생강(Zingiber officinale Roscoe) 추출물의 항산화 및 A2058 흑색종세포 사멸 효과 (Effect of Zingiber officinale Roscoe Extract on Antioxidant and Apoptosis in A2058 Human Melanoma Cells)

  • This study investigated the effects of ginger (Zingiber officinale Roscoe) on antioxidant and antiproliferative activities in A2058 human melanoma cells. The antioxidant and antiproliferative activities of 70% ethanol extracts of Zingiber officinale Roscoe were identified based on DPPH and ABTS free radical scavenging capacities. Treatment of cells with Zingiber officinale Roscoe at concentrations of 0, 0.2, and 0.4 mg/mL for 24 hours significantly reduced cell viability as determined by Hoechst 33258 nuclear staining, apoptosis analysis, and Western blotting analysis, respectively. In our study, 70% ethanol extracts of Zingiber officinale Roscoe exhibited antioxidant activity and inhibited A2058 cell growth in a dose-dependent manner. Concomitant activation of the mitochondria-dependent apoptotic pathway of A2058 human melanoma cells by Zingiber officinale Roscoe extracts was mediated via modulation of Bax and Bcl-2 expression, which activated cleavage of caspases-3, caspases-9, and poly ADP-ribose polymerase. The findings of study indicate that Zingiber officinale Roscoe extracts induce apoptosis in A2058 human melanoma cells, and this phenomenon occurs via the death receptor-mediated and intrinsic pathways.

  • As an attempt to develop new functional health beverage by using medicinal herb, green ginger(Zingiber officinale Roscoe), we investigated the effect of Zingiber officinale on blood pressure and regional cerebral blood flow (rCBF) of rats with Zingiber officinale extracts. Zingiber officinale extract increase rCBF significantly. The drink produced consisted of Zingiber officinale extract 1.825%, maltitol syrup 17.0%, citric aicd 0.06%, ascorbic acid 0.02%, stevioside 0.001%, ginger flavor 0.11% and water. Brix, pH and acidity of product were 13.7, 4.4 and 0.09, respectively. This drink had good score by the sensory evaluation. The above results showed that development of such functional beverage using Zingiber officinale can used as a functional material improving blood circulation in beverage industry.

생강과 톳 추출물이 마우스의 대식 세포에서 Nitric Oxide(NO) 생성에 미치는 영향 (Effect of Zingiber officinale and Hizikia fusiforme Water Extracts on NO Production in Macrophage of Mice)

  • Zingiber officinale and Hizikia fusiforme(sea weed fusiforme) have long been used for food sources in Korea. The present study was performed to investigate the ex vivo effect of Zingiber officinale and Hizikia fusiforme on NO production in macrophage of mice. Seven to eight week old mices(Balb/c) were fed chew diet ad libitum and water extract of Zingiber officinale and Hizikia fusiforme was administrated orally at two different concentrations (50 and 500 mg/kg B.W.). every other day for two or four weeks NO(nitric oxide) production by activated macrophage was assessed by measuring nitrite, the stable NO metabolite, using Griess reaction assay. NO production were significantly enhanced in Zingiber officinale group at 500 mg/kg B.W. and in Hizikia fusiforme group at 50 mg/kg B.W. compared to the coresponding control groups. In conclusion, this study may suggest that Zingiber officinale and Hizikia fusiforme(sea weed fusiforme) extracts enhance the immune function by regulating NO production in macrophages of mice.

생강(生薑)의 약리효능(藥理効能)에 대한 최근(最近) 국내(國內) 연구(硏究) 동향(動向) (Recent Research Trends in the Pharmacological Effects of Zingiber officinale in Korea)

  • Objectives: This study aims to analyze recent research on the pharmacological effects of Zingiber officinale. Methods: We searched for papers from databases such as ScienceON, RISS, DBPia, and NaNet. The papers were classified according to pharmacological effects, and the selected studies were analyzed. Results: Six studies were finally included in the study. 1. Four studies mainly focused on the effects of anti-inflammation using in vitro or in vivo experiments. 2. Two studies mainly focused on the effects of antioxidants using in vitro experiments. 3. Other pharmacological effects, including improvement of gastrointestinal function, inhibition of body temperature reduction, and anti-aging, were investigated using in vitro or in vivo studies. Conclusion: This study shows that Z. officinale has several pharmacological effects, including anti-inflammation and antioxidant.

