Physicochemical Properties of Shade-cultivated Powdered Green Teas (차광재배 가루녹차의 이화학적 품질 특성)

  • The Daecha-12 and Yabukita varieties of powdered green teas were grown under 85% shade-cultivated condition, and their physicochemical properties were analyzed. Total catechin content was not significantly different but, non-gallated catechin content in Daecha-12 was significantly higher than that in Yabukita. Theanine (32%), caffeine (14%), lutein (15%) and total chlorophyll (28%) levels were significantly higher in Daecha-12 than those in Yabukita. The results of a color analysis showed that the lightness L-value of Yabukita was higher than that of Daecha-12 but, that the greenness negative a-value and b-values of Daecha-12 were higher than those of Yabukita. The Daecha-12 cultivar had lower catechin content but higher content of theanine, caffeine, theobromine, lutein, chlorophyll, and a negative a-value than those of the Yabukita cultivar. Thus, the Daecha-12 cultivar is suitable to prepare a high-quality powdered green tea product.

  • The significant chemical constituents estimating the quality of green tea were compared and analyzed in the tea shoots of native Korean tea plants. The tea shoots of different varieties among native tea plants were plucked in Waun-ri, Yongjang-ri, others in eight tea-growing places, and Yabukita, for the comparison, which is excellent Japanese variety. The contents of Yongjang-ri tea shoots were 0.55% lower in total nitrogen, 41.44mg% lower in vitamin C and 0.56% higher in tannin than the average of the other eight places. The contents of ash, caffeine and water extract showed no difference between the tea shoots. Tea shoots of Waunri had similar compositions compared with those of Yabukita and other eight places in the chemical constituents. It is considered that the tea loaves in Yongjang-ri would be different variety comparing with other eight places in the view of characters and constituents. And it is thought that tea loaves in Waun-ri would be the large leaf variety of same genealogy because tea loaves in Waun-ri was different from the other eight places in characters, but was similar to in constituents.

Catechin and Caffeine Concentration Variations in Jeju Green Tea Varieties Harvested Over a Seven-Month Period

  • Caffeine and catechins from the Yabukita, Yutakamidori, Saemidori, Okumidori, and Fushun varieties of tea leaves picked during different harvesting seasons from April to October were evaluated using HPLC. Total content of catechins increased greatly with the later harvesting time of tea leaves (i.e., picking the leaves in September versus in April) and decreased slightly after September. Yabukita tea leaves picked in August contained 43.1 mg% catechins including EGC, EC, ECG, and EGCG, with the ECGC levels constituting greater than 50% of those four compounds. Yutakamidori and Okumidori varieties picked in September contained the highest catechin values, at 43.6 mg% and 31.0 mg%, respectively. Fushun and Saemidori varieties contained lower catechin concentrations of 14.5 mg% (July) and 11.7 mg% (August) compared to other varieties. The EGCG levels gradually decreased in the late harvesting season, while levels of the other catechins, EC, EGC, and ECG, gradually increased. All varieties of green tea showed a gradual decrease in caffeine content toward the end of our harvesting efforts in October, with levels of 58~68 mg% in April and 28~57 mg% in October. Yabukita, Saemidori, and Okumidori varieties reached their highest caffeine levels in late spring/early summer, with Yabukita and Okumidori varieties reaching a high of 73.4% and 63.5% caffeine, respectively, in May, and Saemidori at 64.0% in June. In particular, Fushun still contained high caffeine of 66.8 mg% (September) during the late harvesting season.

Seasonal Occurrence and Development of Gray Blight of Tea Plants in Korea

  • Disease occurrence and development of gray blight of tea (Camellia sinensis) were investigated. Higher incidences and more severe damage by gray blight were found in Japanese tea variety Yabukita than the Korean local variety. In Yabukita, Pestalotiopsis longiseta was more frequently observed on the diseased leaves than P. theae but vice versa in the Korean local variety. This indicates that there was the varietal difference in the distribution of fungal species of gray blight pathogens. Both varieties were most severely damaged during the third harvest period with weather conditions of high temperature and humidity favorable to the disease. Presence of the tea brown blight fungus Glomerella cingulata on the margin of gray blight lesion at the late stahe suggested that the pathogenic fungi of tea gray blight were replaced by the brown blight fungus during the disease development.

Chemical Variability of Leaf Cuticular Waxes According to Leaf Position in Tea Tree

  • Cuticular waxes on tea (Camellia sinensis L.) loaves consisted mainly of alkanes, fatty acids, primary alcohols, triterpenes, and a group of unknown compounds, dominated by primary alcohols and triterpenes. Tea tree accessions used in this study were M-1, M-2, Sakimidori, and Yabukita. For all accessions, the alkane, fatty acid, and primary alcohol constituents consisted of a homologues series, and the major constituents of primary alcohol class were the C28 and C30 homologues. Triterpenes consisted of friedelin, $\beta-amyrin$, and three unidentified ones and friedelin was the most abundant. Leaf area and the total amounts of cuticular waxes per leaf increased with lower leaf position from the apical bud in Yabukita variety. With different leaf position, total wax amount per unit leaf area on the youngest leaves of P1 (the uppermost leaf position) showed the largest amount $(12.80{\mu}g/cm^2)$, and on mature loaves of P2 to P6 ranged from 7.08 to $7.77{\mu}g/cm^2$, and then on the oldest loaves of P7 (the lowest leaf position) remained at an increased level $(17.53{\mu}g/cm^2)$. During leaf development (lower leaf position), the amount of primary alcohols decreased from P1 to P6 and increased at P7, whereas that of triterpenes increased from P1 to P7. The percentage of each wax class in the total wax amount occurred a decrease in primary alcohol and an increase in triterpene, with leaf age.

