Groundwater contamination by nitrate exceeding water quality criteria (10 mg $NO_3{^-}-N/L$) occurs frequently. Fumarate, acetate, formate, lactate, propionate, ethanol, methane and hydrogen gas were evaluated for their nitrate removal efficiencies and removal rates for in situ bioremediation of nitrate contaminated groundwater. Denitrification rate for each substrate was in the order of: fumarate > hydrogen > formate/lactate > ethanol > propionate > methanol > acetate. Microcosm studies were performed with fumarate and acetate. When fumarate was used as a substrate, nitrate was removed 100 percent with rate of 0.66 mmol/day while conversion rate from nitrate to nitrogen gas or another by-product was 87 percent. 42 mg of fumarate was needed to remove 30 mg $NO_3{^-}-N/L$. When using acetate as carbon source, 31 percent of nitrate was removed during initial adjustment period. Among removed fraction, however, 83 percent of nitrate removed by cell growth. Overall nitrate removal rate was 0.37 mmol/day. Acetate showed longer lag time in consumption compared to that of nitrate, which implying that acetate would be better carbon source compared to fumarate as more amount was utilized for nitrate removal than cell growth.
Cup-plant (Silphium perfoliatum L.) has potential to produce high biomass and highly digestible forage in the wetlands where other productive forages do not grow or produce well. However, high moisture content at harvest is a considerable disadvantage of cup-plant for the production of high quality silage. This study was conducted to determine the effect of moisture content on the characteristics of cup-plant silage. Harvested cup-plant was ensiled in farm scale plastic bag silos and laboratory silos. In the plastic bag silos, first growth (FG) and regrowth (RG) cup-plant was harvested, wilted and ensiled. Dry matter content of FG and RG was 280 g/kg and 320 g/kg after 48 hr of wilting. The silage made with FG had pH 5.3 and 5.63 g/kg DM of acetate as a major volatile fatty acid. The composition of lactate, butyrate and acetate production was 1.0: 0.9: 2.3. The pH of silage made with RG was 4.5 and lactate was a major fermentation end product (16.8 g/kg DM). In the laboratory silos, wilted and unwilted first growth cup-plant material was ensiled to compare the early fermentation end products at days 2, 4, 11, and 40. Wilting increased dry matter content by 42% in the harvested material. Wilted silage showed about one unit lower pH until day 11. The contents of ammonia nitrogen and acetate were higher in un wilted silage, while that of lactate was higher in wilted silage (p<0.05). Butyrate and propionate were not detected in the wilted silage until day 40. We conclude from the results that moisture control is essential for the production of high quality cup-plant silage and high pH of cup-plant silage is due to low concentrations of fermentation end products.
Fifty-four, mixed-sex, halothane-carrier crossbred (Yorkshire${\times}$Landrace) pigs with an average initial BW of $108.2{\pm}0.8$ kg were randomly allotted to one of three dietary treatments for 5 d before slaughter: i) a control corn-soybean meal finisher diet devoid of supplemental magnesium; ii) a diet supplemented with 1.5 g/kg of elemental Mg from magnesium acetate; and iii) a diet supplemented with 1.5 g/kg of elemental Mg from magnesium sulfate heptahydrate. Serum creatine kinase (CK), lactate and glucose were analyzed at slaughter. Muscles from longissimus (LM) were packaged and stored to simulate display storage for muscle lactate and glycogen determinations at 0, 1, 2, 3, and 4 d. Mg supplementation reduced (p<0.05) serum CK and lactate concentration, but had no effect (p>0.05) on serum glucose. Daily change of muscle lactate concentration linearly increased (p<0.01), while glucose concentration linearly decreased (p<0.05) as storage time increased in all treatments. However, dietary Mg acetate and Mg sulfate supplementation in pigs elevated (p<0.05) muscle glycogen and reduced (p<0.05) muscle lactate concentrations, especially during the first 2 d of display, compared with pigs fed the control diet. This study suggests that short-term feeding of magnesium acetate and magnesium sulfate to heterozygous carriers of the halothane gene has beneficial effects on stress response and pork quality by improving blood and muscle biochemical indexes.

Electrochemical control of the metabolic flux of W. kimchii sk10 on glucose and pyruvate was studied. The growing cell of W. kimchii sk10 produced 87.4 mM lactate, 69.3 mM ethanol, and 4.9mM lactate from 83.1mM glucose under oxidation condition of the anode compartment, but 98.9 mM lactate, 84.3mM ethanol, and 0.2 mM acetate were produced from 90.8 mM glucose under reduction condition of the cathode compartment for 24 h, respectively. The resting cell of W. kimchii sk10 produced 15.9 mM lactate and 15.2 mM acetate from 32.1 mM pyruvate under oxidation condition of the anode compartment, and 71.3 mM lactate and 3.8 mM acetate from 79.8mM pyruvate under reduction condition of the cathode compartment. The redox balance (NADH/$NAD^+$) of metabolites electrochemically produced from pyruvate was 1.05 and 18.76 under oxidation and reduction conditions, respectively. On the basis of these results, we suggest that the neutral red (NR) immobilized in bacterial membrane can function as an electron channel for the electron transfer between electrode and cytoplasm without dissipation of membrane potential, and that the bacterial fermentation of W. kimchii sk10 can be shifted to oxidized or reduced pathways by the electrochemical oxidation or reduction, respectively.

