정양단(精養丹)의 항염증, 상처 치유효능에 대한 연구 (Wound Healing-Enhancing and Anti-inflammatory effects of five Korean Traditional Herbal Medicines, Jeong Yang-dan)

  • Objectives : The aim of the present study is to evaluate the wound healing-enhancing and anti-inflammatory effects of Pinus densiflora, Cornus officinalis, Zingiber officinale, Ganoderma japonicum and Scutellaria baikalensison human keratinocyte, HaCaT cells. Methods : We adopted in vitro wound healing assay to measure the proliferation-and migration-enhancing effects in HaCaT cells. The expressions of cytokine genes were measured in HaCaT cells using real-time PCR analysis. Results : The extracts of Pinus densiflora, Cornus officinalis, Zingiber officinale, Ganoderma japonicum and Scutellaria baikalensis enhanced the proliferation and migration of HaCaTcells. The expression of keratinocyte growth factor receptor(FGFR2-IIIb) gene was also induced. The extracts inhibited iNOS, IL-$1{\beta}$ and TNF-$\alpha$ gene expression. Conclusions : The extract of Pinus densiflora, Cornus officinalis, Zingiber officinale, Ganoderma japonicum and Scutellaria baikalensis has wound healing-enhancing effects and anti-inflammatory effects.

생강(Zingiber officinale R.)근경으로부터 항산화 활성물질의 분리 (Isolation of Natural Antioxidants from the Root of Zingiber officinale R.)

  • 건강을 80% MeOH수용액으로 추출하고, 얻어진 추출물을 EtOAc, n-BuOH및 $H_2O$로 용매 분획하였다. 그 중에서 뚜렷한 활성을 보인 EtOAc 분획으로부터 DPPH(1-diphenyl-2-picryl hydrazyl) radical 포획법으로 항산화 활성을 추적하여 항산화 활성을 갖는 화합물 2종을 분리하였다. 각각을 NMR 및 MS 데이터의 해석과 문헌 자료를 조사하여 6-shogaol 및 12-shogaol로 구조를 결정하였다. 각 화합물의 $EC_{50}$값은 26.4 ${\mu}M$과 22.5 ${\mu}M$로 상용항산화제인 BHA, BHT 및 ${\alpha}-tocopherol$보다 뚜렷이 높은 활성을 나타내었다.

Quantitative analysis method for zingiber officinale water extract using high-performance liquid chromatography

  • Quantitative analysis of the Zingiber Officinale sample using subcritical water extraction (SWE) was developed employing High-Performance Liquid Chromatography (HPLC) to bolster the advancement of this innovative green extraction process. This research focuses on three principal ginger bioactive compounds: 6-gingerol, 6-shagoal, and 10-gingerol. Various stages were undertaken to establish the quantitative analysis method, encompassing the optimisation of HPLC operating conditions and the formulation of standard calibration curves, yielding individual compound equations. A robust correlation within the calibration curve was achieved, exhibiting an r2 value of 0.9814 and an RSD of 5.00%. A simultaneous, swift, and dependable method was established with an injection time of 20 minutes and an 8-minute delay between injections, in contrast to the previous HPLC analysis requiring a 45-minute injection time for detecting and quantifying all components. Notably, no post-treatment was applied after the SWE process. This advancement allows for potential future online measurement of Zingiber Officinale bioactive compounds extracted using subcritical water extraction through this technology.

