  • 오늘날에는 HGP(Human Genome Project)로 인해 인간과 같은 고등생물은 높은 비율로, 단백질을 만들어낼 때 유전자 개수를 늘려 나가는 것이 아니라 유전자의 활용도를 높임으로써 다양한 단백질을 만들어낸다 새로운 사실이 밝혀졌다. 이로 인해 alternative splicing에 대한 관심이 높아지고 있다. Alternative splicing의 비중이 높아지며 이에 따라 이를 찾아내기 위한 다양한 방법들이 생겨나고, 이러한 방법 중 하나가 splice junction을 찾아내는 것이다. 본 논문에서는 splice junction 탐색을 위한 도구를 개발하기에 앞서 이미 기존에 존재하는 도구들을 조사하여 해당 도구들이 어떠한 사양과 알고리즘을 사용하는지를 분석 및 비교하였다.

Effect of MoSe2 on Contact Resistance of ZnO/Mo Junction in Cu(In,Ga)Se2 Thin Film Solar Module (MoSe2가 Cu(In,Ga)Se2 박막 태양전지 모듈의 ZnO/Mo 접합의 접촉 저항에 미치는 영향)

  • In this paper, the effect of MoSe2 on the contact resistance (RC) of the transparent conducting oxide (TCO) and Mo junction in the scribed P2 region of the Cu(In,Ga)Se2 (CIGS) solar module was analyzed. The CIGS/Mo junction becomes ohmic-contact by MoSe2, so the formation of the MoSe2 layer is essential. However, the CIGS solar module has a TCO/MoSe2/Mo junction in the P2 region due to structural differences from the cell. The contact resistance (RC) of the P2 region was calculated using the transmission line method, and MoSe2 was confirmed to increase RC of the TCO/Mo junction. B doped ZnO (BZO) was used as TCO, and when BZO/MoSe2 junction was formed, conduction band offset (CBO) of 0.6 eV was generated due to the difference in their electron affinities. It is expected that this CBO acts as a carrier transport barrier that disturbs the flow of current, resulting in increased RC. In order to reduce the RC caused by CBO, MoSe2 must be made thin in a CIGS solar module.

The Variation of Channel Bed by Location of Pier near 90° Channel Junction (90° 합류부를 지닌 수로에서 교각 위치에 따른 하상변동)

  • The variation of channel bed which shows the complex hydraulic characteristics at channel junction was investigated by variation of discharge ratio and location of pier. As discharge ratio increase, the depth and width of erosion region become bigger to point of 63% of channel width in case of 1:0.5 discharge ratio. It was observed that the maximum scour depth at the point of 0.5 times of the channel width in the channel junction as 2.5 times bigger than straight channel. It means that the maximum scour depth at the channel junction is 2 times greater than by experimental formulas which are widely used in practical engineering, location of pier should be determined when it is installed in channel junction.

Design of Low Pass Filters Using Corrugated Waveguide for Satellite Communications (코르게이트 도파관을 사용한 위성통신용 저역통과 여파기의 설계)

  • In this paper, low pass filters(LPF) using corrugated waveguide for satellite communication is proposed. To design the proposed LPF, the corrugated waveguide and impedance transformers are combined. The corrugated waveguide is formed by arrangement of T-junction. To obtain low insertion loss and high isolation propriety, T-junction is designed by using Chebyshev function. Impedance transformers is designed by combining T-junction with different heights at both ends of the corrugated waveguide to get high return loss. The measured results of the proposed LPF have a return loss of over 35.4 dB, a insertion loss of less than 0.1 dB from 12.25 GHz to 12.75 GHz and a isolation propriety of over 54.5 dB from 14 GHz to 14.5 GHz. From these results, it is confirmed that the proposed waveguide LPF can be used for satellite communication.

