An Evaluation on Electrochemical Polarization Characteristics and Material Degradation for Cr-Mo-V Steel (Cr-Mo-V강의 전기화학적 분극 특성과 재질열화 평가)

  • This research is described on the applicability of a electrochemical techniques for evaluating nondestructive material degradation with various polarization characteristics for Cr-Mo-V steel. The applied electrochemical technique is anodic polarization test which are widely used to evaluate the corrosion rate and/or sensitization at depleted zone of strengthening elements mainly caused by thermal experience for stainless steels. The evaluation of material degradation is performed by small punch test which has been well known as micromechanics test method using specimen size of $10{\times}10{\times}0.5mm$. The 1,000hrs aged material at $630^{\circ}C$ shows the highest material degradation$({\Delta}[DBTT]_{SP})$, but the 2,000hrs and 3,000hrs aged materials show the decrease of ${\Delta}[DBTT]_{SP}$ as aging time increases. It is observed that the difference of current density $({\Delta}I_{FP}\;and\;{\Delta}I_{SP})$.

Studies on Antioxidant Activity and Inhibition of Nitric Oxide Synthesis of Germinated Brown Rice Soaked in Mycelial Culture Broth of Phellinus linteus (상황버섯균사체배양액에 침지한 발아현미의 항산화 및 nitric oxide 합성저해에 관한 연구)

  • This study investigated the effects on the biological activities of germinated brown rice soaked in mycelial culture broth of Phellinus linteus. The level of free amino acid was higher in the GBRP extract than those of BR and GBR. The major free amino acids were alanine, valine, isoleucine and methionine in both extracts. The level of ${\gamma}$-aminobutyric acid (GABA) was also increased significantly in the GBR and GBRP. Antioxidant activities of methanol extract of BR, GBR and GBRP were measured by using DPPH radical scavenging and SOD-like activity. Antioxidant activities showed the highest level of 83% and 76% when 100 mg/ml GBR and GBRP, respectively. Stimulation of the macrophages RAW264.7 cells with lipopolysaccharide (LPS) resulted in increased production of nitric oxide (NO) in the medium. However, the methanol extract of GBR and GBRP showed marked inhibition of NO synthesis in a does-dependant manner. These results showed that GBR and GBRP were significant role for activation of immune system in the pathogenesis of inflammatory diseases.

Use of Electromagnetic Inductance for Salinity Measurement in Reclaimed Saline Land (전자장 유도 장치를 이용한 간척지 토양의 염농도 측정)

  • Mapping of salinity distribution in the reclaimed lands was attempted by using the electromagnetic inductance technique. Field study was conducted to monitor ground conductivity with an electromagnetic inductance, EM 38 (Geonics), and electrical conductivity of the saturated extract, ECe of the soils, at the Daeho reclaimed land. EM values of horizontal mode, EMh, and vertical mode, EMv, mode were recorded at the interval of $2m{\times}2m$ from the ground. Soil samples were taken through the profile down to 100cm for calibration. ECe of poor drained area of Daeho, were in the range of $19.50-91.50ds\;m^{-1}$, while ECe of well-drained area ranged from $1.10-34.40ds\;m^{-1}$. Multiple regression equations for the measured EMv, EMh, and ECe were highly significant. The EMh showed higher correlation with ECe than EMv. With the multiple equation, ECgM could be calculated. Correlation between ECe and ECgM was the highest ($r=0.753^{***}$), when EMI readings were taken on the ground. The relationships were highly significant below 30 cm height of measurement, With the EM38 measurement, the salinity distribution was effectively expressed for the experimental filed in Daeho reclaimed land.

Safety Evaluation from Aflatoxin risk of Korean Angelicae Gigantis Radix Based on Critical Control Points (중점관리기준에 기초한 국내생산 당귀의 산지 수확 후 아플라톡신의 안전성 평가 연구)

  • HACCP methodology was applied in the post-harvest processing and storage of domestic medicinal produces. Particularly in terms of mold and mycotoxin contamination, candidate critical control points (CCP) in the conventional practice in Korean farms were selected and monitored by comparing with on the standard guided processing and storage. When each processing of Angelicae Gigantis Radix were assessed for their safety, the drying steps such as the sun drying or the thermal drying depending on each farm made differences in mold contamination. Moreover, the storage conditions before or after the processing were another critical determinant in the fungal contamination. In other words, storage under $4{\circ}C$ rather than at room temperature was favorable for reducing mold growth in the harvested crops. Occurrence rate of Aflatoxin $B_1 \;(AFB_1)$ in Angelicae Gigantis Radix were 12.8%, but amount of $AFB_1$ in all the collected samples were below 10 ppb regulatory limit allowed in Korea. However, for a few samples of Angelicae Gigantis Radix, still relatively high levels of total amount of the major aflatoxins (aflatoxin $B_1\; +\; B_2\; +\; G_1\; +\; G_2$) were observed around 0.18~49.94 ppb, which is not regulated presently in Korea. It thus can be suggested that post-harvest processing and storage of Korean medicinal crops need further investigation and monitoring to establish the Good Agricultural Practice (GAP), particularly to minimize microbial risk including mold and mycotoxin contamination under the changing climate. Additionally, it is also warranted for new enacting of regulatory limits for total aflatoxins in the medicinal crops.

