Enzyme Inhibition Activities of Ethanol Extracts from Germinating Rough Rice (Oryza sativar L.) (발아기간에 따른 벼(Oryza sativa L.)의 부위별 효소저해활성)

  • This study investigated the enzyme inhibitory effects of ethanol extracts from the different parts of rough rice (Oryza sativar L.) from its germination period. Rough rice was germinated at $37^{\circ}C$ for 6 days, then separated into hull+sprout and brown rice. ${\alpha}$-Glucosidase inhibitory activity had the highest value (39.38%) in hull+sprout extracts after 5 days of germination. ${\alpha}$-Amylase and DPP-4 inhibitory activity had the highest values (75.32% and 47.77%, respectively) in hull+sprout extracts after germination for 5 days. ACE inhibitory activity of hull+ sprout extracts increased from 43.16% at the beginning to 58.60% at 5 days, while brown rice extracts increased this activity from 0.88% at the beginning to 14.50% at 4 days. The xanthine oxidase inhibitory activity of hull+ sprout extracts increased from 62.02% at the beginning to 64.49% at one day, and then decreased. Lipase inhibitory activity had its highest value (55.17%) in hull+sprout extracts after germination for 5 days. These results indicate that the optimal germination period for increasing enzyme inhibitory activities may be 5 days, and that hull+sprout extracts have a higher enzyme inhibition activity than brown rice.

Evaluation of Biological Activities of Wheat Sprouts with Different Extraction Solvents (새싹 밀의 추출용매에 따른 생리활성 평가)

  • The purpose of this study was to examine biological activities, including total contents of polyphenol, antioxidant activities, inhibitory activities of tyrosinase, and protective effect against oxidative stress in the HepG2 cells of ethanol extracts from wheat sprout. The antioxidant activity of extracts was determined by ABTS and DPPH radical scavenging activities. Ethanol extracts were tested using different ethanol concentrations (0%, 30%, 50%, 80% and 95%, respectively). The highest amount of total polyphenol was extracted by 50% and 80% ethanol which was 26.3 and 26.8 mg gallic acid equivalents/g sample, respectively. High levels of ABTS and DPPH radical scavenging activity were found in 50% ethanol (26.7 and 15.0 mg TEAC/g sample, respectively) and 80% (24.3 and 16.1 mg TEAC/g sample, respectively) ethanol extracts. Also, 50% and 80% ethanol extracts indicated higher inhibitory activities of tyrosinase compared with other extracts. In the cell-based assay, pre-treatment of the HepG2 cells with wheat sprout extracts prevented the cell damage induced by TBHP (tert-butyl hydroperoxide). The results of this study indicate that wheat sprout has significantly higher diverse biological activities and apparently has significant health benefits.

Anti-oxidative and Anti-hyperglycemia Effects of Triticum aestivum Wheat Sprout Water Extracts on the Streptozotocin-induced Diabetic Mice (밀순 물추출물의 항산화 효과 및 Streptozotocin으로 유발한 당뇨 흰쥐에서 혈당강하에 미치는 영향)

  • This study was performed to investigate the anti-hyperglycemia effects of the Triticum aestivum wheat sprout (TAWS) water extracts in the diabetic mice. Diabetic experimental model was established by intraperitoneal injection of streptozotocin into male Balb/c mice. Mice were divided into five groups: normal (CON), diabetic control (DM), and three experimental groups (DM-100, diabetes with TAWS extracts 100mg/kg; DM-50, diabetes with TAWS extracts 50 mg/kg; DM-25, diabetes with TAWS extracts 25 mg/kg). TAWS extracts were administered orally in diabetic mice. Body weight, food intake, and blood glucose levels were recorded for 12 days and blood insulin levels were measured at the day 12. Oral administration of TAWS extracts reduced slightly food intake and induced a little body weight gain in DM-100 groups. The blood level of glucose was decreased in the dose-dependent manner; 55% in the DM-100 group and 39.7% in the DM-50 group. The blood level of insulin also was improved 10 folds in the DM-100 group and 3.6 folds in the DM-50 group compared to the DM group. The contents of total phenolic compounds and total flavonoids in 1 g dry mass of TAWS extracts were 6.6 mg of tannic acid equivalents and 1.0 mg of 8-hydroquinolline equivalents, respectively. In addition, the antioxidant and DPPH radical scavenging activity of TAWS extracts were 1.2 mM and 1.8 mM ascorbic acid equivalents, respectively. These results suggest that TAWS water extracts could contribute to attenuate clinical symptoms of diabetes mellitus.

