The purpose of this study was to investigate the gene profiles that were up- or down-regulated in the livers of high-fat diet-induced obese mice and $db_-/db_-$ mice with deficient leptin receptor. C57/BL6 normal mice and $db_-/db_-$ mice, respectively, were divided into two groups and fed a standard or high-fat diet for four weeks. Liver weight was unchanged in the normal mice but the high-fat diet led to a 10% weight increase in the $db_-/db_-$mice. Adipose tissue mass increased by about 88% in the normal mice that were fed a high-fat diet and by about 17% in the $db_-/db_-$mice on the high-fat diet. In terms of serum lipids, total cholesterol significantly increased in mice on the high-fat diet. Microarray analysis was carried out using total RNA isolated from the livers of standard or high-fat diet-fed mice of the normal and $db_-/db_-$ strains. The change of gene expression was confirmed by RT-PCR. About 1.6% and 6.8% of total genes, respectively, showed different expression patterns in the normal mice fed the high-fat diet and $db_-/db_-$ mice. As a result of microarray, many genes involved in metabolism and signal pathways were shown to have different expression patterns. Expression of Mgst3 gene increased in the livers of normal and $db_-/db_-$ mice that were fed a high-fat diet. Wnt7b and Ptk9l were down-regulated in the livers of the normal mice and $db_-/db_-$ mice that were fed a high-fat diet. In conclusion, a high-fat diet induced obesity and affected gene expression involved in metabolism and signal pathway.
Using MCF7 breast cancer cells, we tested the anticancer activity of metabolites from 130 strains of myxobacteria newly isolated in South Korea. Of these, three strains whose metabolites had high anticancer activity and low cell toxicity were selected and identified by their fruiting body morphology, cell morphology, and 16S rRNA sequence. Strains KYC4030 and KYC4048 were determined to be Myxococcus fulvus, whereas strain KYC4081 was identified as Corallococcus coralloides. We found that metabolites of M. fulvus KYC4048 demonstrated no toxicity in normal cells but specifically induced cancer cell death by suppressing the expression of WNT2B. This discovery highlights the value of assessing the metabolic and biomedical potential of myxobacteria, even those that are already known but were isolated from new areas, and the possible use of metabolites from M. fulvus KYC4048 in cancer treatment.
Objective : As more and more people are interested in appearance in modern society, the increasing number of hair loss population can have an important impact on psychological and social problems such as depression and inappropriate interpersonal symptoms. Therefore, much research is being done on treatments for alopecia using herbal extracts with relatively few side effects. This study was investigated about the effect of Achyranthis Radix (AR) extract with ethanol solvent on hair growth. Methods : We determined the promoting efficacy of AR-ethanol extract compared with minoxidil (MNXD) on the growth of human hair dermal papilla cells (HDPCs). Cell viability was measured by MTT assay and cell proliferation was confirmed by cell cycle analysis from flow cytometry in HDPCs. Also, we monitored the safe concentration range through MTT assay. And protein expression of hair growth-related genes (insulin-like growth factor 1 (IGF-1), Wnt3a, Protein kinase B (Akt), Extracellular signal-regulated kinase (Erk)) was monitored by western blot. Results : On cell cycle analysis, the G2/M phase was higher than that of the DW group in AR ethanol extract group at 0.05 and 0.1 mg/㎖. All protein expression levels of HDPCs were increased in AR ethanol extract groups and the MNXD group, compared to the DW group, respectively. Conclusion : As mentioned above, AR extract increased cell proliferation and the protein expression of IGF-1, Wnt3a, Akt, Erk in HDPCs. These results suggest that AR ethanol extract has promoted hair growth and it might be potential hair growth supplement.
