우귀음이 Zinc에 의한 신경교세포의 고사(Apoptosis)에 미치는 영향 (Protective Effects of Ukyium(우귀음, Yougui-yin) in Zinc-induced Apoptosis of C6 Glial Cells)

  • Objectives : The objective of the current study is to determine the protective effect of Ukyium(Yougui-yin) on the apoptosis induced by zinc. Methods : Zinc is known to generate reactive oxygen species (ROS), including superoxide anion ($O_2$) and hydrogen peroxide ($H_2O_2$), which eventually contribute to cytotoxicity in a variety of cell types. We investigated the viablity of cells, $H_2O_2$ generation, chromatin condensation and nuclear fragmentation in Hoechst dye staining and $IkB-{\alpha}$ degradation in C6 glial cells of $ZnCl_2$ between pretreatment- and not pretreatment-group with Ukyium. The former methods were researched by Time- and Dose-dependent manners. Results : We demonstrated that pretreatment with Ukyium prevented zinc-induced cell death of C6 glial cells and apoptotic characteristics including chromatin condensation and nuclear fragmentation. Ukyium also prevented $H_2O_2-induced$ cell death. We further confirmed that Ukyium decreased zinc-induced generation of $H_2O_2$ and inhibited degradation of $IkB-{\alpha}$ by zinc in C6 glial ceHs. Conclusions : These data indicated that Ukyium (Yougui-yin) prevents zinc-induced apoptotic death of C6 glial cells via inhibition of ROS generation, such as $H_2O_2$ as well as inhibition of $IkB-{\alpha}$ degradation.

  • 황산 용액에서 Zinc-ferrite의 용해에 대한 반응속도론을 황산 용액의 반응온도와 농도 변화에 대해 조사하였다. 반응율(R)과 겉보기 반응 속도상수(K)는 황산 용액의 온도와 농도가 클수록 증가한다. Zinc-ferrite의 반응속도는 반응초기에서 $1-(1-K)^{1/3}=Kt$와 같은 속도식을 적용할 수 있다. 용해에 대한 활성화 에너지는 황산 용액의 농도에 관계없이 약 16.3kcal/mole 이다. Zinc-ferrite가 황산 용액에서 용해할 때는 Zinc-ferrite의 화학 양론적 조성으로 용해되며, Fe 또는 Zn의 단독으로는 용해되지 않는다.

아연 섭취 수준이 고지방 식이로 유도된 비만쥐의 면역 기능에 미치는 영향 (Effects of Zn Intake on Immune Responses in High Fat Diet-induced Obese Rats)

  • The purpose of this study was to investigate the effect of zinc intake on immune responses in high fat diet induced obese rats. The immune status was assessed by the measurements of immunoglobulins (IgG, A, and M) production by SRBC(sheep red blood cell) with mitogen in vivo. The delayed type hypersensitivity(DTH) response was also measured as an index of cell mediated immunity. The re sults are summarized as follows : 1) There were no significant differences in the feed intake, weight gain and feed efficiency ratio of obese rats by the different dietary zinc levels. 2) White blood cell (WBC) counts were significantly affected as the dietary zinc levels decreased. The capacity of Ig M production in obese rats was significantly higher in normal zinc group than that of low and high zinc group. Cell mediated immune response evaluated by means of DTH testing has also been found to be highly impaired by zinc deficiency and overload. From these observations, it was suggested that adequate levels of zinc may promote the immune function of obese individuals. The relationship and its functional role of the zinc in obesity remains to be further studied.

Zinc Restored the Decreased Vascular Smooth Muscle Cell Viability under Atherosclerotic Calcification Conditions

  • Zinc is considered to be involved in maintaining healthy vascular condition. Atherosclerotic calcification of vascular smooth muscle cells (VSMCs) occurs via the mechanism of cell death; therefore, cell viability is a critical factor for preventing VSMC calcification. In this study, we tested whether zinc affected VSMC viability under both normal physiological non-calcifying (0 mM P) and atherosclerotic calcifying conditions (3 and 5 mM P), since VSMC physiological characters change during the VSMC calcification process. The study results showed that an optimal zinc level ($15{\mu}M$) restored the decreased VSMC viability which was induced under low zinc levels (0 and $1{\mu}M$) and calcifying conditions (3 and 5 mM P) at 9 and 15 days culture. This zinc-protecting effect for VSMC viability is more prominent under atherosclerotic calcifying condition (3 and 5 mM P) than normal condition (0 mM P). Also, the increased VSMC viability was consistent with the decreased Ca and P accumulation in VSMC cell layers. The results suggested that zinc could be an effective biomineral for preventing VSMC calcification under atherosclerotic calcifying conditions.

