대두 사포닌이 Aflatoxin B1으로 유도된 세포돌연변이에 미치는 영향에 관한 연구 (Effect of Soybean Saponins on Aflatoxin B1-induced Mutagenicity)

  • Free radicals formed during the metabolism of environmental chemicals are known to induce mutagenicity, while different types of antioxidants suppress this event. The purpose of this study was to determine the antioxidative and antimutagenic effects of soybean saponins, and to examine the relationship between these two effects for the elucidation of mechanisms involved in the anticarcinogenicity of soybean saponins. Also, antioxidative and antimutagenic effects of soybean saponins were compared with those of kinown antioxidants. For the measurement of antioxidative capacity, soybean saponins, L-ascorbic acid, $\alpha$-tocophoerol, and BHT at concentrations between 005 and 1.0mg/ml were tested for their ability to donate hydrogens and to reduce the formation of thiobarbituric substances(TBARS). Antimutagenic activity was examined using the Ames salmonella test system at concentrations of 600, 900 or 1200ug/ml. Study results showed soybean saponins and all of the other antioxidants tested possessed dose-dependent antioxidative activities. The ability of hydrogen-donation to DPPH was in the order of L-ascorbic acid>$\alpha$-tocopherol=>BHT>soybean saponins. TBARS formation was also inhibited by these compounds, in the order of BHT>$\alpha$-tocopherol=L-ascorbic acid>soybean saponins. Soybean saponins and other antioxidants also showed antimutagenicity in a dose-dependent manner. Especially, soybean saponins and BHT were excellent antioxidants compounds, inhibiting near 80% of the mutagenic effects at a concentration of 1200ug/ml. The correlation coefficients between antioxidative capacity and antimutagenicity for each compund was statistically significant at p<0.05. These results indicate that soybean saponins possess antioxidative and antimutagenic capacities. Also, antimutagenicity of saponins and other antioxidats is partly due to their antioxidative activities.

  • 예로부터 식품으로 사용되어온 산돌배나무 (Pyrus ussuriensis) 열매를 대상으로 항산화 활성 성분을 찾기 위하여 그 MeOH추출물의 EtOAc분획을 대상으로 column chromatography를 실시하여 1, 4-dibenzenediol, chlorogenic acid 및 quercitrin등 세종의 화합물을 분리하고 DPPH free radical소거법을 이용하여 항산화활성을 측정하였다. 이중 quercitrin은 a-tocopherol 및 BHA보다 낮은 활성 $(RC_{50}=20{\mu}g)$을 보였으나 1, 4-dibenzenediol $(RC_{50}=0.4{\mu}g)$ 와 chlorogenic acid $(RC_{50}=4{\mu}g)$는 강한 활성을 보였다.

Taurine Possesses In vitro Antimutagenic Activity Comparable to Major Antioxidants

  • Taurine is known to suppress oxidant-induced tissue injury by stabilizing biomembrane and scavanging free radicals. The purpose of this study was to determne the antioxidative and antimutabenic acitvities of taurine, ad to compare those acitivities with major antioxidants. For the measurement of antioxidative capacity, 0.05 , 0.1,0.5 and 1.0mg/ml of taurine, L-Ascorbic acid, alpha-tocopherol, and BHT (dibuty hydroxiy toluene)were prepared and tested for their ability to donate electrons to DPPH (1,1-diphenyl-2-picryl-hydrazyl). Antimutagenic acitivity was examined using the Ames salmonela test system at concentrations of 600, 900 and 1200ug/ml. Results indicated that taurine possesses electron-donating capacity, however, the degree of donation was very weak compared to the major antioxidants tested. However, taurine was evaluated as a potent mutation suppressor. Antimutagenic capacity was in increasing order BHT>taurine>L-ascorbic acid>alpha-tocopherol at concentrations of 600 and 900ug/ml. There was a dose-dependent increase in antimutabenicity of these compounds , however, antimutagenity of the 900ug taurie/plate was not significantly differently from that of 1200ug taurine/plate. These results indicate that taurine effectively suppresses the mutagenicity of AFB1 without noticeable elelctron donating ability.

밤나무 잎으로부터 항산화 활성물질의 분리 (Antioxidative Compounds in Leaves of Castanea crenata S. et Z.)