전 세계적인 인구 고령화 현상으로 인하여 골다공증은 주요한 질병으로 대두되고 있다. 골다공증은 뼈의 질량과 강도가 감소하여 골절의 위험이 증가하는 질환으로 조골세포의 골 형성 및 파골세포의 골 흡수의 불균형으로 인해 발생하는 질환이다. 조골세포에 의한 골 형성은 BMP, RUNX2, $Wnt/{\beta}-catenin$ 경로 등을 통하여 활성화 되며, 파골세포에 의한 골 흡수는 RANKL과 RANK의 결합에 의해서 시작된다. 폐경기 여성은 호르몬 불균형에 의해 여러 질병의 위험에 처해 있으며, 폐경기 여성의 약 30%에서 관찰되는 골다공증은 폐경기 여성에게서 발생되는 가장 흔한 대사성 질환이기도 하다. Estrogen이 부족할 때 파골세포의 골 흡수가 촉진되므로, 특히 폐경 여성에서 골다공증의 발생위험이 증가하게 된다. 호르몬대체요법은 폐경기 증후군의 증상을 경감시키거나 치료하기 위해 널리 사용되어 왔으나, 호르몬 치료를 장기간 실시할 경우 유방암, 난소암, 자궁암 등의 부작용 위험성이 매우 높은 것으로 알려져 있다. 따라서 최근 들어 여러 부작용을 보완하기 위해 폐경기 증후군 증상에 대처할 수 있는 estrogen과 유사한 활성을 지닌 식물성 estrogen인 phytoestrogen에 대한 연구가 활발히 진행되고 있다. 따라서, 본 총설에서는 조골세포 및 파골세포의 분화 기전에 대한 선행연구를 알아보고 골 대사에서의 estrogen의 역할 및 phytoestrogen과 관련한 연구들에 대해서도 살펴보았다.
Inflammatory bowel disease (IBD) is a disease strongly associated with colorectal cancer (CRC) as a well-known precancerous condition. Alterations in DNA methylation and mutation in K-ras are believed to play an early etiopathogenic role in CRC and may also an initiating event through deregulation of molecular signaling. Epigenetic silencing of APC and SFRP2 in the WNT signaling pathway may also be involved in IBD-CRC. The role of aberrant DNA methylation in precancerous state of colorectal cancer (CRC) is under intensive investigation worldwide. The aim of this study was to investigate the status of promoter methylation of MGMT-B, APC1A and SFRP2 genes, in inflamed and normal colon tissues of patients with IBD compared with control normal tissues. A total of 52 IBD tissues as well as corresponding normal tissues and 30 samples from healthy participants were obtained. We determined promoter methylation status of MGMT-B, SFRP2 and APC1A genes by chemical treatment with sodium bisulfite and subsequent MSP. The most frequently methylated locus was MGMT-B (71%; 34 of 48), followed by SFRP2 (66.6 %; 32 of 48), and APC1A (43.7%; 21 of 48). Our study demonstrated for the first time that hypermethylation of the MGMT-B and the SFRP2 gene promoter regions might be involved in IBD development. Methylation of MGMT-B and SFRP2 in IBD patients may provide a method for early detection of IBD-associated neoplasia.
In order to testify the urate lowering effects of Daihwangmudan-tang(DMT), ICR mice were injected monosodium urate into the abdominal cavity and then DMT was administered on 2 and 4 days after Injection. Uric acid and triglyceride were measured as hematological indices of gout, and some genes related with this change were identified by ACP based GeneFishing PCR method and direct sequencing. From this experiment, DMT highly decreased the blood levels of uric acid and significantly suppressed and lowered the acute increment of triglyceride level. There were 11 differentially expressed genes(DEG) having relations with positive actions of DMT, and 4 major genes in the middle of DEGs were sequenced; Mfap 2, jagged 2, Hsd17b7, DkkI-1, These genes were supposed that several mechanisms through interleukin 1 and T-cell anergy, LDL cholesterol metabolism, wnt pathway would be related with the anti-inflammation effect against gout.
