골절된 사자 견치의 근관치료 및 Crown 장착 1례 (A Case of Root Canal Treatment and Crown Therapy on Fractured Canine Teeth of Lion)

  • Seven years old male lion in Everland Zoo has three fractured canine tooth. To avoid the risk of a possible fracture with pulp exposure after restroration of largest abration defects, endodontic therapy was performed. The pulp chamber was filled with calcium hydroxide, zinc oxide cement and gutta percha. We also installed the Crown prosthesis was installed in fractured canine tooth to protect the tooth and to improve the cosmetics and function of the tooth. The lion could eat in one day and there were no any signs of dental problem. It is the obvious prophylactic procedure for wild animal is the inclusion of an oral examination at every opportunity to handle the animal. It is considered that crown prosthesis after endodontic therapy was useful for maintaining normal physiological function and for provention of additional fracture or complication.

A novel CaAbsi1 gene induced by early-abiotic stresses in pepper

  • The full-length cDNA of CaAbsi1 encodes a presumptive protein of 134 amino acid residues that has homology to a putative zinc finger protein in its C-terminus. The deduced amino acid sequence has 50% homology to Oryza sativa NP001049-274, the function of which is unknown. Expression of CaAbsi1 was reduced in response to inoculation of non-host pathogens. On the other hand it was induced one hour after exposure to high concentrations of NaCl or mannitol, and six hours after transfer to low temperature. Induction also occurred in response to oxidative stress, methyl viologen, hydrogen peroxide and abscisic acid. Our results suggest that CaAbsi1 plays a role in multiple responses to wounding and abiotic stresses.

Mancozeb의 급성노출이 마우스의 면역기능에 미치는 영향 (Effects of Acute Oral Administration of Mancozeb on the Immune Function in Mice)

  • Mancozeb, a polymeric complex of zinc and manganese salts of ethylene bisthiocarbamate (EBDC), is used widely in agriculture as fungicides, insecticides, and herbicides. Mancozeb can be occupationally and environmentally exposed to human and has been reported to induce estrogenic activity, therein it is considered as an endocrine disrupter, We investigated the effects of acute exposure of Mancozeb on the immune function in mice. After single oral administration of Mancozeb to female ICR mice, the immunopathological parameters (body- and organ-weight, splenic cellularity hematological parameters), mitogen (Con A, PHA+IL-2, LPS)-induced splenocyte proliferation (SP) and splenic IgM plaque forming cell (PFC). WBC and splenic cellularity were decreased, but liver-, kidney-, and spleen-weight were increased when compared with control group. Splenic IgM PFC against SRBC was slightly lowered. Mitogen-induced proliferation of spleen cells from Mancozeb-treated mice was not signifcantly changed ex vivo, however, SP in vitro were significantly lowered in concentration dependent manner. These results indicate that Mancozeb could affect the immune function in mice.

Over-expression of Cu/ZnSOD Increases Cadmium Tolerance in Arabidopsis thaliana

  • Over-expression of a copper/zinc superoxide dismutase (Cu/ZnSOD) resulted in substantially increased tolerance to cadmium exposure in Arabidopsis thaliana. Lower lipid peroxidation and $H_2O_2$ accumulation and the higher activities of $H_2O_2$ scavenging enzymes, including catalase (CAT) and ascorbate peroxidase (APX) in transformants (CuZnSOD-tr) compared to untransformed controls (wt) indicated that oxidative stress was the key factor in cadmium tolerance. Although progressive reductions in the dark-adapted photochemical efficiency (Fv/Fm) and quantum efficiency yield were observed with increasing cadmium levels, the chlorophyll fluorescence parameters were less marked in CuZnSOD-tr than in wi. These observations indicate that oxidative stress in the photosynthetic apparatus is a principal cause of Cd-induced phytotoxicity, and that Cu/ZnSOD plays a critical role in protection against Cd-induced oxidative stress.

개인용 공기포집기를 이용한 기중 연농도와 생물학적 연 폭로지표와의 관련성 (Relationship between the Biological Lead Exposure Indices and Air lead Concentrations measured by Personal Air Samplers)

