흰쥐의 피부상처 동물모델에서 고삼(苦蔘)이 신생혈관형성에 미치는 영향 (Enhancement of Angiogenesis by Sophorae Radix in Full-thickness Skin Wound of Rats)

  • Wound healing is the restoration in injured skin tissue and one of the most important therapeutic targets. The process consists of inflammation, proliferation, and remodeling. There have been reported multiple factors that accelerate and delay wound healing. In this study we tested the hypothesis that Sophorae Radix extract would improve the recovery of wound healing after full-thickness skin wound in rats. Twenty young male Sprague-Dawley rats were used for the studies. A full-thickness skin wound was made on the dorsal skin of the rats. Either Sophorae Radix water extract (SR) or saline (Control) was orally administrated every day. The wound area was measured and the percentages of wound contraction, wound healed and wound epithelization were evaluated. After 15 days, the skin tissues were excised and examined by histopathological and immunohistochemistrical method. In results, wound area in SR group was significantly decreased to compared with Control. SR group showed the significant enhancements in the percentages of wound contraction, wound healed and wound epithelization. Histopathological examination revealed that SR induces neo-vascularization potential in wound healing process. SR treatment in rats significantly accelerated cutaneous wound healing in the angiogenesis process by increasing VEGF and TGF-${\beta}1$ synthesis. These results suggest that Sophorae Radix enhance skin wound repair by increasing the angiogenic agents such as VEGF and TGF-${\beta}1$.

Enhancing Dermal Matrix Regeneration and Biomechanical Properties of $2^{nd}$ Degree-Burn Wounds by EGF-Impregnated Collagen Sponge Dressing

  • To better define the relationship between dermal regeneration and wound contraction and scar formation, the effects of epidermal growth factor (EGF) loaded in collagen sponge matrix on the fibroblast cell proliferation rate and the dermal mechanical strength were investigated. Collagen sponges with acid-soluble fraction of pig skin were prepared and incorporated with EGF at 0, 4, and 8 $\mu$g/1.7 $cm^{2}$. Dermal fibroblasts were cultured to 80$\%$ confluence using DMEM, treated with the samples submerged, and the cell viability was estimated using MTT assay. A deep, $2^{nd}$ degree- burn of diameter 1 cm was prepared on the rabbit ear and the tested dressings were applied twice during the 15-day, post burn period. The processes of re-epithelialization and dermal regeneration were investigated until the complete wound closure day and histological analysis was performed with H-E staining. EGF increased the fibroblast cell proliferation rate. The histology showed well developed, weave-like collagen bundles and fibroblasts in EGF-treated wounds while open wounds showed irregular collagen bundles and impaired fibroblast growth. The breaking strength (944.1 $\pm$ 35.6 vs. 411.5 $\pm$ 57.0 Fmax, $gmm^{-2}$) and skin resilience (11.3 $\pm$ 1.4 vs. 6.5 $\pm$ 0.6 mJ/$mm^{2}$) were significantly increased with EGF­treated wounds as compared with open wounds, suggesting that EGF enhanced the dermal matrix formation and improved the wound mechanical strength. In conclusion, EGF-improved dermal matrix formation is related with a lower wound contraction rate. The impaired dermal regeneration observed in the open wounds could contribute to the formation of wound contraction and scar tissue development. An extraneous supply of EGF in the collagen dressing on deep, $2^{nd}$ degree-burns enhanced the dermal matrix formation.

  • This study was performed to investigate the wound healing effects of Allium cepa L. on Staphylococcus intermedius contaminated wound. 4 full-thickness skin wounds ($2\;cm{\times}2\;cm$) were made in the back and Staphylococcus intermedius was inoculated in seven dogs. In each dog, the wounds were treated with onion extract (experimental group) and normal saline (control group), respectively. The wound contraction rate of the experimental group was higher than that of the control group during the experimental period. The occupation rate showed that the wounds of the experimental groups contracted more vigorously than that of the control group in the early period of wound healing. The mean value of tensile strength on the 22 day after wounding was $4,741{\pm}848\;g/cm^2$ in the experimental group and $2,400{\pm}397\;g/cm^2$ in the control group. The number of S. intermedius in the experimental groups was decreased more rapidly than that of the control group. These results suggest that application of activated onion extract in like full-thickness skin wounds promote wound healing.

