Urea-Formaldehyde 수지가공포에 있어 Resorcinol의 유리 Formaldehyde 포착효과 (Effect of Resorcinol as Free Formaldehyde Scavenger for Fabric Finished with Urea-formaldehyde Precondensate.)

  • To control free formaldehyde release from fabric finished with N-methylol compounds, resin finished cotton fabric was treated with resorcinol solution, dried and cured. Factors affecting to control formaldehyde release have been investigated. It was shown that the aftertreatment with resorcinol greatly suppressed the free formaldehyde release. Up to concentration of about 5% of resorcinol, the concentration of resorcinol effected on the control of free and evolved formaldehyde. And at high concentration of resorcinol, however, the concentration became rather insensitive to contol formaldehyde release. Addition of some salt catalysts such as ammonium chloride, zinc nitrate, sodium acetate and ammonium acetate, was effective in decreasing formaldehyde release. Considering the effect on the control of formaldehyde and crease recovery, ammonium acetate was concidered to be the best catalyst. It was observed that the optimum curing temperature for the resorcinol treatment was about 15$0^{\circ}C$, and that the curing time did not affected formaldehyde release over three minutes. Although the treatment of resorcinol had a little adverse effect on crease recovery of resin finished fabric, this effect could be negligible.

Fenton Reaction을 이용한 Phenanthrene 오염 토양 처리에서 Acetate의 영향 (Influence of Acetate on the Removal of Phenanthrene from Contaminated Soil using Fenton Reaction)

  • 오염토양에 펜톤 반응을 적용함에 있어서 과산화수소의 빠른 소모로 과량의 과산화수소가 요구되어지는 것이 단점으로 지적되고 있다. 이에 본 연구에서는 대표적인 PAHs물질인 phenanthrene으로 오염된 토양 처리를 위해 철의 리간드로 acetate를 사용하여 과산화수소의 안정성 확보를 통하여 공정의 효율을 높이고자 하였다. Acetate는 철의 몰농도에 대비하여 0.5배에서 3배(2${\sim}$12 mM)까지 주입하였고, 과산화수소는 분해 효율에 미치는 영향을 배제하기 위해 낮은 농도인 0.7%를 주입하였다. Acetate가 주입되어 과산화수소의 잔류시간은 최대 50배 이상 증가하였으며, 과산화수소의 안정성이 확보됨에 따라 phenanthrene의 제거율도 70%까지 향상되었다. 반응 중에 철은 2가와 3가로 산화환원을 반복하였고 과산화수소가 모두 분해되는 시점부터 $HO_2^-$에 의해 2가철로 환원이 이루어졌다. 과산화수소의 영향으로 반응중의 pH는 산성영역을 나타냈으며, acetate가 8 mM 이상 주입되었을때 4${\sim}$5범위 내에 머무르는 것을 확인하였다. Fenton reaction에 의해 철의 리간드로 사용된 acetate 역시 분해가 이루어지는 것을 확인할 수 있었으며, phenanthrene의 비해 분해되는 시점이 느린 것으로 나타나 분해되어지는 경쟁관계에서 phenanthrene이 우세한 것으로 판단된다. 이상의 연구결과를 통해 오염 토양처리에 기존 Fenton reaction의 효율성과 경제성을 향상시킬 수 있는 가능성을 확인할 수 있었으며, 중성영역의 pH로 확장된 연구 등을 통해 좀 더 현실적인 공법으로 발전할 수 있으리라 판단된다.

소리쟁이 분획물의 지방세포 분화 억제 효과 (Inhibitory Effect of Rumex Crispus L. Fraction on Adipocyte Differentiation in 3T3-L1 Cells)

  • 소리쟁이 추출물 및 분획물의 3T3-L1 전지방세포의 분화에 미치는 영향을 확인한 결과, 소리쟁이의 hexane, ethyl acetate 분획물에 의해 지방세포의 lipid droplet의 형성이 유의적으로 억제되었으며, 소리쟁이 에탄올 추출물과 모든 분획물 10 ${\mu}g/mL$의 농도에서 중성지질(triglyceride)의 함량이 유의적으로 감소되었다. 또한 지방세포의 분화에 관여하는 전사인자인 $PPAR{\gamma}$, $C/EBP{\alpha}$의 단백질 발현은 10 ${\mu}g/mL$의 ethyl acetate, butanol 분획물에 의해 현저하게 감소되었다. 따라서 소리쟁이 추출물과 분획물 중 가장 활성이 우수한 ethyl acetate 분획물을 이용하여 지방분화에 관여하는 전사 인자인 $PPAR{\gamma}$, $C/EBP{\alpha}$, 그리고 SREBP1c 및 지질의 합성, 수송, 저장에 관여하는 ACS, FAS, FATP1, FABP4, Perilipin의 발현에 미치는 영향을 관찰한 결과, ethyl acetate 분획물은 유의적으로 모든 유전자의 발현을 억제시켰다. 따라서 소리쟁이의 ethyl acetate 분획물은 지방세포의 분화에 관여하는 전사인자 및 유전자들의 발현을 감소시킴으로써 항비만 효과가 있는 천연물 소재로 이용 가능할 것으로 생각된다.