  • This study was carried out to evaluate the relationship between the biological lead exposure indices and air lead concentrations measured by personal air samplers. The 72 occupationally lead exposed workers were observed and the bioiogical lead Exposure indices chosen for this study were blood lead(PbB), urine lead(PbU), zinc protoporphyrin in whole blood(ZPP), $\delta$-aminolevulinic acid in urine(ALAU), $\delta$-aminolevulinic acid dehydratase activity (ALAD), coproporphyrin in urine(CPU) and hemoglobin(Hb). The workers were divided into four groups by air lead concentrations: Group I; under $0.05mg/m^3$, Group II; $0.05-0.10mg/m^3$, Group III; $0.10-0.15mg/m^3$ and Group IV; and over $0.15mg/m^3$. For evaluation the relationship between the biological lead exposure indices and air lead concentrations was used as correlation coefficients. The results obtained were as follows: 1. In Group I, II, III and IV, the mean value of PbB were $25.45{\pm}1.84{\mu}g/dl,\;27.87{\pm}3.53{\mu}g/dl,\;31.21{\pm}1.76{\mu}g/dl\;and\;47.02{\pm}13.96{\mu}g/dl$. Between Group IV and other groups showed statistically significant difference(p<0.05). 2. There was an increasing tendency of PbB, PbU, ALAU and ZPP according to the increase the mean air lead concentration, while ALAD has decreasing tendency. CPU and Hb did not show any constant tendency. 3. Correlation coefficients between PbB, PbU, ZPP, ALAU, ALAD, CPU, Hb and air lead concentration were 0.95, 0.83, 0.89, 0.72, -0.83, 0.51 and -0.45 respectively, and regression coefficient between PbB(Y) and PbA(X) was Y=126.8746X+16.9996(p<0.01).

아연 축적으로 인한 붕어(Carassius auratus) 조직의 변화 (Changes of Carassius auratus Tissues by Zn Accumulation)

  • 본 연구는 Zn의 노출조건에서 Zn의 농도가 어류의 조직과 생리에 미치는 영향을 조사하기 위하여 붕어의 아가미, 선장, 간, 뼈 및 근육 조직을 사용하였다. 아가미, 뼈와 근육 조직에서 Zn의 축적 농도는 노출 40일에 증가하였다. 또한 아가미 조직 내 Zn의 축적량은 다른 조직에 비해 높았으며, 모든 조직에서 노출기간이 걸어질수록 증가하였다. Zn에 노출된 아가미, 신장, 간과 근육 조직의 항산화효소의 활성은 노출기간이 걸어질수록 증가하였다. 또한 Zn에 노출된 아가미, 선장, 간과 근육 조직의 항산화효소 활성은 아가미와 간 조직에서 높았으며, 신장 조직에서 가장 낮은 활성을 보였다. 농도가 높고 노출기간이 걸어질수록 아가미 조직에서는 이차새변의 간격이 불규칙해지고, 점액 세포 수가 증가하였다. 곤봉화, 부종, 상피세포의 박리가 이차새변에서 관찰되었다. 또한 40 일간 Zn에 노출된 아가미 조직 내 미토콘드리아와 핵에서는 막의 손상이 관찰되었다. 신장 조직 내 사구체는 수축되어 보우만 주머니의 공간이 넓게 관찰되었다. 이러한 결과로 보아 고농도의 Zn 노출은 어류의 호흡과 배설 장애등 심각한 조직, 생리적 장애를 가져 올 수 있을 것으로 사료된다.

카드뮴과 아연 노출에 따른 야외종 장수깔따구(Chironomus plumosus)의 생태독성학적 반응 (Ecotoxicological Response of Cd and Zn Exposure to a Field Dominant Species, Chironomus plumosus)

  • 인간의 인위적 활동으로 인해 발생하는 중금속 중 카드뮴(Cd)과 아연(Zn)은 다양한 경로를 통해 하천으로 유입되어 서식생물에게 유해 영향을 준다. 본 연구에서는 도심 하천에서 우점하는 장수깔따구 (Chironomus plumosus)를 이용하여 Cd와 Zn 노출에 따른 생태독성학적 반응을 연구하였다. 생활사를 고려하여, 생존율, 성장율, 우화율, 성비와 하순기절 기형을 관찰하였다. 장수깔따구 생존율은 Cd와 Zn 노출에 따라 시간, 농도의존적인 경향을 나타냈다. 성장율은 Cd 노출 후 Day 4와 Day 7, 농도의존적인 감소를 보였으며, 고농도인 $50mg\;L^{-1}$에서 대조군에 비해 급격한 감소를 나타냈다. 또한, 우화율은 10, $50mg\;L^{-1}$ Cd에서 대조군에 비해 감소함을 관찰하였다. 성비 변화는 $100mg\;L^{-1}$ Zn에서 암컷의 비율이 증가였으나 농도가 높아짐에 따라 수컷의 비율이 높아짐을 보이며 성비불균형이 관찰되었다. 게다가, 장수깔따구의 하순기절에서는 대조군에 비해 Cd와 Zn 노출된 장수깔따구에서 기형이 관찰되었다. 이와 같은 결과는 수생태계로 유입되는 중금속이 하천의 하상저층에 서식하는 저서무척추동물에게 유해한 영향을 주며, 중금속 노출에 따른 생물학적 분석을 위한 현장 지표종으로서의 가능성을 보여주었다.

