Identification and Functional Analysis of LsMNPV Anti-apoptosis Genes

  • Three anti-apoptosis genes, Ls-iap2, iap3 and p49 were found in Leucania separata multiple nuclear polyhedrovirus. Amino acid sequence homology of Ls-IAP2 and Ls-IAP3 with Op-IAP2 and Op-IAP3 from Orgyia pseddotsugata MNPV were 20% and 42%, while that of Ls-P49 is 28% with Sl-P49 from Spodoptera littorolis MNPV. Ls-IAP2 contains one baculoviral IAP repeat (BIR) domain followed by a RING domain, while Ls-IAP3 contains two BIRs and a RING. Ls-P49 contains a reactive site loop, predicted cleavage site (KKLD$^{74}{\downarrow}$G) that is different from Sl-P49 (TVID$^{94}{\downarrow}$G). Expressed Ls-iap3 or Ls-p49 under presence of actinomycin D in SF9 cells, DNA ladder assayrevealed that Ls- IAP3 or Ls-P49 could block the apoptosis of SF9 cells induced by actinomycin D. Replication of p35 deficient-mutant Autographa californica MNPV in SF9 cells was also rescued when Ls-iap3 or Ls-p49 was expressed transiently. No anti-apoptotic activity was observed for Ls-IAP2. The results showed that both of Ls-IAP3 and Ls-P49 were functional apoptotic suppressors in SF9 cells.

Nonlinear creep model based on shear creep test of granite

  • The creep characteristics of rock is of great significance for the study of long-term stability of engineering, so it is necessary to carry out indoor creep test and creep model of rock. First of all, in different water-bearing state and different positive pressure conditions, the granite is graded loaded to conduct indoor shear creep test. Through the test, the shear creep characteristics of granite are obtained. According to the test results, the stress-strain isochronous curve is obtained, and then the long-term strength of granite under different conditions is determined. Then, the fractional-order calculus software element is introduced, and it is connected in series with the spring element and the nonlinear viscoplastic body considering the creep acceleration start time to form a nonlinear viscoplastic creep model with fewer elements and fewer parameters. Finally, based on the shear creep test data of granite, using the nonlinear curve fitting of Origin software and Levenberg-Marquardt optimization algorithm, the parameter fitting and comparative analysis of the nonlinear creep model are carried out. The results show that the test data and the model curve have a high degree of fitting, which further explains the rationality and applicability of the established nonlinear visco-elastoplastic creep model. The research in this paper can provide certain reference significance and reference value for the study of nonlinear creep model of rock in the future.

Differencing Multiuser Detection Using Error Feedback Filter for MIMO DS-UWB System in Nakagami Fading Channel

  • A differencing multiuser detection (MUD) method is proposed for multiple-input multiple-output (MIMO) direct sequence (DS) ultra-wideband (UWB) system to cope with the multiple access interference (MAI) and the computational efficiency in Nakagami fading channel. The method, which combines a multiuser-interference-cancellation-based decision feedback equalizer using error feedback filter (MIC DFE-EFF), a coefficient optimization algorithm (COA) and a differencing algorithm (DA), is termed as MIC DFE-EFF (COA) with DA for short. In the paper, the proposed MUD method is illuminated from the rudimental MIC DFE-EFF to the advanced MIC DFE-EFF (COA) with DA step by step. Firstly, the MIC DFE-EFF system performance is analyzed by minimum mean square error criterion. Secondly, the COA is investigated for optimization of each filter coefficient. Finally, the DA is introduced to reduce the computational complexity while sacrificing little performance. Simulations show a significant performance gain can be achieved by using the MIC DFE-EFF (COA) with DA detector. The proposed MIC DFE-EFF (COA) with DA improves both bit error rate performance and computational efficiency relative to DFE, DFE-EFF, parallel interference cancellation (PIC), MIC DFE-EFF and MIC DFE-EFF with DA, though it sacrifices little system performance, compared with MIC DFE-EFF (COA) without DA.

