A Study on the Rural Landscape Planning in Rural Village Development Projects - Yesan O-chon District - (농촌마을종합개발사업 경관계획수립에 관한 연구 - 예산 오촌권역 -)

  • This study has been performed with the purpose of making Rural Village Development Project Landscape Plan in which landscape characteristics in rural villages are reflected so that landscape in the medium to long term shall be established and managed. The issue that has been brought up is that procedures of Rural Landscape Plan and Rural Village Development Project Landscape Plan have been focused on facilities. The study carries out resources survey in agriculture, daily life and history and culture to make Rural Village Development Project Landscape Plan in O-chon district. Through landscape evaluation map participated by local residents, landscape management areas which they can form and manage have been established. The study also suggests Projects and guidelines appropriate to management areas. The study has a different signification from the existing Rural Village Development Project Landscape Plan which is fundamentally devoted to specific projects. The study shows Landscape Plan in the medium to long term and suggests guidelines available to be used by residents.

Full Bloom Stage of Prunus mume and Prunus armeniaca var. ansu - Plant Genetic Resource of ex situ Conservation in Yesan - (매실나무와 살구나무 유전자원의 만개기 특성 - 예산지역 이식의 현지외보전 식물자원 -)

  • 본 연구는 예산지역에서 현지외보전을 하는 매실나무(220여점) 및 살구나무(9점) 유전자원에 대한 개화 특성평가를 위하여 개화기를 지난 만개기를 연차별로 조사하였다. 대한민국에서 수집한 매실나무의 만개기는 이식 초기의 2012년에 4월 중순이었으나 이식 3년 후부터 4월 초순으로 약간 앞당겨졌다. 만개기는 2012년의 경우 모두 4월 11일 이후이었으나 2015년 이후는 4월 1~10일까지 70% 이상이었다. 연도별 보면 2015년과 2016년은 4월 1~5일에 가장 많이 만개하였고, 2017년과 2018년은 4월 6~10일에 가장 많이 만개하였다. 또한 일본에서 수집한 매실나무의 만개기도 대한민국 수집의 자원처럼 4월 초순이 대부분이었는데, 연도별로는 2015년, 2016년 및 2017년의 경우 4월 1~5일에 가장 많이 만개하였고, 2018년은 4월 6~10일에 가장 많이 만개하였다. 그러나 일본 수집의 매실나무 만개기는 3월말에서 4월 초순에 집중되어 3월 중순에서 4월 중순까지의 범위를 나타낸 대한민국 수집 자원과 약간의 차이를 보였다. 그리고 예산지역 현지외보전 살구나무(대한민국 수집)의 만개기는 2015년부터 2018년까지 모두 4월 1일부터 10일까지이었으며, 3월 및 4월 중순 이후에 만개한 꽃이 나타나지 않았다.

  • 본 연구는 대한민국 매실나무 열매의 특성평가를 위하여 청매실의 무게를 대, 중, 소로 구분하여 발효액의 수율, 당도, 산도 및 당산비를 조사하였고, 청매실과 황매실의 발효액 특성을 비교하였다. 발효액의 수율은 82~132%의 범위로 매우 넓었으며, 대 > 중 > 소의 순으로 적은 경향을 보였다. 당도는 55.4~59.1%의 범위이었으며, 무게에 따른 차이는 나타나지 않았다. 산도는 1.73~2.71%의 범위이었으며, 무게에 따른 차이는 나타나지 않았다. 당산비는 20.6~31.3의 범위로 매우 크게 나타나 자원의 다양한 특성이 존재함을 나타내었으며, 무게에 따른 차이는 나타나지 않았다. 무게에 따른 발효액 수율, 당도, 산도 및 당산비는 처리간 차이를 나타내지 않았으나 전체적인 자원간에는 큰 차이를 나타내어 다양하였다. 발효액의 변이계수는 당산비 > 산도 > 당도 > 수율의 순이었다. 청매실과 황매실의 발효액은 수율의 경우 황매실이 높았고, 당도와 산도는 청매실이 높았으며, 당산비는 차이가 없었다.

