Studies on the Cellulase Producing Microorganisms(Part I) -Isolation of the Cellulase Producing Molds and their Cultural Conditions- (Cellulase 생성균(生成菌)에 관(關)한 연구(硏究) (제1보(第1報)) -유용균(有用菌)의 분리(分離) 및 그 배양조건(培養條件)에 대(對)하여-)

  • 94 Cellulase producing strains were isoated from soils, composts, rotten woods and straws, and gastric contents and feces of herbivorous animals in various places. Among them, the strain MC-9, MC-10, MC-53 and MC-61 were found to be highly active in the degradation of carboxy methyl cellulose. Their cultural conditions adequate for the cellulase formation and effects of inorganic salts and various organic substances added to the wheat bran media were investigated. The results obtained are as follows; 1. Optimum conditions for the cellulase formation were MC-9: pH 5.5, temp. $35^{\circ}C$, incubation time 5 days, MC-10: pH 5.5-6.0, temp. $30^{\circ}C$, incubation time 5 days, MC-53: pH 3.5, temp. $30^{\circ}C$, incubation time 5 days, MC-61: pH 3.5-4.0, temp. 30-$35^{\circ}C$, incubation time 5 days. 2. Their cellulase activity in their optimum conditions were MC-9: CMC-LP(liquefying power). 87.7%, CMC-SP(saccharifying power) 3.20 glucose mg./gm. of the cultures/min., MC-10: CMC-LP 82.9%, CMC-SP 2.48 glucose mg./gm. of the cultures/min., MC-53: CMC-LP 72.4%, CMC-SP 1.76 glucose mg./gm. of the cultures/min., MC-61: CMC-LP 87.1%, CMC-SP 2.08 glucose mg./gm. of the cultures/min. 3. Additions of inorganic salts to the wheat bran media were not significant for the cellulase formation, but additions of soybean film and orange-peel pomace promoted the CMC-liquefying power 3 to 5 percent in wheat bran cultures of the strains.

Optimization of Solid-State Fermentation Condition Using Distiller's Dried Grain (주정박을 이용한 고체발효 조건의 최적화)

  • To enhance the value as a feedstuff of distiller's dried grain (DDG) and develop fermented feedstuff, we investigated the effects of the culture conditions affecting glucoamylase activity, such as pH in submerged culture and moisture content in solid-state culture. Also, we investigated the optimal mixing ratio of DDG and wheat bran for the production of fermented feedstuff containing high content of amino acids. In culture conditions for high fermented activity, pH and moisture were optimum at pH 4 and 60%, respectively. In the case of mixing ratio, the glucoamylase activity was decreased with increase of DDG content. On the other hand, the content of crude protein was increased slowly. For the development of fermented feedstuff, the optimal mixing ratio of DDG and wheat bran was 1 to 4. Finally, we could produce approximately 1 ton (dry matter) of trial product in incubator of pilot-scale. The glucoamylase activity and the crude protein content were 1,024 U/g and 33.6%, respectively.

Production and Characterization of α-Galactosidases from Two Bacillus licheniformis Isolates (Bacillus licheniformis 분리균 2종의 α-Galactosidase 생산성과 효소특성)

  • Two bacterial strains, Bacillus licheniformis YB-1413 and YB-1414, producing extracellular α-galactosidase, were obtained from homemade Doenjang. On the basis of their biochemical properties, 16S rRNA sequences and random amplified polymorphic DNA patterns by polymerase chain reaction, they were found to be somewhat different from one another. α-Galactosidase productivities of the two isolates were increased by wheat bran, but drastically decreased by melibiose, raffinose and sucrose which were used as carbon sources. The enzyme productivities were increased by yeast extract as a nitrogen source with maximum levels of 1.87 U/ml for YB-1413 and 1.69 U/ml for YB-1414, respectively. The enzymes of both isolates exhibited maximum activity for hydrolysis of para-nitrophenyl-α-D-galactopyranoside (pNP-αGal) under reaction conditions of pH 6.0 and 45℃. Their hydrolyzing activities for pNP-αGal were drastically decreased by the addition of low concentrations of ribose and galactose. They were capable of hydrolyzing completely α-1,6 linked galactosyl residue in melibiose, raffinose and stachyose, which are known to be anti-nutritional factors in products of soybean and legume. In relation to the latter, the isolates YB-1413 and YB-1414 have potential applicability in improving soybean-fermented foods and the nutritional value of soybean feed.

