Antioxidant and Antimicrobial Effects of Solvent Fractions from Smilax china L. Leaves (청미래덩굴 잎(Smilax china L.) 용매 분획물의 항산화 및 항균 효과)

  • To utilize Smilax china L. leaves as a natural preservatives, solvent fractions from crude methanol extract were prepared and investigated their antioxidant and antimicrobial activities against Staphylococcus aureus. Antioxidant activities were examined by 1,1-diphenyl-1-picrylhydrazyl radical scavenging and ferric ion reducing antioxidant power assays. Ethyl acetate fraction from Smilax china L. leaves exhibited the strongest antioxidant and antimicrobial activities among the fractions (P<0.05), as well as the highest total phenolic and total flavonoid contents. Caffeic acid, ferulic acid, quercetin, and kaempferol contents in the ethyl acetate fraction from Smilax china L. leaves were determined by TLC and HPLC. In aqueous phase, as well as the n-butanol and n-hexane fractions, quercetin, ferulic acid, and kaempferol were detected in small amounts. Ferulic acid, which showed the highest content, is the major active compound from Smilax china L. leaves.

Efficiency of Eluting Solvents for Solid-Phase Extraction During Multiresidue Analysis of 16 Pesticides in Cucumber (고상추출카트리지를 이용한 잔류농약 다중분석과정에서 정제용매에 따른 회수율 비교)

  • A study was carried out to optimize the cleanup step using a solid-phase extraction (SPE) cartridge, $SupelClean^{TM}$ Florisil, for multiresidue analysis of 16 pesticides in cucumber matrix. Eluting efficiencies of two solvent systems including acetone/n-hexane and ethyl acetate/n-hexane mixtures were critically evaluated by recoveries of target anaytes from the SPE Florisil column. Based on the recovery as a measure of eluting efficiency, an acetone/n-hexane (20/80,v/v) mixture provided more than 80% recovery for 15 pesticides except bifenthrin. In case of ethyl acetate/n-hexane, 14 pesticides showed recoveries higher than 75% while those far alachlor and bifenthrin were less than 30%.

Analysis of the Free Amino Acids and Volatile-Flavor Compounds in the Commercial Brown-Rice Vinegar Prepared via Static Acetic-Acid Fermentation (시판 정치배양 현미식초의 유리아미노산 및 휘발성 향기성분 특성 분석)

  • This study was performed to evaluate the quality of the commercial brown-rice vinegars of South Korean and Japanese origins, which were prepared via static acetic-acid fermentation. The major free amino acids of Korean brown-rice vinegars were proline, glutamic acid and phenylalanine, while those of the Japanese were proline, valine, phenylalanine, lysine, ${\gamma}$-aminobutyric acid, alanine and isoleucine. The ${\gamma}$-aminobutyric acid (GABA) and total amino acids (3686.37~4212.27 mg%) contents were found to be significantly higher in the Japanese than in the Korean brown-rice vinegars. The key volatile compounds of the Korean brown-rice vinegars, analyzed with GC-MS, were acetic acid, benzaldehyde, phenethyl alcohol and phenethyl acetate while those of the Japanese brown-rice vinegars were acetic acid, ethyl acetate, ethyl alcohol, isoamyl acetate, phenethyl acetate and benzaldehyde. The volatile patterns of the Korean and Japanese commercial brown-rice vinegars were effectively distinguished from each other using an electronic nose, through which it was also elucidated that the volatiles profiles were similar among the Japanese vinegars but were different among the Korean vinegars.

The Effects of Sedum sarmentosum Bunge on Serum Lipid Concentration in Ovariectomized Rats (돌나물이 난소 절제한 흰쥐의 혈 중 지질 함량에 미치는 영향)

