잡초성벼의 superoxide dismutase cDNA cloning과 재배벼로의 형질전환 (Isolation of Superoxide Dismutase cDNAS from an Weedy Rice Variety and Transformation of a Cultivated Rice Variety)

  • 냉해나 한발등의 환경스트레스에 대해 저항성을 유발하는 유전자를 환경스트레스에 강한 잡초성벼로부터 선발하고 이들 유전자를 재배벼에 도입하여 도입유전자 산물의 과량 발현을 통해 냉해나 한발 등에 대한 저항성이 향상된 벼를 선발하고자 하였다. 잡초성벼인 Bhutan 14Ad로부터 냉해 및 한발 저항성 유전자로 알려진 superoxide dismutase (SOD) cDNA를 분리하고자 mRNA를 분리하고 이 분리된 mRNA를 이용해 reverse transcriptase PCR방법으로 SOD cDNA를 cloning 하였다. 그 결과 2종의 SOD cDNA가 cloning되어 SOD-A, SOD-B로 명명하였다. 이들 cDNA의 염기서열을 결정한 결과 이들은 아미노산 서열 상동성이 88.4%를 나타내었으며, SOD-A는 Oryza sativa 계열의 Cu/Zn SOD유전자인 GenBank accession No. L36320와 99.3% 동일하였으며, SOD-B는 accession No. D01000과 100% 동일하였다. 이들 SOD-A와 SOD-B cDNA를 재배벼인 낙동벼에 형질전환하여 형질전환체 벼를 선발하였으며, 이들 형질전환체 벼의 냉해저항성및 한발저항성 검정을 통해 저항성이 향상된 형질전환체 벼를 선발하고 있다.

표준기록관리시스템의 활용현황 연구 (A Survey on Usage of Korean Standard Records Management System)

  • 이 연구는 표준기록관리시스템의 활용현황을 확인하고자 하는 목적으로 수행되었다. 인수, 보존, 평가, 이관, 기준관리, 접근과 감사추적, 검색활용, 공개관리 등 9개 대기능에 속한 240개 세부기능별 활용정도를 묻는 설문도구를 구성하였다. 중앙부처나 광역교육청, 그리고 광역자치단체는 전수를 대상으로 하였고, 기초자치단체는 두 곳의 광역단체에 속한 기초단체 전체를 대상으로 설문지를 배포하였다. 총 101개 기관 중 65곳이 응답하여 최종 응답률은 63.73%였다. 그동안 표준RMS에 대해서는 '불편해서 쓸 수가 없다'는 생산기관 전문요원의 입장과 '잘 쓴다고 하는 곳도 꽤 있다'는 기록원의 입장이 대립해 왔다. 처음으로 실증적인 데이터를 기반으로 어떤 기능이 어느 정도 활용되는지를 확인한 이 연구를 기점으로 해서, 이후로는 표준RMS의 개선을 위한 생산적인 논의가 펼쳐지기를 기대한다.

  • Investigated kinds and utilization frequency for traditional food and Particular food to systematize Gwangju city and Chollanamdo area traditional food culture. Awareness for traditional food had pride because taste is various, and did that must use much when was festive day. Investigation subject were feeling necessity about accession, development of traditional food, and manufacture of traditional food, succession did that must take charge in home. By utilization rate of food-tasting food subdivisions of the season, used much by rice-cake soup, fermented rice punch, order of cake made from glutinous rice New Year's Day, by herbs, Gimgui, Ogokbap's order January Full Moon Day on the lunar calendar. By a cake made in the shape of a flower, azalea honeyed juice mixed with fruits as a punch order to Samjitnal, by beans panbroiling, dropwort raw order Buddha's Birthday, Tano Festival uses Charyunbyeong and used Tteoksudan in Yudu and used much by young chicken soup with ginseng and other fruits, watermelon order period of midsummer heat. Used songpyon and fermented rice punch in Full-moon Harvest Day, and Junggujeol used Chrysanthemum griddle cakes made in flower Pattern, and red-bean gruel taken on the winter solstice, and Nappyeongjeolsik was utilizing Goldongban. Pyebaek food utilization ratio was high the utilization rate by chestnut, jujube, chicken, wine order. The contributiveness food utilization rate was high the utilization rate by rice cake, dried croaker, fruit, oil-and-honey pastry order. The large table food utilization rate was high the utilization rate by fermented rice punch, fruit, steamed short-ribs order.