Comparison of Antioxidative Activities of Fuji Apples Parts according to Production Region (후지 사과의 산지에 따른 부위별 항산화 활성 비교)

  • To observe the functionality of Fuji apples, this study compared and analyzed the general and anti-oxidative components of apples based on production region. This study found that DPPH radical scavenging activities among parts of apple from the Chungju region were 82.84% in peels, 26.98% in peel-flesh, and 18.89% in apple flesh, and these values were lower than those from other regions (P<0.01). Antioxidative was 48.64% in the apple core, which was higher than those in peel-flesh and apple flesh. ABTS radical scavenging activity was highest (79.80%) in peels of apples from the Andong region, whereas values in peel-flesh and apple flesh were highest in apples from the Yesan region (P<0.01). For antioxidative activities in the apple core, apples from the Chungju region showed more than twice the value (52.34%) of other regions. Phenol contents in peels were significantly high [12.03 mg gallic acid equivalent (GAE)/g] in apples from the Muju region, whereas phenol contents in peel-flesh were high (6.01 mg GAE/g) in those from the Andong region. Antioxidative activity in apple flesh was significantly high (5.57 mg GAE/g) in apples from the Yesan region. For antioxidative activities in the apple core, apples from Chungju region showed a relatively high value (6.53 mg GAE/g) (P<0.01), although values were low in apple peel, peel-flesh, and apple flesh. For the combined amount of flavonoids, values in apples from the Yesan region were high in apple peel, peel-flesh, and apple core [56.23 mg quercetin equivalent (QE)/g (P<0.01), 4.05 mg QE/g (P<0.05), and 4.00 mg QE/g (P<0.01), respectively], whereas flavonoid content in apples from the Andong region was high in apple flesh [4.35 mg QE/g (P<0.01)]. The results show that anti-oxidative activities were relatively higher in apple peel than flesh.

아까시나무(Robinia pseudo-acacia)종자 단백질의 전기 영동 변이

  • In order to study the ecotypic variation of Rohinia pseudo-acacia L. distributed in southern area of Korean peninsula, 15 local populations(Daejin, Sokcho, Kangneung, Mt. Surak, Hongcheon, Kwangneung, Namhansanseong, Chungju, Yesan, Andong, Jeonju, Dalseong, Changweon, Mokpo and Wando), located from $34^{\circ}18'N\;to\;38^{\circ}36'N$, were selected based on the latitudes and geographical distances. Seeds of these populations were collected and protein contents of seeds and their band patterns were investigated. The seed proteins of all populations were electrophoresed on SDS-polyacrylamide gel. Total number of protein bands were 35, whose molecular weights ranged from 17, 258 daltons to 142, 232 daltons. The number of bands of seed proteins was 23 in Dalseong and Hongcheon and was 32 in Daejin and Sokcho, showing an increasing tendency in the number of bands as the latitude goes high. The local populations were classified into 3 local types based on protein analysis: the middle north east coastal type(Daejin, Sokcho. Kangneung), the central type (Mt. Surak, Hongcheon, Kwangneung, Namhansanseong, Chungju) and the southern type(Yesan, Andong, Jeonju, Dalseong, Changweon, Mokpo, Wando). According to the results of cluster analysis by UPGMA based on the similarity index(c0efficient of Jaccard) of the patterns, 3 local types were subdivided further into 6 types: the middle north east coastal type(Sokcho, Kangneung), the north central type I (Mt. Surak, Hongcheon), the north central type II (Narnhansanseong, Chungju, Daejin), the north central type III (Kwangneung), the south central type (Yesan, Dalseong, Jeonju) and the southern type(Andong, Changweon, Mokpo, Dalseong, Wando). The No. 12 band of the separated seed proteins showed the highest colored density in the preparations from all the populations. The No. 11~13 and No. 23~28 bands also showed high densities. As a whole, southern type populations (Changweon, Mokpo, Wando) showed high protein contents and high colored density. Total protein contents of the seeds in each population were variable from 9. 68mg / g (Mt. Surak) to 17.30mg/g (Jeonju), showing an increasing trends toward low latitudes.