Anti-inflammatory effects of low-level laser in burn wound models in rats

  • Objective: The anti-inflammatory effects of low-level laser in burn wound model in rats were investigated. Design: Randomized controlled trial. Methods: The rats were assigned to three experimental groups. Group I received second-degree burn wounds; Group II received dressing film and low-level laser ($1.2J/cm^2$) treatment after a burn wound; Group III received dressing film and low-level laser ($2.3J/cm^2$) treatment after a burn wound. After inducing a deep second-degree burn wound, the wound was observed every day and the burn area diameter and retraction quantification at 1, 7, and 14 days were evaluated. Low-level laser was investigated on hematological parameters after 14 days. Effects of low-level laser on the inflammatory cytokines (tumor necrosis $factor-{\alpha}$ [$TNF-{\alpha}$] and interleukin-6 [IL-6]) concentrations in the serum were evaluated using immunosorbent assay kits. Results: Group III showed a significant difference in wound size on days 7 and 14 compared to Group I (p<0.05). Group II showed a significant difference in wound size on day 14 compared to Group I (p<0.05). For wound contraction percentage, both laser therapy treatment groups showed a significant difference compared with Group I (p<0.05). There was also a significant difference in wound contraction percentage in Group III compared to Group II (p<0.05). Compared with the model control group, decreased $TNF-{\alpha}$ and IL-6 levels in the serum was observed at 14 days after burn wound induction. Conclusions: The results of this study suggest that low-level laser therapy can assist in burn wound healing, which might be associated with decreased concentrations of $TNF-{\alpha}$ and IL-6 related proinflammatory cytokines.

Therapeutic Effects of Amnion-Conjugated Chitosan-Alginate Membranes on Diabetic Wounds in an Induced Diabetic Swine Model: An In Vitro and In Vivo Study

  • Background Chitosan (CS) is a well-known antimicrobial dressing material. Moreover, widely used amniotic membranes contain growth factors beneficial for wound healing. Herein, we created a novel amnion-conjugated CS-alginate membrane dressing and tested its wound healing potency in a diabetic swine model. Methods The bovine amniotic powder growth factor contents were evaluated by protein assay, and the powder's wound healing effects were assessed in vitro by HaCaT cell scratch closure. In vivo, two minipigs developed streptozotocin-induced diabetes. Serial serum glucose measurements and intravenous glucose tolerance tests were performed to confirm their diabetic status. Twelve square-shaped wounds created on each pig's back were randomly divided into control (n = 4), CS (n = 4), and amnion-CS (AC; n = 4) groups and treated accordingly with different dressings. Wound healing in each group was assessed by measuring wound contraction over time, capturing wound perfusion with indocyanine green (ICG) angiography, and histologically analyzing inflammatory markers. Results Amniotic powder elution promoted HaCaT cell migration in the scratch wound model, suggesting its beneficial in vitro wound healing effects. In vivo, the CS and AC groups showed earlier wound contraction initiation and reepithelialization and earlier wound perfusion improvement by ICG angiography than the control group. Additionally, the wound size of the AC group at week 3 was significantly smaller than those in the control group. There was no significant difference in the numbers of acute and chronic inflammatory cells between the groups. Conclusion The amnion-conjugated CS-alginate membrane, as well as CS dressing alone, could be a favorable dressing option for diabetic wounds.

  • Objectives : In this study, we investigated whether Astragali Radix Pharmacocupuncture (ARP) has an effective on the full thickness defect wound healing process of mouse. Methods : A total of 50 mice (ICR mouse, 7 week-old male) were divided into control group and ARP group. A single full thickness skin defect was made on the dorsal side of the each mouse using an 8mm diameter biopsy punch. Control group were treated with 0.2㎖ saline and ARP group were treat with 0.2ml ARP at 8 points around the wound every three days total 4 times during the experimental period. The change in wound size, contraction rate, healing rate, and epithelization rate was measured by digital images taken on days 3, 6, 9, and 13, and evaluated using a digital image analysis program. Tissues were collected for histological analysis, RT-PCR, and Western blot on days 3, 6, 9, and 15. Results : The results are as follows. ARP group accelerated the rate of wound contraction, wound healing and epithelization compared to the control group. ARP group showed the decrease of inflammatory cells in early inflammatory phase compared to the control group. ARP group upregulated PECAM-1 mRNA and protein expression compared to the control group. ARP group inhibited the scar width and area compared to the control group. Conclusions: ARP showed positive effects on wound healing through the inhibition of inflammatory reaction and increase of PECAM-1 expression related to the wound healing process.