  • 밤나무(Castanea crenat)의 지상부를 대상으로 DPPH free radical 소거법을 이용하여 2종의 항산화 활성물질을 분리하였다. 분리된 활성물질은 NMR과 mass 분석에 의하여 quercitrin, isoquercitrin으로 밝혀졌다. 두 화합물의 DPPH free radical 소거법에 의한 항산화 활성 $(RC_{50}\;:\;12{\mu}g)$은 BHA $(14\;{\mu}g)$${\alpha}-tocopherol\;(12{\mu}g)$과 비슷하였다.

녹차, 오룡차 및 홍차 추출물의 항산화효과 (Antioxidative Effect of Tea Extracts from Green Tea, Oolong Tea and Black Tea)

  • Antioxidative effect of tea extracts from green tea(steamed, roasted), oolong tea and black tea was investigated. Antioxidative activities of the crude catechin fraction were the most effective in oolong tea which contained the highest level of (-)-epigallocatechin gallate(EGCg). The water-soluble fraction obtained from oolong tea exhibited binding with more ferrous than copper ions. It showed a synergistic effect when used with an antioxidant such as BHT(0.5mg) and $\alpha$-tocopherol(2mg). Especially, this synergistic effect was exhibited more with BHT than with $\alpha$-tocopherol. Also steamed green tea, roasted green tea and oolong tea showed remarkable free radical scavenging action. The SC50(the concentration of a water-soluble fraction which is required to scavenge 50% of 100$\mu$M 1, 1-diphenyl-2-picryl hydrazyl radicals) of steamed and roasted green tea, oolong tea and black tea extracts were 11.3$\mu\textrm{g}$/ml, 11.4$\mu\textrm{g}$/ml, 12.7 $\mu\textrm{g}$/ml and 14.9$\mu\textrm{g}$/ml, respectively. It is assumed that the antioxidative activity of tea extracts is due to inhibition of peroxidation free radical scavenging and binding action of ferrous ions by mainly tea polyphenol compounds.

Effects of Multivitamin-Mineral Supplementation, at Nutritional Doses, on Plasma Antioxidant Status, Erythrocyte Antioxidant Enzyme Activities and Lipid Peroxidation during Pregnancy

  • The aim of this study was to evalute the effect of multivitamin-mineral supplementation during pregnancy on plasma levels of antioxidants, erythrocyte antioxidant enzyme activities, and lipid peroxidation. A controlled, semi-randomized, prospective trial was performed by comparing the supplement group, which received multivitamin-mineral tables once daily for 10 weeks, with the control group. Plasma levels of $\beta$-carotene, tocopherol, coenzyme Q10, ascorbate, folate, zinc, and selenium and malondialdehyde (MDA), as well as the activities of superocxide dismutase(SOD) and glutathione peroxidase(GSH-Px) in erythrocytes were measured initially (20 wk gestation) and at the end of the intervention (34 wk gestation). In the control group, plasma ascorbate and selenium levels decreased and tocopherol levels increased. In the supplement group, a significant increase in plasma $\beta$-carotene(46%), conenzyme Q10 (42%), and zinc (24%) was observed after 10 weeks of supplementation. No changes were observed in the plasma levels of MDA, and erythrocyte GSH-Px activity, while SOD activity increased in both control group and the supplement group during the intervention. These data suggest that multivitamin-mineral supplementation during pregnancy produced moderate increases in plasma $\beta$-carotens, coenzyme Q10, and zinc concentrations but the enhancement of those plasma antioxidants had on direct on the plasma level of MDA, erythrocytes SOD or GSH-Px activities.