  • Dried $Compositae$ flowers have traditionally been used for the treatment of anti-inflammatory and anti-oxidative stress in Korea. This paper investigates the effects of $Compositae$ extracts on the inhibition of proliferation and apoptosis of human gastric cancer AGS cells, human breast cancer MDA-MB-231 cells, and SK-BR-3 cells. The proliferation of AGS cells, MDA-MB-231 cells, and SK-BR-3 cells were determined by MTT assay. Several $Compositae$ extracts inhibited proliferation of AGS cells, MDA-MB-231 cells, and SK-BR-3 cells in a dose-dependent manner. To assess the apoptosis of $Compositae$ extracts, the nuclei of MDA-MB-231 cells were stained with DAPI. The presence of chromatin condensation in the $Compositae$ extract-treated cells was detected on a fluorescent microscope (${\times}200$). We conducted Western blot analysis of changes in Bcl-2, Bax, and p53 protein expression levels. Apoptosis by $Chrysanthemum$ $zawadskii$ subsp. coreanum, $Chrysanthemum$ $zawadskii$ var. $tenuisectum$ and $Rudbeckia$ $laciniata$ var. $hortensis$ treatment created a decrease in Bcl-2 expression, whereas the expression of Bax and p53 were increased. These results indicate that $Chrysanthemum$ $zawadskii$ $subsp.$ $coreanum$, $Chrysanthemum$ $zawadskii$ var. $tenuisectum$ and $Rudbeckia$ $laciniata$ var. $hortensis$ inhibit breast cancer cell growth through the induction of apoptosis.

Genetic Comparison Between Crucian Carp (Carassius auratus Linnaeus) and Crucian Carp (C. cuvieri Temminck and Schlegel) (붕어(Carassius auratus Linnaeus)와 떡붕어(C. cuvieri Temminck and Schlegel)의 유전적 비교)

  • Genomic DNA isolated from two crucian carp species obtained from Yesan (Carassius auratus) and Dangjin (Carassius cuvieri) in Korea were amplified at several times by polymerase chain reaction (PCR). The amplified products were separated by agarose gel electrophoresis (AGE) with oligonucleotides decamer primer and stained with ethidium bromide. The seven arbitrarily selected primers OPC-11, OPC-14, OPC-18, OPD- 02, OPD-11, OPD-15 and OPD-20 generated the shared loci by each species, the polymorphic and specific loci. The seven primers generated the total 458 loci that can be scored from the crucian carp obtained in C. auratus species. 358 fragments were generated from the species obtained in C. cuvieri species. The size of DNA fragments varies from 150 to 1,600bp. The complexity of the banding patterns varies dramatically between the primers and two locations. In this study, 458 loci were identified in the crucian carp species from Yesan and 358 in the crucian carp species from Dangjin: 84 polymorphic loci (18.3%) in the C. auratus species and 48 (13.4%) in the C. cuvieri species. 154 shared loci by each species, the average 22 per primer, were observed in the C. auratus species and 187 loci, the average 26.7 per primer, in the Dangjin species. Based on the average bandsharing (BS) values of all samples, the similarity matrix ranged from 0.434 to 0.868 in the C. auratus species and from 0.449 to 0.924 in the C. cuvieri species. The average BS value was 0.641±0.013 within the C. auratus species and 0.684±0.013 within the C. cuvieri species. The average BS value between two crucian carp species 0.484 ± 0.007, ranged from 0.307 to 0.682. The BS value between the individual No. 09 and No. 16 was 0.682, which was the highest between two crucian carp species. Compared separately, the BS value of individuals within the C. cuvieri species was higher than the C. auratus species. The dendrogram obtained by the seven primers, indicates three genetic clusters: cluster 1 (AURATUS No. 01, 02, 03, 04, 05, 06, 07, 08, 09, 10 and 11), cluster 2 (CUVIERI No. 12, 13, 14, 15, 16, 17, 18, 19, 20 and 21) and cluster 3 (CUVIERI no. 22). The shortest genetic distance displaying significant molecular difference was between the individual AURATUS No. 09 and AURATUS No. 08 from Yesan (genetic distance=0.064). The longest genetic distance displaying significant molecular differences was between the individual CUVIERI No. 17 and AURATUS No. 11 between two crucian carp species (0.477). RAPD-PCR analysis has revealed the significant genetic distance between two crucian carp species pairs.(Key words: Carassius auratus, Carassius cuvieri, Crucian Carp, DNA Polymorphism, Genetic Distance)