Anti-oxidative effects of broccoli (Brassica oleracea var. italica) sprout extract in RAW 264.7 cell and cisplatin-induced testicular damage

  • Background: Brassica oleracea var. italica (broccoli), a rich source of antioxidants, can prevent various diseases and improve human health. In this study, we investigated the antioxidative effects of broccoli sprout extract on oxidative stress induced by lipopolysaccharide and cisplatin in cell and organ tissue models. Methods: Antioxidative effect of BSE was evaluated using DPPH and ABTS in RAW 364.7 cells, and effects of BSE on testes were investigated using Cisplatin-induced testicular damage model with an in vitro organ culture system. Results: The DPPH assay showed that the antioxidant activity of the alcoholic broccoli sprout extract was higher than that of the water extract. Additionally, the expression levels of antioxidation-related genes, Nrf2, Gsr, HO-1, and catalase, were significantly increased in broccoli sprout extract-treated RAW 264.7 cells, and the extract suppressed lipopolysaccharide-induced mitochondrial dysfunction. Based on the results in the RAW 264.7 cell culture, the antioxidative effects of the extracts were investigated in a mouse testis fragment culture. The expression of Nrf2, HO-1, and Ddx4 was clearly decreased in cisplatin-treated mouse testis fragments and not in both broccoli sprout extract- and cisplatin-treated mouse testis fragments. In addition, the oxidative marker O-HdG was strongly detected in cisplatin-treated mouse testis fragments, and these signals were reduced by broccoli sprout extract treatment. Conclusions: The results of this study show that broccoli sprout extracts could serve as potential nutraceutical agents as they possess antioxidant effects in the testes.

Comparison of antioxidant activities of ethanol extracts from seven species of wild edible plants in Korea (국내 야생식용식물 7종 에탄올 추출물의 항산화 활성 비교)

  • In this study, we investigated the potential of 70% ethanol extracts from wild edible plants (Pueraria lobata sprout, Rosa multiflora sprout, Artemisia princeps leaf, Diospyros kaki leaf, Morus alba leaf, Robinia pseudoacacia flower, Inula britannica var. japonica flower), as natural antioxidants. The antioxidant contents and activities of extracts were examined using various methods. The measurements of total polyphenol content revealed that Rosa multiflora sprout extract had the highest value and total flavonoid content showed that Diospyros kaki leaf extract had the highest value. Antioxidant activities were the highest in Rosa multiflora sprout for DPPH (IC50 232.52 ㎍/mL), ABTS+ (IC50 470.10 ㎍/mL), superoxide- (IC50 431.88 ㎍/mL), nitrite (IC50 363.38 ㎍/mL) scavenging activity, and reducing power (2.47 O.D.). These results suggest that the ethanolic extract of Rosa multiflora sprout is a potential source of natural antioxidants.

Antioxidant activities of commonly used Brassica spp. sprout vegetables in Korea (국내 다소비 십자화과 새싹채소 추출물의 항산화 활성)

  • Brassica spp. vegetables have been known to have biological activities such as anti-cancer and anti-inflammatory activities. In this study, we investigated the in vitro physicochemical characteristics and antioxidant activities of commonly used Brassica spp. sprout vegetables such as kohlabi (Brassica oleracea var. gongyloides), red radish (Raphanussativus L. var. sativus), broccoli (B. oleracea var. italica), cabbage (B. rapavar. glabra Regel), rape (B. napus), radish (R. sativus), and tatsoi (B. campestris var. narinosa) sprouts. Our results showed that the vegetables with the highest total phenolics contents were the radish sprout ($24.40{\pm}1.24mg\;TAE/g$) and kohlabi sprout extracts ($23.97{\pm}0.46mg\;TAE/g$). Furthermore, the vegetable with the highest total flavonoid content was the radish sprout extract ($15.30{\pm}1.35mg\;CE/g$). However, the kohlabi sprout extract showed the highest DPPH radical scavenging value ($IC_{50}=1.95mg/mL$) and ORAC value (79.03 mM TE/g). In addition,the six kinds of Brassica spp. sprout vegetable extracts, except tatsoi, significantly inhibited the reactive oxygen species (ROS) production and showed that intracellular oxidative stress is closely related tothe accumulation of differentiated adipocytes and fat during the adipogenesis of 3T3-L1 preadipocytes. These results suggest that Brassica spp. sprout vegetables, especially kohlabi and radish sprout extracts, can be used to develop natural antioxidants.

