New Data on the Synthetic $Pt_{34}Sb_7$ and $Pt_3Sb$ Phases (합성화합물 $Pt_{34}Sb_7$$Pt_3Sb$에 대한 새로운 자료)

  • In the process of re-checking the phase relations of the Pt-Sb system the previously reported Pt4+Sb phase did not occur. Instead, Pt34Sb7 phase with an average chemical composition (wt%) Pt 89.3, Sb 11.4, total 100.7, has been newly found to exist, and the formula Pt34Sb7 has been tentatively assigned to it, It is pale brownish grey to yellowish grey under refloecting microscope and non-bireflectant, VHN100=225 (206-243). X-ray powder patterns are (d(I/Io)): 2.318(100), 2.293(100), 2.110(30), 1.440(50), 1.390(15), 1.283(5), 1.203(70) and 1.192(40)$\AA$. They are indexable on the basis of tertragonal cell with $\alpha$=3.948(3), c=16.85(1)$\AA$. A question whether the tetragonal Pt34Sb7 is the new phase or a polymorph of the cubic Pt4+Sb phase remains unclear and awaits better X-ray diffraction, electron microprobe analysis and DTA for the Pt4+Sb phase. The Pt3Sb phase reported to be of tetragonal symmetry has been confirmed to exist. It is greyish yellow in air and in oil, very weakly bireflectant, and weakly anisotropic. VHN25=216 (183-240). Its X-ray powder data have been successively indexed on a tetragonal cell with $\alpha$=3.9455(7), c=16.959(5)$\AA$.

  • In this presented study, we established a method for diagnosis of porcine epidemic diarrhea(PED) by in situ hybridization(ISH), which made distinct progress in diagnostic pathology. We also carried out the retrospective diagnosis through ISH to assume the exact time of the first outbreak and incidence of PED in Korea. The outbreak of PED in Korea reported in 1992. However, since the end of 1980's, some researches of pig-industry have already suspected the outbreak of PED, not transmissible gastroenteritis(TGE). In this experiment, we performed the ISH using 80 formalin-fixed and paraffin-embedded tissues of porcine intestine which were requests for pathological diagnosis from 63 farms whose primary sign was diarrhea from 1984 to 1991. We prepared biotinylated cDNA probe(492base pairs) for ISH by nick translation method and carried out the ISH, using $Microprobe^{TM}$ capillary action system(Fisher $Biotech^R$). We detected PED virus in intestinal mucosa of 2 cases in 1992, 1 case in 1988, and 1 case in 1987. As a result, we assume that the outbreak of PED in Korea have already started since 1987.

Phases and phase Equilibria of the Pt-Sb System (백금-안티모니 계의 식평구 및 화합흉에 대한 연구)

  • The phase diagram of the Pt-Sb system was reinvestigated, using the conventional sealedcapsule technique. The identification of phases present in the reaction products was made by reflected light microscopy, X-ray diffraction and electron microprobe analysis. The existence of compounds, Pt5Sb, Pt3Sb, Pt3Sb2, PtSb and PtSb5 was confirmed. A new phase, Pt5Sb with a composition of 83at% Pt and tetragonal structure of the lattice parameters a=3.948(3), c=16.85(1)A, was found. The X-ray powder data of Ptsb may be indexable on a tetragonal cell with a=3.9455(7), c=16.959(5)A. PtSb is stoichiometric up to 800t and becomes Pt-deficient as much as lat% at 1000t. Solid solubility limits of Sb in Pt were determined to be 7.5,10.0 and 6.1at% at 1000˚,800˚ and 600˚ , respectively. The earlier reported Pts Sbf was not found in this study. The liquidus curve between the Ptsb2 and Sb phases was revised.

Serpentinization of the Ultramafic Rock in the Yesan-Gongju-Cheongyang Area, Korea (충남 예산-공주-청양지역의 초염기성암의 사문암화 작용)

  • Serpentinite of the Yesan-Gongju-Cheongyang area has been formed by serpentinization of ultramafic rocks. The ultramafic rock might be composed mainly of oilvine with minor pyroxene and amphibole. Olivine has a considerably restricted chemical compositional ranging from Fo90 to Fo93. Fresh serpentinite containing large amount of oilvine is usually massive in occurrence and dark green to black in color. Serpentine minerals occur not only as major mineral of serpentinite, but also as remnants in the talc ore which was formed from serpentinite. XRD study indicates that antigorie is the most abundant serpentine mineral of the serpentinite. Serpentinite consisting of antigorite usually shows non-pseudomorphic texture, whereas that consisting of lizardite shows pseudomorphic texture. Antigorite is found along the margins or fractures of olivine grains resulting in the formation of network of magnetite which was formed at the time of serpentinization. Lizardite, subordinate constituent mineral of serpentinite, frequently shows pseudomorphic mesh-texture after olivine. The chemical differences between antigorite and lizardite/chrysotile are small, so both minerals are not easily discernible with the electron microprobe. Antigorite occuers as elongate blades, flakes, or plates forming interpenetrating texture to obliterate previous textures. SEM study also shows that most serpentine minerals occur in platy or tabular form rather than in asbestiform. Fractures formed after main serpentinization are observed within the pseudomorphic central olivine grain. Careful observation of the serpentine pseudomorphs gives a great deal of data on the pre-serpentinization nature of the serpentine pseudomorphs gives a great deal of data on the pre-serpentinization nature of the ultramafic rocks. It is inferred that the serpentinization took place after the emplacement of ultramafic body into the relatively wet environment ceased and the cooling intrusive body crossed into the stability field of serpentine. It is inferred that the final pervasive serpentinization took place over a long time, by hydrothermal water supplied through the fracture system produced during emplacement of ultramafic rock.

