Effect of Bacterial Wilt on Fungal Community Composition in Rhizosphere Soil of Tobaccos in Tropical Yunnan

  • Bacterial wilt, which is a major soil-borne disease with widespread occurrence, poses a severe danger in the field of tobacco production. However, there is very limited knowledge on bacterial wilt-induced microecological changes in the tobacco root system and on the interaction between Ralstonia solanacearum and fungal communities in the rhizosphere soil. Thus, in this study, changes in fungal communities in the rhizosphere soil of tobaccos with bacterial wilt were studied by 18S rRNA gene sequencing. The community composition of fungi in bacterial wilt-infected soil and healthy soil in two tobacco areas (Gengma and Boshang, Lincang City, Yunnan Province, China) was studied through the paired comparison method in July 2019. The results showed that there were significant differences in fungal community composition between the rhizosphere soil of diseased plants and healthy plants. The changes in the composition and diversity of fungal communities in the rhizosphere soil of tobaccos are vital characteristics of tobaccos with bacterial wilt, and the imbalance in the rhizosphere microecosystem of tobacco plants may further aggravate the disease.

Water Extract from Spent Mushroom Substrate of Hericium erinaceus Suppresses Bacterial Wilt Disease of Tomato

  • Culture filtrates of six different edible mushroom species were screened for antimicrobial activity against tomato wilt bacteria Ralstonia solanacearum B3. Hericium erinaceus, Lentinula edodes (Sanjo 701), Grifola frondosa, and Hypsizygus marmoreus showed antibacterial activity against the bacteria. Water, n-butanol, and ethyl acetate extracts of spent mushroom substrate (SMS) of H. erinaceus exhibited high antibacterial activity against different phytopathogenic bacteria: Pectobacterium carotovorum subsp. carotovorum, Agrobacterium tumefaciens, R. solanacearum, Xanthomonas oryzae pv. oryzae, X. campestris pv. campestris, X. axonopodis pv. vesicatoria, X. axonopodis pv. citiri, and X. axonopodis pv. glycine. Quantitative real-time PCR revealed that water extracts of SMS (WESMS) of H. erinaceus induced expressions of plant defense genes encoding ${\beta}$-1,3-glucanase (GluA) and pathogenesis-related protein-1a (PR-1a), associated with systemic acquired resistance. Furthermore, WESMS also suppressed tomato wilt disease caused by R. solanacearum by 85% in seedlings and promoted growth (height, leaf number, and fresh weight of the root and shoot) of tomato plants. These findings suggest the WESMS of H. erinaceus has the potential to suppress bacterial wilt disease of tomato through multiple effects including antibacterial activity, plant growth promotion, and defense gene induction.

소나무림 숲가꾸기 종류가 소나무재선충병의 제어에 미치는 영향 (Effects on Control of Pine Wilt Disease (Bursaphelenchus xylophilus) by Thinning Methods in Red Pine(Pinus densiflora) Forest)

  • 소나무재선충병 방제법 중 임업적 방제법을 개발하기 위한 일환으로 위생간벌, 간벌, 어린소나무 가꾸기가 소나무재선충병의 제어에 미치는 영향을 구명하기 위하여 야외 그물망 케이지에 소나무재선충을 보유한 솔수염하늘소를 방사하였다. 대조구, 위생간벌, 간벌 처리구 모두에서 공시목의 50% 이상이 고사되었으며, 처리구 간에는 유의성이 나타나지 않았다. 그리고 처리구 내 방치한 숲가꾸기 산물이 소나무재선충병을 확산시키는 촉매제로 작용하였다. 어린 소나무림 가꾸기의 경우 산물을 방치한 처리구가 공시목의 고사율이 90% 이상으로 가장 높았으며, 산물을 케이지 밖으로 들어낸 처리구와 숲가꾸기를 하지 않고 솔수염하늘소의 성충을 방사한 처리구에서는 10% 내외의 고사율을 나타내었다. 솔수염하늘소 우화 당년도인 4월에 위생간벌, 간벌을 실행한 장령목의 벌근에서는 솔수염하늘소의 유충이 출현하지 않았으나, 솔수염하늘소의 우화 당년도인 5월에 숲가꾸기 한 어린나무의 벌근에서는 유충이 출현하였다. 본 실험 결과, 장령 소나무림과 어린소나무림 내에 소나무재선충병이 만연하고 있는 임분의 경우 솔수염하늘소의 우화 당년도인 4월과 5월에 실행한 숲가꾸기는 소나무재선충병의 제어에 아무런 효과가 없었으며 방치한 숲가꾸기 산물은 오히려 소나무 재선충병을 확산시키는 촉매제로 작용하였다.