  • The engineering design process is a creative process, and the designers must repeatedly apply Undo/Redo operations to modify CAD models to explore new solutions. Undo/Redo has become one of most important functions in interactive graphics and CAD systems. Undo/Redo in a collaborative CAD system is also very helpful for collaborative awareness among a group of cooperative designers to eliminate misunderstanding and to recover from design error. However, Undo/Redo in a collaborative CAD system is much more complicated. This is because a single erroneous operation is propagated to other remote sites, and operations are interleaved at different sites. This paper presents a multi-user selective Undo/Redo approach in full distributed collaborative CAD systems. We use site ID and State Vectors to locate the Undo/Redo target at each site. By analyzing the composition of the complex CAD model, a tree-like structure called Feature Combination Hierarchy is presented to describe the decomposition of a CAD model. Based on this structure, the dependency relationship among features is clarified. B-Rep re-evaluation is simplified with the assistance of the Feature Combination Hierarchy. It can be proven that the proposed Undo/Redo approach satisfies the intention preservation and consistency maintenance correctness criteria for collaborative systems.

Role of span length in the adaptation of implant-supported cobalt chromium frameworks fabricated by three techniques

  • PURPOSE. This study evaluated the effect of span length on the adaptation of implant-supported cobalt chromium frameworks fabricated by three techniques. MATERIALS AND METHODS. Models with two solid abutment analogs at different inter-abutment distances were digitized using a laboratory scanner. Frameworks of two-, three-, and four-unit fixed prostheses were designed by a computer. Six dots with a diameter of 0.2 mm were preset on the surface of each framework. A total of 54 implant-supported cobalt chromium frameworks were fabricated by milling, selective laser melting (SLM), and cast techniques. The frameworks were scanned and exported as Stereolithography files. Distances between two dots in X, Y, and Z coordinates were measured in both the designed and fabricated frameworks. Marginal gaps between the framework and the abutments were also evaluated by impression replica method. RESULTS. In terms of distance measurement, significant differences were found between three- and four-unit frameworks, as well as between two- and four-unit frameworks prepared by milling technique (P<.05). Significant differences were also noted between two- and three-unit frameworks, as well as between two- and four-unit frameworks prepared by cast technique (P<.05). The milling technique presented smaller differences than the SLM technique, and the SLM technique showed smaller differences than the cast technique at any unit prostheses (P<.05). Evaluation with the impression replica method indicated significant differences among the span lengths for any fabrication method (P<.05), as well as among the fabrication methods at any unit prostheses (P<.05). CONCLUSION. The adaptation of implant-supported cobalt chromium frameworks was affected by the span length and fabrication method.

The actuation equation of macro-fiber composite coupled plate and its active control over the vibration of plate and shell

  • Plate and shell structure is widely applied in engineering, i.e. building roofs, aircraft wings, ship platforms, and satellite solar arrays. Its vibration problem has become increasingly prominent due to the tendency of lightening, upsizing and flexibility. As a new smart material with great actuating force and toughness, macro-fiber composite (MFC) is composed of piezoelectric fiber and epoxy resin basal body, which can be directly pasted onto the surface of plate and shell and is suitable for vibration control. This paper deduces the actuation equation of MFC coupled plate in different boundary conditions, an equivalent finite element modeling method is proposed which uses MFC actuating force as the applied excitation, and on this basis the active control simulation and experiment of MFC over plate and shell structure vibration are accomplished. The results indicate that MFC is able to implement effective control over plate and shell structure vibration in multi-band range. The comparison between experiment and simulation proves that the actuation equation deduced herein, effective and practicable, can be applied into the simulation calculation of MFC vibration control over plate and shell structure.

Glutathione S-transferase M1 Null Genotype and Hepatocellular Carcinoma Susceptibility in China and India: Evidence from an Updated Meta-analysis

  • Background: Glutathione S-transferase M1 (GSTM1) have been reported to be associated with hepatocellular carcinoma. However, the effect of the GSTMl null genotype was divergent in the literature and we therefore performed the present meta-analysis to explore the relationship in detail. Materials and Metbods: Reported studies were searched from 1990 to March 1, 2014 in PubMed and Wanfang Med Online. The total odds oatio (OR) and 95% CI were calculated and analyzed by Review Manager 5.1 and STATE 12. Results: Total OR was calculated from 26 articles with 3,769 cases and 5,517 controls and the association proved significant (OR [95%CI]=1.50 [1.25, 1.80], P<0.05) in the Chinese population. However, there was no significant association between hepatocellular carcinoma risk among subjects carrying the GSTM1 null genotype (OR [95%CI]=1.20 [0.88-1.64], P=0.24) in subgroups of publication in English and in Indian populations (OR [95%CI]=1.80 [0.80-4.20], P=0.15). Conclusions: The GSTM1 deletion polymorphism might not have a significant effect on the susceptibility of hepatocellular carcinoma overall.

