  • Argyreia capitata (Vahl) Choisy is a species of the genus Argyreia. Although many studies have analyzed the biological activity of A. nervosa, A. speciasa, and A. acuta, the anti-inflammatory effect of A. capitata extract (ACE) remains unclear. In this study, we evaluated the anti-inflammatory effect of ACE using lipopolysaccharide (LPS)-induced inflammatory markers in RAW 264.7 cells. We confirmed that the ACE inhibited the LPS-induced NO (nitric oxide) and iNOS (inducible nitric oxide synthase) expression in RAW 264.7 cells. ACE suppressed not only the LPS-induced phosphorylation of IKK, IκB, and p65 but also IL-1β expression. Collectively, these results suggest that ACE is a novel anti-inflammatory agent that suppresses iNOS expression, NO production, and the NF-κB signaling pathways.

Molecular Characterization of AceB, a Gene Encoding Malate Synthase in Corynebacterium glutamicum

  • The aceB gene, encoding for malate synthase, one of the key enzymes of glyoxylate bypass, was isolated from a pMT1-based Corynebacterium glutamicum gene library via complementation of an Escherichia coli aceB mutant on an acetate minimal medium. The aceB gene was closely linked to aceA, separated by 598 base pairs, and transcribed in divergent direction. The aceB expressed a protein product of Mr 83, 000 in Corynebacterium glutamicum which was unusually large compared with those of other malate synthases. A DNA-sequence analysis of the cloned DNA identified an open-reading frame of 2, 217 base pairs which encodes a protein with the molecular weight of 82, 311 comprising 739 aminoo acids. The putative protein product showed only limited amino acid-sequence homology to its counteliparts in other organisms. The N-terminal region of the protein, which shows no apparent homology with the known sequences of other malate synthases, appeared to be responsible for the protein s unusually large size. A potential calciumbinding domain of EF-hand structure found among eukaryotes was detected in the N-terminal region of the deduced protein.

  • This study was performed to determine the protein profiles using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and Angiotensin-I-converting enzyme(ACE) inhibitory activity (IC50) as affected by the various meat cuts, digestion times with pepsin. Hydrolysates having the protein concentration of 10 ug/mL had approximately 36∼39% ACE inhibitory activities, regardless of meat cut and digestion time. Protein concentration and ACE inhibitory activity of the diluted hydrolysate increased after 1-hr digestion. In original hydrolysates, ACE inhibitory activities of loin had higher than those of round (P<0.05). In addition, non-heated hydrolysates had higher ACE inhibitory activities than heated counterparts. When myosin B was digested by pepsin more than 1 hr, improved ACE inhibitory activities were observed as compared to the non-digested control.

Gene Amplification of aceA and aceB in Lysine-producing Corynebacterium glutamicum ssp. lactofermentum ATCC21799

  • The role of glyoxylate bypass in lysine production by Corynebacterium glutamicum ssp. lactofermentum ATCC21799 was analyzed by using cloned aceA and aceB genes which encode enzymes catalyzing the bypass. Introduction of a plasmid carrying aceA and aceB to the strain increased enzyme activities of the bypass to approximately 5 fold on acetate minimal medium. The strain with amplified glyoxylate bypass excreted 25% more lysine to the growth medium than the parental strain, apparently due to the increased availability of intracellular oxaloacetate. The final cell yield was lower in the strain with amplified glyoxylate bypass. These changes were specific to the lysine-producing C. glutamicum ssp. lactofermentum ATCC21799, since the lysine-nonproducing wild type Corynebacterium glutamicum strain grew faster and achieved higher cell yield when the glyoxylate bypass was amplified. These findings suggest that the lysine producing C. glutamicum ssp. lactofermentum ATCC21799 has the ability to efficiently channel oxaloacetate, the TCA cycle intermediate, to the lysine biosynthesis pathway whereas lysine-nonproducing strains do not. Our results show that amplification of the glyoxylate bypass efficiently increases the intracellular oxaloacetate in lysine producing Corynebacterium species and thus results in increased lysine production.