Runoff and soil loss on newly reclaimed upland (야산개발지(野山開發地)의 토양침식(土壤侵蝕)에 관(關)하여)

  • In order to investigate inherent erodibility of the soil, which is a major factor is soil erosion prediction, a survey on runoff and soil loss of reclaimed upland soil was carried out by using a portable rainulator. The relations of soil loss and some physical properties of the soil were discussed. The soil erodibility factor for Universal soil loss equation was calculated and compared with that of Wischmeier's nomograph. The result were as follows: 1. Total runoff increased for finer textured soil in order of Jeonnam silty clay loam, Songjeong clay loam, Yesan loam, Samgag and Sangju sandy loam. Total soil loss and soil content in runoff were not correspondently related with textural characteristic in order of Jeonnam, Samgag, Sangju, Yesan, and Songjeong. Total runoff, soil loss, and soil content in runoff were increased for steeper slope. 2. Soil loss and soil content in runoff negatively correlated with organic matter content of surface soil, while positively correlated with dispersion ratio, clay ratio, silt content, and significantly correlated with Middleton erosion ratio for coarser textured soil but not correctly related for finer textured soil. 3. The soil erodibilty factor K values for Universal soil loss equation were 0.32 for Jeonnam, 0.22 for Samgag, 0.17 for Sangju, 0.15 for Yesan, and 0.13 for Songjeong respectively. These values were close to those from Wischmeier's nomograph. So, it seems that the nomograph is useful for estimation of soil loss in Korea.

Time Series Change Characteristics of Unconfined Groundwater Wells Temperatures for Agricultural Water Use (농업용수 활용을 위한 비피압지하수관정 수온의 시계열 변동특성)

  • There is a need to analyze unconfined groundwater behavior since the demand of groundwater use has been increasing. While unconfined groundwater temperature is tend to be affected by air temperature, it is hard to find an empirical study in South Korea. In this research, we try to determine the relationship between daily average air temperature and daily average groundwater temperature by time-sequential analysis of groundwater monitoring wells in Galshin basin in Yesan-Gun, Chungcheongnam-Do. In addition, models to estimate groundwater temperature from air temperature were developed. In this research 101-day moving average method with measured air temperature is used to estimate groundwater temperature. To verify the developed model, estimated values of average groundwater temperature with 101 moving average are compared to the measured data from September 10 2007 to September 9 2008. And, Nash-Stucliff Efficiency and Coefficient of Determination were 0.970 and 0.976, therefore it was concluded that the model allowing groundwater temperature estimation from air temperature is with reasonable applicability.

Occurrence of Entomophthora muscae Isolated from Delia platura in South Korea (Delia platura로부터 분리된 Entomophthora muscae의 특성)

  • In June 2023, at the oat field on Kongju National University's Yesan Campus, flies infected with an entomopathogenic fungus were discovered. The flies were hanging with their wings folded backward. Fungal mycelia had sprouted on the abdomens of the flies. An analysis of the 28S ribosomal RNA sequence confirmed that the fungus was Entomophthora muscae. Moreover, cytochrome oxidase sequence analysis of the host insect identified it as Delia platura.
