Hepatic microRNAome reveals potential microRNA-mRNA pairs association with lipid metabolism in pigs

  • Objective: As one of the most important metabolic organs, the liver plays vital roles in modulating the lipid metabolism. This study was to compare miRNA expression profiles of the Large White liver between two different developmental periods and to identify candidate miRNAs for lipid metabolism. Methods: Eight liver samples were collected from White Large of 70-day fetus (P70) and of 70-day piglets (D70) (with 4 biological repeats at each development period) to construct sRNA libraries. Then the eight prepared sRNA libraries were sequenced using Illumina next-generation sequencing technology on HiSeq 2500 platform. Results: As a result, we obtained 346 known and 187 novel miRNAs. Compared with the D70, 55 down- and 61 up-regulated miRNAs were shown to be significantly differentially expressed (DE). Gene ontology and Kyoto encyclopedia of genes and genomes enrichment analysis indicated that these DE miRNAs were mainly involved in growth, development and diverse metabolic processes. They were predicted to regulate lipid metabolism through adipocytokine signaling pathway, mitogen-activated protein kinase, AMP-activated protein kinase, cyclic adenosine monophosphate, phosphatidylinositol 3 kinase/protein kinase B, and Notch signaling pathway. The four most abundantly expressed miRNAs were miR-122, miR-26a and miR-30a-5p (miR-122 only in P70), which play important roles in lipid metabolism. Integration analysis (details of mRNAs sequencing data were shown in another unpublished paper) revealed that many target genes of the DE miRNAs (miR-181b, miR-145-5p, miR-199a-5p, and miR-98) might be critical regulators in lipid metabolic process, including acyl-CoA synthetase long chain family member 4, ATP-binding casette A4, and stearyl-CoA desaturase. Thus, these miRNAs were the promising candidates for lipid metabolism. Conclusion: Our study provides the main differences in the Large White at miRNA level between two different developmental stages. It supplies a valuable database for the further function and mechanism elucidation of miRNAs in porcine liver development and lipid metabolism.

Dietary effect of energy levels on growth performance and carcass characteristics of White Pekin duck over 21 days

  • This experiment was conducted to determine the maximum dietary energy levels on growth performance and carcass characteristics of White Pekin duck. the Six dietary treatments were formulated based on their apparent metabolizable energy (AME) concentrations from 2,700 to 3,200 kcal/kg with a 100 kcal/kg gap to evaluate the accurate dietary AME requirement to address current knowledge and further issues for fulfilling the genetic potential of meat-type white Pekin ducklings. A total of 432 one-day-old male White Pekin ducklings were randomly allocated into one of six dietary treatments with six replicates (12 birds per pen). The diets were formulated as corn-soybean meal-based diets to meet or exceed the Nutrient Requirement of Poultry specification for meat-type ducks. Growth performance indices (i.e. average daily gain [ADG], average daily feed intake, feed conversion ratio) were measured weekly. Medium body weight (BW) ducklings from each pen were sacrificed to analyze the carcass traits and abdominal fat content on day 21. Obtained data were analyzed to estimate significant effect using the one-way ANOVA of IBM SPSS Statistics (Version, 25). If the p-value of the results were significant, differences in means among treatments were separated by Tukey's post hoc test. Significant differences were then analyzed with a linear and quadratic broken model to estimate the accurate concentration of AME. Ducklings fed higher dietary AME diets increased (p < 0.05) BW, ADG. Ducklings fed higher AME than 2,900 kcal/kg diets increased abdominal fat accumulation and leg meat portion. The estimated requirement by linear plateau method showed from 3,000.00 kcal/kg to 3,173.03 kcal/kg whereas the requirement by quadratic plateau method indicated from 3,100.00 kcal/kg to 3,306.26 kcal/kg. Collectively, estimated dietary requirements exhibit diverse results based on the measured traits and analysis methods. All the estimated requirements in this experiment present higher than previous research, the maximum requirement for the next diet formulation should be selected by the purpose of the diet.

