13-acetate (PMA). Therefore, these results suggest that G-Rb1 may have differential modulatory function on cell adhesion events, surface molecule expression and morphological changes responsible for immune responses.

Effects of sulfiting on the indigenous yeast flora and physicochemical properties during the fermentation of Campbell Early wine (아황산의 처리가 캠벨얼리 와인의 자연발효 시 야생효모의 변화 및 발효 특성에 미치는 영향)

  • Campbell Early grapes were spontaneously fermented with and without sulfiting to investigate the effect of sulfiting on the fermentation characteristics and physicochemical properties of Campbell Early wine. During the fermentation, the increase in the alcohol and the decrease in the soluble solid contents were faster without sulfiting, as was the increase in the yeast viable counts compared to those with sulfiting. However, the final alcohol and soluble solid contents reached similar levels with and without sulfiting. The PCR-RFLP analysis of the yeast in the ITS I-5.8S-ITS II region revealed that the increase in the S. cerevisiae was faster in the initial fermentation stage and reached a slightly higher level in the late stage with sulfiting than without sulfiting. The wine prepared after the fermentation with sulfiting showed higher malic and tartaric acid contents, as well as methanol, acetaldehyde, and n-propanol contents, than the wine prepared without sulfiting. The ethyl acetate content of the wine without sulfiting was 375.5 mg/L, which was 5.3 times higher than that (70.5 mg/L) with sulfiting. In the sensory evaluation, the wine without sulfiting obtained higher scores in flavor and overall preference than that with sulfiting.

Anti-inflammatory and Anti-allergic Effects of Phlorofucofuroeckol A and Dieckol Isolated from Ecklonia cava (감태에서 분리된 플로로탄닌의 염증 및 알러지 억제 효과)

  • The anti-inflammatory effect of brown alga Ecklonia cava is well known, and several phlorotannins have also been reported. In this study, major active components for anti-allergy and anti-inflammation were identified by NMR and MS analysis, and the levels of effectiveness were compared. Six major phlorotannins-phloroglucinol, eckol, eckstolonol, triphlorethol-A, phlorofucofuroeckol A, and dieckol-were isolated from the ethyl acetate fraction of E. cava. In order to analyze the major active substances in E. cava, antioxidant, anti-inflammatory, and anti-allergic effects were evaluated for the six separate substances. Antioxidant capacities of each phlorotannin were evaluated using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radicals, where phlorofucofuroeckol A and triphlorethol-A had the highest radical scavenging capacity in respective radical scavenging assays. Phlorofucofuroeckol A exhibited the highest inhibition of nitric oxide production in LPS-stimulated RAW 264.7 cells among phlorotannins tested. Dieckol inhibited the release of ${\beta}-hexosaminidase$, a marker for the release of histamine in mast cells, in a dose-dependent manner in antigen-stimulated RBL-2H3 mast cells. Additionally, no cytotoxicities were observed at 1 and $2{\mu}g/ml$ in both phlorofucofuroeckol A and dieckol. These results suggest that phlorofucofuroeckol A and dieckol may play a key role in allergic inflammatory reactions.

Bioequivalence Evaluation of Two Brands of Zolpidem Tartrate 10 mg Tablets(Zanilo and Stilnox) in Healthy Male Volunteers

  • The purpose of the present study was to evaluate the bioequivalence of two zolpidem tartrate tablets, Stilnox tablet(Sanofi-aventis Korea, reference product) and Zanilo tablet(ChoDang Pharm Co., Ltd., Korea, test product), according to the guidelines of Korea Food and Drug Administration(KFDA). After adding an internal standard(cimetropium), 250 ${\mu}L$ plasma samples were extracted using 1.3 mL of ethyl acetate. Extracted compounds were analyzed by HPLC with triple-quadrupole mass spectrometry. This method for determination of zolpidem is proved accurate and reproducible with the limit of quantitation of 1 ng/mL in human plasma. Twenty-four healthy male Korean volunteers received each medicine at the zolpidem tartrate dose of 10 mg in a $2{\times}2$ crossover study. There was one-week washout period between the doses. Plasma concentrations of zolpidem were monitored for over a period of 8 hr after the administration. $AUC_{0-t}$(the area under the plasma concentration-time curve) was calculated by the linear trapezoidal rule. $C_{max}$(maximum plasma drug concentration) and $T_{max}$(time to reach $C_{max}$) were compiled from the plasma concentration-time data. Analysis of variance was carried out using logarithmically transformed $AUC_{0-t}$ and $C_{max}$. No significant sequence effect was found for all of the bio-availability parameters indicating that the crossover design was properly performed. The 90% confidence intervals for the log transformed data were acceptable range of log 0.8 to log 1.25(e.g., log 0.92-log 1.06 for $AUC_{0-t}$, log 0.96-log 1.13 for $C_{max}$). The major parameters, $AUC_{0-t}$ and $C_{max}$ met the criteria of KFDA for bioequivalence indicating that Zanilo tablet is bioequivalent to Stilnox tablet.

Phosphorylated Proteins of Mitogen Stimulated-Rat Peripheral Blood Lymphocytes (분열유발인자에 의한 흰쥐 림프구 단백의 인산화)

  • This study was done to classify the proteins involved in the specific phosphorylation using the rat peripheral blood lymphocytes (rPBL) stimulated with mitogens, phorbol 12-myristate 13-acetate (PMA) and concanavalin A (Con A). The lymphocytes were incubated with $^{32}P-orthophosphate$ before PMA or Con A stimulation. The migration patterns of the phosphorylated proteins of mitogen-treated rPBL in two dimensional electrophoretic fields were analyzed after autoradiography. The stimulation of the lymphocytes with PMA and Con A increased the phosphorylation of thirteen protein fractions. The phosphorylation intensities of the protein spots differ to the treatments of the cells with specific kinase inhibitors, H-7 and W-7. These protein fractions were grouped into 3 classes, namely, PKC-mediated, CaM kinase-mediated, and other kinase mediated proteins. The effect of the duration of the stimulation on the phosphorylated behaviors occurred concurrently, not sequentially, although each individual protein fraction had a different time for the peak phosphorylation during the stimulation period upto 30 minutes. The phosphoproteins found in the cytosolic soluble fraction were phosphorylated prior to those in the pellet, whose phosphorylations were sustained at a high level for over 10 minutes. The above results suggest that the early events in lymphocyte activation involve 3 different sets of proteins which are phosphorylated by CaM kinase, PKC and other kinases, and those kinases do not work sequentially, but rather, independently or cooperatively.

Analysis of Microbial Community during the Anaerobic Dechlorination of PCE/TCE by DGGE (DGGE를 이용한 PCE 및 TCE의 혐기적 탈염소화 군집의 미생물 군집분석)

  • This study investigated the effect of PCE and TCE as electron acceptors on the bacterial composition of dechlorinating communities. The enrichment cultures reductively dechlorinating PCE and TCE were developed from three environment samples using acetate as electron donor. The cultures were prepared by sequential enrichment, which was seeded with sediment and dredged soil. Denatured gradient gel electrophresis (DGGE) of 16S rRNA gene fragment was used to compare the microbial communities of these three enrichment cultures. After incubation for 4 weeks, the removal efficiencies of PCE and TCE were highest from Yeocheon site (87.37% and 84.46%, respectively). PCE and TCE as electron acceptors affected the bacterial diversity and community profiles in the enrichment cultures. DGGE analysis showed that the dominant bacteria in PCE and TCE enrichment were belonged to Clostridium sp., Desulfotomaculum sp., and uncultured bacteria.
