Comparative Analysis of Microcystin during Water Treatment Process between Real-Time PCR and LC/MS (Real-Time PCR법과 LC/MS법을 이용한 수계중의 마이크로시스틴 검출방법 비교연구)

  • We performed a comparative analysis using a Real-time PCR (Polymerase Chain Reaction) and LC/MS (Liquid-Chromatograph/Mass Spectrometer) method in order to detect microcystin in environmental sources. Among the three different primer sets tested for microcystin using three positive strains of Microcystis aeruginosa by Real-time PCR assay, only TOX2P/TOX2M primer pairs were able to detect Microcystis aeruginosa. According to the results of a survey carried out from June 2009 to September 2009, 11 out of 11 (100%) raw water samples were were found to have microcystin when the Real-Time PCR and LC/MS method was used, with total microcystin concentration ranging from 5.98~219.0 ${\mu}g/l$. A microcystin removal treatment process was used to ensure entire removal, by passing it through a BAC filtration step. It was concluded that real-time PCR assay can be used to estimate micrucystin detection more rapidly and easily than the LC/MS method.

New test method for real-time measurement of SCC initiation of thin disk specimen in high-temperature primary water environment

  • In this study, a new rupture disk corrosion test (RDCT) method was developed for real-time detection of stress corrosion cracking (SCC) initiation of Alloy 600 in a primary water environment of pressurized water reactors. In the RDCT method, one side of a disk specimen was exposed to a simulated primary water at high temperature and pressure while the other side was maintained at ambient pressure, inducing a dome-shaped deformation and tensile stress on the specimen. When SCC occurs in the primary water environment, it leads to the specimen rupture or water leakage through the specimen, which can be detected in real-time using a pressure gauge. The tensile stress applied to the disk specimen was calculated using a finite element analysis. The tensile stress was calculated to increase as the specimen thickness decreased. The SCC initiation time of the specimen was evaluated by the RDCT method, from which result it was found that the crack initiation time decreased with the decrease of specimen thickness owing to the increase of applied stress. After the SCC initiation test, many cracks were observed on the specimen surface in an intergranular fracture mode, which is a typical characteristic of SCC in the primary water environment.

  • Miami Isopycnic Coordinate Ocean Model is applied to the East Sea to estimate the renewal time of the upper Intermediate Water The model gives about 10 years of renewal time. Extrapolating this result to the whole water mass below, including the upper Intermediate Water, leads to about 81.4 years of renewal time, which is quite comparable to that obtained by Kim and Kim (1997) based on the recent observations. Deep winter mixing occurs in the north of the basin. The areas of the largest water mass conversion, from the upper mixed to the intermediate below, are along the periphery of the deep mixing zone. Large portion of the renewed Intermediate Water then advects along the Korean and Japanese coasts. It is concluded that the high-oxygen content Intermediate Water found off the Korean coast (Kim and Chung, 1984) is in part locally formed but mostly advected from the deep mixing zone.

  • In order to establish the methodology for estimating an emergency drinking water supply in Korea, overseas cases and accidents history of cutting off water supply were investigated, and questionnaire was conducted. Investigating accidents history of cutting off water supply in Korea, actual cutting off times of most cases were less than 13hours. Also, cases related with water quality and facility failure have been not enough to derive useful information for estimating an emergency water. From the results of questionnaire and cross-tabulation analysis, about 1,066 lpcd(liter per capita${\cdot}$day) as an emergency water and 14 days as tolerable outages time could be estimated. The results of water quality simulation could tell us that it might take 5-16 days for pollutant matter to travel from 15 contamination points to source water intake point in the selected reservoir(D-dam). This travel time was in good accordance with the estimated tolerable outage time, 14days.

Characterization of gas-water flow in tight sandstone based on authentic sandstone micro-model

  • Eight tight sandstone reservoir samples from $He_8$ and $Shan_1$ Formations of the Sulige Gas field were selected to perform gas-water micro-displacement experiment based on authentic sandstone micro-model. The gas pressure-relief experiment was proposed for the first time to simulate the pressure change and gas-water percolation characteristics in the process of gas exploitation. The experiment results show that: (1) In the process of gas accumulation, the gas preferentially flows into the well-connected pores and throats with large radius, but rarely flows into the area without pores and throats. (2) Under sufficient gas drive, the water in pores and throats usually exists in the forms of 'thin water film', 'thick water film', and 'water column', but under insufficient gas drive, gas fails to flow into new pathways in time, so that the reservoirs with large pores and throats are high in water cut. (3) Under the same water saturation, the reservoirs with better petrophysical properties has higher gas recovery factor within unit time. Under the same petrophysical conditions, the reservoirs with lower water saturation show higher gas recovery factor within unit time. The higher the permeability, the stronger the liquid carrying capacity of reservoirs.

