Feeding Ecology of Sillago japonica in an Eelgrass (Zostera marina) Bed

  • Feeding habits of Sillago japonica collected from in an eelgrass (Zostera marina) bed in Jindong Bay, Korea were studied. S. japonica was a carnivore which consumed mainly gammarid amphipods, polychaetes, bivalves, caridean shrimps and crabs. Its diets also included a small amount of fishes, copepods and caprellid amphipods. The diet of S. japonica underwent significant size-related changes; small individuals (<5cm SL) fed mainly on gammarid amphipods and crab larvae, while proportion of polychaetes and bivalves increased with increasing fish size and gammarid amphipods were also important prey for medium size individuals (5.1-9.9cm SL). The large individuals (>10cm SL) ate polychaetes, caridean shrimps and crabs. The dietary breadth of S. japonica were varied with size. The diet of S. japonica also underwent seasonal changes that could be related to differences in prey availability; gammarid amphipods were mainly consumed in spring and polychaetes in summer.

  • Feeding habits of Acentrogobius pellidebilis collected from in an eelgrass(Zostera marina) bed from January to December 2006 were studied. A. pellidebilis ranged form 1.6 to 6.9 cm in standard length were determined. A. pellidebilis was a carnivore which consumed mainly copepods, polychaetes, and amphipods. Its diets included small quantities of isopods, seaweeds, nematods, and gastropods. It showed ontogenetic changes in feeding habits. Smaller individuals (<2.0cm SL) fed mainly on copepods. While the consumption of copepods decreased with increasing fish size, the consumption of polychaetes and amphipods were increased. Dietary breadth index was varied with fish size.

  • Apigenin-7-O-${\beta}$-D-glucoside, chrysoeriol, and luteolin were isolated from the aqueous ethanolic extract of Zostera marina L. leaves as the scavengers of reactive oxygen species (ROS) with the SC$\_$50/ values of 0.18 mM, 0.68 mM, and 0.18 mM against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 0.04 mM, 0.03 mM, and 0.01 mM against superoxide radicals in the xanthine/xanthine oxidase system, respectively. The luteolin suppressed the expression of matrix metalloproteinase-1 (MMP-1) up to 44% at 4.0 ${\mu}$M. (omitted)

Inhibitory Effect of Zostera japonica on Growth of Human Cancer Cells (애기거머리말 추출물의 암세포 성장 억제효과)

  • In this study, crude extracts of the marine eelgrass Zostera japonica and their solvent-partitioned fractions were evaluated for their inhibitory effect against AGS, HT-1080 and MCF-7 human cancer cells using MTT assay. Each of the crude extracts (acetone/methylene, chloride, and methanol) of Z. japonica showed a significant inhibitory effect on the growth of human cancer cells. The combined crude extracts were partitioned between $CH_2Cl_2$ and water. The organic layer was further partitioned between 85% aq. MeOH and n-hexane, and the aqueous layer was then fractionated into n-BuOH and $H_2O$, successively. Growth inhibition effects of solvent-partitioned fractions from Z. japonica on human cancer cells increased in a dose-dependent manner. Among these tested samples, the 85% aq. MeOH fraction revealed good inhibitory effects on the growth of AGS and HT-1080 human cancer cells, while the n-hexane fraction exhibited good inhibitory effects on the growth of AGS and MCF-7 human cancer cells. In addition, 85% aq. MeOH and n-hexane fractions enhanced mRNA expression of p53 gene. These results suggest that there is further scope for the isolation of active compounds from Z. japonica, which should show much stronger anticancer activity.

A Comparison of Methods for Estimating the Productivity of Zostera marina

  • Because seagrass production significantly contributes to the biodiversity and production of coastal and estuarine ecosystems, accurate estimation of seagrass productivity is a critical step toward understanding the ecological roles of seagrass in these ecosystems. To develop an accurate and effective method of measuring seagrass productivity, we estimated leaf productivity of eelgrass (Zostera marina) on the southern coast of Korea using three methods, the conventional leaf marking method, the elongation-mass method (Short '87 method), and the plastochrone method. In each season, shoots were pierced through the bundle sheath using a hypodermic needle and were collected after 2-4 weeks had elapsed to estimate their productivity. The leaf elongation and the leaf plastochrone intervals varied significantly among seasons. On an annual basis, the conventional leaf marking method showed the lowest leaf productivity estimates compared to the elongation-mass method and the plastochrone method, suggesting that the conventional leaf marking method underestimated leaf productivity as it ignored leaf maturation processes and new leaf growth within the sheath. Since the elongation-mass method considered leaf maturation processes, this method produced higher leaf productivity estimates than the conventional leaf marking method. On an annual basis, the plastochrone method produced the highest leaf productivity estimates. Below-ground productivity, which can be easily estimated using the plastochrone method, ranged between 3.29 and 5.73 (mg dry weight $shoot^{-1}\;day^{-1}$) and accounted for about 17.8% to 30.3% of total productivity. Because of the high contributions of below-ground productivity to total seagrass production, we suggest that the plastochrone method is an effective and simple technique for assessing both above- and below-ground productivities.