Energy Balance and Methane Production of Hanwoo Cows Fed Various Kind of Roughage (다양한 조사료 급여조건에서 한우 암소 경산우의 에너지분배 및 메탄배출량)

  • This experiment was conducted to determine methane production and emission factors for a range of roughage diets fed to Hanwoo cows at a level of maintenance energy requirement. Seven cows were fed mixed hay only, 12 cows were fed mixed hay containing 30~50% rice straw and 6 cows were fed a mixed hay diet supplemented with 1.0 kg of wheat bran. Each cow was placed in a metabolic crate for 10 consecutive days, including last 7 days for sampling of feces and urine. At the end of the sampling period, the cows were transferred to an open-circuit respiration chamber for 24 consecutive hours. Methane conversion rate (5.5~6.2%, mean value = 5.8%) and emission factor (33.6~38.6 kg/head/year, mean value = 35.2) were not significantly affected by the diets although the mixed hay only diet resulted in 11 and 15% higher respective rates than the other two. However, in light of the many assumptions that forage with good quality might reduce methane production, additional experiments should be required for participation in the greenhouse gas emission trading system.

  • We investigated the effect of different media components on the characteristics and productivity of Lentinula edodes cultivated in sawdust. Ten substrates were used for this study. Soybean and cotton seed meal were unsuitable substrates as fungal mycelia did not grow to maturity during the incubation period. Lentinula edodes (NIFoS culture number 2462) was grown on the ten substrates, and soybean hull, mixed medium, cotton seed hull and corn grain showed greater effect on the productivity than wheat bran. However, wheat bran induced higher productivity in NIFoS 2778 than the other substrates. Sawdust medium with soybean meal produced smaller fruiting bodies than the other substrates. In contrast, corn meal media produced larger mushrooms than the other substrates. The external characteristics of the mushrooms varied based on the substrates upon which they were grown. This is not surprising given that the substrates differed with respect to carbohydrate and protein content, e.g., cotton seed and soybean meal contained a higher crude protein and crude ash than the other substrates tested.

  • Cultural characteristics of a strain of Streptomyces sp. K-14 (KFCC 35051) producing glucose isomerase were demonstrated. The glucose isomerase was produced when the strain was grown in the medium containing pure xylan or xylan.containing materials such as wheat bran or com cob. The optimum condition was attained in a culture medium composed of 3 % wheat bran or com cob, 2 % com steep liquor, 0.1% $MgSO_4$$7H_2O$ and 0.012 % $CoSO_4$$7H_2O$ for the production of the glucose isomera,e. The production of the enzyme reached to a maximum level when the strain was cultured for 40 hrs $30^{\circ}C$ and pH 7.0.

Effect of Dietary Alfalfa Meal on Egg Yolk Cholesterol Content and Productivity in Laying Hens (산란계 사료에 대한 Alfalfa Meal의 첨가가 난황 콜레스테롤 및 생산성에 미치는 영향)

  • This study was conducted to estimate the effect of dietary alfalfa meal on the contents of egg yolk cholesterol, the levels of egg yolk pigmentation and the performance of laying hens. Alfalfa meal which was alternated wheat bran was supplied 0, 3.5 and 7%, respectively. Total 192 brown laying pullets of 47 weeks old were randomly assigned to the wire cages from July 23 to October 7, 1991 for 10 weeks. The results obtained were summarized as follows : 1. The egg production was highest at 3.5% alfalfa meal level during the entire period. But there was no significant difference among treatments. 2. Egg weight increased in treatments 3.5 and 7% alfalfa meal supplementation significantly. (P<0.01). 3. Even though there was no significant difference, the daily egg mass was the highest at 3.5% of alfalfa meal level. 4. Daily feed intake was the highest at 3.5% of alfalfa meal level. However, feed conversion rate was the lowest among treatments. 5. The feed cost per kg egg mass in the treatment of 3.5% alfalfa meal was significantly lower than the other treatments. (P<0.05). 6. Shell thickness and shell weight in terms of egg shell quality were increased as the levels of alfalfa meal were increased. But there was no significant difference among treatments. 7, The egg yolk Pigmentation was improved significantly as the level of alfalfa meal was increased (P<0.01). 8.899 yolk cholesterol contents were decreased in the treatments of 3.5 and 7% alfalfa meal supplementation compared with those in the treatment of wheat bran only(P >0.05). The results of this study indicate that 3.5% alfalfa meal level was most effective and economical in laying hen.

