The physiological roles of brain angiotensin II in mediating water deprivation-induced drinking and in regulating renal renin release were assessed in male Sprague-Dawley rats. Specific $AT_1$ receptor antagonists, losartan and SK 1080, and antisense oligonucleotide (AS-ODN) directed to $AT_1$ receptor mRNA were intracerebroventricularly (i.c.v.) administered in conscious unrestrained rats. When water was given 20 min after i.c.v. injection of $AT_1$ receptor antagonists in 48-h water-deprived rats, losartan and SK 1080 produced approximatly 20% and 50% decrease in 1-h water intake, respectively. In contrast, i.c.v. treatment of the AS-ODN to $AT_1$ receptor mRNA for 24-h did not alter 1-h water intake in 24-h water-deprived rats, but prevented the increase in overnight water intake after 24-h water-deprivation. Six-day i.c.v. treatment of AS-ODN did not alter either the basal plasma renin concentration or renal cortical levels of renin and renin mRNA. The present results suggest that endogenous brain Ang II plays an important role in thirst and water intake through $AT_1$ receptors, but further studies are required to elucidate its regulatory role in renal renin synthesis.
The water extract of Hwansodan has been traditionally used for treatment of ischemic brain damage in oriental medicine. However, little is known about the mechanism by which the water extract of Hwansodan rescues cells from neurodegenerative disease. PC12 pheochromocytoma cells have been used extensively as a model for studying the cellular and molecular mechanisms of neuronal cell damages. Under deprivation of growth factor and ischemic injury, PC12 cells spontaneously undergoes apoptotic cell death. Serum and glucose deprivation markedly decreased the viability of PC12 cells, which was characterized with apparent apoptotic features such as membrane blebbing as well as fragmentation of genomic DNA and nuclei. However, the aqueous extract of Hwansodan significantly reduced serum and glucose deprivation-induced cell death and apoptotic characteristics through reduction of intracellular peroxide generation. Pretreatment of Hwansodan also ingibited the activation of caspase-3, in turn, degradation of ICAD/DFF45 was completely abolished in serum and glucose deprivated cells. Furthermore, pretreatment of Hwansodan obviously increased heme oxygenase 1 (HO-1) expression in PC12 cells. Taken together, the data suggest that the protective effects of Hwansodan against serum and glucose deprivation induced oxidative injuries may be achieved through the scavenging of reactive oxygene species accompanying with HO-1 induction.
Under sulfur deprived conditions, PS II and photosynthetic $O_2$ evolution by Chlamydomonas reinhardtii UTEX 90 are inactivated, resulting in shift from aerobic to anaerobic condition. This is followed by hydrogen production catalyzed by hydrogenase. We hypothesized that the photosynthetic capacity and the accumulation of endogenous substrates such as starch for hydrogen production might be different according to cell age. Accordingly, we investigated (a) the relationships between hydrogen production, induction time of sulfur deprivation, increase of chlorophyll after sulfur deprivation, and residual PS II activity, and (b) the effect of initial cell density upon sulfur deprivation. The maximum production volume of hydrogen was 151 ml $H_2$/l with 0.91 g/l of cell density in the late-exponential phase. We suggest that the effects of induction time and initial cell density at sulfur deprivation on hydrogen production, up to an optimal concentration, are due to an increase of chlorophyll under sulfur deprivation.
Objective: Combination of two stressors on alteration of mineral footprints in animals needs due attention to meet maximum production and welfare, particularly in grazing sheep. This study tested whether ewes (Ovis aries) exposed to water deprivation and thermal-humidity stressors had altered mineral footprints in their wool, serum, urine, and feces. Methods: Nine ewes (age = 3 years; mean body weight = 41±3.5 kg) were divided among a control group with free access to water, and treatment groups with water deprivation lasting either 2 h (2hWD) or 3 h (3hWD) after feeding. Using a 3×3 Latin square design, animals were assigned to treatment groups for three sampling periods of 21 days each (n = 9). Blood was collected by jugular venipuncture. Wool was collected at the end of periods 2 and 3. Metabolic crates designed with metal grated floors were used for urine and feces collection. We measured sodium (Na), magnesium (Mg), phosphorus (P), chloride (Cl), calcium (Ca), manganese (Mn), copper (Cu), iron (Fe), and zinc (Zn). Results: The wool mineral levels did not differ between the treatment groups, although K was marginally lower (p = 0.10) in the 2hWD group. The serum and urine mineral levels did not differ between the treatments (p>0.05). Fecal K was significantly lower in the 2hWD group than in the other groups (p≤0.05). Conclusion: In conclusion, water deprivation and thermal-humidity exposure altered the excretion of K, but not of other minerals, in the wool, urine, feces, or serum of ewes. Thus, no additional mineral supplementation is needed for water deprived ewes during thermalhumidity exposure.