  • The aim of this study was to investigate in an in vivo model the effects of ethanol extract and ethyl ether and ethyl acetate fractions of Sedum sarmentosum Bunge (SS). To investigate the effects of ethanol extract and ethyl ether and ethyl acetate fractions, Sprage-Dawley female rats were randomly assigned to groups as follows : Sham, OVX-control and ovariectomized rats supplemented with SS at 50 mg/kg bw/day, with SS at 100 mg/kg bw/day, with SS at 200 mg/kg bw/day, with ethyl ether fraction of SS at 10 mg/ka bw/day and with ethyl acetate fraction of SS at 10 mg/kg bw/day. Food intake and food efficiency ratio were not significantly different in groups. But body weight gain was decreased by supplementation of ethyl acetate fraction of SS compared to OVX-control. The levels of total cholesterol and triglyceride in serum were higher in ovariectomized control rats than Sham-operated rats, but supplementation of SS ethanol extracts at 200 mg/kg bw/day decreased the level of the lipid in serum. The level of HDL-cholesterol in serum increased by supplementation of SS ethanol extracts at 200 mg/kg bw/day (p<0.01). The levels of total cholesterol and triglyceride in serum were decreased by supplementation of ethyl acetate fraction of SS (p<0.01). On the other hand, the level of HDL-cholesterol in serum was increased by supplementation of ethyl acetate fraction of SS (p<0.05). On conclusion, it might be expected that ethanol extract and ethyl acetate fraction of SS is believed to be a possible protective effects for the fatty serum increasing serum lipid.

Biosynthesis of Saponins in Panax ginseng (고려인삼 사포닌류의 생합성에 관한 연구)

  • Biosynthesis of saponins from acetate, mevalonate and squalene using root slices of panax ginseng C.A. Meyer was investigated. The sliced roots (2g) were incubated with the reaction mixture containing 20 M sodium acetate ($500\mu$Ci [U-$^{l4}C$]-acetate),10 mM mevalonate ($25\mu$Ci [2-$^{l4}C$]-mevalonate) or 10 mM swidme ($10\mu$Ci [4,8,12,13,17,21-$^3H$]-squalene) respectively at $30^{\circ}C$ for 72 hours. Biosynthesis of labelled ginseng saponine from [U-l4C]-acetate, [2-$^{l4}C$]-mevalonate and [4,8,12,13,17,21-$^3H$]-squalene was confirmed by autoradiography. Analysis of the products from [U-$^{l4}C$]-acetate by T.L.C. showed that the % radioactivities in panaxadiol, panaxatriol, squalene and mevalonate were found to be 2.1%, 2.7%, 2.6% and 0.2% respectively. Some of the sugars were also highly labelled. Analysis of the products from [2-$^{l4}C$]-mevalonate by T.L.C. showed that squalene was highly labelled and the products from [4,8,12,13,17,21-$^3H$]-squalene showed that panaxadiol, panaxatriol and sterol were highly labelled. From the above results, it was suggested that saponine might be synthesized from acetate via mevalonate-squalene route as expected in ginseng root.

Effects of Some Chemials on Ethylene Evolution and Abscission of Fruits and Leaves in Oriental Pera (수종(數種)의 약제(藥劑)가 배나무의 낙과(落果) 및 에틸렌 발생(發生)에 미치는 영향(影響))

  • In order to examine the relationship of fruit abscission and ethylene evolution in 'Jojuro' and 'Imamuraaki' pear trees, the thinning chemicals, ${\beta}$-naphthalene acetic acid (NAA), O, O-dimethyl-O-(3-methyl-4 nitrophenyl) posphorothioate (MEP), 1-naphthyl N-methyl carbamate (carbaryl), and 2-chloroethyl phosphonic acid (ethephon) were applied. Concurrently the effect of calcium acetate on the control of fruit abscission were examined when calcium acetate was added to ethephon and carbaryl. 1. All the applied chemicals (MEP, NAA, carbaryl and ethephon) was effective to abscise fruits in 'Jojuro', and MEP and carbaryl in 'Imamuraaki's. 2. The application of ethephon increased the ethylene evolution but other chemicals did not increase it. 3. There were no significant differences in total sugar contents of fruits by MEP, NAA, carbaryl and ethephon treatments. 4. When the calcium acetate was added to the ethephon and carbaryl, the thinning effects were offsetted. 5. The rate of defoliation due to ethephon treatment was controlled by addition of calcium acetate at 0.1~0.25 M. It was clarified that fruit abscission induced by thinning chemicals except ethephon was not directly related to ethylene production and that degree of fruit thinning can be regulated by addition of calcium acetate to thinning chemicals.

  • 7-keto-dehydroepiandrosterone-acetate (7-keto-DHEA-acetate) is an anabolic steroids, and we studied basically to the metabolites of it after human dosing. We tested the matrix effect from human urine to detect the 7-keto-DHEA-acetate. And LC/ESI/MS and GC/MSD was used to detect the metabolites in dosed urine. We found the some unknown compound from dosed urine (M1, M2, M3, M4 and M5), and from these results, we supposed that these compounds have the more than 3 hydroxyl and/or ketone group. Metabolite M1 was supposed that molecular weight is 302 and 3-,17-diketone and 7-hydroxyl compound (7-OH-androstendione). Metabolite M2 was supposed that the molecular weight was same to M1 and 7-,17-diketone and 3-hydroxyl compound (7-keto-DHEA).