양하, 양하근경 및 생강분말이 고지혈증 유발 흰쥐의 지질성분에 미치는 영향 (Effects of Zingiber mioga, Zingiber mioga Root and Zingiber officinale on the Lipid Concentration in Hyperlipidemic Rats)

  • 고지혈증 유발 식이와 양하, 양하근경 및 생강분말의 혼합급이가 흰쥐의 혈청 및 간장의 지질대사에 미치는 영향을 검토하고자 식이에 각각 3%씩 첨가하여 4주간 실험사육하였다. 혈청 총콜레스테롤 농도는 대조군에 비하여 전 실험군이 유의적으로 낮았으며, HDL-콜레스테롤 농도는 양하와 양하근경 급여군이 대조군에 비하여 높게 나타났다. 유리콜레스테롤 및 콜레스테릴 에스테르의 농도는 전 실험군이 대조군에 비하여 유의적으로 낮았고, 콜레스테릴 에스테르 농도비는 대조군이 72.36%, 실험군에서 62.14~65.33%였다. Chylomicron, LDL 및 VLDL 농도는 대조군에 비하여 전 실험군에서 유의적으로 낮았다. 중성지질 및 인지질은 대조군에 비하여 전 실험군이 유의적으로 낮았다. 간장 중의 총콜레스테롤, 중성지질, 유리콜레스테롤 및 콜레스테릴 에스테르의 농도는 대조군에 비하여 각 실험군에서 유의적인 감소를 나타내었으나 인지질의 농도는 유의적인 차이가 없었다.

  • Recently many investigators have initiated searches for immunomodulating substances from natural food sources. Ginger (Zingiber officinale Roscoe) has been used as a raw material in many traditional preparations since the ancient time. This study was performed to investigate the immunomodulative effects of Zingiber officinale Roscoe in mice, using ex vivo experiments. In order to elucidate the immunomodulative effects of Ginger, water extracts of the plant were orally administrated into mice, and isolated splenocytes and macrophages were used as experimental model. In order to identify its ex vivo effect six to seven week old Balb/c mice were fed ad libitum on a chow diet and water extracts of ginger were orally administrated every other day for two weeks at two different concentrations (50 and 500 mg/kg b.w.). After preparing the single cell suspension, the proliferation of splenocytes was determined by MTT assay. The result of ex vivo study showed that the highest proliferation of splenocytes and macrophage activatation was seen in the mice orally administrated at the concentration of 500 mg/kg b. w. of ginger water extracts. In conclusion, this study suggests that ginger extracts nay enhance the immune function by regulating the splenocyte proliferation and cytokine prodution capacity by activated macrophages in mice.

  • Ginger (Zingiber officinale) belonging to the family Zingiberaceae is a perennial herb. It is widely distributed in tropical Asia. In India, it is cultivated mainly in Kerala, Andhia Pradesh, Uttar Pradesh, West Bengal and Maharashtra. It is one of the most common spices, which is in use since centuries for its versatile medicinal actions like antiemetic, stomachic, expectorant, anti-inflammatory, aphrodisiac etc in traditional system of medicine (Unani, Ayurveda, and Chinese medicine). It is useful for the treatment of various gastrointestinal, pulmonary, cardiovascular and sexual disorders. The phytochemical study of ginger showed the presence of many volatile oils and oleo-resins like gingerol, zinger one, zingiberol etc. Numerous experimental and clinical trials have proven ginger for its range of therapeutic activities such as antibacterial, antidiabetic, antiemetic, hypolipidaemic, hepatoprotective etc properties. The present article aims to explore traditional Unani and pharmacological activities of this herb reported till date.

Isolation, Identification and Determination of Antioxidant in Ginger (Zingiber officinale) Rhizome

  • The antioxidative compounds and antioxidant contents of ginger (Zingiber officinale) rhizomes were determined. Substances reextracted using ethyl acetate from crude methanol extract of fresh ginger rhizome were separated through thin layer chromatography. Ten phenolic antioxidative bands were visualized through color reactions using ferric chloride-potassium ferricyanide and 1,1-diphenyl-2-picrylbydrazyl (DPPH). The antioxidative compounds were purified through preparative TLC and high performance liquid chromatography (HPLC), among which, five antioxidants were identified as 4-, 6-, 8-. and 10-gingerols and 6-shogaol on the basis of their molecular weights determined through LC-MS. As shown in experiments using DPPH free radicals, 6-Gingerol and PT4-HP8 (unknown) were revealed to be more efficient than BHT (butylated hydroxy toluene). Contents of gingerols were determined through reverse phase HPLC. Total gingerol contents (sum of 6-,8-, and 10-gingerols) in rhizomes of different ginger varieties varied significantly. The HG55 (collected at Wanju district in Korea) and the HG52 (imported from Brazil) showed the highest gingerol contents.