Reduction of causative organisms associated with dental caries and alteration of oral hygiene status in connection with toothbrushing instruction for preschool children (미취학 아동의 칫솔질 교육에 따른 치아우식원인균 감소와 구강위생상태 변화)

  • Objectives: This study was conducted to reveal the biological basis of dental plaque and preschool children's dental health status through repetitive and continuous toothbrushing instruction to preschool children, using quantitative criteria, and to emphasize the importance of the role of an oral health manager for the prevention of dental caries. Methods: After IRB approval, toothbrushing instruction and oral microbial specimen collection were conducted with children of preschool age at the Daycare Center attached to D. University, Busan. Specimens from 27 children were characterized by analysis with real-time multi-chain enzymatic polymerization reaction technique at an agency specializing in genetic analysis. Results: In a survey of the children's dental health behaviors, 48.1% responded that the toothbrushing time is 91 to 150 seconds; 66.7% responded that the frequency of toothbrushing is more than three times per day; 81.5% stated that they performed tongue brushing; and 81.5% brushed their teeth. Regarding levels of dental cariogenic bacteria, after children received continuous toothbrushing instruction, there were reductions in both Streptococcus mutans and Streptococcus sobrinus. Regarding toothbrushing time, the time increased in relation to the presence of instruction. Regarding Quigley Hein Index, the index decreased in the early and middle periods upon addition of instruction, then increased in the middle and later periods. Conclusions: Through persistent and repetitive toothbrushing instruction, healthy behavior can be positively established in children of preschool age; it is necessary to prepare institutional measures so opportunities for dental health education can be consistently provided and maintained. In the future, it will be necessary to conduct a follow-up study to verify the factors affecting toothbrushing time and volume of oral pathogens.

Studies on Antioxidant Activity and Inhibition of Nitric Oxide Synthesis from Codium fragile (청각추출물의 항산화 및 일산화질소 합성 저해 연구)

  • This study was carried out to investigate the biological effects from Codium fragile. Methanol extract of Codium fragile increased two times at 2500 ㎍/ml the growth of Lactobacillus plantarum that associated with probiotic properties of lactic acid bacteria of Kimchi. Ethyl acetate extract of Codium fragile inhibited the cellulase activity up to approximately 60% at $2500\;{\mu}g/ml$. Methanol extract of Codium fragile was fractionated into several subfractions and their antioxidant activities were measured by using DPPH radical scavenging and SOD-like activity. Especially the antioxidative activity of ethyl acetate fraction was shown higher than that of other fractions and its fraction showed higher contents of total phenolic compounds, indicating the positive relationship between DPPH radical scavenging effect and total polyphenol content. Stimulation of the macrophages RAW264.7 cells with lipopolysaccharide (LPS) resulted in increased production of nitric oxide (NO) in the medium. However, the methanol extract of Codium fragile showed marked inhibition of NO synthesis in a dose-dependent manner. This result suggest that Codium fragile plays significant role for activation of immune system in the pathogenesis of inflammatory diseases.

Development of Loop-Mediated Isothermal Amplification Targeting 18S Ribosomal DNA for Rapid Detection of Azumiobodo hoyamushi (Kinetoplastea)

  • Ascidian soft tunic syndrome (AsSTS) caused by Azumiobodo hoyamushi (A. hoyamushi) is a serious aquaculture problem that results in mass mortality of ascidians. Accordingly, the early and accurate detection of A. hoyamushi would contribute substantially to disease management and prevention of transmission. Recently, the loop-mediated isothermal amplification (LAMP) method was adopted for clinical diagnosis of a range of infectious diseases. Here, the authors describe a rapid and efficient LAMP-based method targeting the 18S rDNA gene for detection of A. hoyamushi using ascidian DNA for the diagnosis of AsSTS. A. hoyamushi LAMP assay amplified the DNA of 0.01 parasites per reaction and detected A. hoyamushi in 10 ng of ascidian DNA. To validate A. hoyamushi 18S rDNA LAMP assays, AsSTS-suspected and non-diseased ascidians were examined by microscopy, PCR, and by using the LAMP assay. When PCR was used as a gold standard, the LAMP assay showed good agreement in terms of sensitivity, positive predictive value (PPV), and negative predictive value (NPV). In the present study, a LAMP assay based on directly heat-treated samples was found to be as efficient as DNA extraction using a commercial kit for detecting A. hoyamushi. Taken together, this study shows the devised A. hoyamushi LAMP assay could be used to diagnose AsSTS in a straightforward, sensitive, and specific manner, that it could be used for forecasting, surveillance, and quarantine of AsSTS.

