Ten to thirty percent of ginko nut shell(GNS) were added to poplar sawdust to investigate mycelial growth. Mycelial growth on the medium with GNS was 93~97(mm/28days) and was slower than that of the control without GNS as that of 101(mm/28 days). Mycelial growth time on medium of sawdust with 10% GNS was same with control without GNS as 28 days but that on medium with 20 to 30% GNS was delayed for 2 days. The time of pinhead formation of sawdust with 10 to 30% GNS was 7 days, the growth time to harvest was 5 days. Yields per volume of GNS added treatments were 80.5g to 91.8g, which was increased 14 to 30% compared to 70.8g of the control without GNS.
1. Aflatoxin free and highly active new type kokja was made with strong two strains of yeast (Y-16, Y-21) and two strains of highly saccharifying active Aspergillus (A-32, A-11). And they were isolated from many kinds of kokja (over hundred) collected from all over Korea. 2. Activities of new type kokja was compared with old type kokja, and results were as following. (i) Saccharifying activity; New type kokja B, C and D were excellent (over 8. 0%), and home made old type kokja were poor quality. (ii) Alcohol fermentation; New type kokja C and D were over 12% and the old type K-12 and K-161 were low activity (less than 10%). (iii) Acid fermentation; Old type kokja K-2o and K-156 (industrial use) were better than the new type C and D. (iv) Fusel oil fermentation; Old type kokja K-23, K-26 (industrial use) and K-12 (home made) fermented high fusel oil (0.07%) and new type kokja C, D and B were the lowest (0.04%). 3. Adding food preservatives (AF-2, D.E.P.C.) and heat sterilizing of wheat bran for kokja had little concerning with the activity of new type kokja. 4. In brewing test, takju made from kokja C and D were more excellent in alcohol and fusel oil concentration than those from old type kokja K-1 and K-23. Taste and odour of takju from new type kokja was good enough to drink.
Mutational experiments were performed to imporve the cellulase productivity of Aspergillus phoenicis KU175, isolated from the southern part of Korea, as a high cellulase producer. By treatment ultra-violet light nad 4-NQO(4-Nitroquinoline-N-Oxide), mutation waas induced, and treatment ultra-violet light and 4-NQO (4-Nitroquinoline-N-Oxide), mutation was induced, and A.phoenicis KU175-115 was finally selected for its highest avicelase production. Avicelase production of the mutant was increased about 2 times compared with those of the wild strain. However, activities of other hydrolytic enzymes, such as amylase, protease and nuclease, of the mutant strain didn't show a marked difference compared with those of the nuclease, of the mutant strain didn't show a marked difference compared with the wild strain, except slight increase in ribonuclease activity and slight decrease in glucoamylase activity. Avicelases from the mutant strain selected were purified from wheat bran culture by successive salting out, followed by dialysis and column chromatography, and their charcteristics were compared with thosw of the wild strain. Avicelase was separated into three peaks in the mutant strain as well as in the case of wild strain. Avicelase II activity of the mutant strain was prominently higher than that of the wild strain, while avicelase I and III activities of those were equivalent. The optimal pH ranges and stability of avicelase II from the mutant strain were pH4-5 and pH3.5-6.0, respectively, as well as in the case of the wild strain. The optimal temperature and thermal stability of avicelase II from the mutant strain were $40{\sim}50^{\circ}C\;and\;20{\sim}55^{\circ}C$, respectively. These results were same as those of the wild strain. By the using of Eadie-Hofastee plot, $K_m\;and\;V_{max}$ of avicelase II from the mutant and the wild strain were calculated to be 2.29mg/ml and $4.84{\mu}g$ reducing sugar as glucose per min equally, from the line fitted to the data by the least square method. Activity of avicelase II from the mutant strain was slightly activated by $Mg^{++}\;but\;inhibited\;by\;Cu^{++}, \;Mn^{++}\;and\;Zn^{++}$, as well as in the case of the wild strain. Therefore, it was concluded that the mutant didn't induce the formation of another avicelase isozyme, or the changes in the properties of avicelase, but induce the changes in the productively of the same avicelase II by the action of regulatory gane.