Cultural and Rainfall Factors Involved in Disease Development of Fusarium Wilt of Sweet Potato

  • Environmental factors such as soil moisture, land management, and weather conditions affecting Fusarium wilt of sweet potato were investigated in major sweet potato cultivation regions in Korea. Fusarium wilt occurred mainly in reclaimed terracing lands, which are flattened and located in hilly to mountainous areas at the base of the mountain, in early seasonal cultivation regions. Disease severity was lower in reclaimed fields with natural slope. The development of Fusarium wilt in the fields was highly correlated with precipitation during planting period (r=-0.96**). Fusarium wilt was more severe in fields with less than 20 cm of available soil depth than in fields with over 20 cm of available soil depth. Greenhouse studies were consistent with field studies that less soil moisture content caused severe Fusarium wilt of sweet potato. These results indicate that low rainfall and moisture of soil with low effective soil depth during planting period are important environmental factors influencing the development of Fusarium wilt.

STABILITY ANALYSIS OF A HOST-VECTOR TRANSMISSION MODEL FOR PINE WILT DISEASE WITH ASYMPTOMATIC CARRIER TREES

  • A deterministic model for the spread of pine wilt disease with asymptomatic carrier trees in the host pine population is designed and rigorously analyzed. We have taken four different classes for the trees, namely susceptible, exposed, asymptomatic carrier and infected, and two different classes for the vector population, namely susceptible and infected. A complete global analysis of the model is given, which reveals that the global dynamics of the disease is completely determined by the associated basic reproduction number, denoted by $\mathcal{R}_0$. If $\mathcal{R}_0$ is less than one, the disease-free equilibrium is globally asymptotically stable, and in such a case, the endemic equilibrium does not exist. If $\mathcal{R}_0$ is greater than one, the disease persists and the unique endemic equilibrium is globally asymptotically stable.

Virulence of Xanthomonas translucens pv. poae Isolated from Poa annua

  • Bacterial wilt is a vascular wilt disease caused by Xanthomonas translucens pv. poae that infects Poa annua, a grass that is commonly found on golf course greens throughout the world. Bacterial wilt causes symptoms of etiolation, wilting, and foliar necrosis. The damage is most prevalent during the summer and the pathogen can kill turf under conditions optimal for disease development. Fifteen isolates of X. translucens pv. poae were collected from northern regions in the United States and tested for virulence against P. annua. All 15 isolates were pathogenic on P. annua, but demonstrated variable levels of virulence when inoculated onto P. annua under greenhouse conditions. The isolates were divided into two virulence groups. The first group containing four isolates generally resulted in less than 40% mortality following inoculation. The second group, containing the other eleven isolates, produced between 90 and 100% mortality following inoculation. These results suggest that differences in the virulence of bacterial populations present on a golf course may result in more or less severe amounts of observed disease.