Experimental study on the mechanical property of coal and its application

  • Brazilian splitting tests, uniaxial compression tests and triaxial compression tests are carried out on the coal samples cored from Shanxi group $II_1$ coal seam of Jiaozuo coal mine, Henan province, China, to obtain their property parameters. Considering the bedding has notable effect on the property parameter of coal, the samples with different bedding angles are prepared. The effects of bedding on the anisotropic characteristics of the coal seam are investigated. A geological geomechanical model is built based on the geology characteristics of the Jiaozuo coal mine target reservoir to study the effects of bedding on the fracture propagations during hydraulic fracturing. The effects of injection pressure, well completion method, in-situ stress difference coefficient, and fracturing fluid displacement on the fracture propagations are investigated. Results show bedding has notable effects on the property parameters of coal, which is the key factor affecting the anisotropy of coal. The hydraulic cracks trends to bifurcate and swerve at the bedding due to its low strength. Induced fractures are produced easily at the locations around the bedding. The bedding is beneficial to form a complicated fracture network. Experimental and numerical simulations can help to understand the effects of bedding on hydraulic fracturing in coalbed methane reservoirs.

Alternative Splicing of Breast Cancer Associated Gene BRCA1 from Breast Cancer Cell Line

  • Breast cancer is the most common malignancy among women, and mutations in the BRCA1 gene produce increased susceptibility to these malignancies in certain families. In this study, the forward 1-13 exons of breast cancer associated gene BRCA1 were cloned from breast cancer cell line ZR-75-30 by RT-PCR method. Sequence analysis showed that nine BRCA1 splice forms were isolated and characterized, compared with wild-type BRCA1 gene, five splice forms of which were novel. These splice isoforms were produced from the molecular mechanism of 5' and 3' alternative splicing. All these splice forms deleting exon 11b and the locations of alternative splicing were focused on two parts:one was exons 2 and 3, and the other was exons 9 and 10. These splice forms accorded with GT-AG rule. Most these BRCA1 splice variants still kept the original reading frame. Western blot analysis indicated that some BRCA1 splice variants were expressed in ZR-75-30 cell line at the protein level. In addition, we confirmed the presence of these new transcripts of BRCA1 gene in MDA-MB-435S, K562, Hela, HLA, HIC, H9, Jurkat and human fetus samples by RT-PCR analysis. These results suggested that breast cancer associated gene BRCA1 may have unexpectedly a large number of splice variants. We hypothesized that alternative splicing of BRCA1 possibly plays a major role in the tumorigenesis of breast and/or ovarian cancer. Thus, the identification of cancer-specific splice forms will provide a novel source for the discovery of diagnostic or prognostic biomarkers and tumor antigens suitable as targets for therapeutic intervention.

Mangiferin ameliorates cardiac fibrosis in D-galactose-induced aging rats by inhibiting TGF-β/p38/MK2 signaling pathway

  • Aging is the process spontaneously occurred in living organisms. Cardiac fibrosis is a pathophysiological process of cardiac aging. Mangiferin is a well-known C-glucoside xanthone in mango leaves with lots of beneficial properties. In this study, rat model of cardiac fibrosis was induced by injected with 150 mg/kg/d D-galactose for 8 weeks. The age-related cardiac decline was estimated by detecting the relative weight of heart, the serum levels of cardiac injury indicators and the expression of hypertrophic biomakers. Cardiac oxidative stress and local inflammation were measured by detecting the levels of malondialdehyde, enzymatic antioxidant status and proinflammatory cytokines. Cardiac fibrosis was evaluated by observing collagen deposition via masson and sirius red staining, as well as by examining the expression of extracellular matrix proteins via Western blot analysis. The cardiac activity of profibrotic TGF-β1/p38/MK2 signaling pathway was assessed by measuring the expression of TGF-β1 and the phosphorylation levels of p38 and MK2. It was observed that mangiferin ameliorated D-galactose-induced cardiac aging, attenuated cardiac oxidative stress, inflammation and fibrosis, as well as inhibited the activation of TGF-β1/p38/MK2 signaling pathway. These results showed that mangiferin could ameliorate cardiac fibrosis in D-galactose-induced aging rats possibly via inhibiting TGF-β/p38/MK2 signaling pathway.
