In this study, the genetic diversity and the population structure of 77 wild strains and 23 cultivars of Lentinula edodes from Korea were analyzed using 20 genomic SSRs, and their genetic relationship was investigated. The tested strains of L. edodes were divided into three sub-groups consisting of only wild strains, mainly wild strains and several cultivars, and mainly cultivars and several wild strains by distance-based analysis. Using model-based analysis, L. edodes strains were divided into two subpopulations; the first one consisting of only wild strains and the second one with mainly cultivars and several wild strains. Moreover, AMOVA analysis revealed that the genetic variation in the cultivars was higher than that in the wild strains. The expected and observed heterozygosity and values indicating the polymorphic information content of L. edodes cultivars from Korea were also higher than that of the wild strains. Based on these results, we presume that the cultivars in Korea have developed by using numerous strains from other countries. In conclusion, the usage of wild strains for the development of new cultivars could improve the adaptability of L. edodes to biotic and abiotic stress.
90 wild Agaricus strains from China, including 44 Agaricus bisporus strains identified preliminarily by isozyme electrophoresis, were studied by the techniques of SRAP and ISSR. 18 special SRAP bands and 12 special ISSR bands were analyzed, the strains were clustered and a demdrogram was obtained. The results showed that the strains were divided into 2 groups, wild A. bisporus group and the other Agaricus group. It is similar to the result of isozyme electrophoresis. 41 wild A. bisporus strains from Sichuan and Tibet were divided into 4 groups based on their growing places, suggesting the regionally difference of the strains to be quite obvious. Some white wild A. bisporus strains from Xinjiang and Tibet had special patterns, resulting in lower coefficient values with other wild A. bisporus strains. The biological characteristics of three wild A. bisporus strains were analyzed, and the results showed: 1. The wild strains grew slowly on PDA medium with weak appressed mycelia, and grew normally in kernel or fermented cottonseed shell substrate. 2. They grew faster than control strain As2796 under lower temperature of $16^{\circ}C$, and higher temperature of $32^{\circ}C$, with optimum growing temperature of $20-24^{\circ}C$, which was $4^{\circ}C$ lower than that of control strain. 3. In the cultivation with manure compost via twice fermentation, the mycelia grew normally in compost and quite slowly in casing soil, and the fruitbodies occurred less and late with easily opening and low production. 4. The fruitbody was off-white with flat and scaled cap, long stipe and dark gill. The bisporus basidia occupied 70-80% and trisporus basidia 20-30% of the total basidia. 5. Heterokaryotic monospore isolates could fruit in cultivation, and the homokaryotic isolates could cross with those derived from overseas wild A.bisporus strains. 6. The electrophoresis phenotype of isozymes such as esterase etc. belonged to high production type (H type). 7. The RAPD patterns made much difference from those of high production, good quality or hybrid strains, which indicated that the wild strains produce a new kind of RAPD type.
This study was conducted to preserve of mushroom resources and utility useful wild mushrooms by segregation and identification from 2005 to 2007. The mushroom strains were collected a center of native mushroom wild growth place of Chungbuk Province. The obtained results from this study were summarized as follows ; We collected 79 wild mushroom strains, and the collected wild mushrooms were composition of 32 strains of edible mushrooms, 3 strains of medicinal use mushrooms, 15 strains of poisonous mushrooms, and 29 strains indistinct mushrooms. The 28 strains were segregated and identified from 32 strains of edible mushrooms. The present preservation strains are 15 strains, and other 13 strains were damaged in tissue culture and preservation. We made specimen of wild mushroom by alcohol, and have preserved perennial mushrooms by drying. We photographed 79 strains of wild mushrooms.
The morphometrical characteristics such as external measurements and mandible size assessment in mice and rats have to be highly heritable and sufficiently variable between strains in order to calculate a strain specific profiles. The coat color of Korean wild rats were observed and morphometric analysis of external measurements were carried out on Korean wild rats compared to laboratory strains in order to clarify the genetic characteristics of Korean wild rats and to establish background data as a domestication of Korean wild rats for new laboratory strain. Korean wild rats were captured from Chunchon and Hoengsong. 4 inbred and 1 outbred strains of rats were used in this study for the comparison of genetic characteristic of Korean wild rats. Total body length, head length, tail length, hind foot length and ear length were measured and then statistical analysis were carried out by discrimiant analysis. The coat color of Korean wild rat showed golden white in ventral portion and dark agouti in dorsal portion. Korean wild rats could be distinguished from the other laboratory strains distinctly by morphogenetical analysis. There was significant variations among Korean wild rat compared to those of the other laboratory strains of rat. This study may provide that Korean wild rats have a unique genetic characterization compared to those of other inbred strains of rats based on morphogenetical characteristics by external measurements.

The diversity of A mating type in wild strains of Lentinula edodes was extensively analyzed to characterize and utilize them for developing new cultivars. One hundred twenty-three A mating type alleles, including 67 newly discovered alleles, were identified from 106 wild strains collected for the past four decades in Korea. Based on previous studies and current findings, a total of 130 A mating type alleles have been found, 124 of which were discovered from wild strains, indicating the hyper-variability of A mating type alleles of L. edodes. About half of the A mating type alleles in wild strains were found in more than two strains, whereas the other half of the alleles were found in only one strain. About 90% of A mating type combinations in dikaryotic wild strains showed a single occurrence. Geographically, diverse A mating type alleles were intensively located in the central region of the Korean peninsula, whereas only allele A17 was observed throughout Korea. We also found the conservation of the TCCCAC motif in addition to the previously reported motifs, including ATTGT, ACAAT, and GCGGAG, in the intergenic regions of A mating loci. Sequence comparison among some alleles indicated that accumulated mutation and recombination would contribute to the diversification of A mating type alleles in L. edodes. Our data support the rapid evolution of A mating locus in L. edodes, and would help to understand the characteristics of A mating loci of wild strains in Korea and help to utilize them for developing new cultivars.
