Identification of a Cellular Protein Interacting with Murine Retrovirus Gag Polyproteins

  • The retroviral Gag polyprotein directs the assembly of virion particles and plays an important role in some events after entry into a host cell. The Gag polyprotein of a virus mixture is responsible for inducing murine acquired immunodeficiency syndrome (MAIDS) when injected into susceptible strains of mice. In order to identify the host cellular proteins which interact with the MAIDS virus Gag proteins and possibly mediate the function of the Gag proteins, mouse T-cell leukemic cDNA expression library was screened using the yeast GAL4 two hybrid system. Of 11 individual positive clones, the clone Y1 was selected for the study of protein-protein interaction. Its DNA sequence revealed that it was an exact match to the murine SH3 domain-containing protein SH3P8. It is expressed as 2.4 kbp transcripts in testis at higher levels and in various tissues tested at lower levels. Glutathione S-transferase-Y1 fusion protein binds tightly to $Pr60^{def-gag}$ as well as $Pr65^{eco-gag}$.

  • This paper deals with the WASPIFA (Web-based Assistant System for Protein-protein Interaction and Function Analysis) system that can provide the comprehensive information on Protein-protein interaction and function concerned with function analysis. Different from existing systems for protein function and protein-protein interaction analysis, which provide fragmentary information restricted to specific field, our system furnishes end-user with comprehensive and synthetic information on the input sequence to be analyzed, including function and annotation information, domain information, and interaction relationship information. The synthetic information that our system contains as local databases has been extracted from many resources related to function, annotation, motif and domain by various pre-processing. Employing our system, end-users can evaluate and judge the synthetic results to do protein interaction and function analysis effectively. In addition, the WASPIFA system is equipped with automatic system management and data update function that facilitates system manager to maintain and manage it efficiently.

Effects of Milk Protein levels and Casein/Whey Ratios on Organ Growth and Protein Metabolism in Early Weaned Rats (조기 이유한 흰쥐에서 유단백질의 섭취수준과 조성비가 기관성장과 단백질대사에 미치는 영향)

  • This study was conducted to investigate the effects of protein levels and casein/whey ratios on organ growth and protein metabolism in early weaned rats. Premature rats weaned by the 17th day were fed six semipurified synthetic, isocaloric and gel diets that contained three levels (low, medium and high) and two different combinations(casein/whey ; 80 : 20 or 20 : 80) of milk protein for 8 days. On the 25th day postpartum, frest weigth and DNA, RNA and milk protein contents in brain, liver, kidney and muscle were determined to ascertain organ and cellular growth. Futher, with a view to ascertain protein metabolism and renal functions, serum total protein, $\alpha$-amino N, urea N, and creatinine and creatinine and urinary urea N, creatinine and hydroxproline were determined. Total DNA contents of brain, liver and kidney, which may represent as an index of cell numbers in those organs were significantly decreased in the rats fed diets containing low level protein regardless of casein/whey ratio. However, as fat as the rats fed high protein diets were concerned, their fresh weight, protein contents and GFR of kidney were significantly increased. Furthermore, nitrogen components, $\alpha$-amino N, urea N and creatinie in serum and urine were also increassed. Another observation was that high casein/whey ratio significantly facilitated accumulation of porteins in muscle and kidney and urinary hydorxyproline excretion, not affecting the DNA content of those organs. This study showed that low(8%) or high(32%) contents of protein had less desirable effects either on protein metabolism or on organ cellular growth in prematurely weaned rats, whereas there were no effects on general growth and bone strength.

  • Representing protein three-dimensional structure by concatenating a sequence of protein fragments gives an efficient application in analysis, modeling, search, and prediction of protein structures. This paper investigated the effective combination of distance measures, which can exploit large protein structure database, in order to construct a protein fragment library representing native protein structures accurately. Clustering method was used to construct a protein fragment library. Initial clustering stage used inter alpha-carbon distance having low time complexity, and cluster extension stage used the combination of inter alpha-carbon distance, Binet-Cauchy distance, and root mean square deviation. Protein fragment library was constructed by leveraging large protein structure database using the proposed combination of distance measures. This library gives low root mean square deviation in the experiments representing protein structures with protein fragments.

