Medium components for maximum activity of NAD$^{+}$-dependent formate dehydrogenase (EC 1.2.1.2; FDH) were optimized with a methanol-assimilating yeast Hansenula sp. MS-364, preserved by our laboratory. The maximum activity of the enzyme was obtained when the strain was cultivated at 30$circ$C for 24 hours in a medium containing methanol 3%(v/v), yeast extract 0.8%(w/v), K$_{2}$HPO$_{4}$, 0.1%(w/v), KH$_{2}$PO$_{4}$ 0.1%(W/V), MgSO$_{4}$, 7H$_{2}$O 0.05%(w/v), and the pH of the culture broth was adjusted at 5.0.
Fermentation strategies for recombinant protein production in Pichia pastoris have been investigated and are reviewed here. Characteristics of the expression system, such as phenotypes and carbon utilization, are summarized. Recently reported results such as growth model establishment, app58lication of a methanol sensor, optimization of substrate feeding strategy, DOstat controller design, mixed feed technology, and perfusion and continuous culture are discussed in detail.
Arthrobacter sp. L-3로부터 glucose isomerase의 생산성을 검토하였다. 배지의 탄소원으로는 glucose와 xylose를 혼합해서 공급한 것이 효소 생산성이 가장 높았다. 질소원으로는 yeast extract가 가장 높은 효소 생산성을 나타내었다. Glucose isomerase의 생산성은 배양시간이 40시간일 때 가장 높게 나타났다.
Several yeast strains degrading malic acid as a sole carbon and energy source were isolated from Korean wine pomace after enrichment culture in the presence of malic acid. Among them, the strain designated as KMBL 5774 showed the highest malic acid degrading ability. It was identified as Issatchenkia orientalis based on its morphological and physiological characteristics as well as the nucleotide sequences of the internal transcribed spacer (ITS) 1-5.8S rDNA-ITS II region. Phylogenetic analysis of the ITS I-5.8S rDNA-ITS II sequences showed that the KMBL 5774 is the closest to I. orientalis zhuan 192. Identity of the sequences of the KMBL 5774 was 99.5% with those of I. orientalis zhuan 192. The optimal pH of the media for the growth and malic acid degradation by the yeast was between 2.0 and 3.0, suggesting that the strain is an acidophile. Under the optimized conditions, the yeast could degrade 95.5% of the malic acid after 24 h of incubation at $30^{\circ}C$ in YNB media containing 2% malic acid as a sole carbon and energy source.
The objective of this work was to evaluate a thermotolerant yeast Issatchenkia orientalis DY252 as a microbial feed additive for ruminants. In the present study, the influence of volatile fatty acids (VFA) and temperature on oxygen uptake rate by I. orientalis DY 252 was investigated. It was evident that the oxygen uptake rate was decreased gradually as the VFA concentrations increased in a range of 30 to 120 mM. Although the oxygen uptake rate was not greatly affected by temperature in the range 37 to $43^{\circ}C$, a maximum value of $0.45mg\;O_2/g$ cell/ min was obtained at $39^{\circ}C$. With regard to the oxygen uptake rate by yeast, viability was found to be less important than the metabolic activity of yeast.
효모를 Na-alginate에 고정화한 후 연속반응기를 이용한 glucose 발효로 에탄올을 생산하였다. 그 결과 고정화 효모의 활성화 시간은 20~25시간이었다. 연속발효에서 고정화효모의 온도안정성은 30~$37^{\circ}C$였으며 pH 안정성은 pH 4.0~pH 8.0, 최적 희석속도는 $0.2h^[-1}$ 이었고 에탄올생산 최적 당농도는 15%였다. 최적조건에서 에탄올수율은 0.23, 생산된 에탄올 농도는 33.90g/l 그리고 에탄올 생산성은 7.12g/$l\cdot h$로 각각 나타났다.
BACKGROUND: Identification and characterization of endophytic yeasts inhabiting the roots of Ulmus parvifolia Jacq. and Quercus salicina Blume require biotechnological and culture-based techniques. METHODS AND RESULTS: Homogenized U. parvifolia and Q. salicina root samples were spread onto four types of agar medium containing ancgtibiotics, L-sorbose, and Triton X-100. In total, 25 yeast strains were isolated and subjected to phylogenetic analysis based on their internal transcribed spacer region sequences. The results revealed that the yeast genera Cyberlindnera (12 isolates) and Cryptococcus (1 isolate) were associated with roots of U. parvifolia; and the genera Rhodotorula (8 isolates), Trichosporon (3 isolates), and Kluyveromyces (1 isolate) were associated with roots of Q. salicina. Additionally, a Kluyveromyces isolate produced a detectable level of bioethanol. The yeast strains reported herein may be used in industrial production of biosurfactants and bioethanol. CONCLUSION: Our findings revealed that the endophytic yeast genera Cyberlindnera and Cryptococcus predominated in roots of U. parvifolia; and the genera Rhodotorula (8 isolates), Trichosporon (3 isolates), and Kluyveromyces (1 isolate) predominated in roots of Q. salicina. Additionally, Kluyveromyces isolates produced a detectable level of bioethanol.
The use of yeast assist kefir fermentation, but also can cause food spoilage if uncontrolled. Hence, in this study, the microbial composition of an existing commercial kefir starter was modified to produce a functional starter, where Lactobacillus acidophilus KCNU and Lactobacillus brevis Bmb6 were used to replace yeast in the original starter to produce non-yeast kefir-like fermented milk. The functional starter containing L. acidophilus KCNU and L. brevis Bmb6 demonstrated excellent stability with 1010 CFU/g of total viable cells throughout the 12 weeks low-temperature storage. The newly developed functional starter also displayed a similar fermentation efficacy as the yeast-containing control starter, by completing the milk fermentation within 12 h, with a comparable total number of viable cells (108 CFU/mL) in the final products, as in control. Sensory evaluation revealed that the functional starter-fermented milk highly resembled the flavor of the control kefir, with enhanced sourness. Furthermore, oral administration of functional starter-fermented milk significantly improved the disease activity index score by preventing drastic weight-loss and further deterioration of disease symptoms in DSS-induced mice. Altogether, L. acidophilus KCNU and L. brevis Bmb6 have successfully replaced yeast in a commercial starter pack to produce a kefir-like fermented milk beverage with additional health benefits. The outcome of this study provides an insight that the specific role of yeast in the fermentation process could be replaced with suitable probiotic candidates.
Biological removal capacities for volatile organic compounds (VOCs) were determined using a yeast strain, Candida tropicalis. In this study, VOCs including toluene, benzene, p-xylene, and styrene as single substrates or mixtures were tested in the batch culture of the yeast strain. In addition, a kinetic model was applied to evaluate substrate interactions between the VOCs. The yeast strain was able to biodegrade each VOC effectively as a growth substrate, implying it could applied to wide range of VOCs. When the yeast strain was subjected to VOCs in mixtures, the biodegradation rate of one substrate were either increased (stimulated) or decreased (inhibited) by the presence of the others. Both benzene and toluene were inhibited by the other VOCs, and substrate interaction parameters estimated in the model indicated that styrene was the strongest inhibitor for the benzene and toluene biodegradation. Meanwhile, the biodegradation of p-xylene and styrene was stimulated by the presence of either benzene or toluene. The biodegradation rate of p-xylene was significantly increased especially by the presence of toluene, and the styrene biodegradation was enhanced greatly by the benzene addition. The results of the substrate interaction by the yeast strain suggest that the biodegradation rates for the VOCs in mixtures should be carefully evaluated.