LPS로 유도된 RAW 264.7 대식세포에 대한 미역(Undaria pinnatifida) Ethyl Acetate 분획물의 항염증 효과 (Anti-Inflammatory Effect of Ethyl Acetate Fraction Isolated from Undaria pinnatifida on Lipopolysaccharides-Stimulated RAW 264.7 Cells)

  • An ethanolic extract of Undaria pinnatifida was fractionated using several solvents. Of the fractions, the ethyl acetate fraction had the greatest inhibitory effect on lipopolysaccharide (LPS)-induced nitric oxide (NO) production in RAW 264.7 macrophage cells. Using this fraction (U. pinnatifida ethyl acetate extract, UPE), we investigated the molecular mechanism underlying its inhibitory effect on LPS-stimulated RAW 264.7 cells. Pretreatment of the cells with up to $100{\mu}g/mL$ UPE significantly inhibited NO production and inducible nitric oxide synthase (iNOS) expression, in a dose-dependent manner. Similarly, UPE treatment markedly reduced the production of pro-inflammatory cytokines, such as interleukin (IL)-1, IL-6 and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), while it strongly suppressed the nuclear translocation of nuclear factor-kappa B (NF-${\kappa}B$) by preventing proteolytic degradation of inhibitor of nuclear factor ${\kappa}B$ $(I{\kappa}B)-{\alpha}$. Moreover, UPE treatment significantly reduced the phosphorylation of phosphatidylinositol 3-kinase (PI3K)/Akt and mitogen-activated protein kinase (MAPK) in LPS-stimulated cells. These results indicate that UPE contains anti-inflammatory compounds and suggest that it might be used as a functional food material that assists in prevention of inflammatory diseases.

Effect of Alpinia officinarum on the Formation of Advanced Glycation Endproducts and Aldose Reductase Activity

  • The inhibition effects of an Alpinia officinarum (AO, Zingiberaceae) on the formation of advanced glycation end products, aldose reductase, and scavenging effect on 1,1-diphenyl-2-picryl-hydrazil (DPPH) radical for the prevention and/or treatment of diabetic complications were investigated. The ethyl acetate fraction of AO was the most effective among all fractions. Through the tests with electron impact-mass spectrometry and nuclear magnetic resonance, two compounds (compound 1 and 2) finally obtained from the ethyl acetate fraction of AO were identified as galangin (1) and kaempferide (2), respectively. In addition, the compound 1 and 2 and the ethyl acetate fraction were compared for the prevention effect on advanced glycation end products, aldose reductase, and the scavenging effect on DPPH radical. The ethyl acetate fraction was significantly more effective than the 2 compounds for those preventive activities.

  • The Synthesis of E, E, E-12-fluorofarnesol and E, Z-6-fluorofarnesol which are key intermediates for the study of biosynthesis of some sesquiterpenes, is described. E, E-Farnesyl acetate is treated with selenium dioxide to give E, E, E-12-hydroxy farnesyl acetate, whih is transformed by DAST into E, E, E-12-hydroxy farnesyl acetate, which is transformed by DAST into E, E, E,-12-fluorofarnesylacetate. The latter compound is hydrolyzed to E, E, E,-12-fluorofarnesol. The reformatsky reaction of 6-methyl-5-hepten-2-one with ethyl bromofluoroacetate affords ethyl 2-fluoro-3-hydroxy-3, 7 dimethyl-6-octanoate. This ester is acetylated and eliminated to give ethyl (Z)-2-fluoro-3, 7-dimethylocta-2, 6-dienoate, which is transformed to allyl bromide via allylic alcohol. The allyl bromide is treated with dianion of methyl acetate to give-keto ester. The $\beta$-keto ester is converted to diethyl phosphoryloxy compound. The conjugate addition of lithium dimethylcuprate to the latter compound gives fluoro ester, which is treated with DIBAL to afford E, Z-6-fluorofarnesol.

쑥 추출물의 Tyrosinase 효소활성에 미치는 억제효과 (Inhibitory Effect of Mugwort Extracts on Tyrosinase Activity)

  • Melanin 생합성에 관련된 tyrosinase 저해활성올 생 쑥과 건조 쑥 용매 추출물로부터 검색하였다. 쑥은 ethanol, hexane, chloroform, ethyl acetate 용매 및 열수 추출을 하였고, 생 쑥의 ethanol 추출물의 ethyl acetate 분획과 건조 쑥의 ethan야 추출물의 hexane 분획에서 회수올은 각각 15.2%와 15.5%로 가장 높았다. 각 용매 추출물의 tyrosinase 활성 저해율은 건조 쑥 추출불보다 생 쑥 추출울이 더 높았고, 생 쑥의 ethanol 추출물의 chloroform 분획과 hexane 분획에서 각각 98.9%와 9 96.7%로 큰 저해활성을 가졌다.