중국(中國)의 WTO가맹(加盟)에 의한 일본기업(日本企業)의 대중투자(對中投資)에 관한 연구(硏究) (Japanese Companies Investment in China After Chinas Entry into WTO)

  • Since China was admitted to the World Trade Organization (WTO) on December 11, 2001, China has sped up its active participation in the competition of the world economy by lifting its trade and investment restrictions and limits. Its trading system is getting more and more free and its huge market with a population of 1.3 billion has been getting more and more open. With Chinas WTO accession, China has been honoring its commitments to its WTO members by complying with international business practices and WTO rules. For example, it will cut down its import tariffs for automobiles from the present 80% to 25% by 2006. Its financial sector and service industries are open to foreign investors now. As Chinas biggest business partner, Japan has benefited tremendously from Chinas open policies and deepening economic reform. In order to reduce the costs of production, a large number of enterprises from Japan have set up production bases in China since a Chinese workers wages accounts for only one tenth of those of a Japanese worker on the average. Japan has become one of Chinas biggest investors on a par with the United States of America (USA). How are the Japanese enterprises in Japan and in China adjusting themselves to the rapid changes of the Chinese market? What characteristics do the Japanese enterprises in China have? What effects has Chinas WTO accession had on those Japanese enterprises that have invested in China? This paper finds answers to the above questions. It at first reviews the Sino-Japanese economic and trade relations since 1980s, then analyses the situation and characteristics of Japanese-invested enterprises in China, and explores their problems in China. Finally it puts forward some suggestions for the Japanese enterprises in China.

16S rRNA를 이용한 다금바리, 자바리, 능성어 판별법 개발 (Development of Detection Method for Niphon spinosus, Epinephelus bruneus, and Epinephelus septemfasciatus using 16S rRNA Gene)

  • 다금바리, 자바리 및 능성어는 농어목(Perciformes Order) 바리과(Serranidae Family)에 속하며, 고급횟감으로 인기가 높은 어종이다. 자바리 및 능성어의 경우 몸통부분에 줄무늬가 선명하게 있으나 성어가 되면서 점차 불분명해지는 특징이 있어 무늬의 유무로 인하여 다금바리와 구별하기는 쉽지 않다. 또한 자바리는 제주도에서 다금바리라는 명칭으로 불리고 있으며, 일부 유통업자들이 자바리 및 능성어를 다금바리로 유통시키는 사례가 있어 종 특이 프라이머를 이용한 판별법을 마련하게 되었다. 종 특이 프라이머를 설계하기 위하여 유전자은행(www.ncbi.nlm.nih.gov)에 등록되어있는 다금바리(Accession No. AY947575), 자바리(Accession No. JN603832), 능성어(Accession No. AY947559), 우럭(Accession No. DQ678295) 등의 16S DNA부위의 염기서열을 대상으로 하였으며 비교 및 분석에는 소프트웨어인 BioEdit ver. 7.0.9.0 프로그램을 사용하였다. 그 결과 다금바리, 자바리, 능성어를 판별할 수 있는 각각의 NS-003-F/NS-005-R(136 bp), EB-001-F/EB-002-R(181 bp), ES-001-F/ES-001-R(123 bp) 프라이머를 개발하였으며, PCR 조건을 확립하였다. 또한 적용성 검토를 위하여 우럭, 참돔을 대상으로 실시한 결과 비 특이적 밴드가 형성되지 않는 것을 확인하였다. 따라서 본 연구에서 개발된 다금바리 등에 대한 판별법은 시중에 불법적으로 유통 가능성이 있는 제품을 신속하고 과학적으로 판별할 수 있어 식품안전관리에 활용도가 매우 클 것으로 기대된다.

국가도서관과 지역대표도서관의 도서기호 현황 분석 및 개선방안에 관한 연구 (A Study on the Use of Book Numbers in the National Libraries and Regional Central Libraries in Korea)

  • 본 연구는 우리나라 2개 국가도서관과 9개 지역대표도서관, 서울시립정독도서관을 대상으로 담당 사서들의 도서기호에 대한 인식 및 문제점을 조사하고 각 도서관에서 사용하고 있는 도서기호의 현황을 분석하여 도서기호에 대한 개선방안을 제시하였다. 도서기호의 가장 큰 문제점은 도서기호의 중복으로 인해 길어지는 도서기호의 길이로 나타났으며 도서기호 현황 분석을 통해 도서기호의 중복과 함께 직관성 부족이라는 문제점을 도출하였다. 이에 본 연구는 이용자 친화적이면서 간단하게 부여할 수 있는 저자명순과 연대순 방식을 조합한 새로운 도서기호법의 구성 요소를 제안하였다.

