The mature wheat embryo is arguably one of the best explants for genetic transformation because of its unlimited availability and lack of growth season restriction. However, an efficient regeneration system using mature wheat embryos (Triticum aestivum L.) is still not available. To identify genes related to the tissue culture response (TCR) of wheat, QTLs for callus induction from mature embryos and callus regeneration were mapped using an RIL population derived from the cross of 'Wangshuibai' with 'Nanda2419', which has a good TCR. By whole genome scanning we identified five, four and four chromosome regions conditioning, respectively, percent embryos forming a callus (PEFC), percent calli regenerating plantlets (PCRP), and number of plantlets per regenerating callus (NPRC). The major QTLs QPefc.nau-2A and QPcrp.nau-2A were mapped to the long arm of chromosome 2A, explaining up to 22.8% and 17.6% of the respective phenotypic variance. Moreover, two major QTLs for NPRC were detected on chromosomes 2D and 5D; these together explained 51.6% of the phenotypic variance. We found that chromosomes 2A, 2D, 5A, 5B and 5D were associated via different intervals with at least two of the three TCR indexes used. Based on this study and other reports, the TCRs of different explant types of wheat may be under the control of shared or tightly linked genes, while different genes or gene combinations may govern the stages from callus induction to plantlet regeneration. The importance of group 2 and 5 chromosomes in controlling the TCRs of Triticeae crops and the likely conservation of the corresponding genes in cereals are discussed.

Objective: Wheat is an alternative to corn silage for ruminant feeding in northern China. This study examined the effects of harvest time and added molasses on nutritional content, ensiling characteristics and in vitro degradation of whole crop wheat (WCW). Methods: Fresh WCW at the milk-ripe stage was harvested at 0700 h (i.e., in the morning [Mo]) and 1700 h (i.e., in the afternoon [Af]), and then immediately used to prepare silage and make hay. Commercial molasses was added to Af WCW at 0%, 2%, 4%, and 6% (fresh weight) proportions. The WCW treated with molasses was mixed thoroughly prior to ensiling. Results: Dry matter (DM), neutral detergent fiber, water soluble carbohydrate (WSC) content (p<0.01), accumulative gas production in 72 h ($GP_{72h}$, 77.46 mL/g vs 95.15 mL/g) and dry matter disappearance in vitro (69.15% vs 76.77%) were lower (p<0.05), while crude protein (CP) content was higher for WCW silage (WCWS) compared to WCW (p<0.01). The propionic acid and butyric acid concentrations in WCWS from Mo WCW were 1.47% and 0.26%, respectively. However, the propionic and butyric acid concentrations were negligible, while the ammonia nitrogen/total nitrogen ($NH_3-N/TN$, p<0.01) concentration was lower and the rate of gas production at 50% of the maximum (17.05 mL/h vs 13.94 mL/h, p<0.05) was higher for Af WCWS compared to Mo WCWS. The incubation fluid's $NH_3-N$ concentration was lower in WCWS and Af WCW compared to Mo WCW (p<0.05). The CP and WSC content increased with increasing molasses levels (p<0.05). Furthermore, the pH (p<0.01) and time when gas production was 50% of the maximum (2.78 h vs 3.05 h, p<0.05) were lower in silage treated with 4% molasses than silage without molasses. Conclusion: Harvesting wheat crops in the afternoon and adding molasses at 4% level to WCW optimally improved ensiling characteristics, leading to well-preserved silage.

Cereal grains are rich in phenolic compounds that give beneficial effect in human health. Although several research works have been reported on the effects of phytochemicals of plant origin, such as fruits, vegetables, few studies have examined the antioxidative effects of whole cereal grains. The objective of this study was to determine total antioxidant capacity of 80% ethanolic extracts of cereal grains by testing the ability of the extracts to inhibit UV -induced lipid peroxidation in vitro using linoleic acid in comparison to well-known antioxidant such as ascorbic acid and tannic acid. The total phenolic content of the cereal grain (80% ethanolic extracts) investigated in this study varied from 2.1 mg/g (wheat cv. Olgeurumil) to 10.4 mg/g (barely cv. Seodunchalbori). Highly positive relationship between total phenol compounds and antioxidant activity was found. When the antioxidant activities of all investigated extracts were measured with application of same quantity of phenol compounds, oat grain extracts showed similar antioxidant activity of barely cultivars. However, barely extract appeared as the most potent antioxidant activity of inhibition of UV -induced lipid peroxidation. This indicated that factors such as phenolic compound composition and their individual antioxidant activity could playa crucial role in the total antioxidant activity of cereal grains.