Detection of bovine viral diarrhea virus by In situ hybridization (In situ hybridization에 의한 소 바이러스성 설사증 바이러스의 검출)

  • Detection and distribution of bovine viral diarrhea virus(BVDV) was studied in formalin-fixed, paraffin-embedded tissues from two naturally infected cattle by in situ hybridization with a non-radioactive biotinylated probe. A 600 base pair cDNA probe from BVDV B-25 strain was used for probe. The whole procedure of ISH to diagnose was carried out within 1~2 hours in $Microprobe^{TM}$ capillary action system. The biotin-labelled probe was demonstrated after hybridization under standard conditions by the application of streptoavidin and biotinylated alkaline phosphatase. Alkaline phosphatase was visualized using a fast red TR/naphthol phosphatase and the sections were counterstained with hematoxylin. We have obtained the result of positive reactions in digestive tract(sm1.all intestine and colon) and epidermis of tongue in the state of the intact tissues. The result suggested that in situ hybridization method can be considered as a useful diagnostic technique for detection of specific nucleic acid sequences of BVDV.

  • In situ hybridization was performed to detect rat heumocwstis ca4nii in the lung sections. Rats were immunosuppressed by weekly subcutaneous injection of 10 mg/kg methylprednisolone. On the 6th, 8th and 9th week of immunosuppression, the lungs were removed and fled in 10% neutral formalin. A 22 base oligonucleotide probe complementary to p. carinii 5S ribosomal RMh was commercially synhesized and its 3' terminal was labeled wiH biotin. In situ hybridization was performed utilizing manual capillary action technolog)r on the Microprobe system. p. cnrinii were detected along the luminal surface of alveolar pneumocytes, in exudate of alveolar cavities, and also in secretory material of bronchioles. In the 6th week group, positive reaction was observed focally in the peripheral region of the lung sections, but the reaction was observed diffusely in the 8th or 9th week groups. In comparison with Grocott's methenamine silver stain, in situ hybridization technique can detect the organism rapidly, and can detect trophic forms very well. Furthermore, no nonspecific reaction with other pathogenic fungi and protozoa was recognized. Therefore, in situ hybridization can be a good technique to detect p. carinii in the lungs of infected rats.

  • Background : A presumptive histopathologic diagnosis of tuberculosis is commonly based on the finding of acid-fast bacilli upon microscopic examination of a diagnostic specimens. Although this traditional histochemical staining method is satisfactory, it is time-consuming and not species-specific. For more specific assessment, in situ hybridization assay with oligonucleotide probes is introduced. Methods : The human surgical specimens were obtained from tuberculosis patients, and experimental specimens were made by injecting cultured M. tuberculosis organisms into fresh rat liver. Oligonucleotide probes complementary to ribosomal RNA portion were synthesized and labeled with multiple biotin molecules. For a rapid detection, all procedures were carried out using manual capillary action technology on the Microprobe staining system. Results : The in situ hybridization assay produced a positive reaction in experimental specimens (80-90% sensitivity) after pepsin-HCl pre-treatment for a good permeabilization of probes, but reliable result was not obtained from human surgical specimens. Conclusion : It is, therefore, suggested that biotin-labeled oligonucleotide probes have considerable potential for identification and in situ detection of M. tuberculosis but, there are some barriers to overcome for the diagnostic use of this method.

SHRIMP Zircon U-Pb Ages of Basement Rocks in the Danyang National Geopark (단양 국가지질공원 기반암류의 SHRIMP 저어콘 U-Pb 연령)

  • We carried out the U-Pb age dating of zircon from basement rocks in the southern part of the Danyang National Geopark. Two migmatitic gneisses composed of biotite±sillimanite±garnet+feldspar+quartz were dated. Leucosomes in the samples were clearly distinguished from their melanosomes. The U-Pb isotopic compositions of zircon from sillimanite- and garnet-bearing migmatitic samples were measured using a secondary ion microprobe, yielding metamorphic ages, 1870±10 Ma (2σ)와 1863±6 Ma (2σ), respectively. 1.87~1.86 Ga metamorphic ages are consistent with those of the Paleoproterozoic low-P and high-T regional metamorphism (1.87~1.85 Ga) in the Yeongnam Massif. The maximum depositional age based upon the apparent 207Pb/206Pb ages of detrital zircon in the samples was estimated as 2.06 Ga, and thus sedimentation age of the protolith of the migmatitic gneisses ranges between 2.06 and 1.87 Ga.