Characteristics of Angiotensin Converting Enzyme Inhibitory Peptides from Salt-fermented Squid Liver Sauce (오징어 간 액젓으로부터 분리된 Angiotensin Converting Enzyme 저해 Peptide의 특성)

  • In order to utilize squid liver by-products, which is normally discarded as industrial waste in the process of squid manufacturing, salt-fermented squid liver sauce was prepared experimentally and also tested for inhibitory activity against angiotensin converting enzyme (ACE). ACE inhibitory activity of squid liver sauce was increased with the elapse of fermentation days until 12 months, followed by a constant level of inhibitory activity thereafter. 15-month-old sauce ($IC_{50}=29.66\;{\mu}g$) was filtered through PM-10 membrane (M.W. cut-off 10,000 Da) to obtain the peptides fractions with ACE inhibition activity. Filtered fractions were applied to a Bio-gel P-2 column and three active fractions (A, B and C) were collected. Among them, fraction B applied to a SuperQ-Toyopearl 650S column chromatography lead to the isolation of active B-1 fraction. It has the ACE inhibitory activity ($IC_{50}=5.46\;{\mu}g$). The main composition of its amino acids is lysine, glycine and proline, which cover about 85% of the total amino acids.

In-silico Studies of Boerhavia diffusa (Purnarnava) Phytoconstituents as ACE II Inhibitor: Strategies to Combat COVID-19 and Associated Diseases

  • COVID-19 caused a catastrophe in human health. People infected with COVID-19 also suffer from various clinical illnesses during and after the infection. The Boerhavia diffusa plant is well known for its antihypertensive activity. ACE-II inhibitors and calcium channel blockers are reported as mechanisms for the antihypertensive activity of B. diffusa phytoconstituents. Various studies have said ACE-II is the virus's binding site to attack host cells. COVID-19 treatment commonly employs a variety of synthetic antiviral and steroidal drugs. As a result, other clinical illnesses, such as hypertension and hyperglycemia, emerge as serious complications. Safe and effective drug delivery is a prime objective of the drug development process. COVID-19 is treated with various herbal treatments; however, they are not widely used due to their low potency. Many herbal plants and formulations are used to treat COVID-19 infection, in which B. diffusa is the most widely used plant. The current study relies on discovering active phytoconstituents with ACE-II inhibitory activity in the B. diffusa plant. As a result, it can be used as a treatment option for patients with COVID-19 and related diseases. Different phytoconstituents of the B. diffusa plant were selected from the reported literature. The activity of phytoconstituents against ACE-II proteins has been studied. Molecular docking and ligand-protein interaction computation tools are used in the in-silico experiment. Physicochemical, drug-likeness, water solubility, lipophilicity, and pharmacokinetic parameters are used to evaluate phytoconstituents. Liriodenine has the best drug-likeness, bioactivity, and binding score characteristics among the selected ligands. The in-silico study aims to find the therapeutic potential of B. diffusa phytoconstituents against ACE-II. Targeting ACE-II also shows an effect against SARS-CoV-2. It can serve as a rationale for designing a drug for patient infected with COVID-19 and associated diseases.

한우에서 추출한 Myosin B의 Pepsin 가수분해물의 Ultrafilteration에 의한 Angiotensin Converting Enzyme(ACE) 저해 활성 분석

  • 한우의 Myosin B 단백질을 단백질 가수분해 효소인 pepsin으로 처리한 다음 단백질의 함량과 혈압상승 펩티드 생성효소인 angiotensin-I converting enzyme(ACE)에 대한 저해활성을 측정하였다. 등심이 우둔에 비해 단백질의 함량이 높았으며, 가수분해 처리 후 우둔과 다르게 등심은 3시간 가수분해 처리구에서 단백질의 함량이 높게 나타났다. ACE 저해활성은 등심에서는 3시간, 우둔에서는 6시간동안 가수분해시켰을 때 ACE 저해율이 유의적으로 가장 높게 나타났으며, 3, 6시간동안 가수분해시켰을 경우 부위 별로 유의적인 차가 있었으나(p<0.05), 0, 1시간동안 가수분해 시켰을 때는 부위간의 유의적인 차는 없었다(p>0.05). ACE 저해율이 가장 좋은 가수분해 처리구를 ultrafiltration시킨 결과, 저분자 peptide 상태의 가수분해물이 고분자에 비하여 ACE 저해율이 높은 것으로 나타났다. 차후 ACE 억제활성도가 높은 단백질을 분리하여 가장 우수한 분획을 찾아 아미노산 염기서열을 밝혀 고혈압 억제제로 합성 개발하는 연구를 추진할 예정이다.