LIMK1/2 are required for actin filament and cell junction assembly in porcine embryos developing in vitro

  • Objective: This study was conducted to investigate the roles of LIM kinases (LIMK1 and LIMK2) during porcine early embryo development. We checked the mRNA expression patterns and localization of LIMK1/2 to evaluate their characterization. We further explored the function of LIMK1/2 in developmental competence and their relationship between actin assembly and cell junction integrity, specifically during the first cleavage and compaction. Methods: Pig ovaries were transferred from a local slaughterhouse within 1 h and cumulus oocyte complexes (COCs) were collected. COCs were matured in in vitro maturation medium in a CO2 incubator. Metaphase II oocytes were activated using an Electro Cell Manipulator 2001 and microinjected to insert LIMK1/2 dsRNA into the cytoplasm. To confirm the roles of LIMK1/2 during compaction and subsequent blastocyst formation, we employed a LIMK inhibitor (LIMKi3). Results: LIMK1/2 was localized in cytoplasm in embryos and co-localized with actin in cell-to-cell boundaries after the morula stage. LIMK1/2 knockdown using LIMK1/2 dsRNA significantly decreased the cleavage rate, compared to the control group. Protein levels of E-cadherin and β-catenin, present in adherens junctions, were reduced at the cell-to-cell boundaries in the LIMK1/2 knockdown embryos. Embryos treated with LIMKi3 at the morula stage failed to undergo compaction and could not develop into blastocysts. Actin intensity at the cortical region was considerably reduced in LIMKi3-treated embryos. LIMKi3-induced decrease in cortical actin levels was attributed to the disruption of adherens junction and tight junction assembly. Phosphorylation of cofilin was also reduced in LIMKi3-treated embryos. Conclusion: The above results suggest that LIMK1/2 is crucial for cleavage and compaction through regulation of actin organization and cell junction assembly.

Ultrastructural Study on Connective Tissue-Epithelial Junctions in Anagen Hair Follicle of Human Fetus (사람태아 성장기 모낭에서 결합조직-상피 경계부의 미세구조에 관한 연구)

  • The dermal papilla is known to playa major role in influencing the form and dynamics of the hair follicle, which probably involves regulatory substances crossing the basal lamina. But little is known about the junctions between the dermal papilla and the surrounding epithelial cells of the hair bulb, or between the connective tissue and the epithelial cells on the outside of the hair follicle. This study was performed to identify the ultrastructural differences between dermoepidermal junction of the skin and connective tissue-epithelial junctions on the outside of the hair follicle and around the dermal papilla of normal anagen hair follicles in the human fetal scalp skin. Electron microscopic findings of dermoepidermal junction in scalp skin showed that basal lamina was very irregular and undulated, and it contained many attachment plaques of hemidesmosomes with sub-basal dense plates, tonofilaments, and anchoring filaments. Also invaginations of plasma membrane of basal keratinocytes were seen. There were clear differences both on the outside of the follicle and around the dermal papilla as compared with similar junction in the skin. In particular, neither hemidesmosomes nor tonofilaments, as seen in dermoepidermal junction, were observed in the dermal papilla. Also attachment plaque, sub-basal dense plate and anchoring filaments were not observed at the junction on the outside of the follicle and the dermal papilla. There were some differences between connective tissue-epithelial junctions on the outside of the hair follicle and around the dermal papilla, ie, smoothness of basal lamina and orthogonal arrangement of collagen fibers were seen in the outside of hair follicle, but not in the dermal papilla. These results indicate that the mechanical connection between the hair follicle and the connective tissue component is much weaker than that between the corresponding components in skin, and it reflects the dynamic processes during the anagen phase of the hair follicle compared to the relatively permanent state of the epidermis.

Junction Area Dependence of Tunneling Magnetoresistance in Spin-dependent Tunneling Junction with Natural $Al_2O_3$Barrier (자연산화 $Al_2O_3$장벽층을 갖는 스핀의존 터널링 접합에서 자기저항특성의 접합면적 의존성)

  • Spin dependent tunneling (SDT) junction devices of Ta/NiFe/Ta/NiFe/FeMn/NiFe/AlOx/CoFe/NiFe/Al with in-situ naturally oxidized Al barrier were fabricated using ion beam deposition and dc sputtering in UHV chamber of 10$^{-9}$ Torr. The maximum tunneling magnetoresistance (TMR) and the product resistance by junction (R$_{j}$ A) are 16-17% and 50-60 $\Omega$${\mu}{\textrm}{m}$$^2$, respectively. The values of TMR and (R$_{j}$ A) with field annealing were slightly increased. The TMR and (R$_{j}$ A) dependence versus the junction area size was observed. These results were explained by using sheet resistance effect of bottom electrode and spin channel effects.