Wild soybean plants(Glycine ussuriensis) collected from Korea(47 strains) and abroad(41 strains) were grown under field condition and classified according to the growth habit. Seeds were analyzed for protein content. The results were as follows: 1. Wild soybean plants were classified into three groups each by seed size, growth habit of stem, leaf size and flowering time. 2. Seed protein was higher in the strains with large leaves, tendril, stem or late flowering. 3. Glycine gracilis, supposed to be the intermediate species between cultivated and wild soybean(according to the stem growth habit, straight or semitendril, and seed size) was medium in protein content of seeds. 4. The average protein content of Glycine ussuriensis, 43.2%, was highest in comparison with those of others such as G. gracilis, 37.5%, or G. max, 36.2%. 5. One hundred seed weight of Amphicarpaea trisperm, similar to the wild soybean, was almost same as Glycine ussuriensis, but protein content of this species was lower than wild soybeans.
Some streptomycin resistant strains of Rhizobium trifolii having nodulation ability were selected, and their nitrogenase activities, symbiotic effects on plant growth, and nodule electronmicroscope were compared with those of the wild type. After NTG treatment, as a mutagen, at the concentration exhibiting 99.7% lethal rate, 5 strains of streptomycin resistant mutant having nodulating ability were selected. Among these nodulating mutant strains, 3 strains produced more nodules and 2 strains showed less nodules than wild type. But their nitrogenase activities were decreased significantly, and nodule formation time was also delay compared with those of the wild type, and there was no remarkable difference in effects on plant growth. Microstructure of nodules by electronmicroscopy had mant distinctive differences between red clover nodules inoculated with wild type and mutants.
The goal of this study was to isolate and identify the wild yeast strains in intestine of Othius punctulatus collected in Cheongju-si, Chungcheongbuk-do, Korea and characterize their unrecored yeast strains. We isolated 16 wild yeast strains from 5 intestine samples from Othius punctulatus. The 16 wild yeast strains were included three strains in the genus Aureobasidium, four strains in the genus Cystofilobasidium, one strain in the genus Mrakia, three strains in the genus Naganishia, two strains in the genus Saitozyma, two strains in the genus Sampaiozyma and one strain in the genus Scheffersomyces. Among them Cystofilobasidium capitatum YP53, Cystofilobasidium capitatum YP71, Cystofilobasidium macerans YS620, Cystofilobasidium macerans YS622, Mrakia aquatica YP158 and Scheffersomyces stipitis YI5 were recorded for the first time in Korea. The microbiological characteristics and carbon assimilation of these previously unrecorded yeasts were investigated. Almost all of them were oval-shaped. And, glycerol, L-arabinose, xylitol, and inositol are not utilized.

The goal of this study was to investigate the diversity present among wild yeasts obtained from soils of spice fields and from mountain soils, and to further, characterize previously unrecorded novel wild yeast strains. In total, 36 strains from 17 different species of wild yeasts were isolated from 35 soil samples obtained from garlic fields of Geumsan, Chungcheongnam-do, Korea. Among these, six yeast strains of Trichosporon moniliiforme, and four strains each of Papiliotrema flavescens and Candida melibiosica species were isolated. Additionally, 22 strains of 18 different species of wild yeasts were isolated from 32 soil samples collected from the ballonflower and ginger fields of Geumsan, Korea. Finally, 46 strains of wild yeasts were isolated from 35 soil samples obtained from Mt. Daedun in Geumsan, Korea. Among the total of 106 isolated wild yeast strains, 10 strains, including Debaryomyces vindobonensis GHY31-3 represented novel yeast strains which were previously unrecorded. All the 10 previously unrecorded yeasts were oval or global in shape, and five strains, including Filobasidium stepposum SFG1-4 formed ascospores. Three strains, including Pseudozyma alboarmeniaca CD 23-5 grew well in vitamin-free medium. Cell-free extract obtained from Filobasidium magnum SFG1-3 indicated 28.6% of xanthine oxidase inhibitory activity.
The purpose of this study was to isolate and identify wild yeasts from soil collected in Daegu City and Cheongyang County, Republic of Korea. Among 11 strains isolated in this study, nine strains were previously reported and two strains were unreported in Republic of Korea. To identify wild yeast strains, pairwise sequence comparisons of the D1/D2 region of the 26S rRNA gene sequence were done using Basic Local Alignment Search Tool (BLAST). The cell morphologies were observed by phase contrast microscope and assimilation test are done using API 20C AUX kit. All strains were assigned to the phylum Basidiomycota. Of the two unrecorded yeast strains, CY-9-10C belongs to the genus Mrakia (family Mrakiaceae, order Cystofilobasidiales, class Tremellomycetes) and PG3-4-10C belongs to the genus Slooffia (family Chrysozymaceae, order Microbotryomycetes incertae sedis, class Microbotryomycetes). Both strains had oval-shaped and polar budding cells. This research described the morphological and biochemical properties of the two unreported yeast species that had not officially reported in Korea.