The objectives of this study were to evaluate the different cutting dates on the changes of feed quality among five cereals (barley, wheat, rye, triticale and oat) for whole crop silage. Field trials were conducted at paddy field in Yesan, Chungnam Province and the aerial parts were clipped 10 days from 15 March to 15 June. Changes of acid detergent fiber (ADF) content in relation to different cutting dates was described by a quadratic curve for 5 winter cereals crops. ADF content reached a maximum at 5 days after heading in barley cultivar 'Youngyang', 7 days in wheat 'Keumkang', 18 days in rye 'Gogu', 1 days in triticale 'Shinyoung' and 10 days in oat 'Samhan'. Neutral detergent fiber (NDF) content were linearly increased as growing after overwintering and stagnated or slightly decreased after heading. The crude protein were linearly decreased throughout the growth period of 5 whole crop cereals. Digestible dry matter (DDM) content were decreased from early stages to heading and subsequently increased as grain filling. Relative feed value (RFV) for 5 crops were decreased as growing and subsequently increased as grain filling after heading. Barley cultivar for only forage use 'Youngyang' were lower at ADF and NDF content and higher at DDM and RFV after heading than those of other cereals for forage use. So, barley for whole crop silage was a good crop with high feed quality and high proportion of spikes compared with other winter cereal crops. Wheat cultivar for grain 'Keumkang' were higher at crude protein than those of other four cereals from overwintering to maturing and were higher at DDM and RFV after heading than those of rye, triticale and oat. Rye cultivar with cold tolerant and high fresh yielding 'Gogu' were highest at ADF and NDF content and lowest at DDM content and RFV. So, rye was a crop with low quality for forage use compared to other winter cereal crops. Triticale cultivar with flourishing and high yielding 'Shinyoung' was intermediated between barley and rye, and were linearly increased at DDM yield by different cutting dates. Oat cultivar with cold tolerant and high tillering 'Samhan' were lower at ADF and NDF content and higher at crude protein before heading, but after heading, there are not especially advantages compared to barley, wheat or triticale.
Polyphenol oxidase (PPO) is implicated in discoloration of white salted noodles and other wheat based foods. PPO activity was evaluated to determine the effect on discoloration of noodle dough sheets prepared from 25 Korean wheat flours during storage and to screen experimental lines with low PPO activity in 52 Korean wheats. PPO activity was assayed with whole-seed and performed with L-dihydroxyphenylalanine (L-DOPA) as substrates. Absorbance by L-DOPA assay of 25 Korean wheats was from 0.285 to 1.368 at 475 nm. PPO activity was significantly related with grain characteristics, including 1000-kernel weight and grain colors. In flour characteristics, PPO activity positively correlated with ash and protein content (r = 0.658, P < 0.001 and r = 0.424, P < 0.05, respectively) and negatively correlated with $L^*$ value of flour (r = 0.412, P < 0.05). In the changes of color of noodle dough sheet, $L^*$ and $b^*$ values consistently decreased and $a^*$ value increased during storage. PPO activity negatively correlated with $L^*$ value of noodle dough sheet during storage (r = 0.566, P < 0.01 at 2 hr, r = 0.547, P < 0.01 at 24 hr, and r = 0.509, P < 0.01 at 48 hr). But, no significant relationship was found in between PPO activity, $a^*$ and $b^*$ values during storage. The 52 Korean wheat lines examined in this study were divided into 3 different groups, low (< 0.500), medium (0.501-0.999) and high level (> 1.000), on the basis of the level of PPO activity. Twenty two Korean wheat lines showed low level of PPO activity and Suwon 252, 277 and 280 showed lower PPO activity (< 0.200) than others.
Recently, an interest has developed in the use of whole green grains as functional food materials. The present study was conducted to provide the baseline data for the stable production of whole green grains in 20 cultivars of wheat by investigating the greenness of grain with maturation (from $20^{th}$ to $41^{st}$ day after heading, at an interval of 3 days). On the $20^{th}$ day after heading, the grains were dark green with a wrinkled long-oval shape. After the $35^{th}$ day of heading, the grains turned almost yellow with an oval shape. Their redness ('a' value of chromaticity) increased from the $20^{th}$ to $41^{st}$ days after heading, indicating a negative value up to the $32^{nd}$ day after heading. A significant decrease in their chlorophyll content was observed with maturation. The yield of whole green wheat grain (including greenish yellow grain) was the highest from the $32^{nd}$ to $35^{th}$ after heading. Therefore, we concluded that the optimal harvesting period for whole green wheat grain was from the $32^{nd}$ to $35^{th}$ day after heading. The heading time of various cultivars ranged from April 28 to May 5, the time of Jopummil cultivar grew the fastest among them, such as Gurumil, Alchanmil, but Dahongmil got the latest in heading time. The greenness of seven cultivars (Jeokjungmil, Keumkangmil, Jogyeongmil, Jopummil, Baekjungmil, Yeonbaekmil, and Milseongmil) was relatively higher than that of the others. The yield of greenish whole grain was relatively high in six cultivars (Alchanmil, Baekjungmil, Eunpamil, Yeonbaekmil, Dahongmil, and Urimil). Based on their greenness and yield, the Baekjungmil and Yeonbaekmil cultivars have been considered to be optimal for the production of whole green wheat grain.