Protective Effects of $\alpha$-Tocopherol and Ischemic Preconditioning on Hepatic Reperfusion Injury

  • This study evaluated the effect of $\alpha$-tocopherol ($\alpha$-TC), ischemic preconditioning (IPC) or a combination on the extent of mitochondrial injury caused by hepatic ischemia/reperfusion (I/R). Rats were pretreated with $\alpha$-TC (20 mg/kg per day, i.p.) for 3 days before sustained ischemia. A rat liver was preconditioned with 10 min of ischemia and 10 min of reperfusion, and was then subjected to 90 min of ischemia followed by 5 h or 24 h of reperfusion. I/R increased the aminotransferase activity and mitochondrial lipid peroxidation, whereas it decreased the mitochondrial glutamate dehydrogenase activity. $\alpha$-TC and IPC individually attenuated these changes. $\alpha$-TC combined with IPC ($\alpha$-TC+IPC) did not further attenuate the changes. The mitochondrial glutathione content decreased after 5 h reperfusion. This decrease was attenuated by $\alpha$-TC, IPC, and $\alpha$-TC+IPC. The significant production of peroxides observed after 10 min reperfusion subsequent to sustained ischemia was attenuated by $\alpha$-TC, IPC, and $\alpha$-TC+IPC. The mitochondria isolated after I/R were rapidly swollen. However, this swelling rate was reduced by $\alpha$­TC, IPC, and $\alpha$-TC+IPC. These results suggest that either $\alpha$-TC or IPC reduces the level of mitochondrial damage associated with oxidative stress caused by hepatic I/R, but $\alpha$- TC combined with IPC offers no significant additional protection.

  • Acornic powder was extracted with methanol and ethylacetate to obtain a crude acornic compounds. And they were examined concerning their antioxidant activities for linoleic acid. The results were as follows: 1. Hydrogen donating activity for DPPH was higher in 100ppm acornic compound than 100ppm BHT and 100ppm ${\alpha}$-tocopherol. 2. When the 100ppm acornic compound was added to linoleic acid, which was heated at $50^{\circ}C$ for 48 hours, antioxidant activities by POV and TBA was higher than that of 100ppm ${\alpha}$-tocopherol, but the effect was almost the same as the 100ppm BHT. 3. Antioxiodant activity of acornic compound showed synergistic effect along with malic acid, citric acid, tartaric acid, alanine, arginine, histidine, lysine-HCl, galactose, maltose, glucose and sucrose. 4. Acornic compound inhibited peroxidation of linoleic acid induced by heavy metals.

들깨의 발아가 들깨지방질의 특성에 미치는 영향 (Effects of Seed Germination on Characteristics of Perilla Seed Lipids)

  • Color, lipid and fatty acid composition, and tocopherols and polyphenols contents of perilla seed lipids in response to seed germination were studied. Perilla seeds were germinated at $30^{\circ}C$ in the dark for 12, 36, or 48 h, after which total lipids were extracted by the Folch method using chloroform and methanol (2:1, v/v). Seed germination resulted in a decrease in yellowness and greenness in perilla seed lipids, but there were no significant changes in composition of the lipids including major neutral lipids (>90%). Contents of phosphatidylcholine and phosphatidylethanolamine in the perilla seed lipids significantly increased in response to germination. Linolenic acid (>63%) was the most abundant fatty acid. Seed germination tended to decrease the relative content of linolenic acid and increase the contents of oleic and stearic acids. Contents of antioxidants, especially ${\alpha}$-tocopherol and polyphenols, increased in response to seed germination. As the germination period was extended, the antioxidant content increased. Therefore, increases in useful components, phosphatidylcholine, phosphatidylethanolamine, ${\alpha}$-tocopherol, and polyphenols contents by seed germination can contribute to the improvement of perilla seed utilization in food industry.

Antioxidative and Radical Scavenging Properties of Extracts from Geum japonicum

  • Extracts from Geum japonicum (Bammu in Korean) were analyzed for their antioxidative activities and scavenging effects on free radicals. The ethyl acetate fraction of G. japonicum methanol extract (EFGJ) showed a remarkable scavenging activity on the 1,1-diphenyl-2-picrylhydrazyl radical. EFGJ also showed excellent antioxidative activity on linoleic acid during long-period storage and on rat liver microsome peroxidation system, and good anti-peroxidation effect on lipid in Rancimat system using lard, palm oil, and perilla oil, as compared with BHT and ${\alpha}$-tocopherol. Varying effects of antioxidative activity of the EFGJ on various fatty acids were observed. The prevention of linoleic acid and linolenic acid peroxidation was superior to ${\alpha}$-tocopherol, but inferior to BHT. The methanol extract of G. japonicum did not show mutagenicity as revealed through SOS chromotest. Based on the results, the extracts of G. japonicum may be useful source as natural antioxidants.