An experiment was conducted to study the effect of post-hatch feed deprivation on yolk sac utilization andsubsequent performance of young broiler chickens (280) up to 35 days of age. The experimental treatments included access to feed at 8 h intervals after hatch, up to 48 h (0, 8, 16, 24, 32, 40 or 48 h). Water was offered ad libitum to all the groups immediately after placement. Results indicated that chicks with access to feed immediately after hatch used up the residual yolk more quickly. Access to feed between 8-24 h post-hatch, supported faster utilization of residual yolk compared to those chicks that remained unfed for 40-48 h (p<0.05). Further, deprivation of feed up to 24 h did not alter the lipid and protein contents in residual yolk, but fasting of chicks beyond 24 h (32, 40 and 48 h) led to retention of higher lipid and lower protein content in the yolk sac (p<0.05). At 7 days of age, the weights of proventiculus and gizzard were not affected by feed deprivation up to 48 h. However, the liver, pancreas and jejunum recorded significantly (p<0.05) heavier weights in chicks that were fed during the initial 24 h period compared to delayed feeding (32-48 h). Chicks fed within 24 h after hatch gained significantly (p<0.05) higher weight at 5 weeks of age than those that received feed between 32 and 48 h. Feed deprivation for 48 h was more detrimental to growth than 24-40 h. This study revealed the significance of early posthatch feeding (<24 h) on faster utilization of yolk sac nutrients and optimum development of intestines and organs, culminating in improved weight gain (>10.5%) of broilers at 5 weeks of age.
Chlamydomonas reinhardtii is a green algae that can use light energy and water to produce hydrogen under anaerobic condition. This work reports the effect of limiting factors on hydrogen production in sulfur deprived anaerobic C. reinhardtii culture. In order to confirm the relationship between hydrogen production and limiting factors such as residual PSII activity and endogenic substrate degradation, the increase in chlorophyll concentration and the decrease in starch concentration was investigated during sulfur deprivation. The overall hydrogen production increased depending on cell density in range of $0.4{\sim}0.96\;g$ DCW/l. At this time, the increase in chlorophyll concentration during 24 h after sulfur deprivation increased in proportion to hydrogen production, however, the decrease in starch concentration was not proportional to that. Therefore, hydrogen production under sulfur deprivation using green alga was closely associated with the residual PSII activity than the endogenic substrate degradation.

Purpose: The aim of this study was to identify the effects of environmental factors on depressive-like behavior and memory function during adolescence. We performed behavior tests in adolescent rats exposed to environmental enrichment, handling, and social deprivation for eight weeks. Methods: Wistar rats were randomly assigned to control, environmental enrichment, handling, and social deprivation groups at the age of four weeks. Results: In the forced swim test, the immobility time in the environmental enrichment group was decreased than that in the control group (p=.038), while the immobility time in the social deprivation group was increased than that in the control group (p=.035), the environmental enrichment group (p<.001), and the handling group (p=.001). In the Morris water maze test, the social deprivation group had an increased latency time than the control group (p=.013) and the environmental enrichment group (p=.001). In the passive avoidance test, the environmental enrichment group had an increased latency time than the control group (p=.005). However, the social deprivation group had reduced latency time than the socially housed groups (control: p=.030; environmental enrichment: p<.001; handling: p<.001). Conclusion: These findings suggest that environmental factors play an important role in emotion and memory function during adolescence.
Objectives : This work was designed to investigate the effect of The root of Achyranthes japonica Nakai (AJN) water extract on serum deprivation reperfusion-induced apoptosis in PC-12 cells and transient middle cerebral artery occlusion (tMCAO)-induced ischemic brains of rats. Methods : Apoptosis in PC12 cells was induced by serum deprivation and reperfusion. The cells were treated with AJN water extract at doses of 0.5 and 1.0 mg/ml for 24 hr after inducing the apoptosis. Cell viability was determined by WST-1 assay. The expression of caspase-3 protein was determined by Western blot. Ischemic brains were prepared from tMCAO-induced ischemic rats after oral administration with AJN at dose of 50 and 100 mg/kg, and then brain infarction was measured by TTC staining. Results : AJN significantly increased the cell viability in apoptocic-induced PC-12 cells, and also decreased the expression of caspase-3 protein. Furthermore, the administration of AJN significantly inhibited tMCAO-induced brain infarction in rats. Conclusions : Our results suggest that AJN extract has a neuroprotective property via suppressing the apoptosis in PC12 cells and the infarction of ischemic brains.
Objectives : This study was designed to investigate the neuroprotective effect of Hwansodan(Huanshao-dan) on the apoptosis induced by withdrawal of neurotrophic support. Methods : PCl2 pheochromocytoma cells have been used extensively as a model for studying the cellular and molecular effects of neuronal cells. The viability of cells was measured by MTT assay. We used DNA fragmentation and caspase 3-like protease activation assay. Results : The water extract of Hwansodan(Huanshao-dan) significantly showed protective effects on serum and glucose deprivation-induced apoptotic death. Hwansodan(Huanshao-dan) also prevents DNA fragmentation and caspase 3-like protease activation, representing typical neuronal apoptotic phenomena in PCl2 pheochromocytoma cells and induces tyrosine phosphorylation of proteins around 44 kDa, which was identified as ERK1 with electrophoretic gel mobility shift by Western blot. In addition, MAPK kinase(MEK) inhibitor PD98059 and Ras inactivator, ${\alpha}-hydroxyfarnesylphosphonic$ acid attenuated the neuroprotective effects of Hwansodan(Huanshao-dan) in serum-deprived PCl2 cells. Conclusions : These results indicate that Ras/MEK/ERK signaling pathway plays a key role in neuroprotective effects of Hwansodan(Huanshao-dan) in serum-deprived PCl2 cells. Taken together, we suggest the possibility that Hwansodan(Huanshao-dan) might provide a neurotrophic-like activity in PCl2 cells.