오이풀 아세톤 추출물을 이용한 용매 분획물의 항산화 효과 (Antioxidant Effects of Solvent Fraction from Sanguisorbae officinalis L. with Acetone)

  • 오이풀 아세톤 추출물로부터 용매분획물의 기능성 화장품 소재로 활용하기 위하여 항산화 효과를 측정한 후 화장품 소재로서의 가능성을 검증하였다. 항산화 효과를 확인하기 위하여 electron donating ability을 측정한 결과 ethyl actate 층과 n-butyl alcohol 층이 전 농도에서 대조군 BHA와 유사한 활성을 나타내었으며, ABTS radical cation decolorization assay 결과 ethyl acetate 층, n-butyl alcohol 층, water 층 모두 전 농도에서 99% 이상의 효과를 나타내어 BHA와 유사하였다. hydrogen peroxide scavenging assay 결과 ethyl acetate 층, n-butyl alcohol 층이 대조군 ascorbic acid 보다 효과가 높았다. Superoxide dismutase (SOD) 유사 활성 측정 결과, n-butyl alcohol 층이 1,000 ${\mu}g/mL$ 농도에서 50% 이상의 효능이 있었으며, Superoxide anion 라디칼 소거능은 n-butyl alcohol 층의 1,000 ${\mu}g/mL$에서 45%의 효과가 있었다. 이상의 결과로 미루어 보아 오이풀 아세톤 추출물의 분획물 중 ethyl acetate 층과 n-butyl alcohol 층에서 항산화 효과가 있었으며, 새로운 항산화 화장품의 소재로서 가능성을 확인 할 수 있었다.

포도종자 에텔아세테이트 분획물로부터 분리한 카테킨 화합물의 항산화 및 지질과산화 억제효과 (Antioxidant and Lipid Peroxidation Inhibition Effects of Catechin Compounds Isolated from Ethyl Acetate Fraction of Grape Seed Ethanol Extract)

  • 본 연구는 캠벨종 포도 종자 ethyl acetate 분획물로 부터 생리활성 물질을 분리$\cdot$동정하고 분리물질의 생리활성을 검토하기 위하여 수행되었다. 캠벨종 종자 ethyl acetate분획물로부터 sephadex LP-20 column chromatography를 이용하여 활성물질을 단리하고 ${1}^H$${13}^C$) NMR 분석을 통하여 (+)-catechin과 (-)-epicatechin을 동정하였다. HPLC를 이용하여 정량 분석한 결과, (+)-catechin은 45.7$\%$의, (-)-epicatechin은 $35.1\%$의 함량을 나타내었으며 이들 화합물이 포도종자 중의 주요한 생리 활성물질로 사료된다. 이들 화합물의 단독 또는 병용 처리 시 항산화 활성, 지질과산화 억제효과 및 자외선 차단 효과를 살펴본 결과, DPPH free radical 소거 활성에서는 (+)-catechin과 (-)-epicatechin 모두가 $RC_{50}$= 11.1 ${\mu}g/mL$$RC_{50}$= 10.4 ${\mu}g/mL$으로 높은 free radical 소거 활성을 나타내었으나, 지질 과산화 억제는 (+)-catechin이 나 (-)-epicatechin 단독처리보다는 ethyl acetate 분획물에서 훨씬 효과가 좋은 것으로 나타났다.

갈근 및 녹차추출물의 아질산염 소거작용 및 간독성 보호효과 (Nitrite scavenging activity and protective effect of the Puerariae Radix and green tea extract on lead acetate and cadmium-induced liver damage in mice)

  • This study was designed to evaluated the nitrite scavenging activity and protective effect of the Puerariae Radix and green tea extract on lead acetate and cadmium-induced liver damage in mice. The quantitative analytical method for major antioxidants, isoflavones, puerarin, catechine and caffeine in galgun (Puerariae Radix) and green tera extract were established by HPLC. Contents of isoflavones, such as daidzin, genistin, daidzein and genistein were 4.23g/100g, 0.13g/100g, 0.07g/100g, and 0.03g/100g, and puerarin contents was 8.99g/100g, respectively. The total catechins and caffeine contents of green tea extract were 49.24g/100g and 6.53g/100g. The nitrite scavenging ability of galgun extract (pH 1.2, 100mg/ml) was 98.07% and it was higher than those of other extracts. It was the highest at the pH 1.2 and more than 64% in 25~100mg green tea extract, and was dependents on pH and concentration of the samples. The hepatoprotective effects of an aqueous extract from the root of gal gun and standard puerarin were evaluated against lead acetate and cadmium-induced liver damage in mice. Galgun extract and standard at a dose of 100mg/kg and 10mg/kg, 50mg/kg were administered orally once daily for successive 5 days and then a lead acetate and cadmium were orally at 3 hrs after the every day administration of galgun. The substantially elevated serum enzymatic activities of alanine and aspartate aminotransferase were due to lead acetate and cadmium treatment was dose dependently restored to the near normal level. In addition, galgun extract also significantly prevented the elevation of hepatic malon-dialdehyde formation in the liver of lead acetate and cadmium intoxicated mice in a dose-dependent manner. The results of this study clearly indicated that green tea and galgun extracts had nitrite scavenging ability and galgun extract had potent hepato-protective effects against lead acetate and cadmium-induced hepatic damage in mice and standard puerarin was also showed similar to the results of the galgun extracts.