As a part of our research project for in-situ bioremediation of nitrate contaminated. groundwater, screening studies to determine an effective electron donor (EO) and/or carbon source (CS) such as acetate, ethanol, formate, fumarate, lactate, and propionate were conducted. To evaluate the feasibility for the biological degradation of nitrate, soil microcosm studies using nitrate-contaminated soil and groundwater were performed. The nitrate removal percentage in the order from the highest to the lowest was: formate, fumarate, and ethanol > lactate > propionate. Essentially no nitrate consumption was observed In acetate-fed microcosms. The order of nitrate removal rate from the highest to lowest was fumarate, formate, lactate, ethanol, and propionate. These results suggest that fumarate and formate are promising EDs/CSs for in-situ bioremediation of nitrate - contaminated oxygenated groundwater.
Lactate and malate dehydrogenase isozymes in crude extracts of various tissues from 28 avian species were investigated by means of celluose acetate electrophoresis. In a given species the presence of two types of the malate dehydrogenase isozymes could be revealed in somatic tissues; one was characterized by fast electrophoretic mobility, and the other by slow mobility. Although the bird lactate dehydrogenase isozymes existed in a variety of species-specific molecular forms, they seemed to show the same pattern within the same genus and family.
Using the synchronous culture method and the manometric technique, changes in respiratory activities, utilization of some organic acids (succinate, malate, lactate and acetate etc.) and its effect on glucose metabolism in Chlorella cells at different growing stages were measured. 1) Endogenous respiration of the cells was not active at growing stage and was almost constant throughout the early ripening, maturing and division stages. 2) Lactate was utilized as respiratory substrate better than other organic acids tested. Exogenous respiration of glucose was most active at growing and maturing stages and was decreased strikingly at division stage. 3) Succinate and citrate inhibited endogenous and glucose respiration of the cells throughout the all life cycle. 4) Malate and acetate were utilized in the cells at early growing and division stages better, and malate enhanced the glucose respiration while in case of acetate it was depressed. 5) Calcium ion inhibited not only permeability of respiratory substrate but endogenous respiration itself.
The growth of Bifidobacterium breve strain HP2 was completely inhibited by the addition of lactate higher than 4.0% but not by the addition of acetate. Two kinds of lactate-tolerant mutants were isolated by the nitrosoguanidine treatment, enrichment on a liquid medium with 5% lactate, and selection on agar plates with 5% lactate. The mutants were not only able to grow in the presence of 5% lactate but also improved in viable cell stability in the acidic pH range. In a pH-controlled fermentor, mutant N-1-5 grew at a rate slower than that of the wild type but its growth yield was higher. Notably, mutants were more halotolerant and more osmotolerant than the wild type and they were able to grow in the presence of 3% NaCl or 25% lactose at which the wild type entirely stopped the growth. The enzyme activities involved in the lactose metabolism in B. breve were measured to elucidate the biochemical basis for lactate tolerance. In the mutants, activities of several enzymes including phosphoglucomutase decreased compared to the wild-type, which may explain their lower growth rate. However, the activity of lactate dehydrogenase or its nature of inhibition by lactate was not altered.
The effects of energy-yielding substrates on coronary circulation, myocardial oxygen metabolism, and intramyocytic adenylates of perfused Wistar control rat(WC) and spontaneously hypertensive rat(SHR) hearts were examined under basal and $\beta$-adrenergic stimulation conditions. The perfusion medium (1.0mM Ca2+) contained 5mM glucose (+5U/l insulin) in combination with 5mM pyruvate, 5mM lacate, 5mM acetate, or 5mM octanoate as energy substrates. Hearts were perfused with each substrate buffer for 20min under basal conditions. Coronary functinal hyperemia was induced by infusing for 20min isoproterenol (ISO, 1uM), a $\beta$-receptor agonist. Cardiac adenylates, glycolytic intermediates, and coronary venous lactate were measured by using an enzymatic analysis technique. Under basal conditions, acetate and octanoate significantly increased coronary flow(CF) of WC in parallel with myocardial oxygen consumption. However, CF of SHR was partly attenuated by coronary vasoconstriction despite metabolic acidosis. In addition, pyruvate and lactate depressd ISO-induced coronary functional hyperemia in SHR. It should be noted that octanoate exhibited coronary dysfunction under ISO conditions. On the other hand, fat substrates depleted myocardial high energy phosphate pool and accumulated breakdown intermediates. In SHR with coronary vasoconstriction under basal conditions, and with depressed coronary functional hyperemia, high energy phosphates were greatly depleted. These results suggest that energy substrates in the myocardium and coronary smooth muscle alter remarkably coronary circulation, and that coronary circulatory function is associated with a reserve of high energy phosphates and a balance between breakdown and nono synthesis of energy phosphates. These findings could be explained by alterations in the cytosolic redox state manipulated by LDH and hence in the cytosolic phosphorylation potential, which might be involved in hypertension of SHR.
In batch cultures of Lactobacillus delbrueckii, cell growth and lactic acid production were affected by two main factors, inhibition by lactic acid and limitation by nutritional components. In order to increase th productivity significantly, a continuous stirred tank reactor with cell recycle was employed. A cell desnity of 145g dry weight/l and a volumetric productivity of 73 g/l$\cdot $h were obtained with an effluent concentration of 85 g/l lactic acid. The productivity achieved by this system was 23-fold higher than those obtained by the corresponding batch cultivations. Once the lactic acid concentration reached the steady steady state, lowering the yeast extract concentration caused the reduction of the lactic acid concentration without affection the biomass concentration. Finally, the formation of D-lactate was investgated. During the various cultures, a small amount of D-lactate always formed, even thought a majority of lactate was L-isomer, It was supposed that the relative amount of the D-lactate was affected by glucose limitation, and there seems to exist a certain relationship between the concentration of D-lactate and acetate.