Studies on in vivo Wound Healing Activity of Cassia fistula Linn. Leaves (Leguminosae) in Rats

  • Cassia fistula commonly known as Sundali was selected to evaluate its wound healing potentials based on traditional use and literature references. Methanol extract of C. fistula leaves were examined for its wound healing property in the form of an ointment in two types of wound models in rats: i) Excision wound model and ii) Incision wound model. The ointment of the leaf extract of two different concentrations (5% and 10% w/w ointment of leaves extract in simple ointment base) responded significantly in both models of wounds tested. The results were also comparable to that of standard drug, nitrofurazone in terms of wound contraction ability, epithelisation period, tensile strength and regeneration of tissue at wound area.

소양막이식편이 개의 전층 피부 창상치유에 미치는 효과 (Effects of bovine amniotic membrane graft on healing of full-thickness skin wound in dogs)

  • The purpose of the present study was to investigate the effect of bovine amniotic membrane grafts on healing of full-thickness skin wound in dogs. Two $3cm{\times}3cm$ area-matched full-thickness skin wounds were induced bilaterally on the dorsolateral aspect of the trunk of 15 dogs. Chlorhexidine-treated amnion, dried amnion, silver sulfadiazine and 0.9% sterile saline solution were applied on the wound area and examined grossly and histopathologically. Begining 14 days after wounding, amnion applied group had appreciably less amount of inflammatory exudate and hemorrhage than sulfadiazine and saline treated groups. From 14 days after wounding, the degree of wound contraction in amnion groups, especially in the dried amnion group was greater than that of the sulfadiazine and saline treated groups. The percentages of wounds completely healed on 28 days after wounding in saline treated group, chlorhexidine-treated amnion group, dried amnion group and sulfadiazine treated group were 33%, 50%, 83% and 50%, respectively. Microscopically neovascularization and fibrosis were first noticed on 5 days after wounding in the dried amnion group and sulfadiazine treated group, on 7 days in the chlorhexidine-treated amnion group and on 14 days in the saline treated group. Epithelialization in the dried amnion and sulfadiazine treated groups was first noticed on 9 days after wounding, which was faster than that in the other groups. The present study suggests that bovine amniotic membrane, especially dried bovine amnion is effective on healing of full-thickness skin wound in dogs through both wound contraction and epithelialization.

광물성 미네랄이 흰쥐 전층 피부창상 치유에 미치는 효과 (Effect of Ore Minerals on the Healing of Full-Thickness Skin Injury Model of Rat)

  • The oriental ore minerals, which mainly consisted of talc, actinolite, sericite, and halloysite were developed, and then used to examine the healing effect on the skin wound in rats. Full-thickness square wounds were formed on the backs of rats after the hairs on the dorsal surface were shaved. The ore minerals were applied to examine the healing effect from day 0 to 15 after wounding. Notable wound healings in terms of congestion around the wound, wound contraction and epithelialization were found in ore mineral-treated groups. Moreover, microscopic results revealed the formation of epithelial layer, hair follicles and progressive angiogenesis in ore mineral-treated groups, while complete epithelial layer could not be found in the control. These results suggest that ore minerals from Korean indigenous ores may have wound healing effect on the skin injury in rats.

Anti-ulcer and wound healing activity of Ruta graveolens

  • The effects of ethanol extract of Ruta graveolens on ulceration induced by ethanol and wound healing properties were investigated in mice. Oral administration of the extract reduced the incidence of ulcer, ulcer index and ulcer length produced by ethanol. The gastroprotective effects of R. graveolens were observed in a dose-dependent manner. However, this activity was statistically less potent than the anti-ulcer drug, omeprazole. When the extract applied on the wound, it promoted wound healing in mice. The wound contraction was faster in these mice when compared to untreated wounds. Based on these present findings, R. graveolens possesses anti-ulcer activity and promotes incision wound healing in mice.