First Report on Gray Blight of Tea Plant Caused by Pestalotiopsis theae in Korea

  • A fungus associated with gray blight on tea plant (Camellia sinensis) was identifed as Pestalotiopsis theae based on the mycological characteristics. Mycelial growth on potato dextrose agar and size and shape of conidia of P. theae were similar to those of P. longiseta, but P. theae was different from P. longiseta in the color of three median cells and the number of apical appendages. Artificial inoculation of conidial suspension or mycelial mats on the wounded leaves and shoots of healthy plants induced the same disease, respectively. The Korean native variety was relatively. The Korean native variety was relatively more resistat to P. theae than a Japanese variety‘Yabukita’which has bee recently introduced and planted in large areas of southern parts of Korea. Here, we report the report the first record of gary blight caused by P. theae on tea plant in Korea.

Changes of the Chemical Constituents and Antioxidant Activity During Microbial-fermented Tea (Camellia sinensis L.) Processing (미생물발효차(Camellia sinensis L.) 제조과정 중의 품질특성 변화)

  • Microbial-fermented tea (MFT), which is made by microorganisms through fermentation, is a popular beverage in Asia, especially in the Yunnam province, China. In this study, changes of the chemical constituents and antioxidant activity during the manufacturing process of MFT were investigated. MFT were respectively prepared from fresh leaves of three different tea species (Yabukita, Daecha, and Korean wild cultivar) and a processed green tea (Korean wild cultivar). The color of the tea infusions gradually changed to red and yellow as a function of fermentation time. Total nitrogen and caffeine contents were not significantly changed. Whereas, the chlorophyll, tannin, and total catechins contents gradually decreased. Interestingly, the epicatechin and epigallocatechin contents increased up to 25 days of fermentation and then decreased. Change of the chemical constituents of all samples showed the same patterns. The antioxidant activity of MFT from Daecha and Yabukita slightly decreased as increasing fermentation time. However, the range over which the antioxidant activity of MFT from Korean wild cultivar and green tea were not changed. This research suggests that it may be possible to manufacturing possibility of MFT using Korean wild cultivar and processed green tea.

Disease Progress of Gray Blight on Tea Plant and Selection of a Biocontrol Agent from Phylloplanes of the Plant (차나무 겹둥근무늬병의 발생소장 및 엽권 길항미생물 선발)

  • Disease progress of gray blight of tea (Camellia sinensis O. Kuntze) cv. Yabukita was investigated during the growing season of 2004 at Boseong Tea Experiment Station, Jeonnam Agriculture Research and Extension Service, Boseong, Jeonnam. The disease began to occur from late June and peaked in late July. Antagonistic bacteria against Pestalotiopsis longiseta, the causal pathogen of causing gray blight of tea plants were isolated from phylloplanes of tea plants. An isolate BD0310 which showed the strongest antifungal activity against the pathogen but nonpathogenic to tea plants was selected as a biocontrol agent for the gray blight. The isolate was identified as Bacillus subtilis based on its cultural, morphological, and biochemical characterization and 16S rDNA sequence analysis.

Studies on Growth Enviromental and Inorganic Components of Korean Native Tea Plants(Camellia sinensis O. kuntze) (한국(韓國) 자생차(自生茶)의 생육지 토양과 엽중 무기성분 함량)

  • To characterize the growth environment, inorganic composition and morphological chracteristics of leaves of Korean tea plant, soil and tea leaf samples were collected from 15 locations and analyzed. The chemical characteristics of soils were in range of pH 4.09~6.15, OM 23.9~72.6g/kg, available phosphate less than 300mg/kg, K $0.8{\sim}2.5cmol^+/kg$, Na $tr{\sim}0.17cmol^+/kg$, Ca $1.0{\sim}6.2cmol^+/kg$, and Mg $0.3{\sim}2.1cmol^+/kg$. The contents of Ni. Cr, Zn. Cu, Pb, and Cd were at the level less than natural content in upland soil. Most of the sample soils were sandy loamy and loamy texture. The native tea plants were mainly grown in bamboo thicket or in forest. The leaf sizes of tea plants were $6.85{\pm}1.75{\times}2.6{\pm}0.5cm$, lateral vein number $14.2{\pm}2.7$, and crenated number $58.5{\pm}11.2$, and the leaf color was thin to dark green. The contents of $NH_4{^+}$, $Na^+$, $K^+$, $Mg^{2+}$ and $Ca^{2+}$ in tea leaves were in range of 30.5~47.7, 45.5~164.5, 16,998~25,431, 1.590~2,392 and 1,085~1,958mg/kg, respectively. The contents of $F^-$, $Cl^-$, $NO_3{^-}$, $PO_4{^{3-}}$ and $SO_4{^{2-}}$ were in range of 21.2~63.2, 126.4~257.9, 108.5~185.9, 1,270~1.819, and $954{\sim}1,670mg/kg$, respectively. The leaf size of native tea plant grown widlly in Shunchun Changchun-ri, Hwasun Ssangbongsa, Kuryoi Chonunsa, Bosong Daewonsa and Namhae Boriam was as large as those of Yabukita. Japan cultiver, grown at Kangjin Jangwon tea field.