비타민 E 첨가가 아연이 결핍된 흰쥐의 혈액과 간의 지질과산화물 수준에 미치는 영향 (Effect of Vitamin E Supplementation on Lipid Peroxide Levels of Blood and Liver in Zinc Deficient Rats)

  • To study antioxidant role of zinc, the effects of dietary zinc deficiency and vitamin E supplementation on lipid peroxidation were studied. Levels of zinc and vitamin E in blood and liver were also measured. Forty Sprague-Dawley male rats aging 8 weeks old were used as experimental animals. Zinc deficient diet (Zn, 0 ppm), zinc normal diet (Zn,36.5 ppm), and vitamin E supplemented diet (1,000 IU ${\alpha}$-tocopherol/kg of diet) were used as experimental diet. During the first three weeks, rats were divided into zinc normal (ZnN, 8 animals) and zinc deficient (ZnD, 32 animals) group. Eight rats from each group were sacrificed to get blood and liver after 3 weeks of experiment. The remaining 24 zinc deficient rat were then divided into zinc normal (ZnDN), zinc deficient (ZnDD), vitamin E supplemented (ZnDE) diet groups. After another 3 weeks of experiment, all animals were sacrificed as well. Thiobarbituric acid reactive substanc (TBARS) levels in plasma and liver, conjugated diene levels in liver were measured as lipid peroxidation index. There were no significant differences in food intake, body weight gain, and food efficiency ratio among groups. Weights of liver per 100 g body weight were not significantly different. There were no significant differences in Zn levels in serum. Plasma and liver TBARS level, and liver conjugated diene level were significantly lower in ZnDE than in ZnDN or ZnDD, and significantly higher in ZnDD than in ZnDN. Therefore, it seems that lipid peroxidation is accelerated by dietary zinc deficiency and recovered partly by vitamin E supplementation.

Ni-Zn 합금도금에 미치는 초음파의 영향(I) (Effects of Ultrasonic Waves on Electrodeposition on Nickel-Zinc Alloys(I))

  • The nickel-zinc alloy depositions have been studied in nickel chloride added chloride baths, to find out the effects of ultrasonic irradiation for the electrodeposition processes. The compositions of deposited alloys, the current efficiencies and the metallographic appearances in various conditions of Electrodeposition were investigated, in the range of ultrasonic irradiation of 50,500 and 1,000 Kc/s respectively. The results obtained are as follows; 1. Generally the nickel deposition process is more preferably activated than that of zinc by the ultrasonic irradiation. 2. The radios of nickel to zinc in the deposit are higher according to increase of nickel ion concentration and bath temperatures in irradiated baths. 3. The current efficiencies are also higher in the irradiated baths, so that the depolarization effect is noticeable. 4. The brightness and leveling effect of the deposits are appreciably better in the irradiated baths than in non-irradiated in 0.3M and 0.6M of nickel chloride and zinc chloride solutions and the current density of 3A/$dm^2$. 5. The mechanism of alloy deposition has been tentatively suggested in the case of ultrasonic irradiation.

Effect of Sodium Chloride on Weight Loss of AA1100 Aluminum Alloy and SGACD Zinc coated Steel Lap Joint

  • This research aims to study an effect of sodium chloride solution concentration on the corrosion rate of AA1100 aluminium alloy and SGACD zinc coated steel lap joint with a test duration of 30 days and a test temperature of $45^{\circ}$. The summarized results are as follows. Increase of the NaCl solution concentration increased the weight loss of Al, corrosion rate of Al, weight loss of Fe and also decreased the corrosion rate of Fe. Increase of the test duration affected to increase the weight loss and corrosion rate of Al and also decrease the weight loss and corrosion rate of Fe. The corrosion that was formed in a lap joint consisted of the uniform corrosion on the surface of the metals and the galvanic corrosion in the lap area of the joint. The maximum weight loss of AA 1100 aluminium and SGACD zinc coated steel that was occurred in the sodium chloride with 3.25% was 2.203% and 3.208%, respectively.. The maximum corrosion rate of AA 1100 aluminium and SGACD zinc coated steel that was occurred in 4.00% and 3.5% sodium chloride solution was 0.156 mm/year and 0.479 mm/year, respectively.

