Orchargrass 우점초지에서 초장에 의한 생산량과 사료가치 추정 (Estimation of Pasture Production and Nutritive Vallue in Relation to Plant Heigth of Orchardgrass (Dactylis glomerata L. ))

  • For simple estimation of pasture production and nutritive value in relation to height of orchardgrass (Ductylis glomumtu L.), the experiment was carried out in a pasture mixture dominated by orchardgrass. The samples of grasses were harvested at the heights of about 30, 40, 50, 70 and 90 cm at the first harvest under different percentage of 100 (grass coverage): 0 (bareland), 80 : 20, 60 : 40, 40 : 60 and 20 : 80, respectively. As the height of orchardgrass was increased, fresh and dry matter (DM) yield were significantly increased. The regression equation between DM yield (Y) and height (X) was Y=ll. 5390 X- 146.60 (r=O. 9414 * * ). Also the higher bareland, the lower pasture production was observed. As the plant height at the first harvest was increased, the contents of crude (C) protein, C. fat, C. ash and relative feed value were markedly decreased. On the other hand, the contents of C. fiber, NFE, NDF. ADF, cellulose and lignin were significantly increased, respectively. The higher height, the lower DM digestibility was found. The equation between digestibility (Y) and height (X) was Y=- 0.2193 X+ 80.99 (r = - 0.9862**). Considering DM yield, digestibility and crude protein, the optimum height of orchardgrass at the first harvest was 59 to 67crn for better pasture production and nutritive value.

인삼 Saponin이 양신장에서 정제한 $Na^+$ $K^+$-ATPase의 활성, 인산화 및 $[^3H]$Ouabain결합에 미치는 영향 (Effect of Ginseng Saponin on the Activity, Phosphorylation, $[^3H]$Ouabain Binding of Purified$Na^+$ $K^+$-ATPase Isolated from the Outer Medulla of Sheep Kidney)

  • The effects of ginseng saponin on the activity, phosphorylation, [$^{3}$H] ouabain binding and light scattering (disruption) of purified $Na^{+}$ ,$K^{+}$ -ATPase isolated from the outer medulla of sheep kidney were compared to those of gypsophila saponin, sodium dodecylsulfate (SDS), and Triton X-100 on the same parameters. $Na^{+}$ , $K^{+}$ -ATPase activity, phosphorylation, and [$^{3}H$] ouabain binding were inhibited by ginseng saponin (triol>total>diol), SDS, or Triton X-100, but increased by gypsophila saponin. Low doses of ginseng saponin (3.mu.g saponin/.mu.g protein) decreased phosphorylation sites and ouabain binding site concentration (Bmax) without any change of turnover number and affinity for ouabain binding which were decreased by high dose of ginseng saponin (over 10.mu.g saponin/.mu.g protein), SDS or Triton X-100. On the other hand, gypsophila saponin increased the affinity without any change of Bmax for ouabain binding. Inhibition of $Na^{+}$ ,$K^{+}$ -ATPase activity by ginseng saponin and SDS or Triton X-100 appeared before and after decrease in light scattering, respectively. These data suggest that ginseng saponins (total, diol, triol saponin) inhibit $Na^{+}$ , $K^{+}$ -ATPase activity by specific direct and general detergent action at low and high concentrations, respectively, and this inhibitory action of ginseng sapornin to $Na^{+}$ , $K^{+}$ -ATPase is not general action of all saponins.

대두를 이용한 이유식 제조에 관한 연구(제 1보) -효소를 이용한 대두단백질 분해 적정 조건결정 및 조제에 관하여- (Studies on the Preparation of Weanling Food from Soybean (Part 1) -Conditions for the digestion of soybean protein by Eezyme from Aspergillus-)