A Shared Electronic Medical Record for Lung Cancer Clinic (폐암 클리닉을 위한 공유 전자의무기록)

  • Since the year 2000, lung cancer has become the leading cause of cancer death in South Korea as in many other parts of the world. The current multidisciplinary approach for lung cancer includes a wide range of modalities, not only surgery, radiotherapy, medical drug therapy but also pain control, as well as social and psychological support. Therefore, thoracic surgeons, radiologists, nuclear medicine specialists, anesthetists, psychologist, nurses and social workers as well as medical doctors care for lung cancer patients. Sharing a common treatment protocol and optimal communication are vital aspects of shared care both from a medical and cost-effectiveness point of view. We developed a shared electronic medical record (SEMR) for treating patients with lung cancer in a university hospital to facilitate the sharing protocols and communications between doctors involved in a lung cancer clinic. A SEMR system was developed within a order communication system(OCS) for a lung cancer clinic. The records of radiological, laboratory and pathological studies as well as the records of surgery, chemotherapy, and radiotherapy were stored and presented to all doctors who treat the same patient. Every doctor was allowed to change his/her own records. They could review other doctor s records but could not alter them. With the SEMR, it was expected that the time to complete the medical records for one patient could be reduced because it was easy to review all the data from the other doctors who share the same patient. In addition, the confidence of the doctors who share a common treatment protocol would be higher. Therefore, a shared electronic medical record is expected to improve the quality of patient care.

Purification and Properties of a Novel Extracellular Agarase from Marine Bacterium, Sphingomonas paucimobilis AS-1 (해양미생물 Sphingomonas paucimobilis AS-1이 생산하는 새로운 extracelluar agarase의 정제 및 특성)

  • An agar-degrading marine bacterium, strain AS-1 was isolated from the seawater. The strain AS-1 was identified as Sphingomonas paucimobilis (90% probability) by VITEK. The optimum medium for agarase activity of the isolated strain was determined to be marine medium, marine broth 2216 containing 0.1% agar as carbon source. An extracellular agarase was purified 104-fold from the culture supernatant by ammonium sulfate precipitation, ion exchange chromatography and gel filtration methods. The molecular weight of the purified enzyme was estimated to be 80 kDa by SDS-PAGE. The optimum pH and temperature for activity were 7.0 and $40^{\circ}C$, respectively. Antioxidative activity of the strain AS- was 72% in the supernatant cultured for 12 h. The culture supernatant of the strain AS-1 showed antibacterial activity against bacteria causing putrefaction and food poisoning such as Escherichia coli, Staphylococcus aureus and Proteus vulgaris. However, the cell growth of the lactic aicd forming strain, Lactobacillus plantarium was promoted by the treatment of 10% culture supernatant of an agar-degrading strain.

Antimicrobial and Antioxidant Activities and Inhibition of Nitric Oxide Synthesis of Oak Wood Vinegar (참나무 목초액의 항균 및 항산화 활성과 일산화질소 합성 저해연구)

  • This study was carried out to investigate the biological effects oak wood vinegar. Antimicrobial activity was tested in five microbial species at the concentration of 5 to $50{\mu}l$ of oak wood vinegar by paper disc method. Growth of P. oleovoranse, P. vulgaris, E. coli, S. aureus and Prevotella intermedia was inhibited at a dose of as low as $50{\mu}l$ of oak wood vinegar. Antioxidant activities were measured by using DPPH radical scavenging and SOD-like activity. DPPH radical scavenging and SOD-like activities were 90% and 65% at the concentration of $25{\mu}l\;and\;50{\mu}l$ of oak wood vinegar, respectively. Stimulation of the macrophages RAW264.7 cells with lipopolysaccharide (LPS) resulted in increased production of nitric oxide (NO) in the medium. However, the oak wood vinegar showed marked inhibition of NO synthesis in a dose-dependent manner. This result suggest that oak wood vinegar plays significant role for activation of immune system in the pathogenesis of inflammatory diseases.