Studies on Milk-clotting Enzyme of Dothiorella ribis -Part II. Properties of the Enzyme- (Dothiorella ribis가 생산하는 응유효소에 관한 연구 -제 2 보 응유효소로서의 일반적 성질-)

  • The amount of the milk clotting enzyme which is produced by Dothiorella ribis in wheat bran was 950 Soxhlet units per gram of wheat bran. The milk clotting activity of this enzyme was increased by the elevation of clotting temperature and by the increase of the addition of $CaCl_2$ to milk. It was also increased when the pH of milk was lower than that of the fresh milk. When milk was diluted by distilled water, the milk clotting activity of the enzyme was decreased. And its milk clotting activity was good when milk was pasteurized at low temperature. The enzyme of Dothiorella ribis has larger proteolytic activity per Soxhlet unit than that of the milk clotting enzyme of Mucor pusillus Lindt. This enzyme was rather stable between pH 6 and pH 8 when it was conditioned for ten minutes. The heat stability of enzyme was tested by treating it under the condition for $10{\sim}60$ minutes. And the enzyme was stable under the temperature of $45^{\circ}C$. Also the recovery of protein as a form of curd was 76.2 percent to the total protein content of milk.

  • For efficient and economical production of Btrc,illus tlz~rr.ingi~r1~sstirsa in NT0423 as amicrobial-control agents, a new culture medium and culture condition were developed. Five mediadesignatzd as SWI I , SW14, SW23. SW32 and SW4I were prepared ~ : i t hv arious mixture ratio ofsoybean cake and wheat bran. It was founcl that in terms of the cell growth rate and development ofsporulation of B, thrri.il~girrl.sis strain NT0423 in all SW culture media were more efficient than those inGYS and in LB media. Total cell number in all SW media showed similar values, hut SW32 lnediilm wasthe most efficient in the development of spore, which amo~~ntetod 3.7 x 10XC FUImI. Also. at the pHranging frorn 6.2 to 7.3 in the mediiun~ no ad\:erse effect was not made in the culture of B. thur-ingicnsisstrain NT0423. The optimal volume (%) of SW32 mecliuni in a 5 1 fernientor was determined as 4 8\rolume of total niediuni. resulting ill growth (4.2 x 1OTCFUlrnl) of H. t1~~irir1,yirrz.ssit.vr ain NT0423. Study on brewing of sweet potato starch (고구마 전분질원료를 이용한 주류제조에 관한 연구)

  • We have been studied on brewing sweet starch. We obtained the results as follows ; 1) 5 strains, T-T-2, T-T-4, T-K-2, T-T-18, T-T-1, were the most available in view of fermentative power by capacity of $CO_2$. 2) 5 strains, T-T-4, T-T-2, T-T-1, T-T-3, T-K-2, produced capacity of alcohol more than 5.78%. 3) 6 strains, T-T-2, T-K-2, T-T-4, T-S-2, T-I-3, T-I-1, are available not only taste and flavour, but productive power of alcohol in sweet potato starch. 4) The form of 6 strains are long oval and round and most of them are similar to the other yeast in size. 5) In giant colony the color was cream color and cream buff, and T-K-2 was formed by $15{\times}12mm$ on diameter and by 3.5mm on high. 6) Optimum temperature of most of all strains is 25~ $300^{\circ}C$but T-K-4 is 28-30.deg.C. 7) Optimum pH is 3.4-4.6. 8) T-S-2 was died off at 65.deg.C, the other strains died $60^{\circ}C$. 9( Making Bun-kok with non-heated wheat bran .alpha.-amylase was more increased by 4.5-13.5 mg of glucose in reaction solution and .betha.-amylase more 1.6-3.4ml of N/10-$KMnO_4$ Solution than Bun-kok with heated wheat bran. 10) It seems that mycellium grows better than original in substance containing 0.4 ~ 1.2% of HCl. 11) Making Bun-kok to add 0.8% HCl, .alpha.-amylase was increased 9.93-20.7mg of glucose and .betha.-amylase ws increased 2.6~4.3ml of N/10-$KMnO_4$ solution to reaction solution. 12) 1.2%-HCl, or higher concentration, acts as inhibitor, in the meanwhile the concentration between 0.4~0.8% of HCl acts as activator. 13) We must make Bun-kok for 42 hours, at 28~$30^{\circ}C$) After we made Bun-kok using S-O-II and R-J-I one by one, Bun-kok which mix each other in equal quantity is increased more than original on enzyme acrivity. 15) Oxidation is the best way of refining sweet potato starch in N/10-phosphate buffer solution (pH 7.5). 16) When we prepared sweet potato starch, first pH was 3.0.