Structural and Compositional Characteristics of Skarn Zinc-Lead Deposits in the Yeonhwa-Ulchin Mining District, Southeastern Taebaegsan Region, Korea Part I: The Yeonhwa I Mine

  • The zinc-lead deposits at the Yeonhwa I mine were investigated in terms of ore-forming geologic setting, structural style of ore control, geometry of individual orebodies, zoning, paragenesis and chemical composition of skarn minerals, as well as metal grades and ratios of selected orebodies. The Yeonhwa I mine is characterized by a large swarm of chimney type massive orebodies with thin skarn envelopes, boldly developed through a thick sequence of Pungchon Limestone, the overlying Hwajeol Formation, and the underlying Myobong Slate of Cambrian age. Nearly 20 orebodies of similar shape, but of varying size are arranged in a V-shaped pattern with northwest and northeast trends, clearly indicating an outstanding ore control by a conjugate system of fractures with these trends. Important orebodies are the Wolam 1, 2, 3, and 5 orebodies in the west, and the Namsan 1, 2, 3. and 5 orebodies in the east, among others. The Wolam 1 orebody, which was observed from the -360 level through the -240, -120, and 0 levels to the surface outcrops (totaling a vertical height of about 500m), shows a vertical variation in skarn mineralogy, ranging from pyroxene-garnet zone on the lower levels. through pyroxene (without garnet) zone on the intermediate levels, and finally to rhodochrosite vein on the upper levels and surface. Microprobe analyses of pyroxene and garnet on a total of 14 mineral grains revealed that pyroxenes are manganoan salitic in most samples, with downward increase of Fe and Mn, whereas garnets are highly andraditic, containing fractions of subordinate grossular with downward decrease of Fe. This indicates a reverse relationship of Fe-contents between pyroxene and garnet with depth. Ore minerals are major sphalerite, subordinate galena, and minor chalcopyrite. Sulfide gangue minerals include major pyrrhotite, and minor pyrite and marcasite of later age. Two types of variational trends in metal grades and ratios with depth are present on the plots of assay data from the Wolam orebodies: one is a steady upward increase in Pb, Zn, and Pb:Zn ratios, with a terminal decline at the top of orebody: the other is an irregular or sinusoidal change. The former is characteristic of chimney-type orebodies, whereas the latter is of vein· shaped orebodies. The Pb grades show large variations among orebodies and from level to level, whereas the Zn grades are relatively constand or less variable.

Geochemical Occurrence of Uranium and Radon-222 in Groundwater at Test Borehole Site in the Daejeon area (대전지역 시험용 시추공 지하수내 우라늄 및 라돈-222의 지화학적 산출특성)

  • A drilling project was undertaken to characterize the geochemical relationship and the occurrence of radioactive materials at a test site among public-use groundwaters previously known to have high occurrence of uranium and radon-222 in the Daejeon area. A borehole (121 m deep) was drilled and core rocks mainly consist of two-mica granite, and associated with pegmatite and dykes of intermediate composition. The groundwater samples collected at six different depths in the borehole by a double-packed system showed the pH values ranging from neutral to alkaline (7.10-9.3), and electrical conductivity ranging from 263 to 443 ${\mu}S/cm$. The chemical composition of the borehole groundwaters was of the $Ca-HCO_3(SO_4+Cl)$ type. The uranium and Rn-222 contents in the groundwater were 109-1,020 ppb and 9,190-32,800 pCi/L, respectively. These levels exceed the regulation guidelines of US EPA. The zone of the highest groundwater uranium content occurred at depths of 45 to 55m. The groundwater chemistry in this zone (alkaline, oxidated, and high in bicarbonate) is favorable for the dissolution of uranium into groundwater. The dominant uranium complex in groundwater is likely to be $(UO_2CO_3)^0$ or $(UO_2HCO_3)^+$. Radon-222 content in groundwater shows an increasing trend with depth. The uranium and thorium contents in the core were 0.372-47.42 ppm and 0.388-11.22 ppm, respectively. These levels are higher values than those previously been reported in Korea. Microscopic observations and electron microprobe analysis(EPMA) revealed that the minerals containing U and Th are monazite, apatite, epidote, and feldspar. U and Th in these minerals are likely to substitute for major elements in crystal lattice.