  • 우수한 영양가를 가진 대두를 이용하여 이유식을 제조하기 위하여 가압증자한 대두에 Protease및 Cellulase 역가가 비교적 높은 Asp. niger 및 Asp. sojae균의 피국 추출조효소액을 작용시켜 대두 단백질을 아미노산 내지 Peptide 태로 분해시키는 최적조건을 결정하고 여기서 얻은 분해물을 탈색 농축시키는 효과에 관하여 실험하여 다음과 같은 결과를 얻었다. 1. 대두의 가압증자는 15Ib에서 10분간 처리함이 가장 높은 단백응해율 및 단백분해율을 나타냈다. 2. Asp. sojae enzyme은 pH 6.0, Asp. niger enzyme은 pH 4.4에서 가장 높은 단백용해율과 단백분해율을 나타냈다. Asp. sojae enzyme을 처리한 다음 Asp. niger emzyme을 작용 분해시킨 것이 각 효소 단독으로처리했을때 보다 높은 단백응해율(62.3%) 및 단백분해율을 (56.4%) 나타냈다. 3. 기질에 대한 효소액 첨가량은 원료 대두에 10배의 물을 가한 마쇄기질액에 피국에 대하여 10배의 물로 추출한 효소액 1/2에 해당하는 양 (Asp. sojae enzyme와 Asp. niger emzyme 총량)을 가하는것이 가장 실용적이었다. 4. 분해시간이 길 수록 단백응해율 및 단백분해율이 높아지나 부패 등을 고려할 때 실용분해 시간은 8시간 정도가 적당하다. 5. 탈색 효과는 활성탄으로 처리한 후 음이온교환수지 (Dowex 2-x-8)을 처리한 것이 가장 좋고 단독처리로는 음이온교환수지, 활성탄, 양이온교환 수지(Amberite)의 순으로 효과가 적었다. 6. 이상의 최적 조건으로 대두단백질을 분해하고 $60^{\circ}C$ 이하에서 감압농축하여 얻은 제품은 수분 12.51%, 단백질 66.31%, 지방 4.25%, 탄수화물 12.75%,인 건조분말을 얻었다. 이 연구는 1969년도 문교부 학술연구 조성비로 이루워진 것이며 본 연구에 헌신적인 보조를 아끼지 않았던 박 관화 군에게 감사하는 바이다.

Protein Adsorption on the Nickel-coated Glass Slide for Protein Chips

  • The adsorption of proteins on the surface of glass slides is essential for the construction of protein chips. Here, we report that a Histidine (His)-tagged protein protein has been efficiently adsorbed on glass coated with nickel. A variety of nickel chloride-coated plates were prepared by the spin-coating method and adsorbed to the His-tagged protein. When the protein was adsorbed onto the surface of a variety of nickel chloride-coated glass slides, the efficiency of protein adsorption was dependent upon the coating conditions such as nickel chloride concentration, the spin speed and the drying temperature. The slides appropriate for protein adsorption were obtained when the slides were coated with 11%(w/w) of $NiCl_2$ at the spin speed of 4000 rpm for 20 sec and then dried at higher than 40°C. The physical properties of their nickel chloride thin layer were characterized by scanning electron microscopy. x-ray diffraction and atomic force microscopy, finding that the nickel chloride particles were around 10 nm in diameter and uniformly crystallized at 101 faces. These results show that nickel chloride-coated slides prepared by the spin-coating method are utilizable for the construction of Histagged protein chips.

구조 생물학을 이용한 Antifreeze protein의 최근 연구동향 (Recent Advances in Structural Studies of Antifreeze Proteins)

  • Antifreeze proteins (AFPs) have ice binding affinity, depress freezing temperature and inhibit ice recystallization which protect cellular membranes in polar organisms. Recent structural studies of antifreeze proteins have significantly expanded our understanding of the structure-function relationship and ice crystal growth inhibition. Although AFPs (Type I-IV AFP from fish, insect AFP and Plant AFP) have completely different fold and no sequence homology, they share a common feature of their surface area for ice binding property. The conserved ice-binding sites are relatively flat and hydrophobic. For example, Type I AFP has an amphipathic, single ${\alpha}$-helix and has regularly spaced Thr-Ala residues which make direct interaction with oxygen atoms of ice crystals. Unlike Type I AFP, Type II and III AFP are compact globular proteins that contain a flat ice-binding patch on the surface. Type II and Type III AFP show a remarkable structural similarity with the sugar binding lectin protein and C-terminal domain of sialic acid synthase, respectively. Type IV is assumed to form a four-helix bundle which has sequence similarity with apolipoprotein. The results of our modeling suggest an ice-binding induced structural change of Type IV AFP. Insect AFP has ${\beta}$-helical structure with a regular array of Thr-X-Thr motif. Threonine residues of each Thr-X-Thr motif fit well into the ice crystal lattice and provide a good surface-surface complementarity. This review focuses on the structural characteristics and details of the ice-binding mechanism of antifreeze proteins.