새 연대순 도서기호법의 연구 (A Study of New Chronological Book Numbers)

  • In most of the modern libraries throughout the world, it is popular to adopt the author number which arranges the books or entries in alphabetical order of their author's names within the same ultimate class. However, viewed historically, that was the Western practice never used in the East before. Traditionally chronological order was followed in the East. Book number not only individualizes the books within the same class but also makes it easy for the user to choose and find out the relevant materials. In this respect, chronological book number is decidedly superior to all kinds of book number systems ever have been existed. Especially in these days of rapid obsolescence of documents and with serious problems of storage of documents, the chronological order seems to be the most modern and future-oriented of all other book number systems because it distinguishes clearly new materials from aged materials by the date of publication and controls the stack spaces mechanically and effectively. This writer devised New Chronological Book Numbers adaptable to both the Eastern and Western materials, and the system has been adopted at Yonsei University Library. The features of the system are as follows: (1) It is easy to type the call number in cataloging the Eastern and Western materials. That is because the structure of the chronological number is pure numerical notation by taking the last two figures of the year within twentieth century(e.g. 85 for AD 1985), that is 1900s, and the last three figures of the year from twenty-first century to thirtieth century, that is 2000s. (2) It does not make all classes have chronological number uniformly. Within the classes for individual biographies, genealogies and collections of biography of specific family, books about individual organization, individual literary and artistic works, philosophical works of the famous philosophers, books and its facsmile editions published before 1900, new editions, translations, commentaries, serials, chronological number is assigned to the author number as in the past. (3) It separates the Eastern materials from the Western materials and makes the typing easy in cataloging. That is because the subarrangement within the same chronological book number is made by the accession order which uses 'panjol-ponmun' or the characters of basic syllabic table of Korean alphabet in case of the Eastern materials and Roman alphabet in case of the Western materials.

애기장대 cDNA library로부터 Glutamate Decarboxylase 유전자의 부분 클로닝 및 서열분석 (Cloning and Nucleotide Sequencing of a Partial Glutamate Decarboxylase Gene from Arabidopsis thaliana cDNA Library)

  • In order to study the molecular mechanism of $\gamma$-aminobutyric acid (GABA) production in plants, we cloned and sequenced a partial glutamate decarboxylase (GAD) cDNA from the Arabidopsis thaliana cDNA library, using primers targeted at highly conserved sequences of the petunia GAD gene. The cDNA fragment was inserted into TA cloning vector with T7 promoter and the recombinant plasmid obtained was used to transform E. coli. The plasmid DNA purified from the transformed E. coli was digested with EcoRI and the presence of the insert was confirmed. Nucleotide sequence analysis showed that the fragment is a partial Arabidopsis thaliana GAD gene and that the sequence showed 98% and 78% identity to the region of the putative Arabidopsis thaliana GAD sequences deposited in GenBank, Accession nos: U46665 and U10034, respectively. The amino acid sequence deduced from the partial Arabidopsis thaliana GAD gene showed 99% and 91% identities to the GAD sequences deduced from the genes of the U46665 and U10034, respectively. The partial cDNA sequence determined may facilitate the study of the molecular mechanism of GABA metabolism in plants.

Identification and characterization of a rice blast fungal elicitor-inducible Oshin1 gene

  • In order to understand the molecular interactions that occur between rice and the rice blast fungus during infection, we previously identified a number of rice blast fungal elicitor-responsive genes from rice (Oryza sativa cv. Milyang 117). Here, we report the cloning and characterization of the rice fungal elicitor-inducible gene Oshin1 (GenBank Accession Number AF039532). Sequence analysis revealed that the Oshin1 cDNA is 1067 bp long and contains an open reading frame encoding 205 amino acid residues. The Oshin1 gene shows considerable sequence similarity to the tobacco hin1 and hin2 genes. The predicted Oshin1 protein has a cysteine-rich domain at the N-terminus and is rich in leucine, serine, and alanine residues. Southern blot analysis suggests that Oshin1 gene is a member of a small gene family in the rice genome. To examine the expression of Oshin1, Northern blot analysis was conducted. Expression of the Oshin1 transcript is rapidly induced in suspension-cultured rice cells treated with fungal elicitor, salicylic acid or hydrogen peroxide. In addition, Oshin1 transcript levels are rapidly increased by treatment with $Ca^{2+}$/A23187. The expression of Oshin1 was also elevated in 3-week old leaf tissues upon ethephon application or fungal elicitor treatment. Our results suggest that the Oshin1 gene is involved in plant defense responses to environmental stresses.

Human Ribosomal Protein L18a Interacts with hnRNP E1

  • Heterogeneous nuclear ribonucleoprotein E1(hnRNP E1) is one of the primary pre-mRNA binding proteins in human cells. It consists of 356 amino acid residues and harbors three hnRNP K homology(KH) domains that mediate RNA-binding. The hnRNP E1 protein was shown to play important roles in mRNA stabilization and translational control. In order to enhance our understanding of the cellular functions of hnRNP E1, we searched for interacting proteins through a yeast two-hybrid screening while using HeLa cDNA library as target. One of the cDNA clones was found to be human ribosomal protein L18a cDNA(GenBank accession number BC071920). We demonstrated in this study that human ribosomal protein L18a, a constituent of ribosomal protein large subunit, interacts specifically with hnRNP E1 in the yeast two-hybrid system. Such an interaction was observed for the first time in this study, and was also verified by biochemical assay.
