Hatching Rate and Larval Viability of Cultured Marine Fish Exposed to Water-soluble Fraction of Kuwait Crude Oil during Egg Development (난발생중 원유의 수용성 성분에 노출된 해산 양식어류 수정란의 부화율 및 자어의 생존능력)

  • Exposure experiments during the egg development were conducted to assess the influences of 5 different concentrations (0, 25, 50, 75 and 100%) of water-soluble fraction (WSF) of Kuwait crude oil on the eggs and larvae of black seabream (Acanthopagrus schlegeli), red seabream (Pagrus major) and olive flounder (Paralichthys olivaceus). All experiments were triplicated. Hatching time and hatching rate were examined on the eggs. The median lethal time ($LT_{50}$), morphological abnormality and swimming activity (swimming frequency and speed) of larvae were also investigated. The time and rate of egg hatching were not significantly influenced by WSF on the eggs of the fishes. The larvae exposed to WSF during the egg development were also not significantly influenced on the $LT_{50}$ and swimming activity. But the higher morphological abnormalities of notochord were observed from the larvae in 100% WSF exposure.

  • The musk has been reported to have significant anti-inflammatory activities in clinical use and several animal models and we examined the effects of water soluble fraction(WSF) of the musk on murine peritoneal macrophages. WSF decreased the production of nitric oxide from the lipopolysaccharide(LPS)-treated murine peritoneal macrophages and also reduced the phagocytic activity of macrophages on the opsonized sheep red blood cells(SRBC). Transcriptional expression level of the inducible nitric oxide synthase(iNOS) was also decreased and the viability of the treated macrophages was not affected by WSF, suggesting that the effects could be partly explained by transcriptional regulation. Contrary to down-regulating iNOS expression, WSF slightly increased the release of tumor necrosis factor-alpha$(TNF-{\alpha})$, which implied its selective action on cellular pathways activated by LPS. Our results showed that anti-inflammatory activities of the musk could be partly explained by the inhibitory effects of the water soluble fraction on the macrophageal activation.

Effect of Water Soluble fraction from Japanese Larch Wood on Sawdust Cultivation of Lentinula edodes (일본잎갈나무재의 수용성추출물 첨가가 표고버섯의 톱밥재배에 미치는 영향)

  • The water soluble fractions(WSF) from Japanese larch wood were isolated, purified by anion exchange resin and Sephadex gel filtration and identified its chemical structure by means of periodate oxidation and methylation reactions. Its major components are arabinose and galactose (1 : 3.4). Based on the results of periodate oxidation, methylation and gas chromatographic analysis of purified WSF, main chain is composed of β-1,3-glycosidic linkage among D-galactopyranoses, and two different side chains; β-1,6-glycosidic linkage among 2-3 units of D-galactopyranoses and β-1,6-glycosidic linkage between 1-2 units of D-galactopyranose and L-arabinopyranose. Addition of WSF to culture media of oak mushroom (Lentinula edodes) accelerated the mycelial growth. In the case of PDA cultures, 2 percent addition of WSF in Sanlim No. 6 strain and 4 percent of WSF in Mok-H strain mostly enhanced the mycelial growth of the mushroom. In the case of sawdust cultures, 4 percent addition of WSF in two strains showed the best mycelial growth. High percentages addition of WSF inhibited mycelial growth of the mushroom. Mushroom production was increased with addition of WSF. By the addition of WSF, ergosterol contents in the media were quite high at the colonized stage and rapidly increased at the fruiting stage. Therefore the ergosterol content could be utilized as an indicator to evaluate the culture maturity for the mushroom fruiting.

Effects of the Water Soluble Fraction from Kuwait Crude Oil on the Early Larval Stages of Three Cultured Fish Species (3종 양식어류의 전기자어기에 미치는 원유 수용성 획분(WSF) 의 영향)

  • Bioactivity growth and survival rte of yolk sac larvae in cultured black seabream (Acanthopogrus schlegeli) red seabream (Pagrus major) and olive flounder (Paralichthys olivaceus) to 5 different concentration (0, 25, 50, 75 and 100%) of water soluble fraction (WSF) of Kuwait crude oil were studied. The 75% and 100% of WSF concentration influenced negatively on the length growth of larvae in each species so the growth rates of total length in the concentration of WSF were significantly lower than those of control. The high concentration (50%, 75%, and 100%) of WSF influenced negatively not only on the swimming frequency and speed but also on the morphological abnormality of larvae. Red seabream larvae showed the highest rate of abnormality by WSF among the tested fishes. The fish larvae exposed during pre-and post-hatch (egg and larval stages) and during post-hatch (larval stage only) to the WSF were significantly influenced on their survival rate. The larvae of red seabream were more sensitive to WSF than those of black seabream and olive flounder. The survival rates of larvae exposed to higher concentration (50%, 75%, and 100%) of WSF were significantly lower than those of the control but those of the 25% group were not.

Effect of Crude Oil (WSF) on Early Life Stage of the Pacific Oyster, Crassostrea gigas (원유의 WSF (water soluble fraction)가 참굴의 초기생활사에 미치는 영향)

  • The observed the toxic effect of WSF (Water Soluble Fraction) on development time, development rate, attachment rate, survival rate and growth of the larva during the early stage of the Pacific oyster, Crassostrea gigas. The time required early juvenile from the fertilized eggs exposed to crude oil (WSF) was about 13 hours in control group. But that of the crude oil dose group takes 12 hours, with shorter compared to control group. Development and attachment rate of the oyster significantly higher in 0.4 and 0.8 mg/L WSF compared to control group (P<0.05) but significantly lower more than 1.6 mg/L WSF (P<0.05). Survival rate of the oyster larva significantly higher in 0.4 and 0.8 mg/L WSF compared to control group (P<0.05) but significantly lower more than 2.4 mg/L WSF (P<0.05). Survival rate of spot showed no significantly difference compared to control group (P>0.05). Shell growth of the oyster were significantly lower more than 1.6 mg/L WSF compared control group (P<0.05). The effect concentration of crude oil showed that more 0.4 mg/L in development and attachment rate and more 1.6 mg/L in growth. Also, chronic exposure of crude oil even effect also in low concentration.