생약복합물인 SH21B는 황금(Scutellaria baicalensis Georgi), 행인(Prunus armeniaca Maxim), 마황(Ephedra sinica Stapf), 석창포(Acorus gramineus Soland), 포황(Typha orientalis Presl), 원지(Polygala tenuifolia Willd), 하엽(Nelumbo nucifera Gaertner)의 혼합(비율 3:3:3:3:3:2:2)으로 이루어졌다. SH21B는 예로부터 한의학에서 비만의 치료에 사용되어 왔으나 자세한 분자적 메커니즘과 효능에 대한 연구는 이루어지지 않았다. 본 연구진은 선행연구를 통해 SH21B가 지방세포의 분화에서 adipogenesis (지방세포형성)와 관련된 유전자를 조절하여 중성지방의 축적을 억제함을 밝혔다. 본 연구에서는, microarray 기술을 이용하여 adipogenesis의 in vitro 모델인, 3T3-L1 세포에서 SH21B에 의한 지방세포형성 억제의 분자적 기작을 보다 상세하게 연구하고자 하였다. 전지방세포, 분화된 세포 그리고 SH21B에 의해 분화가 억제된 세포의 각각의 유전자 발현을 분석하기 위해 각 시료들에서 total RNA를 분리하여 cDNA를 합성한 후 microarray에 적용시켰다. 그 결과, 각각의 시료들의 비교에서 2배 이상의 유의한 발현 변화를 가지는 2,568개의 유전자를 확보하였다. 이 유전자들에 대해 Hierarchical clustering과 K-means clustering 분석을 진행하였고 서로 다른 양상을 가지는 9개의 군집(cluster)들을 분류하였다. 그 중, SH21B의 첨가에 의해 뚜렷하게 감소(cluster 4, cluster 6 및 cluster 9)하거나 반대로 뚜렷하게 증가(cluster 7와 cluster 8)하는 양상을 보이는 군집들을 따로 선별하여 그 군집들에 포함되어 있는 유전자들을 분석하였다. 선택 된 5개의 군집에는 지방세포형성과 세포증식에 관련된 유전자가 다수 포함되어 있었다. Cluster 4, cluster 6 그리고 cluster 9에는 peroxisome proliferator activated receptor gamma $\gamma$ ($PPAR{\gamma}$), CCAAT/enhancer binding protein $\alpha$ (C/$EBP{\alpha}$), sterol regulatory element binding transcription factor 1 (SREBF1), adiponectin (ADIPOQ), fatty acid synthase (FASN), lipoprotein lipase (LPL) 등의 지방세포형성 유도 및 관련 인자와 B-cell leukemia/lymphoma6 (BCL6), retinoblastoma 1 (RB1), cyclin-dependent kinase inhibitor 2C (CDKN2c), ras homolog gene family, member B (RHOB) 등의 많은 세포증식 억제 유전자가 포함되었다. 이와는 반대로, cluster 7과 cluster 8에는 $\beta$-catenin, cyclin D1 (CCND1), WNT1 inducible signaling pathway protein 2 (WISP2) 등과 같은 지방 세포형성 억제 조절자와 MARCKS-like1 (MARCKSL1), colony stimulating factor 1 (CSF1), discoidin domain receptor family, member 2 (DDR2), leukemia inhibitory factor receptor (LIFR) 등의 세포증식을 유도하는 조절자가 다수 포함되었다. 결론적으로, 이러한 결과들은 SH21B가 지방세포형성과 관련된 조절자 및 세포증식과 관련 된 조절자들의 유전자 발현을 조절하여 지방세포형성을 억제함을 제시한다.

Biochemical tests such as aspartate aminotransferase (AST) and alanine aminotransferase (ALT) are useful for diagnosing patients with liver disease. In this study, we tested the association between copy number variation and the hepatic biomarkers AST and ALT based on 8,842 samples from population-based cohorts in Korea. We used Affymetrix Genome-Wide Human 5.0 arrays and identified 10,534 CNVs using HelixTree software. Of the CNVs tested using univariate linear regression, 100 CNVs were significant for AST and 16 were significant for ALT (P < 0.05). We identified 39 genes located within the CNV regions. DKK1 and HS3ST3B1 were shown to play roles in heparan sulfate biosynthesis and the Wnt signaling pathway, respectively. NAF1 and NPY1R were associated with glycoprotein processes and neuropeptide Y receptor activity based on GO categories. PTER, SOX14 and TM7SF4 were expressed in liver. DPYS and CTSC were found to be associated with dihydropyrimidinuria and Papillon-Lefevre syndrome phenotypes using OMIM. NPY5R was found to be associated with dyslipidemia using the Genetic Association Database.