생강 추출물 투여가 전구염증성 사이토카인 $IFN-{\gamma}$와 항염증성 사이토카인 IL-10 분비량에 미치는 영향 (The Effects of Zingiber officinale Roscoe Extracts on Mouse $IFN-{\gamma}$ and IL-10 Production)

  • Ginger(Zingiber officinale Roscoe) has been used as a raw material in many various traditional preparations since the ancient times. As a component of traditional health products, ginger is known to be effective as an appetite enhancer, and has anti-cold and anti-inflammatory activities. This study was performed to investigate the immunomodulative effects of Zingiber officinale Roscoe in mice, using ex vivo experiments. In order to elucidate ginger's immunomodulative effects of Ginger, water extracts were orally administered to mice, and isolated macrophages were used as the experimental model. To identify the ex vivo effects, six to seven week old Balb/c mice were fed a chow diet ad libitum and the ginger water extracts were administered orally every other day for two or four weeks at two different concentrations(50 and 500 mg/kg b.w.). The results show that IL-IO and $IFN-{\gamma}$ were detected in the 500 mg/kg b.w. supplemented group with LPS stimulation in all cases. Also, the $IFN-{\gamma}$ /IL-10 ratio ranged from 3~5 with mitogen stimulation such as Con A and LPS. In conclusion, this study suggests that ginger extracts may enhance the immune function by regulating the cytokine(IL-10 and $IFN-{\gamma}$) production capacity of activated macrophages in mice.

생강 분획에 따른 추출물이 마우스 비장세포와 Cytokine (IL-1 ${\beta}$, IL-6, TNF-${\alpha}$)의 생성량에 미치는 영향 (Effect of Zingiber officinale Roscoe Fractionation of Extracts on Mouse Spleen and Macrophage Cells Activation)

  • Ginger(Zingiber officinale Roscoe) has long been used as a food source in Korea, and it is widely used as a dietary condiment throughout the world. The present study focused on the immunomodulative effects of ginger extracts via in vitro experiments. To identify the immune-activation fractions of the plant, we performed the systematic fractionation of ginger with methanol, hexane, chloroform, butanol and water for separation and refining. The results showed that the chloroform fraction had the highest immune cell activation properties. In conclusion, this study suggests that ginger extracts may enhance immune function by regulating the splenocyte proliferation as well as the cytokine production capacity of activated macrophages.

In Vitro Propagation of Zingiberaceae Species with Medicinal Properties

  • Zingiber officinale buds from the rhizomes were used to produce in vitro shoots. These explants produced the largest number of multiple shoots, 9.8 shoots per explant, when were cultured on MS (Murashige and Skoog 1962) medium supplemented with 2.0 mg/L benzyladenine (BA) and 2.0 mg/L indole butyric acid (IBA). This medium was also found to be suitable for in vitro propagation of other Zingiberaceae species: Alpinia conchigera, Alpinia galanga, Curcuma domestica, C. zedoaria and Kaempferia galanga. Both C. domestica and C. zedoaria produced more multiple shoots when were cultured in the liquid proliferation medium, MS medium containing 2.0 mg/L BA and 2.0 mg/L IBA. To maintain the in vitro plantlets of Zingiberaceae species, they were required to subculture every four weeks. After executing proper acclimatization protocol, in vitro plantlets of Alpinia galanga, A. conchigera, Curcuma domestica, C. zedoaria, Kaempferia galanga and Zingiber officinale could be successfully planted in the field with high percentage of survival.