This study was carried out to analyze winter cereal crops for harvest time, proper drying time and haylage quality in order to investigate the possibility of the production of good quality haylage substitute for hay. As a result, in case of productivity and feed value, dry matter yield and TDN yield was increased with late harvest and crude protein and NDF, ADF was decreased (p<0.05). To make haylage for whole crop barley, oats, triticale, whole crop wheat need more than 32 hour wilting time in the milk ripe stage, yellow ripe stage need about 8~24 hour, and wilting time did not need the full ripe stage. Rye does not suitable for making haylage because of difficulty pre-wilting time. In case of fermentative quality on haylage, pH was increased with late harvest on all winter cereal crop forage. Lactic acid content was decreased with late harvest (p<0.05), and highest in the milk ripe stage. Acetic acid was also decreased with late harvest, and butyric acid was not detected. In conclusion, to harvest sooner of winter cereal crops could be an efficient way when making haylage in terms of protein content and fermentation quality. Ray can used as fresh and silage, because difficulty for pre-wilting time in Korea. Whole crop barley, oats, triticale, wheat can be used haylage when harvested yellow ripen stage and pre-wilting time for 8~24 hours.

The overwintering crops, barley, wheat, oat and triticale etc, have been received in korea as high-quality roughage for round-baled silage making as livestock feed. Studies were carried out to evaluated the effects of seed blending on growth, yield and feed value between barley and wheat, triticale and oat, respectively. The results are summarized as follows : The barley was declined in the growth and spike's size as affected by sown as mixed seeding with triticale, but not effected with oat. The total fresh yield were increased by grown as mixed seeding with wheat, triticale and oat, but the dry matter yield were not significantly increased because of the low percentage of dry matter and the decrease of barley's growth. The neutral detergent fiber (NDF) and crude protein content were increased by seed blending, but acid detergent fiber (ADF) and digestible dry matter (DDM) content were decreased. The feed value was improved by seed blending of barley and wheat, but not significant at seed blending of triticale and oat. Because of the flourishing tillers, difference of heading date and abundant leaves of oat, the optimum crop for mixed seeding with barley for increment of forage productivity was oat in middle area of korea.
The objective of this study was to identify the effect of zebularine, a DNA methylation inhibitor, on the chromosomal association between homoeologous chromosomes in the wheat genetic background. Zebularine at a final concentration of 10 µM was used to treat the spikes of the double monosomic wheat addition line (DMA) with one Leymus mollis chromosome and one Leymus racemosus chromosome, both of which were in a homoeologous relationship. In late prophase, zebularine led to chromosome breakage in the Leymus homoeologous chromosomes. Chromosome breakage caused an increase in the frequency of chromosomal associations between the Leymus homoeologous chromosomes. Ordinary DMA showed 65 cells (35.3%) with chromosomal associations and 119 cells (64.7%) with no association, whereas treated DMA showed 102 cells (60.0%) with chromosomal associations and 67 cells (39.4%) with no association. In diakinesis, the Leymus bivalent showed a chromosomal association in the whole euchromatic region. In metaphase, the Leymus bivalent showed association in the whole chromosomal region, unlike other Leymus bivalents with partial chromosomal association. Chromosomal association by chromosome breakage occurred not only between Leymus chromosomes but also between Leymus and wheat chromosomes. The frequency of other chromosomal association (such as fusion and insert) was increased. Chromosome breakage by zebularine treatment is a useful method at the chromosome level as the spores with others are hereditary stable, although the homologous index (h) was not significantly different between ordinary DMA and treated DMA. It is necessary to study how to control zebularine treatment with a more stable concentration for chromosome breakage during meiosis.
This study was investigated the quality characteristics of muffins which were prepared with whole waxy sorghum flours. Muffins were made with different sorghum flour contents of 10, 30, 50 and 100% substitution for wheat flour. The crude protein and fat contents of muffins were not different among samples, but the crude ash content was determined to increase with increasing content of sorghum flour. The specific gravity of the sorghum muffin batter as well as the weight and uniformity index of the sorghum muffins, were not significantly different from those of the control. The volume of muffins was found to decrease significantly when 100% sorghum flour was substituted for wheat flour. In terms of color, the L and b values of muffins were decreased with increasing contents of sorghum flour, and the a value was increased. The sensory test revealed that the hardness and adhesiveness of muffins was increased with the addition of sorghum flour, but springiness and chewiness decreased. According to the results of a sensory evaluation test, the flavor, taste and texture of muffins were reduced with the addition of 50 and 100% sorghum flour. The appearance and overall acceptability of sorghum muffins were not found to be different significantly from those of the control, up to 30% level substitution with sorghum flour.