지리산 싸리의 지질 구성 지방산과 유리 아미노산 및 구성 총 아미노산 조성에 관하여 (Compositions of Fatty Acid, Free Amino Acid and Total Amino Acid of Lespedeza x chiisanensis T. LEE)

  • 지리산 싸리의 유리 아미노산, 총 아미노산 조성과 총 지방산 조성을 규명하기 위한 gas chromatography와 HPLC를 사용하여 분리 정량한 결과는 다음과 같다. 조지방 함량은 11.13%이고, 조단백질 함량은 5.18%로 조성되어 있다. 유리 아미노산의 총 함량은 433.14mg/100g으로 조성되어 있으며, 유리 아미노산 중에서 필수 아미노산의 총 함량은 94.84mg/100g으로 21.90%를 차지하고 있다. 총 아미노산의 함량은 3159.85mg/100g으로 조성되어 있으며, 총 아미노산중에서 필수 아미노산의 함량은 1068.18mg/100g으로 35.80%를 차지하고 있다. 총 지방산 조성은 $C_{18\;:\;2}$가 45.05%로 제일 많이 함유되어 있고, $C_{18\;:\;3}=18.71%,\;C_{19}=14.70%,C_{18\;:\;1}=6.81%,\;C_{16}=4.35%,\;C_{16\;:\;1}=1.59%$순으로 함유되고 있고, 탄소소 홀수 지방산이 14.70% 함유되어 있으며, 불포화 지방산이 72.44%이고, 필수 지방산은 63.76%를 차지하고 있는 결과로 볼 때 유지자원 및 사료의 자원으로 가치가 있다고 판단된다.

안동지역 농촌주부의 영양소 섭취 실태에 관한 조사 연구 (A Study on Nutrient Intake of Rural Housewives)

  • 본 연구는 안동지역 농촌주부 222명을 대상으로 영양소 및 식품섭취 실태를 조사하였고, 영양소 섭취 실태에 영향을 미치는 요인들의 상호관련성을 분석한 결과는 다음과 같다. 조사대상자들의 연령분포는 45~49세가 29.3%로 가장 높은 비율이었고, 평균 연령은 44세였다. 경제수준은 종류가 44.1%로 가장 많았으며, 교육수준은 국민학교 졸업이 37.4%로 가장 많았다. 1일 1인 총식품섭취량은 113g이었으며, 식품군별 섭취량을 보면 곡류가 35.0%로 가장 높았다. 동물성 식품은 전체 식품섭취량중에서 11.6%였고, 동물성 단백질 식품은 총단백질 섭취량의 40.4%로 나타났다. 영양소 섭취 상태는 모든 영양소가 권장량을 충족시켰으며, 특히 단백질 섭취에 있어서 동물성 식품에서 40.4%의 높은 섭취율을 보였다. Beaton의 방법에 의한 영향결핍 가능성은 열량이 19.8%, 단백질이 10%, 칼슘이 14%, 및 철이 25.2%였다. 단백질의 섭취와 소득수준 및 식비와는 양의 상관관계를 나타내었다. 주부의 교육수준(X)와 열량섭취량(Y)과의 회귀분석에서 Y=1890.9+25.14X($r^{2}=21.7$), 그리고 소득수준(X)와 단백질 섭취량(Y)과의 회귀분석에서는 Y=40.6+0.63X($r^{2}=27.6$)을 보여주었다.

Expression, Purification and Characterization of the BLM binding region of human Fanconi Anemia Group J Protein

  • FANCJ is a DNA helicase which contributes genome stability by resolving G-quadruplex DNA from 5' to 3' direction. In addition to main ATPase helicase core, FANCJ has the protein binding region at its C-terminal part. BRCA1 and BLM are the binding partner of FANCJ and these protein-protein interactions contribute genomic stability and the proper response to replication stress. As the first attempt for studying FANCJ-BLM interaction, we prepared BLM binding region of FANCJ and characterized with CD and NMR spectroscopy. FANCJ (881-941) with N-ter 6xHis was purified as the oligomer. Secondary structure prediction based on CD data revealed that FANCJ (881-941) composed with ${\beta}$ sheet, turn and coils.$^1H-^{15}N$ HSQC spectra showed nonhomogeneous peak intensities with less number of peaks comparing than the number of amino acids in the construct. It indicated that optimization should be necessary for detailed further structural studies.
