The crude ethanolic extract of the leaves of Clerodendrum viscosum (Family: Verbenaceae) was evaluated for its antioxidant and analgesic activities to investigate the scientific basis of the traditional uses. The antioxidant property of the extract was assessed by 1, 1-diphenyl -2- picryl hydrazyl (DPPH) free radical scavenging assay. The extract showed prominent antioxidant activity ($IC_{50}about{\sim}16{\mu}g/ml$) which was comparable to standard drug ascorbic acid ($IC_{50}about{\sim}15{\mu}g/ml$).The extract produced significant (P<0.001) writhing inhibition in acetic acid induced writhing in mice at the dose of 125 mg, 250 mg and 500 mg/kg body weight respectively, which were comparable to the standard drug diclofenac sodium. The results tend to suggest that the crude leaves extract at the above doses have antioxidant and analgesic activities and indicate that it might possess biologically active constituents having free radical scavenging and analgesic activities respectably.

The ethanol extract of leaves of the mangrove Aegiceras corniculatum Blanco (Myrsinaceae) was screened for its antinociceptive, antidiarrhoeal and cytotoxic activities. The extract produced significant writhing inhibition in acetic acid-induced writhing in mice at the oral dose of 250 and 500 mg/kg body weight (P < 0.001), which was comparable to the standard drug diclofenac sodium at the dose of 25 mg/kg of body weight. When tested for its antidiarrhoeal effects on castor oil induced diarrhoea in mice, it increased mean latent period and decreased the frequency of defecation significantly at the oral dose of 500 mg/kg body weight (P<0.05; P<0.01) comparable to the standard drug loperamide at the dose of 50 mg/kg of body weight. Moreover, when tested for toxicity using brine shrimp, the extract showed potent activity against the brine shrimp Artemia salina ($LC_{50}$ 10 mg/ml). The overall results tend to suggest the antinociceptive, antidiarrhoeal and cytotoxic activities of the extract.
The extract of Galgun-tang at oral doses of 200 and 400 mg/kg showed remarkable inhibition of 42.5% and 45.0%, respectively, on the carrageenin edema in rat paw. In case of combined oral administration of the extract at each dose of 100, 200 and 400mg/kg with 100mg/kg of aspirin, the inhibitions were 70.5%, 83.6% and 80.7% respectively. These results showed the remarkable effects as compared with those of aspirin which showed 50.3% and 69.4% of inhibition at doses of 100 and 200mg/kg, respectively. On the other hand, on the writhing syndrome in mice induced by 0.7% acetic acid solution, the oral administration of the extract showed remarkable inhibition of 18.5 and 47.7% at doses of 200 and 400mg/kg, respectively. In case of combined oral administration of the extract at each dose of 100, 200 and 400mg/kg with 100mg/kg of aspirin, the inhibition percentages of writhing syndromes were 66.6, 72.3 and 80.8, respectively, whereas the inhibition at doses of 100 and 200mg/kg of aspirin showed 59.9 and 69.5%, respectively.
Objectives : We want to know the effect of Gyejakjimo-tang in mice model of acute pain. Methods : We investigated writhing reflex in mice with acetic acid-induced abdominal pain using mice, we observed c-Fos protien expression by immunohistochemistry dyeing method in the paraventricular nucleus(PVN) and supraoptic nucleus(SON) of the hypothalamus. Results : All of Gyejakjimo-tang treated group suppressed acetic acid-induced writhing response as acetic acid injuction group, but in 100㎎/㎏ Gyejakjimo-tang treated group and 200㎎/㎏ Gyejakjimo-tang treated groups represernted significance. All of Gyejakjimo-tang treated groups(50, 100 and 200㎎/㎏ Gyejakjimo-tang-treated group), Fos-positive cells in PNV significantly decreased as acetic acid injuction group, and All of Gyejakjimo-tang-treated groups Fos-positive cells in SON significantly decreased as acetic acid injuction group. Conclusions : The present results showed that the mice pre-treated with the aqueous extract of Gyejakjimo-tang showed analgesic effect on acetic acid-induced abdominal pain.