Inhibitory Effect of Rumex Crispus L. Fraction on Adipocyte Differentiation in 3T3-L1 Cells (소리쟁이 분획물의 지방세포 분화 억제 효과)

  • The anti-obesity effect of ethanol xtract and their fractions from Rumex Crispus L. on the differentiation of 3T3-L1 pre-adipocytes to adipocytes was investigated by suppressing adipocyte differentiation and lipid accumulation with Oil red O assay, western blot and real-time PCR analysis. Ethyl acetate fraction of Rumex crispus L. significantly inhibited adipocyte differentiation when treated during the adipocyte differentiation process, as assessed by measuring fat accumulation using Oil red O staining. In inducing differentiation of 3T3-L1 preadipocytes in the presence of an adipogenic cocktail, isobutylmethylxanthine (IBMX), dexamethasone- and insulin-along with ethyl acetate fraction residue processing treatment significantly decreased protein expression of obesity-related proteins, such as peroxisome-proliferators-activated-receptor-${\gamma}$ ($PPAR{\gamma}$) and CCAAT enhancer-binding-proteins ${\alpha}$ ($C/EBP{\alpha}$). These results indicate that ethyl acetate fraction of Rumex crispus L. is the most effective candidate for preventing obesity. However further studies will be needed to identify the active compounds that confer the anti-obesity activity of ethyl acetate fraction from Rumex crispus L.

A Study on the Mechanical Properties and Handle of Velvet Fabrics (Velvet직물의 역학적 특성과 태(태))

  • The purpose of this study is to examine and to evaluate the properties of pile materials to produce velvet fabrics which have excellent handle. In order to perform this purpose, the mechanical properties, hand value (H.V.), total hand value (T.H.V.), total appearance value (T.A.V.) of acetate, cuprammonium rayon, cotton materials for velvet as the typical commercial Pile fabric were measured and analyzed by KES-F system. The important results obtained by this study are as follows. 1 For acetate pile, elogational deformation was easy and bending rigidity of weft was high and elastic recovery was excellent compared with the other materials. 2. Cuprammonium rayon velvet was shown that pile bending rigidity and hysterisis were low. Cotton velvet was shown that compressional energy was high and compressinal elasticity was exellent. 3. In the case that specimens were applied by men's winter suit program, H.V was shown that Koshi of acetate velvet was similar to that of cotton. Numeri of cuprammonium rayon velvet was higher than the others and Fukurami values for all of the three fabrics were similar. 4. But in case that specimens were applied by women's winter suit program Koshi of cuprammonium rayon velvet was lower. But Numeri of that was higher than the others and Fukurami for all of the three fabrics were similar and Sofutosa of cotton velvet was lower than the others. T.H.V. of the acetate velvet was the highest in men's winter suit program while cuprammonium rayon velvet was the highest in women's winter suit program T.A.V. of acetate velvet was highest.

A Study on Joining of 3D Thermoset and Biodegradable Polymers (열경화성 3D 프린트 몰드와 생분해성 소재 접합에 관한 연구)

  • Laser heat source was applied on 3D poly urethane model built by 3D printer and cellulous acetate for joining. A diode laser with 808nm wavelength was transmitted through the 3D model and applied on the boundary of ABS/Acetate and 3D poly urethane model. Based on the experimental result, the ABS and 3D built poly-urethane polymers was successfully joined, but the mechanical strength was not enough at the joining boundaries in the range of 6watt to 8watt of laser heat source. However, biodegradable acetate was successfully joined without damaging the 3D built model and mechanical strength was properly achieved. The optimum laser power was found between 5watt and 8watt with scanning speed of 500mm/min, 700mm/min and 1,000mm/min. Based on the SEM analysis the filling mechanism was that the applied pressure on 3D built model squeezed the fluidic thermoplastics, ABS and acetate, into the structure of 3D model. Therefore soundness of joining was strongly depending on the viscosity of thermoplastics in polymers. The developed laser process is expected to increase productivity and minimize the cost for the final products.