Evaluation of Water Supply Adequacy using Real-time Water Level Monitoring System in Paddy Irrigation Canals (실시간 관개수로 수위 모니터링을 활용한 논 관개용수 공급적정성 평가)

  • Appropriate amount of water supply to paddy fields in proper time is important to achieve efficient agricultural water management. The purpose of this study is to evaluate the irrigation water supply adequacy for paddy fields using water level data in irrigation canals. For the evaluation, the real-time water level data were collected from main canals in the Dongjin irrigation district for 2 years. Using the water level data, delivered irrigation water amounts at the distribution points of each canal were calculated. The water balance model for paddy field was designed considering intermittent irrigation and the irrigation water requirement was estimated. Irrigation water supply adequacy was analyzed from main canals to the irrigation blocks based on the comparison between estimated requirement and delivered irrigation water amounts. From the adequacy analysis, irrigation water supply showed poor management condition in 2012 with low efficiency except the Daepyong canal section, and the adequacy in 2013 was good or fair except the Yongsung canal section. When irrigation water for paddy fields was insufficient, water supply adequacy was affected by irrigation area, but when irrigation water was enough to supply, adequacy was affected by distance from main canal to distribution points. These results of the spatial and temporal dimensions of the irrigation adequacy could be utilized for efficient irrigation water management to improve the temporal uniformity and equity in the water distribution for paddy fields.

  • Waste-to-energy facilities including incinerators are known as an efficient method to reduce wastes. In waste-to-energy facilities, more efficient cooling system is still needed for grates as the energy density of waste increased. For better cooling performance with the water-cooled grates, optimal design of cooling water pathways is highly beneficial. We performed numerical investigation on fluid flow and residence time of cooling water with change of the geometry of the cooling water pathway. With addition of round shaped guide vanes in the water pathway, the maximum residence time of flow is reduced(from 4.3 sec. to 2.4 sec.), but there is no significant difference in pressure drop between inlet and outlet, and average residence time at the outlet. Furthermore the flow stagnation region moves to the outlet, as the position of the round shaped guide vanes is located to the neck point of pathways.

  • In this study we analyzed and interpreted the phenomenologlcal characteristics and meanings of water space in religious architecture through field studies and phenomenologlcal approaches, focusing on the religious architecture Introducing water space in contemporary architecture. The phenomenological experience elements of the research ate light, time, place and context. As the results of this study, first, light as a phenomenological element not only induces a contrast and change between light and the shadow in architectural space and various changes of light in inner space, but also operates as an element which expresses building features outward with time. Second, time as a phenomenological perception experience makes us feel changes of the environment and physical properties and experience space visually and physically. The continuity of time shows the change of a day, night and daytime and seasons. Third, water space makes the sense of place more remarkable, connected closely with surrounding nature In outer space, and creates a new space. Fourth, water space is a contextual element which harmonizes with surrounding environment by connecting surrounding natural environment with architectural space. Fifth, water space in religious architecture is a symbolic and central element, and emphasizes the atmosphere of architectural space. In addition, it makes the people who experience water space perceive the place by triggering sensitive response.

Stochastic Properties of Water Quality Variation in Downstream Part of Han River (한강 하류부의 수질변동에 대한 추계학적 특성(I) - 특히 뚝도 및 노량진 지점의 DO, 탁도, 수온의 변동을 중심으로 -)

  • The stochastic variations and structures of time series data on water quality were examined by employing the techniques of autocorrelation function, variance spectrum, Fourier series, autoregressive model and ARIMA model. These time series included hourly and daily observation on DO, turbidity, conductivity pH and water temperature. The measurement was made by automatic recording instrument at Noryangjin and Dook-do located in the downstream part of Han River during 1975 and 1976. Hourly water quality time series varied with the dominant 24-hour periodicity, and the 12-hour periodicity was also observed. An important factor affecting 24-hour periodic variation of DO is believed to be photosynthesis by algae. These phenomena might be attributable to periodic discharges of municipal sewage. Noryangjin site showed the more distinct 12-hour periodicity than Dook-do site did, and tidal effect might be responsible for the difference. The water quality, as measured by DO and turbidity, was better in the afternoon compared with the quality in the morning. This change can be explained by the periodic variation of DO, temperature and the amount of municipal wewage discharge. It was also observed that the water temperature at Noryangjin was higher than the temperature at Dook-do. This difference might have been caused by the pollutants that were added to the section between two sites. The correlation coefficients between some of the variables were fairly high. For example, the coefficient was -0.88 between DO and water temperature, 0.75 between turbidity and river flow, and 0.957 between water temperature and air temperature. The lag time of heat transfer from the air to the water was estimated as 24 days. The first order auto-regressive model was appropriate for explaning standardized hourly DO time series. The ARIMA model of (1, 0, 0) type provided relatively satisfactory results for daily DO time series after the removal of significant harmonic value.

  • In this paper, real-time water quality monitoring system of small water supply facilities based on IEEE 802.15.4e-2012 DSME MAC and IEEE 802.15.4g-2012 PHY standard is presented, which is capable to acquire for highly reliable water quality information in the wide outdoor areas for effective water quality management of small water quality facilities is distributed in the long distance and remote areas. Previously, Long distance transmission is difficult in most water quality sensor module is using RS-485 protocol. But with this system, even in harsh outdoor environment, it is possible to establish a radio wave sensor in a wide area network, and not only water quality sensor shall be connected to the wireless system, but also wireless integrated management system shall provide more effective way of management of the numerous small water supply facilities spread throughout the community, so that the administrator can remotely monitor the data of water turbidity, pH, residual chlorine in the water-supply, water-level, and generate alarm to cope with risks. The management of small water facilities is done by residents will be very effective to notice water quality information of small water facilities to residents.