  • 봉동산 생강(Zingiber officinale Roscoe)의 정유성분조성을 분석코자 액체 이산화탄소 주출법으로 분리하여 silica gel column chromatography범에 의해 탄화수소 화합물 분획과 함산소 화합물 분획으로 분류한 다음 GC 및 GC-MS로 성분을 검토하였다. 추출물의 수율은 6.96%이었으며, SCC에 의해 분리된 분획의 분석결과 mass spectrum에 의해서 잠정적으로 확인된 58종을 포함하여 102종의 향기성분이 확인되었다. 중 탄화수소 화합물 분획에서는 zingiberene, ${\beta}-sesquiphellandrene,\;{\gamma}-bisabolene,\;{\gamma}-cadinene,\;ar-curcumene$ 등이 주성분이었고, 함산소 화합물 분획에서는 gernal, sesquisabinene hydrate, borneol, zingiberenol 등이 주성분 이었다. 총 essential oil의 조성은 $zingiberene,\;citronellol+{\beta}-sesquiphellandrene,\;geranial,\;{\gamma}-bisabolene,\;ar-curcumene+geranyl\;acetate$ 등이 주성분으로 sesquiterpene hydrocarbon의 함량이 높게 나타났다.

Effect of Activated Carbon on Growth of Ginger (Zingiber officinale)

  • Activated carbon(AC) can be utilized as a soil conditioner in agricultural crop areas. This study was conducted to investigate the effect of AC on growth and yield of Ginger(Zingiber officinale) as affected by different amounts of AC. The results obtained are summarized as follows. Growth characteristics including plant height and leaf length were the highest when activated carbon added with $5-10\%$, suggesting that optimum amount of activated carbon was ranged from 5 to $15\%$. Growth and enlargement of the root were improved by $10\%$ AC with higher rhizome length and weight.

  • Zingiber officinale is a spice which is having a strong historical medicinal background and used by different systems of medicine for various ailments. It is also used to maintain the characteristic sharpness and tangy essence of food and beverages and used in carbonated drinks, as a conserve in sugar syrup. Ginger considered as an incredible drug for inflammation, gastrointestinal problems, pain and sexual problems since centuries. There are many studies carried out in recent years, which proves that the active constituents of ginger shows a variety of pharmacological effects like antiemetic, hepatoprotective, anti hyperlipidemic and antibacterial. The most momentous among all the nutraceutical aspects of ginger are its positive control on gastrointestinal tract including digestive stimulant action, anticancer effect and anti-inflammatory effect.

Comparative antidiabetic activity of different fractions of methanolic extract of Zingiber officinale Roscoe in streptozotocin induced NIDDM rats

  • Earlier we have reported the antidiabetic activity of fresh juice of rhizomes of Zingiber officinale (Z. officinale) and its correlation with 5-HT receptor antagonism. Since 6-gingerol the marker compound of Z. officinale is reported to posses 5-HT anatgonistic activity, the present investigation, was undertaken to find out the concentration of 6-gingerol present in methanolic extract of Z. officinale and its different fractions (petroleum ether, toluene and chloroform). We also evaluated these fractions for antidiabetic activity in streptozotocin (STZ)-induced neonatal type 2 diabetic rats. Fasting glucose and insulin levels in non insulin dependent diabetes mellitus (NIDDM) rats were found to be significantly (P < 0.05) higher than control rats and these were significantly decreased by treatment with methanolic extract of Z. officinale and its fractions. The results of oral glucose tolerance test (OGTT) showed that methanolic extract and its fractions significantly (P < 0.05) decreased both STZ-induced increase in $AUC_{glucose}$ and $AUC_{insulin}$ values in NIDDM groups. Treatment with petroleum ether fraction produced a greater reduction in elevated glucose and $AUC_{glucose}$ levels as compared to treatment with other fractions. Treatment with methanolic extract of Z. officinale and its fractions also produced significant reduction in the elevated lipid, serum glutamate oxaloacetate transaminase (SGOT) and serum glutamate pyruvate transaminase (SGPT) levels in NIDDM rats. The effect of petroleum ether fraction on elevated lipid, SGOT and SGPT levels was significantly greater as compared to treatment with other fractions. The concentration of 6-gingerol was found to be maximum in petroleum ether fraction (11.430%) and minimum in chloroform fraction (0.973%). The methanolic extract and toluene fraction was found to contain 3.080% and 2.191 %, 6-gingerol respectively. In conclusion, our data suggest that methonolic extract and its fractions possess significant antidiabetic activity in NIDDM rats. The extent of activity appears to be dependent on the concentration of 6-gingerol present in the extract or its fractions.

  • The content analysis of 6-gingerol, which is an active compound, in Zingiber spp. (Z. officinale and Z. mioga) and their commercial foods (ginger teas and powders) was conducted using high-performance liquid chromatography. A reverse phase system was used, with a gradient solvent system of water and acetonitrile. The 6-gingerol content was highest in the methanol extract of Z. officinale root (17.09 mg/g extract) and ginger powder B (15.92 mg/g extract). The results demonstrated that this method was simple and reliable for the quality control of Zingiber commercial foods.