Protective Effects of Plant Extracts on the Hepatocytes of Rat Treated with Carbon Tetrachloride (사염화탄소를 투여한 흰쥐의 간세포에 대한 식물 추출물들의 보호효과)

  • To investigate the effects of plant extracts on the protection against liver damage by $CCl_4$ in rat, two kinds of experiment were performed, firstly by the primary hepatocyte culture and secondly by the animal feeding. The primary hepatocyte culture with the extracts of pine leaf, soybean sprout and mugwort showed significantly low activities (p<0.01∼0.05) of aspartate aminotransferase (AST) and alanine aminotransferase (ALT), indicating an excellent protective effect against liver damage by $CCl_4$. In the second experiment, the microsomal malondialdehyde (MDA) contents of the above same groups were also significantly lower (p<0.01) than the $CCl_4$-treated group without plant extracts, but shiitake showed less effect. Among four kinds of plant extracts, extracts of pine leaf and mugwort showed also much higher activities of the microsomal cytochrome P-450 in comparison to soybean sprout and shiitake. In the test of xanthine oxidase (XOD) activity, all of three groups except shiitake showed significantly low activities (p<0.01). These consistent results in vitro and in vivo suggest that the extracts of pine leaf, soybean sprout and mugwort may have strong protective effects against liver damage induced by the potential toxicants such as $CCl_4$.

In Vitro Assessment on Biological Activities of Methanol Extracts from Several Compositae Edible Plants

  • Phytotoxicity, antioxidant activity, and cytotoxicity of the aqueous or methanol extracts from the young sprouts of the six Compositae medicinal plants were determined. Aqueous leachates at 40g dry tissue $L^{-1}$ (g $L^{-1}$) Cirsium japonicum and Aster yomena showed the highest inhibitory effect on alfalfa (Medicago sativa L.). Total phenolic content showed the highest amount in methanol extracts from Ixeris dentata, and followed by A. yomena, and Cephalonoplos segetum. Methanol extracts of C. segetum and I. dentata at 25${\mu}g$ m$L^{-1}$ exhibited the highest DPPH radical scavenging activity by 87.2, and 52.8%, respectively. By means of HPLC analysis, MeOH extracts of C. segetum had the highest amount of antioxidant chlorogenic acid. Based on MTT assay, the methanol extracts from Y. sonchifolia ($IC_{50}$ = 65.7${\mu}g$ $mL^{-1}$) showed the highest cytotoxicity against Calu-6. These results suggest that plant extracts had a dose-dependent biological potentials including phytotoxicity, antioxidant activity, and anticancer activity, and that their activities exhibited differently depending on plant species.

Changes in Antioxidant and Whitening Activities of Buckwheat Seeds with Germination Time

  • The purpose of this study was to evaluate the antioxidant and whitening action of Fagopyrum esculentum and F. tataricum buckwheat seeds depending on their germination time. In a previous study, we reported significant changes in sprout yield and phytochemical content in ethanol extracts from F. esculentum and F. tataricum seeds with increase in germination times. DPPH radical scavenging activities of F. tataricum increased with increasing germination time, whereas that of F. esculentum decreased. Next, we investigated anti-melanogenic activities of these species by estimation of melanin content and tyrosinase activity. Inhibition of melanin production in ${\alpha}-MSH$ (${\alpha}$-melanocyte stimulating hormone)-induced B16F10 cells by extracts from these seeds were analyzed. Among the two, F. tataricum extracts were characterized by higher inhibitory activity against melanin production. In addition, when B16F10 cells were incubated with L-DOPA for detection of in situ tyrosinase activity, F. tataricum and F. esculentum extracts were observed to reduce melanin production in cells. Taken together, these results indicate that extracts from buckwheat seeds could influence cellular processes via modulation of tyrosinase activity. Hence, buckwheat seeds could be utilized as whitening agents in the cosmetic industries and as therapeutics for hyperpigmentation disorders in a clinical setting.