Zinc and Selenium Requirements for Glutathione Peroxidase Activity and Cell Survival in Chinese Hamster Ovary Cells Overexpressing Metallothionein

  • Many defined cell culture media were formulated over 3() years ago and may be deficient in certain micronutrients whose essentiality has only subsequently been recognised. The objective of this study was to evaluate whether alpha-minimal essential medium (MEM) supplemented with 10% foetal bovine serum contained sufficient selenium for optimal activity of the selenium containing enzymes cytosolic glutathione peroxidase (cGPx) and phospholipid hydroperoxide glutathione peroxidase (PHGPx) in cultured Chinese hamster ovary (CHO) cells. Additionally, the effect of zinc deficiency and metallothionein (MT) overexpression on cGPx and PHGPx activity was studied. The addition of 100 nM of selenous acid to the culture medium increased cGPx expression by 10-fold and PHGPx by about 2-fold in both wild-type CHO-K1 cells and CHO-K1 cells overexpressing mouse MT-1. Zinc deficiency had no significant effect on enzyme activity, but cells overexpressing mouse MT-1 had higher levels of cGPx activity. Zinc deficiency decreased cell survival but overexpression of MT-1 was partially protective, probably because its presence in quantity favoured the uptake, sequestration and cellular retention of any remaining zinc. This study demonstrates that selenium in complete alpha-MEM is insufficient for optimal cGPx and PHGPx activity and may compromise the cellular response to oxidative stress.

Effect of Zinc-enriched Yeast FF-10 Strain on the Alcoholic Hepatotoxicity in Alcohol Feeding Rats

  • The possible protective effects of highly zinc-containing yeast Saccharomyces cerevisiae, FF-10 strain, isolated from tropical fruit rambutan on acute alcoholic liver injury in rats were evaluated. Zinc concentration in this strain was 30.6mg%. The activities of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), and $\gamma$-glutamyl transpeptidase ($\gamma$-GTP) were highly increased when alcohol was treated, relative to the normal rats. Also, a highly significant increase in the blood alcohol and acetaldehyde levels by alcohol treatment was observed. Administration of FF-10 strain markedly prevented alcohol-induced elevation of the activities of serum ALT, AST, and $\gamma$-GTP, and the levels of blood alcohol and acetaldehyde, and these reduced levels reached to that of normal rats. As compared with alcohol treated control rats, the FF-10 strain supplementation showed highly decreased the triglyceride concentration in serum. Alcohol treatment induced the marked accumulation of small lipid droplets, hepatocytes necrosis, and inflammation, but FF-10 strain administration attenuated to alcohol-induced accumulation of small lipid droplets and hepatocyte necrosis in the liver. Therefore, the current finding suggests that zinc-enriched yeast FF-10 strain isolated from tropical fruit rambutan may have protective effect against alcohol-induced hepatotoxicity.

Investigation of the Effective Range of Cathodic Protection for Concrete Pile Specimens Utilizing Zinc Mesh Anode

  • A zinc mesh sacrificial anode cathodic protection method is recently being developed to protect the reinforced concrete structure in a marine environment. However, comprehensive information regarding the cathodic protection technology applied to reinforced concrete test specimens utilizing zinc mesh sacrificial anodes remains limited. Particularly, no research has investigated the effective range of sacrificial anode cathodic protection in a reinforced concrete structure regarding the transmission of protection current from zinc mesh sacrificial anode to the reinforced concrete structure, particularly concerning effects of temperature variations. This study examined the distribution of potential and current using a long single rebar and several segment reinforcing bars inside a horizontal beam. Vertical pile specimens were applied with a zinc mesh sacrificial anode to simulate concrete bridges or harbor structures. To check the effect of cathodic protection, cathodic protection potential and current of the reinforced concrete specimens were measured and 100 mV depolarization criterion test was performed. It was confirmed that effect of cathodic protection varied depending on resistivity and temperature. The cathodic protection test of pile specimens revealed that the maximum reachable range of cathodic protection current was 10 cm from the waterline as observed in the experiment.