Influence of Spirulina Level in Diet on Skin Color of Red- and White-colored Fancy Carp Cyprinus carpio var. koi (배합사료 내 Spirulina 함량에 따른 비단잉어 홍백 치어의 체색 변화)

  • This study was conducted to investigate the effects of Spirulina level in the diet on growth and skin color of red- and white-colored fancy carp Cyprinus carpio var. koi at different feeding periods. Five experimental diets (designated as Con, S5, S9, S13 and S17) were prepared to contain 0, 5, 9, 13 and 17% Spirulina. Each experimental diet was fed to two replicate groups of fish (6.6 g/fish) to visual satiation three times a day for 12 weeks. Weight gain and feed efficiency of fish were not affected by dietary Spirulina level. The values of redness ($a^*$), lightness ($L^*$) and yellowness ($b^*$) of the fish skin were influenced by dietary Spirulina level at different feeding period. The $L^*$ value of skin in the fish fed S9, S13 and S17 diets tended to decrease with feeding period. The skin $a^*$ value of the fish increased significantly with feeding period and dietary Spirulina level. The skin $a^*$ value of the fish fed the diets containing 5-17% Spirulina was higher than that of fish fed control diet at 6 and 12 weeks of feeding. The skin $a^*$ value and total carotenoids of the fish fed the S17 diet was the highest among groups. Based on the results of this study, it can be concluded that dietary inclusion of Spirulina powder could increase the skin redness of red- and white-colored fancy carp.

Lysine Requirement of Male White Pekin Ducklings from Seven to Twenty-one Days of Age

  • A dose-response experiment with five lysine levels (0.65, 0.80, 0.95, 1.10, and 1.25%) was conducted to evaluate the lysine requirement of male White Pekin ducklings from 7 to 21 days of age. Two hundred and eighty, 7-day-old, male White Pekin ducklings were allocated to 5 experimental treatments, each containing 8 replicate pens with 7 birds per pen. Feed and water were provided ad libitum from 7 to 21 days of age. At 21 days of age, weight gain, feed intake, feed/gain, breast meat weight, and breast meat yield relative to body weight of ducklings from each pen were all measured. As dietary lysine level increased, weight gain, feed intake, feed/gain, breast meat weight, and breast meat yield of ducklings were all improved significantly (p<0.05). According to broken-line regression analysis, the lysine requirement of male White Pekin ducklings from 7 to 21 days of age for weight gain, feed/gain, breast meat weight, and breast meat yield was 0.84, 0.90, 0.97, and 0.98%, respectively. Considering that Pekin duck production is directed to meat production, the lysine requirement of male starter Pekin ducklings during this period is suggested to be 0.98%.

The Effect of the Hot Water Extract from Taraxacum coreanum Nakai on Hepatocarcinogenesis Induced by N-nitrosodiethylamine in Rats (흰민들레 열수추출물이 N-nitrosodiethylamine로 유발된 흰쥐의 간암 발생에 미치는 효과)

  • The present study was carried out to investigate the hepatoprotective effect of hot water extract of Korean white dandelion (Taraxacum coreanum Nakai) on liver cancer induced in rats by a single dose of N-nitrosodiethylamine (NDEA). The animals were divided into three groups. The animals in the normal (Nor) group were fed basal diet. Control (Con) group were administered with NDEA (200 mg/kg, i.p.). Hot water extract of Korean white dandelion (KDDA, KDDB) group treated with hot water extracts of Korean white dandelion (30, 60 mg/kg/day) for 8 weeks after NDEA. Serum AST and ALT were measured, enzyme activity of liver were examined by spectrophotometer and histological alternation of liver were examined by light microscopy. KDDA and KDDB group were decreased the increase of serum AST, ALT level induced by NDEA. The activities of SOD and CAT were significantly increased in KDDA, KDDB then in the Con. In the histological observation, KDDA and KDDB group were structure of liver lobula and nuclear shape was relatively cleared. In addition, the cytoplasm staining was similar to those of the Nor group. vacuolization of cytoplasm was not observed. These results suggest that administration of hot water extract of Korean white dandelion suppress or retard liver cancer induced by NDEA in rats.

Effects of Dietary Inclusion of Spirulina, Astaxanthin, Canthaxanthin or Paprika on the Skin Pigmentation of Red- and White-colored Fancy Carp Cyprinus carpio var. koi (Spirulina, Astaxanthin, Canthaxanthin 및 Paprika 함유 사료가 비단 잉어(Cyprinus carpio var. koi) 홍백 치어의 체색에 미치는 영향)