Effects of Phosphorylation and Acetylation on Functional Properties and Structure of Soy Protein (인산화와 초산화가 대두단백질의 기능특성과 구조에 미치는 영향)

  • Phosphorylation of soy protein by sodium trimetaphosphate and acetylation of soy protein by acetic anhydride were performed. Then, the functional properties of modified soy proteins were compared with that of unmodified soy protein. Isolated soy protein prepared from defatted soybean flake had protein content of 92.7% as moisture-free basis. The phosphorylated soy protein showed higher solubility, foaming properties, and water holding capacity than unmodified soy protein. Acetylation of soy protein increased emulsification activity and foaming properties greatly, whereas decreased the solubility at pH 8.0. Isoelectric pHs of phosphorylated and acetylated soy protein were shifted to acidic regions(pH 3.0 and pH 4.0) from pH 5.0, which was the isoelectric pH of unmodified soy protein. Soy protein seems to be aggregated during phosphorylation and acetylation procedure, judging form Sepharose CL-4B gel filtration profiles. The modified soy proteins showed increased mobilities to anode direction in disc-gel electrophoresis.

  • In the previous studies, one of our author had observed the obligatory nitrogen losses through urine and feces in young Korean male adults, and utilization of whole egg protein in Korean young adults with ordinary intake of energy as a basic study for estimating protein requirements of Korean. This study has tried to determine the protein requirement and utilization when subjects ate the Korean mixed diet at the energy level of 45 kcal per Kg body weight per day. Six healthy Korean young female adults, college students, aged $20{\sim}25$ years old, and $45{\sim}61Kg$ of body weight participated as the study subjects. They were given isocaloric diets with four different protein levels for five days each successively. These diets contained protein levels of 0.45, 0.60, 0.75 and 0.90g per Kg of body weight per day, respectively. It was attempted to observe energy and protein intake, urinary and fecal nitrogen losses, true digestibility of protein, net protein utilization of protein in the body over last two days of period of eating each diet and nitrogen balances were calculated. The body weight change and hematological observation were also performed. The results obtained were summarized as follows ; 1) True digestibility of protein of Korean mixed diets ranged from 83.1% to 86.5% and the average of which was $84.7{\pm}1.7%$. 2) Net protein utilization rate of Korean mixed diets was range of 49% to 55% and the average of which was $52{\pm}3%.$ 3) The body weight of subjects were slightly increased and the values of hemoglobin and hematocrit of the blood were also little increased during the experimental periods of twenty days. 4) Protein requirements of Korean mixed diet with the energy level of 45 kcal/Kg body weight per day of Korean young female adults were estimated as 1.00g/Kg/day on this experimental condition.

A study on the development of high functional food protein ingredient from rice bran (고기능성 쌀단백질 소재 개발 연구)

  • Rice bran proteins from different cultivars(Youngan, Sindongjin, Suwon 511) were extracted with Xylanase using orthogonal analysis method and their functional properties were investigated. The optimum extraction conditions, based on protein content in the extract found to be at 1 wt% xylanase, pH 7 and 50:1, solvent to rice bran ratio(v/w %). Nitrogen solubility indices(NSI) of rice bran protein concentrates were shown a minimum value at pH 4 ranged 2~23%, varied with different cultivars and a maximum (NSI${\geq}$90% for all cultivars) at pH 10. As for water adsorption and fat adsorption capacity, rice bran protein concentrates were shown to be better than Na-caseinate and isolated soy protein, respectively. Emulsifying activities were observed high in order of Na-caseinate>Youngan rice bran protein>Shindongjin rice bran protein>Suwon 511 rice bran protein>isolated soy protein. In general, the surface tension of rice bran protein solution($10^{-3}$ wt%, 5 mM bis-tris, pH 7) was increased with increasing concentrations and found a minimum value near pI. On heating, it was decreased slightly with increasing temperatures up to $70^{\circ}C$ and then increased above $80^{\circ}C$. Addition of sodium chloride was made the surface tension decrease. In conclusion, with Xylanase, rice bran protein concentrate can be successfully extracted from the rice bran of different cultivars and the Youngan rice bran protein was thought to have best functionality among rice cultivars tested. It might be used as a milk protein substitute.