Currently, plastic bags are being used for sawdust cultivation of shiitake mushroom. However, due to serious environmental problems caused by the use of plastic bags, we studied the efficacy of bottle cultivation method to replace the sawdust bag method. Small detachable plastic bottles (400 g capacity) filled with Quercus spp. sawdust and wheat bran (4:1 w/w) media were incubated for 80 and 120 days. The weight loss (%) of the media was higher for the NIFoS 2464 strain at an approximate light intensity of 300 Lux than light intensity of 500 Lux; the light intensity was associated with the loss of sawdust medium-weight during the cultivation period. The highest yield was observed when the strain was cultivated for 80 days under dark conditions, 40 days under 500 Lux light, and air circulation fan speed of 30 rpm. When incubated for 120 days, mushroom yield in the bottle media was higher at 40 days of light exposure than 20 days of light exposure. In the bottle media incubated for 80 days under dark conditions, the mushrooms fruited due to repetitive water spraying on the top of the media and light stimulation during the fruiting period. The media could be separated from the bottles because the media shrank after the first harvest. These separated plastic bottles could be re-used for mushroom cultivation, thereby reducing the amount of plastic waste.
Three edible species, e.g. Lyophyllum sp. and Armillaria mellea introduced from Japan in 1979 and Auriaularia auricula-judae from Taiwan in 1978 were selected for artifcial cultivation. The media, MSDS for Lyophyllum sp, MHS and PDS for A. auricula-judae, and MSDS and MHS for A. mellea were selected. The range of optimum pH of Lyophyllum sp. was 6.0 and pH 8.0 for A. auricula-judae and pH 6.0 to 7.0 for A. mellea. The range of optimum temperature for the mycelial growth of Lyophyllum sp. was 25 to $30^{\circ}C,\;30^{\circ}C$ for A. auricula-judae and $25^{\circ}C$ for A. mellea. When the moisture content 75 to 80% (w/w) in the substrates, the mycelial growth of Lyophyllum sp. and A. auricula-judae, was rapid. 65% (w/w) for A. mellea. As the spawning substrates, wheat for Lyophyllum sp. oak sawdust for A. auricula-judae and poplar sawdust for A. mellea were selected, respectively. In this experiment, yields of Lyophyllum sp. and A. auricula-judae were high on the poplar sawdust added 20% (w/w) of rice bran. Lyophyllum sp. and A. auricula-judae for sources of new edible mushrooms seems to be possible.
Aspergillus niger S-1 was proved to be a good hesperidinase producer which have been selected for naringinase utilization. Enzyme of this strain had good characteristics and purified relative high degree with good recovery by ammonium sulfate or aceton treatment. Results obtained were summarized as follows (1) The enzyme was most active at 60$^{\circ}C$, when the reaction was performed in the pH 4.0 for 30min. Optimum pH for enzyme activity was 5.0 and activity was retained 78% at pH value 3.5. (2) Hesperidinase activity retained 95% of its full activity after treatment at 60$^{\circ}C$ for 30min at pH value 4.0., 70% at 70$^{\circ}C$ and 65% at 80$^{\circ}C$. Most stable pH of this enzyme was showed 5.0 after treatment for 24hr at 4$^{\circ}C$ (3) Only Magnesium ion activated enzyme reaction, while other metallic ions, Cu$\^$++/, Mn$\^$++/, Pb$\^$++/, Mo$\^$++/, Ag$\^$++/, Hg$\^$++/ inhibited. (4) Eleven fold purification with 35% recovery was obtained in the case of 60% aceton treatment and 10-fold purification with 5.6% recovery was showed with 40% aceton comparing to the crude extract Enzyme. (5) Crude enzyme precipitated with 0.4-0.6 saturated ammonium sulfate contained 13f6 of the original enzyme activity with 48-fold increase in specific activity and enzyme has been purified 25 fold with a yield 19% by 0.6-5.8 saturation. (6) Hesperidinase formation was noticeably increased by addition of small amount of orange-peel extraction on the wheat bran medium.