  • A feeding trial was conducted to investigate the effects of dietary carotenoid source and level on growth and skin color of red- and white-colored fancy carp Cyprinus carpio var. koi. Nine experimental diets (designated as CON, SP5, SP10, AS50, AS100, CA50, CA100, PA5 and PA10) were formulated to contain 5-10% Spirulina powder, 5-10% paprika powder, 50-100 ppm astaxanthin and 50-100 ppm canthaxanthin. The CON diet without supplementation of carotenoid source was considered as the control diet. Each experimental diet was fed to three replicate groups of fish (3.6 g/fish) to visual satiation three times a day for 8 weeks. Survival, growth and feed efficiency of fish were not significantly affected by dietary inclusion of carotenoid source and level (P>0.05). After 8 weeks feeding period, the values of $a^*$, $L^*$and $b^*$ of fish skin were significantly changed by dietary carotenoid source (P<0.05), but not carotenoid level. The $a^*$ value of fish fed the diets containing CA and PA was higher than control and SP diets (P<0.05). Based on the results of this study, it can be concluded that dietary inclusion of 50 ppm canthaxanthin or 5% paprika powder could increase the redness of red-and white-colored fancy carp fingerling.

  • This experiment was carried out to investigate the growth responses of white clover (Trifolium repens L.) as affected by prohexadione-calcium foliar application for the growth restraint in the lawn yard, field and golf course etc. The leaf length and leaf weight of white clover were significantly retarded compared to the untreated plot with foliar application of prohexadione-calcium after mowing within 1 day, and the effect was continued up to 50 days. The leaf area of clover with prohexadione-calcium treatment was to be narrowed as compared to untreated plot. The SPAD value of the leaf colour was increased in the treated plots and was to be continued for 40 days more. The coverage rate of clover was decreased with prohexadione-calcium foliar application after mowing in the lawn ground.

  • A mixed Lactobacillus acidophilus and Propionibacterium freudenreichii was cultured in the $10\%$ whey broth with $0.5\%$ yeast extract for 4 days. Viable cell numbers of Lactobacillus acidophilus and Propionibacterium freudenreichii were $2.4\ties10^9$ cfu/mL after 36hr and $9.42\times10^8$ cfu/mL after 96 hr. Consumption rate of lactose during fermentation was $87.5\%$. Propionic acid was produced 18.5g/L and acetic acid was produced 4.8g/L, as a result of that, comparison of propionic acid and acetic acid was 3.8:1 Volume of white pan bread was decreased as added amount of whey was increased. Hardess of white pan bread was decresed as added amount of whey was increased. Preservation period of white pan bread with $10\%$ whey fermented product elongated 2 day compared to control.

Nine experimental diets (designated as Con, CP, PA, SP, OP, MB, TO, BE and PO) were formulated to contain Carophyll Pink, red paprika, Spirulina, Opuntia, mandarin bark, tomato, beet and Porphyra, respectively. Each experimental diet was fed to two replicate groups of fish (22.9 g/fish) to visual satiation three times a day for 8 weeks. Weight gain and feed efficiency of fish fed the diets containing Spirulina or Opuntia were higher than those of fish fed the control diet (P<0.05). The values of $a^*$, $L^*$ and $b^*$ of fish skin were significantly changed by dietary carotenoids sources (P<0.05). The $a^*$ values of fish fed the diets containing Carophyll Pink, red paprika and Spirulina were higher than those of fish fed other diets (P<0.05). The skin total carotenoids of fish fed the diets containing Spirulina and Opuntia were higher than those of others (P>0.05). Based on the results of this study, it can be concluded that dietary inclusion of red paprika and Spirulina pacifica could increase the skin redness of red- and white-colored fancy carp.

Frozen Stability of Proteins Recovered from Fish Muscle by Alkaline Processing (알칼리 공정으로 회수한 어육 단백질의 동결 안정성)

  • Frozen stability of proteins recovered from white croaker and jack mackerel have been tested by measuring oxidation of residual lipid, browning, total plate count, and texture of gel during storage at $-20^{\circ}C$. The oxidation of residual lipid in recovered protein from Jack mackerel increased up to 60 days, and then decreased. Both browning values significantly was increased after 90 days. Total plate count was $1.2{\times}10^4\;CPU/g$ for proteins recovered from white croaker and $3.2{\times}10^4\;CPU/g$ for proteins recovered from jack mackerel in 60 days. The breaking force, deformation, and whiteness of gel formed from proteins recovered from white croaker did not change up th 120 days significantly, while proteins recovered from jack mackerel did not form heat-induced gel in 120 day. Frozen storage of the recovered protein was limited to 90 days for white croaker and to 60 days for jack mackerel considering the gelling ability and textural properties.