서울지역 집비둘기 Columba livia의 서식지별 중금속 오염 (Heavy Metal Contamination of Feral Pigeons Columba livia by Habitat in Seoul)

  • 집비둘기 Columba livia의 서식 환경별로 중금속 오염정도를 알아보기 위하여 서울시 관내의 상업지역, 공업지역, 공원지역 및 주거지역에서 집비둘기를 채집하여 간, 콩팥, 뼈, 근육 등 4개 조직에 함유된 중금속 농도를 측정하고 지역별로 비교하였다. 대부분의 중금속 원소 농도는 서식환경에 따라 유의한 차이를 나타내었다. 주거지역에 서식하는 집비둘기의 간에 함유된 철과 콩팥에 함유된 아연의 농도는 다른 지역보다 현저하게 낮았다. 망간은 모든 조직에서 공업지역이 높았으며, 구리는 모든 조직에서 공원지역이 다른 지역보다 현저히 높았다. 뼈의 납 함량은 주거지역이 가장 낮았고 상업지역이 가장 높았으며, 콩밭의 카드뮴 함량은 공원지역이 다른 지역보다 높은 것으로 나타났다. 특히 일부 조직의 철, 아연 및 납 농도는 교통량과 관련이 있는 것으로 나타났다.

고해상도 CCD카메라를 이용한 Single-Frame PIV 속도장 측정기법 개발 (Development of Single-Frame PIV Velocity Field Measurement Technique Using a High Resolution CCD Camera)

  • Although commercial PIV systems have been widely used for the non-intrusive velocity field measurement of fluid flows, they are still under development and have considerable room for improvement. In this study, a single-frame double-exposure PIV system using a high-resolution CCD camera was developed. A pulsed Nd:Yag laser and high-resolution CCD camera were synchronized by a home-made control circuit. In order to resolve the directional ambiguity problem encountered in the single-frame PIV technique, the second particle image was genuinely shifted in the CCD sensor array during the time interval dt. The velocity vector field was determined by calculating the displacement vector at each interrogation window using cross-correlation with 50% overlapping. In order to check the effect of spatial resolution of CCD camera on the accuracy of PIV velocity field measurement, the developed PIV system with three different resolution modes of the CCD camera (512 ${\times}$ 512, lK ${\times}$ IK, 2K ${\times}$ 2K) was applied to a turbulent flow which simulate the Zn plating process of a steel strip. The experimental model consists of a snout and a moving belt. Aluminum flakes about $1{\mu}m$ diameter were used as scattering particles for the liquid flow in the zinc pot and the gas flow above the zinc surface was seeded with atomized olive oil with an average diameter of 1-$3{\mu}m$. Velocity field measurements were carried out at the strip speed $V_s$=1.0 m/s. The 2K ${\times}$ 2K high-resolution PIV technique was significantly superior compared to the smaller pixel resolution PIV system. For the cases of 512 ${\times}$ 512 and 1K ${\times}$ 1K pixel resolution PIV system, it was difficult to get accurate flow structure of viscous flow near the wall and small vortex structure in the region of large velocity gradient.

Micronucleus Test for the Classification of Chemical Mutagenicity according to Globally Harmonized System

  • To classify the chemical hazard according to globally harmonized system of classification and labeling of chemicals (GHS), we investigated the genotoxicity of three chemicals, methyl myristate, 2-ethylhexanoic acid zinc salt, N,N,N',N'-tetrakis(2-hydroxyethyl) ethylenediamine, using male ICR mice bone marrow cells for the screening of micronucleus induction. Although these three chemicals have already been tested numerous times, a micronucleus test has not been conducted. The seven week-old male ICR mice were tested at three dosages for the three chemicals, respectively. After 24 h of oral administration with the three chemicals, the mice were sacrificed and their bone marrow cells were prepared for smearing slides. As a result of counting the micronucleated polychromatic erythrocyte (MNPCE) of 2,000 polychromatic erythrocytes, all treated groups expressed no statistically significant increase of MNPCE compared to the negative control group. There were no clinical signs related with the oral exposure of these three chemicals. It was concluded that these three chemicals did not induce micronucleus in the bone marrow cells of ICR mice, and there was no direct proportion with dosage. These results indicate that the three chemicals have no mutagenic potential under each test condition, and it is not classified these chemicals as mutagens by GHS.