Use of Quantitative Models to Describe the Efficacy of Inundative Biological Control of Fusarium Wilt of Cucumber

  • Fusarium wilt of cucumber caused by Fusarium oxy-sporum f. sp. cucumerinum is a serious vascular disease worldwide. Biological control of Fusarium wilt in several crops has been accomplished by introducing non-pathogenic Fusarium sup. and other biocontrol agents in soil or in infection courts. In this study, quantitative models were used to determine the biocontrol efficacy of inundatively applied antagonist formulations and the length of their effectiveness in controlling Fusarium wilt of cucumber. Quantitative model of the form [Y=L (1${-exp}^{-kx}$)] best described the relationship between disease incidence (Y, %) and inoculum density (X) of isolates F51 and F55. Isolate F51 was selected as a more virulent isolate based on the extent of its effectiveness in causing the wilt disease. The degree of disease control (Xi/X) obtained with the density of the biocontrol agent (Z), was described by the model [Xi/X=A (1${-exp}^{-cz}$)]. The zeolite-based antagonist formulation amended with chitosan (ZAC) was better at lower rates of application and peaked at around 5 g/ kg of the potting medium, whereas the peat-based antagonist formulation (PA) peaked at around 10 g/kg of the potting medium. ZAC formulation provided significantly better suppression of Fusarium wilt as described by the curvilinear relationship of the type Y= a+bX+c$X^2$, where Y represents percent disease incidence and X represents sustaining effect of the biocontrol agent.

  • 최근, 소나무재선충(Bursaphelenchus xylophilus)에 의한 소나무림의 피해에 대한 사회적 심각성이 크게 대두되고 있다. 소나무 재선충에 의한 산림피해는 피해지 내에서는 매개충인 솔수염하늘소의 자연적인 영역확장에 의해 확산되는 반면, 전국적으로는 감염목의 인위적 반출 및 이동에 의해 확산이 진행되고 있다. 본 연구에서는 부산 대변항의 재선충 피해지내에서 항공사진 및 현지조사에 의해 피해목의 공간적인 위치를 파악하였고, 공간통계학적인 방법을 통하여 피해목의 공간분포유형, 피해발생과 지형인자간의 관계를 분석하였다. 또한, 지형공간자료를 통계학적 Tree 모형에 적용한 CART(Classification and Regression Trees)모형을 이용하여 재선충 피해의 자연적인 확산 예측 지도를 작성하였다. 본 연구를 통해 공간통계학적인 분석과 CART모형이 소나무재선충 피해의 공간분포 및 자연적 확산유형을 파악하는데 유용한 도구로 활용될 수 있음을 확인할 수 있었다.

  • Fusarium wilt of lisianthus occurred severely throughout all cultivation areas, especially in alpoine areas during summer season and the disease incidence was 5 to 30 percent in Korea. The major symptoms of the disease were wilt with chlorosis and water deficiency, stunted plants and scorched leaves. Severe symptoms appeared just after high temperature period around late August in Daekwallyong area. Whit and pale red colored mycelia were developed on stems of infected plants near the soil surface. The causal organism of Fusarium wilt of lisianthus was isolated and identified as Fusarium oxysporum f. sp. eustomae on the basis of pathogenic and cultural characteristics. The causal organism was reisolated from all infected and inoculated stems of plants. This is the first report referring to F. oxysporum f. sp. eustomae of lisianthus in Korea.

Reduced Tomato Bacterial Wilt by Ferrous Chloride Application

  • Exogenous ferrous chloride (FeCl2) suppressed in vitro growth of Ralstonia pseudosolanacearum, causing bacteria for tomato bacterial wilt. More than 50 μM of FeCl2 reduced the in vitro bacterial growth in dosedependent manners. Two to 200 μM of FeCl2 did not affect the fresh weight of detached tomato leaves at 3 and 5 days after the petiole dipping without the bacterial inoculation. The bacterial wilt of the detached tomato leaves was evaluated by inoculating two different inoculum densities of R. pseudosolanacearum (105 and 107 cfu/ml) in the presence of FeCl2. Bacterial wilt in the detached leaves by 105 cfu/ml was efficiently attenuated by 10-200 μM of FeCl2 at 3 and 5 days post-inoculation (dpi), but bacterial wilt by 107 cfu/ml was only reduced by 200 μM of FeCl2 at 3 and 5 dpi. These results suggest that iron nutrients can be included in the integrated disease management of tomato bacterial wilt.