Analysis of Angiotensin I Converting Enzyme Inhibitory Activity of Oligosacchride Extracted from Capsosiphon fulvescens (매생이 유래 올리고당의 추출 분리 및 Angiotensin I Converting Enzyme 저해능 분석)

  • The hydrolysates prepared with various enzyme digestion of Capsosiphon fulvescens were used to measure the inhibitory effects against angiotensin I converting enzyme (ACE). The commercially available enzymes such as Celluclast, Viscozyme, Lysing enzyme, Flavourzyme, Alcalase and Pectinex were used to digest C. fulvescens and produce hydrolysates. The maximum ACE inhibitory activity was observed using Alcalase hydrolysis (72.9%). The optimal conditions of Alcalase extraction were pH 8.0 and extraction time for 12 hr. The hydrolysates were fractionated using preparative-LC and anion-exchange chromatography on DEAE-cellulose and the fraction B and B-2 were isolated. The ACE inhibitory activity of fraction B-2 by anion-exchange chromatography was 82.6%. The molecular weight of fraction B-2 estimated using size exclusion chromatography was about 1 kDa. The monosaccharide composition of the fraction B-2 was determined to be mannose (1.1%), glucuronic acid (1.3%), galactose (1.3%) and glucose (96.3%).

ACE Inhibitory Materials from Raja kenojei (홍어의 항고혈압 활성물질)

  • This study was carried out to investigate the ACE inhibitory materials of Raja kenojei. Raja kenojei was sperated to fillet and viscera, and these were extracted with hot water. Antihypertensive activity was examined by mesearing angiotensin converting enzyme ACE inhibitory activity. ACE inhibitory activity of viscera at the concentration of 2% for Day 0 showed the highest value by 71.0%. But ACE inhibitory activity of fillet at 2% showed by 29%, which was lower antihypertensive activity than viscera. The protein content of viscerial hot water extracts in proximate composition showed the highest. And also, there was a large amount of aromatic and branched aliphatic amino acids in viscera than those in fillet. For the purification of antihypertensive material in visceral hot water extracts, it was separated and collected by Sephadex G-25 gel chromatography. The fraction (B) of 111 to 160 showed the highest ACE inhibitory activity by 65.1% at the concentration of 0.05%. But the other fractions (A and C) showed lower activity than B. These results demonstrate that crude hot water extracts of viscera from Raj kenojei may be useful as functional food ingredient with antihypertensive property.

  • 본 연구는 고혈압을 억제하는 ACE inhibitory peptide가 한우에 함유되어 있는지를 조사하기 위한 기초연구로 한우의 우둔과 등심으로부터 Myosin B를 추출하여 가열한 것과 가열하지 않은 것으로 구분하여 pepsin으로 0, 1, 3, 6시간 동안 $37^{\circ}C$에서 가수분해 시켰다. 가수분해물을 전기영동을 실시하여 pepsin 처리시간에 따른 단백질의 변화를 검토하였고, ACE 저해 활성을 측정하였다. 전기영동의 결과 가열한 것은 가열하지 않은 것에 비해서 단백질의 변성과 소멸이 많이 일어나 밴드의 수가 적었다. Pepsin으로 가수분해한 시간이 길어짐에 일부 단백질이 소실되었고, 저분자의 단백질이 생성되었다. Pepsin으로 가수분해함에 따라 등심과 우둔에서 25, 32, 40 및 44 kDa는 소실되었고, 37 kDa의 분자량을 갖는 밴드는 생성되었으며, 27 kDa의 단백질은 처음상태 그대로 유지되었다. 가수분해물을 이용하여 ACE 저해활성도를 측정한 결과, 등심은 1시간 가수분해시 유의차가 있었으나, 우둔은 가열여부에 따라 유의차가 발견되지 않았다. 반면 등심과 우둔 모두 가수분해 시간이 증가하면 ACE 저해율은 상승하는 경향을 보였다. 이와 같은 결과를 토대로 한우로부터 추출한 Myosin B의 ACE 억제활성이 우수한 단백질분획을 찾아 아미노산 염기서열을 밝히고 고혈압 억제제로 합성 개발하는 연구를 차후 추진할 예정이다.