Spatial Distribution of Macropore Development on a Hillslope (소유역의 사면에서의 대공극발달의 공간적 분포)

  • 사면에서 발생되는 강우유출과정에 기여하는 대공극의 영향은 그 중요성에도 불구하고 잘 알려져 있지 않다. 특히 대공극의 공간적분포특상에 대한 현장측정은 이뤄지지 않았다. 본 연구의 실험지역은 경기도 포천시 광릉수목원에 있는 작은 소유역이다. 이 지역의 정밀한 측량을 하여 수치고도모형(DEM)을 얻었다. 이 수치고도모형을 바탕으로 수치지형분석을 통해 흐름선을 파악하여 총 20지점을 선정하였다. 각 지점에서의 대공극을 통한 수직적인 유동들은 장력침투계를 사용하여 지표면아래 깊이 10cm에서 측정하였다. 공간적 토양의 특성분포를 파악하기 위해 각 지점에서의 체적밀도와 점토함량을 조사하였다. 토양수분의 공간적 분포 특성은 TDR(Time Domain Reflectometry)방식인 TRASE를 이용하여 토양수분 값을 얻었다. 이러한 다양한 공간적 특성들은 대공극발달의 공간적 분포특성을 파악하는 중요자료가 된다. 소유역을 크게 기여사면 면적을 기준으로 상부, 중부, 하부로 나누어 대공극의 유효 공극율과 대공극흐름율을 계산하였다. 상부에서의 유효 대공극율의 평균값과 변동계수는 각각 4.3%, 42.1%이고, 대공극흐름율의 평균값과 변동계수는 각각 45.0%, 26.6%이다. 중부에서는 유효 대공극율의 평균값과 변동계수는 6.8%, 37.3%이고, 대공극흐름율의 평균값과 변동계수는 56.2%, 14.4% 이다. 그리고 하부에서의 유효공극율의 평균값과 변동계수는 12.5%, 58.3% 이고 대공극흐름율의 평균값과 변동계수는 64.5%, 24%이다. 이는 유효 대공극율과 대공극흐름율의 비율은 기여사면 면적이 증가할수록 증가하였다. 이는 대공극을 통한 물 이송 능력이 원두부로 갈수록 증가한다는 것을 보여주고 있다.e, taurine, methionine, phenylalanine은 함량(含量)이 적었다. 5. 일건(日乾)중 총유리아미노산의 변화(變化)는 생시료(生試料)의 경우 2,041.2 mg%였으나 1일(日) 건조(乾燥) 후는 1,784.0 mg%로 감소(減少)하다가 그 이후 계속 증가(增加)하여 20일(日) 건조(乾燥) 후는 5,277.0 mg%였다. 6. 일건(日乾)중 leucine, isoleucine, valine은 대체로 증가(增加)하는 경향(傾向)을 나타내었으나 aspartic acid, proline, taurine은 대체로 감소(減少)하는 경향(傾向)을 나타내었다. 436.59mg%로 가장 많았고 군유산(軍有山) 차엽(茶葉)이 146.94mg%로 가장 적었으며 일반차엽(一般茶葉)의 평균치(平均値)는 264.59mg%, 용장(龍欌) 차엽(茶葉)이 223.10mg%, Yabukita 차엽(茶葉)이 256.49mg%였다. 7) 이상(以上)의 결과(結果)를 종합(綜合)할 때 용장(龍欌) 차엽(茶葉)은 일반차엽(一般茶葉)과 형질(形質) 뿐만 아니라, 성분(成分)도 다르므로 품종(品種)이 다른 수종(樹種)으로 추정(推定)되며 와운(臥雲) 차엽(茶葉)은 일반차엽(一般茶葉)과 형질(形質)은 다르나 성분상(成分上)의 비슷한 점으로 보아 동일계통(同一系統)의 변이(變異)된 대엽종(大葉種)으로 추정(推定)된다.5(${\pm}0.77$0.77) % 의 오차로 크게 감소하였다. 결론: 방사선이 통과하는 경로에 불균질조직인 폐가 존재할 경우에도 불균질조직에 대하여 조직의 밀도를 이용하여 보정하는 방법을 사용하여 투과선량으로부터 종양선량을 계산할 수 있음을 알 수 있었다.X>로 평균$43.26{\m