Studies were made to optimize the simultaneous hydrolysis-fermentation (SSF) process for the production of ethanol from rice straw. Trichoderma sp. KI 7-2 was selected to produced cellulase by solid culture for SSF. Ethanol production was highest when the SSF process utilized koji culture of the fungus grown on a medium of wheat bran-rice straw 3 : 2 mixture with moisture content of 50% adjusted to pH 4.5 for 7 days as the enzyme source. It was found that pretreatment of the substrate is not necessary. To ferment 1g of rice straw by SSF 2.47 units of cellulase were required, and the initial yeast concentration of 2.5$\times$10$^{7}$ cell/$m\ell$ was found to be sufficient. Optimum pH and temperature for the process were 4.5 and 4$0^{\circ}C$, respectively. It was also found that higher ethanol concentration in the broth can be obtained by the addition of substrate or substrate and enzyme to SSF broth.

'Miso', a new variety of oyster mushroom was developed for the bottle culture at the Chungnam Agricultural Research & Extension Services. Its mycelium grew rapidly with $8.4{\sim}8.6\;mm/day $ at $25{\sim}28^{\circ}C$ on PDA medium. The optimum pH of the mycelial growth was pH 5.0. It took 24 days for the primitive primordium formation after inoculation on pine sawdust media mixed with 20% wheat bran. Fruiting body color was white, and the shape of pileus was convex-umbonate. In the bottle culture, the yield was 115.7 g per 850 ml bottle. Stipe length was about 54 mm, the number of stipe per bottle was 18.1, the size of pileus was 28 mm, and gill was crowded. The moisture content of 'Miso' was lower than that of Jangan No.5 and Wonhyeong No.1, and contents of protein, ash and sugar of 'Miso' were higher than those of other varieties, and tannin acid content of 'Miso' was lower than that of others. On the basis of AFLP analysis, the 'Miso' was distinct not only from Wonhyeong No.1, but also from their closest relative, oyster mushrooms.

This study was carried out to develop artificial culture method of Grifola frondosa in polypropylene bag. To find out the favorable substrate formulation of G. frondosa, physicochemical conditions were investigated. The T2 formulation (55 : 25 : 12 : 8) mixing ratio of oak sawdust, oak chip, dried bean-curd refuse and wheat bran showed the shortest time to complete the crop cycle and the highest yield (weight of fresh mushrooms harvested at maturity). Those physicochemical conditions were pH 4.3, 2.4% crude fat contents, 54.1% total carbon, 1.42% total nitrogen, 38.1 C/N ratio, 75.5% porosity and $0.21\;g/cm^3$ bulk density. Among the physicochemical factors, pH, crude fat and total nitrogen may affect mushroom yield. Therefore, development of favorable substrate would be benefit to increase production efficiency of G. frondosa, mushrooms and be commercial potential.
A bacterium producing the extracellular mannanase was isolated from Korean soybean paste. The isolate WL-7 has been identified as Bacillus subtiis on the basis on its 16S rRNA sequence, fatty acid composition, morphology and biochemical properties. The mannanase of culture supernatant was the most active around $55^{\circ}C$ and pH $6.0^{\circ}C$, and retained 90% of its maximum activity at range of pH 5.0∼7.5 and $50∼60^{\circ}C$. The additional carbohydrates including lactose, $\alpha$-cellulose, avicel, locust bean gum (LBG), wheat bran and konjak increased dramatically the mannanase productivity of strain WL-7. Especially, the maximum mannanase productivity was reached to 224 U/ml in LB medium supplemented with both 0.5% LBG and 0.5% konjak, which was approximately 200-folds more than that in LB medium. It was suggested that the increase of mannanase production was owing to induction of mannanase biosynthesis by both LBG and konjak hydrolysates transported following initial hydrolysis by extracellular mannanase during the cell growth.