The Regeneration Effect of Platelet-Rich Plasma on the Treatment of Grade II Furcation Involvement Using Synthetic Bone in Human (2급 치근이개부 병변의 합성골 이식 시 혈소판 농축 혈장의 재생에 관한 효과)

  • The present study evaluated of regeneration effect of platelet rich plasma on the treatment of grade II furcation involvement, with coralline calcium phosphate bone in humans. 30 teeth with grade II furcation defect were selected and 15 teeth(control) were treated with coralline calcium phosphate bone, the others(test) were treated with coralline calcium phosphate bone and platelet rich plasma. Pocket depth, clinical attachment level, width of keratinized gingiva width were measured at baseline, postoperative 3, 6months. from cementoenamel junction to alveolar crest and fundus were measured at baseline, 6months(re-entry). Both groups were statistically analyzed by Wilcoxon signed Ranks Test & Mann-whitney Test using SPSS program(5% significance level). The results were as follows: 1. Pocket depth, clinical attachment level, keratinized gingva width, cementoenamel junction - alveolar crest, cementoenamel junction - fundus were not differ significantly in both groups at baseline 2. The change of pocket depth, clinical attachment level, keratinized gingva width, cementoenamel junction - alveolar crest, cementoenamel junction - fundus in both groups were decreased significantly at 3, 6months(p<0.05). 3. The change of pocket depth, clinical attachment level in test group decreased significantly than control at 3, 6months(p<0.05). 4. The change of keratinized gingiva width, cementoenamel junctional - alveolar crest, cementoenamel junction - fundus were not differ significantly in both groups at 3, 6months. 5. The pocket depth, clinical attachment level, keratinized gingiva width exhibited marked changes at 3 months in both groups. In conclusion, the results of this study suggest that platelet rich plasma have adjunctive clinical treatment effect to periodontal regeneration in grade II furcation defects.

  • If the target volume cannot be included with one field at head and neck cancer, we commonly used two or more field. It is very important to irradiate uniform dose at junction area of the fields. However, according to body shape of patient or general condition of patient, skin junction area can be matched incorrect, So overdose area or underdose area can be appeared in the junction area. This study researched therapy technique which can give uniform dose at skin junction owing to applying the edge block of lateral field at head and neck cancer. We measured the changed distance and rotational angle between central line of anterior supraclavicle lymph node and low margin of right lateral field on simulation process using the shielding block of variable rotation. As a result, the changed distance between central line of anterior supraclavicle lymph node and low margin of right lateral field was below 2mm to ${\pm}$10cm distance at central line of Y axis, changed angle was average 1.28 degree. But by using it the shielding block of variable rotation, the incorrect match at junction can be minimized. We think that this technique is very efficient one to apply this technique at head and neck cancered by the movement of organs can be not included, Therefore we have to pay attention on the process to imput MLC layer

Tight junctional inhibition of entry of Toxoplasma gondii into MDCK cells (MDCK세포의 tight junction 형성이 Toxoplusmu gondii의 숙주세포 침투에 미치는 효과)

  • Various conditions of cultures were performed to investigate the role of tight junctions formed between adjacent MDCK cells on the entry of Toxoplasma. When MDCK cells were cocultured with excess number of Toxoplasma at the seeding density of 1×105, 3×105, and 5×105 cells/ml for 4 days, the number of intracellular parasites decreased rapidly as the host cells reached saturation density, i.e., the formation of tight junctions. When the concentration of calcium in the media (1.8 mM in general) was shifted to $5{\mu}M$ that resulted in the elimination of tight junction, the penetration of Toxoplasma increased about 2-fold(p<0.05) in the saturated culture, while that of non-saturated culture decreased by half. Trypsin-EDTA which was treated to conquer the tight junctions of saturated culture favored the entry of Toxoplasma about 2.5-fold(P<0.05) compared to the non-treated, while that of non- saturated culture decreased to about one fifth. It was suggested that the tight junctions of epithelial cells play a role as a barrier for the entry of Toxoplasma and Toxoplasma penetrate into hoot cells through membrane structure-specific, i.e., certain kind of receptors present on the basolateral rather than apical surface of MDCK cells.