Therapeutic and Prophylactic Effects of Zostera Marina on Dextran Sulfate Sodium-induced Colitis (해대(海帶) 추출물이 Dextran Sulfate Sodium로 유발된 대장염 동물모델에 미치는 치료 및 예방적 효과)

  • Objectives: Inflammatory bowel disease (IBD) is chronic inflammatory disorders of the intestines. Due to limitation of conventional treatment including steroids, herbal medicines have emerged as possible therapeutic options for IBD. The purpose of the current study was to investigate the therapeutic and prophylactic effects and mechanisms of Zostera Marina water extract (ZME) on DSS-induced colitis. Methods: Colitis was induced by DSS in Balb/c mice. In pre-treatment setting, ZME was administered 7 days before DSS treatment and in co-treatment setting, ZME was simultaneously administrated with DSS treatment. In both settings, ZME 100, 300 and 1000 mg/kg were orally administered twice a day, respectively. Mice weight and clinical findings were measured daily. Colon length, macroscopic findings and histological damages of colon mucosa were assessed at the end of experiments. The levels of cytokines including TNF-${\alpha}$, IFN-${\gamma}$, IL-$1{\beta}$, IL-6, IL-10 and IL-17 were measured by Biometric Multiplex Cytokine Profiling method. Results: In a dose dependent manner, ZME significantly inhibited the colon shortening, and improved macroscopic score and histological score. However, there were insignificant changes on inhibition of weight loss and improvement of clinical score. There were no significant differences of effects between co-treatment and pre-treatment settings. ZME 300 and 1000 mg/kg groups significantly inhibited IFN-${\gamma}$. Only ZME 1000 mg/kg group significantly inhibited TNF-${\alpha}$, IL-$1{\beta}$ and IL-6. Conclusions: The current results show the possibility of therapeutic use and its prophylactic application of ZME on inflammatory bowel diseases. Future studies for targeted mechanisms of ZME are needed.

Growth dynamics of the seagrass, Zostera marina in Jindong Bay on the southern coast of Korea

  • Growth dynamics of the seagrass, Zostera marina were examined at the two stations (Myungju and Dagu) in Jindong Bay on the southern coast of Korea. Eelgrass leaf productivities, underwater irradiance, water temperature, dissolved inorganic nitrogen (DIN) in water column and sediments, and tissue carbon (C) and nitrogen (N) content were monitored monthly from March 2002 to January 2004. Underwater irradiance fluctuated highly without a clear seasonal trend, whereas water temperature showed a distinct seasonal trend at both study stations. Water column DIN concentrations were usually less than $5{\mu}M$ at both study sites. Sediment pore water $NH_4{^+}$ and $NO_3{^-}+NO_2{^-}$ concentrations were higher at the Myungju site than at the Dagu site. Eelgrass leaf productivity at both study sites exhibited a distinct seasonality, increasing during spring and decreasing during summer. Seasonal variation of eelgrass productivity was not consistent with seasonal patterns of underwater irradiance, or water temperature. Eelgrass tissue C and N content at both study sites also showed significant seasonal variations. Relationships between tissue C and N content and leaf productivities exhibited usually negative correlations at both study sites. These negative correlations implied that the growth of Z. marina at the study sites was probably limited by C and N supplies during the high growth periods.

Chlorophyll α fluorescence as an indicator of establishment of Zostera marina transplants on the southern coast of Korea

  • To test the feasibility of using chlorophyll ${\alpha}$ fluorescence to assess the establishment success of seagrass transplants, photosynthetic characteristics of eelgrass Zostera marina transplants were measured using a Diving-pulsed amplitude modulation fluorometer in Jindong Bay on the southern coast of Korea. Maximum quantum yield ($F_v/F_m$), photosynthetic efficiency ($\alpha$), saturating irradiance ($E_k$) and maximum electron transport rate ($ETR_{max}$) of transplants and reference plants in a nearby transplant site were measured using the fluorometer for 5 months. Additionally, shoot morphology, individual shoot weight and productivity of transplants and reference plants were also monitored. Shoot height, leaf weight and productivity of transplants were significantly reduced during the first two or three months after transplantation compared to those of reference plants, and then increased to the levels of reference plants Characteristics of chlorophyll a fluorescence, including $F_v/F_m$, $\alpha$, $E_k$ and $ETR_{max}$ of transplants were also significantly reduced in the initial period, but recovered slightly sooner than shoot morphology or leaf productivity. These results indicated that after transplantation, Z. marina transplant photosynthesis recovered faster than shoot morphology, biomass or productivity. Thus, chlorophyll a fluorescence can be used as an indicator for early assessment of the status of eelgrass transplants without destructive sampling.