This study was investigated on anti-inflammatory and analgesic activity on Gu-Mi-Gang-Hwal-Tang which has been used for the treatment of common cold and headache, arthralgia, fever in oriental medicine. The anti-inflammatory activities of Gu-Mi-Gang-Hwal-Tang water extract (GMGHT) on the carrageenin induced edema, acetic acid induced edema, and corton oil induced granuloma pouch in rats were examined. The analgesic effect of the GMGHT was investigated utilizing acetic acid induced writhing syndrome in mice. The antipyretic activity of the GMGHT was also investigated. The oral administration of GMGHT has been shown the anti-inflammatory activities in 1% carrageenin and acetic acid induced edema in rats. The GMGHT has been shown significant inhibitory effects on glanuloma and exudate formation in rats, especially the GMGHT (450 mg/kg) has the efficacy more than the GMGHT (150 mg/kg). The writhing syndromes induced by acetic acid in mice were inhibited, especially analgesic activity of the GMGHT (450 mg/kg) is similar to that of piroxicam and antipyretic activities were recognized in rats. In the present study, the GMGHT is indicated to have the anti-inflammatory, analgesic and antipyretic activities.
For developing plant anti-inflammatory agents, extracts from Lonicera japonica were obtained and evaluated for their anti-inflammatory and analgesic activities using acute/chronic inflammatory models and writhing tests. It was found that the extracts with a modified extraction method showed higher anti-inflammatory and analgesic activities than those of the extracts based on the ancient literatures.
Inflammatory mediators such as interleukin-1 (IL-1), tumor necrosis $factor-{\alpha}\;(TNF-{\alpha}),\;interferon-{\gamma}\;(IFN-{\gamma})$ and lipopolysaccharide (LPS) are thought to play major roles in joint diseases such as a rheumatoid arthritis (RA), and there is considerable evidence playing a role for these cytokines in osteoarthritis (OA). Therefore, we have studied the effects on anti-inflammation and analgesic by ethyl acetate fraction from 70% ethanol extract of Artemisia capillaries (EAC). As a positive control, apigenin, which is known as an anti-inflammatory agent as an iNOS inhibitor, was used and showed the dose-dependent inhibitory effect. EAC showed strong inhibitory efficacy against cytokine-induced proteoglycan degradation, $PGE_2$ production, nitric oxide (NO) production, and matrix-matalloproteinases (MMPs) expression in rabbit articular chondrocyte. In the writhing test induced by acetic acid, EAC $(200{\sim}400\;mg/kg)$ exhibited a dose-dependent inhibition of writhing. The results indicate that EAC have anti-inflammatory and analgesic activities, and could be a good herbal medicine candidate for curing of RA and/or OA.
This study was undertaken to evaluate the antinociceptive and antiinflammatory effects of the heartwood extract of Rhus verniciflua (Anacardiaceae) and the two major components, sulfuretin and fustin. The MeOH extract, its EtOAc-soluble portion and sulfuretin showed significant antinociceptive activity in writhing and hot plate test assays and antiinflammaory effects in carrageenan-induced hind paw edema in rats. In particular, treatment of sulfuretin with 10 mg/kg dose (i.p.) reduced writhing frequency by 48.0% (p<0.01) compared to that of a control group. Further, the treatment of sulfuretin (5, 10 mg/kg, i.p.) for 7 days prevented the carrageenan-induced hind paw edema significantly (p<0.01). The antiinflammatory effect of sulfuretin was also confirmed by microscopic observation of mast cell numbers in muscle. In addition, sulfuretin suppressed the cyclooxygenase- 2 (COX-2) activity $(IC_{50}\;=\;28.7\;{\mu}M)$ in lipopolysaccharide-activated macrophage cells. This result indicates that the inhibitory effect of sulfuretin on COX-2 may be one of the antinociceptive/antiinflammatory mechanism.
Chang-Back-San and Chil-Mi-Chang-Back-San have been used for the treatment of neuralgia and arthritis in traditional medicine. The anti-inflammatory activities of Chang-Back-San and Chil-Mi-Chang-Back-San water extract (CBSE and CCBSE) on the carrageenin induced edema, Corton oil induced granuloma pouch, and adjuvant arthritis in rats were examined. The analgesic effects of the CBSE and CCBSE were also investigated utilizing acetic acid induced writhing syndrome in mice. The oral administration of CBSE and CCBSE showed to have the anti-inflammatory activities in 1% carrageenin induced edema in rats. They also showed significant inhibitory effects on granuloma and exudate formation in rats. In the method of adjuvant arthritis, they orally administered for 19 days, inhibited the hind paw edema in rats from 3rd day to 19th day, especially CCBSE has the efficacy more than CBSE. They significantly decreased the number of writhing syndromes induced by acetic acid in mice. In the present study, CBSE and CCBSE were indicated to have the anti-inflammatory and analgesic activities.