  • After figuring out the function of an amino acid sequence, we can infer the function of the other amino acids that have similar sequence composition. Besides, it is possible that we alter protein whose function we know, into useful protein using genetic engineering method. In this process. an original protein amino sequence produces various protein sequences that have different sequence composition. Here, a systematic technique is needed to manage protein version sequences and reference data of those sequences. Thus, in this paper we proposed a technique of managing protein version sequences based on local sequence alignment and a technique of managing protein historical reference data using Trigger This method automatically determines the similarity between an original sequence and each version sequence while the protein version sequences are stored into database. When this technique is employed, the storage space that stores protein sequences is also reduced. After storing the historical information of protein and analyzing the change of protein sequence, we expect that a new useful protein and drug are able to be discovered based on analysis of version sequence.

Hepatic and Renal cysteine Sulfinic Acid Decarboxylase Activities in Cats Fed Different Levels of Dietary Protein and Taurine

  • In order to evaluate the dietary regulation of cysteine sulfinic acid decarboxylase (EC 4.1.1.29) in cats, acitivity and protein content of CSAD were assessed in the liver and kidney of cats fed different levels of dietary protein, with and without taurine. Four groups of cats were fed one of the follow diets for 5 weeks ; 20% protein and taurine- free diet(LP0T) ; 20% protein and 0.15% taurine diet(LPNT) ; 60% protein and taurine-free diet(HP0T); and 60% protein and 0.15% taurine diet (HPNT). CSAD activity was determined in the liver and kidney of cats by measuring 14C2 released form [1-14C]-L cysteine sulfinic acid. CSAD protein was quantified using an immunochemical method. CSAD activity was extremely low in cat tissues, among which kidney showed the highest activity which was 0.118$\pm$0.050, and 0.377$\pm$0.056 nmol.min-1.mg soluble portein-1 iin animals fed LP0T and HP0T, respectively. Even though renal CSAD protein content was 18~55% of the hepatic CSAD protein content, renal CSAD acitivity was 1.3~6.5 times of the hepatic CSAD activity . Renal CSAD acitivities of cats fed 60% protein were about 1.6~3.2 times those of animals fed 2.% protein , and hepatic CSAD activity was not significantly affected by the dietary level of protein. Taurine depletion significantly elevated both hepatic and renal CSAD activities above the values for cats having normal taurine status most probably as an adaptive response.

A Survey of Protein Nutrition of Rural Adults in Korea and A Correlation between Their Protein Nutrition and Their Food Habits (농촌 성인 남녀의 단백질 섭취 실태 및 급원과 식습관, 기호품과의 관계)

  • The purpose of this study is to provide useful information which will aid in the promotion of nutritional policy in the Korean rural area which are derived from a survey of intake and source of protein in some Korean rural adults and the correlations between their protein nutrition and various nutrients, the uses of tobacco, alcohol, coffee, etc. The survey was conducted from July, 24 through July, 18, 1989. The healthy subjects were 45 males(average age 42.3, average BMI 22.1kg/m2) and 55 females (44 years, 21.2 kg/m2) residing in Sungjoo Kyun, Chulanam-Do, Korea. The subjects were examined for the anthropometric, food Intake and food habits. Their daily diets were measured by 24-hr recall method. The results can be summarized as follows : The daily mean protein intakes of male and female subjects were 9595 of RDA(66, 5g) (16. 7% of total food intake per day) and 102.3Bh of RDA(61g) (14.8% of total intake per day) respectively. The order of sources of animal protein in all subjects was fish(47.9%), meat(29.8%), milk (12%), and egg(10.3%). Among protein sources the intake frequencies of fork and chicken were higher than those of others. The protein nutrition of the subjects showed positive correlations with energy and fat, carbohydrate, fiber at the level of significance of 1%. The protein nutrition of the male subjects showed no correlation with age, BMI, uses of alcohol, coffee, medicine, but the plant protein nutrition showed a positive correlation with smoking and exercise(at 5%). And the protein nutrition of the male subjects showed no correlations with uses of tobacco, alcohol, coffee, medicine and their opinion of their present state of their health, but the age of the male subjects showed negative correlations with milk(at 1%) and egg(at 5%). In conclusion, the daily protein Intake was good and the main sources of animal protein were fishes in rural adults. The protein nutrition of the subjects showed a correlation with energy, fat, carbohydrate and in the case of female, milk and egg intakes showed the negative correlation with age.
