Genetic Transformation of the Mycelia of Tremella fuciformis and Changes of Cytotoxicity (흰목이 균사체 형질전환 및 세포독성의 변화)

  • Tremella fuciformis, as one of higher basidiomycetes, can asexually reproduce yeast-like conidium (YLC) cells by budding. We have developed an efficient method to introduce pCambia1300 plasmid containing hph gene into YLC cells using Agrobacterium. This was successful only when YLC cells were wounded by NaOH treatment before co-cultivation. In average, 40~50 transformants were produced out of $1.0{\times}10^6$ YLC cells investigated. The T-DNA transfer was confirmed by PCR. Methanolic extracts from transformants demonstrated different levels of toxicity against SKOV-3 cervical cancer cells.

Selection of Unnecessary Urine Culture Specimens Using Sysmex UF-5000 Urine Flow Cytometer (Sysmex UF-5000 소변 유세포분석기를 이용한 요배양 불필요 검체의 선별)

  • Background: Urine culture is one of the most frequently requested tests in microbiology. Automated urine analyzers yield much infection-related information. The Sysmex UF-5000 analyzer (Sysmex, Japan) is a new flow cytometry urine analyzer capable of quantifying urinary particles, including bacteria, WBCs, and yeast-like cells (YLCs) and can provide a Gram stainability flag. In this work, we evaluated how many unnecessary urine cultures could be screened out using the UF-5000. Methods: We compared the culture results of 126 urine samples among 453 requested urine cultures (from sources other than the Urology and Nephrology departments) with urinalysis results. Urine cultures were considered positive if bacterial or YLC growth was ${\geq}10^4CFUs/mL$. Results: We used urinalysis cut-off values of $50/{\mu}L$ and $100/{\mu}L$ for bacteria and YLC, respectively. Forty eight of the 126 (38.1%, or 10.6% of 453 requested) cultures were below these cut-off values and did not contain any culture-positive samples. Conclusion: Bacteria and YLC counts generated using the UF-5000 analyzer could be used to screen out negative cultures and reduce urine culture volume by ~10% without sacrificing detection of positive cultures.

Adaptive Quantization of Difference Wavelet Image for Close-Range Low-Bitrate Transmission (근거리 저전송률 통신을 위한 차영상 웨이브릿 적응 양자화)

  • This paper presents a image coding method that is well adaptive to close-range video transmission because of its low titrate and simple coding procedure. At first, it reduces temporal redundancies by performing image DPCM between previous frame and current frame, and makes wavelet transformed image of this difference image. Then, the coefficients are quantized selectively by using the coefficient values of base level and mid-frequency level because inter-level redundancies are widely exists in multi-resolution images. Finally quantized coefficients are made iron the function that implies the target bitrate, the average coefficient energy, and the value of the level. The proposed method shows the effective Performance in the experiments using the continuous motion images and transition images.