Effects of the water soluble fraction of crude oil on the abalone, Haliotis discus hannai (Ino, 1952) early life stage (WSF(Water Soluble Fraction) 가 북방전복, Haliotis discus hannai (Ino, 1952) 의 유생 및 치패발생에 미치는 영향)

  • This study was carried out to investigate the toxic effect of WSF (Water Soluble Fraction) on development time, development rate, attachment rate, survival rate and growth of the larvae during the early life stage of the abalone, Haliotis discus hannai. Also, observed effect of water temperature on the survival rate and histological change of gill in the early young shell. In the abalone, it takes around 12 hours in all experimental groups. Development and attachment rate of the abalone significantly lower more than 0.4 mg/L WSF compared to control group (P < 0.05). Survival rate of abalone larva and spot was significantly lower more than 0.4 mg/L and 2.4 mg/L WSF compared to control group, respectively (P < 0.05). Shell growth of the abalone were significantly lower more than 2.4 mg/L WSF compared control group (P < 0.05). Survival rate lower more than $25^{\circ}C$ exposure group compared water temperature $17^{\circ}C$ exposure group in the early young shell. The gill of abalone exposed water temperature $17^{\circ}C$ and $25^{\circ}C$ was showed atrophy of nucleus and breakdown of the filament, vacuolation of filament epithelial cell.

Stability of Anti-Helicobacter pylori IgY Under Some Condition (Anti-Helicobacter pylori IgY의 처리조건에 따른 안정성에 관한 연구)

  • The aim of this experiment was to evaluate the stability of anti-Helicobacter pylori IgY in water soluble fraction(WSF) of egg yolk according to the heat, pH and digestive enzyme treatment. Anti-H. pylori IgY content of WSF remained 76% after pasteurization(63$^{\circ}C$ for 30 min). The stability of anti-H. pylori IgY at different pH showed a tendency to diminish according to decreasing pH from 7.0 to 1.5(p<0.05). Anti-H. pylori IgY content was 84.4% after treatment for 1 hour at 37$^{\circ}C$ in pH 5.0. There were significantly differences in IgY content between 1 hour and 2 hours at pH 2.0 in 200 units of pepsin treatment(p<0.05). However, IgY was relatively stable at pH 4.0 regardless of the reaction time and the concentration of pepsin. The stability of IgY of egg yolk after the treatment of trypsin was significantly higher than that of water soluble fraction (p<0.05). This results indicated that anti-H. pylori IgY showed relatively a good stability on heat, pH and digestive enzyme.

Effect of Crude Oil on Early Life Stage of the Flounder, Paralicthys olivaceus (원유의 WSF (Water Soluble Fraction)가 넙치, Paralicthys olivaceus의 초기생활사에 미치는 영향)

  • This study was carried out to investigate the toxic effect of WSF (water soluble fraction) in crude oil on the hatch, survival rate, abnormality and physiological activity of fertilized eggs and early larvae in Paralicthys olivaceus. The time required in hatching the fertilized eggs exposure to crude oil was 50.8${\sim}$53.2 hours both in control group and experimental group, showing no significant difference(p>0.05). The hatching rate in the control group was more than 80% in 1.9 ${\mu}g/mL$ WSF concentration, but hatching rate was less than 55.7% in below of 3.9 ${\mu}g/mL$ WSF concentration, showing significant difference (p<0.05). The survival rate on the 3rd day of early larval stage was 61.96% with 1.9 ${\mu}g/mL$ WSF, 11.1% with 3.9 ${\mu}g/mL$ WSF, and they all died with other concentration levels. The oxygen consumption rate in experimental groups was lower than control group. The heart rate (no./min) was 47.4${\sim}$52.8 before hatching and there was no significant difference between control group and experimental group, but heart rate reduced with the decrease of WSF concentration after hatching. The abnormality was 1.1% in the control group whereas 36.7% with 3.9${\mu}g/mL$ WSF, and abnormality was mostly comprised of incomplete spinal formation and spinal curvature. The results of this study suggest that even low concentration to WSF affects the early development of the flounder, Paralicthys olivaceus.

Tainting and Depuration in Fish by Petroleum Hydrocarbon (유류오염이 어체에 미치는 영향과 유성분 제거에 관한 연구)

  • A series of short (8 hours) and long term (96 hours) exposure studies was conducted on a laboratory scale to evaluation (1) the threshold concentrations of petroleum hydrocarbon in seawater which can lead to tainting problems in yellow tail (Seriola quinqueradiata) and (2) the time to recover (depuration period) once the tainting has occurred. The water-soluble fraction (WSF) of crude oil was prepared by stirring the oil with cold seawater. The main component of the WSF were low-boiling aromatics, although these were only al small proportion of the starting oil. From the sencory evaluation it was concluded that the threshold hydrocarbon levels in seawater which will impart a taint in yellow tail fillets within a 8-hour exposure period (short exposure period) are in the range of 0.4 to 1.0 ppm and within a 96-hour exposure period (long exposure period) are in the range of 0.2 to 0.5 ppm. Depuration trials were carried out with WSF from crude oil. The fish were exposed for 48 hours to concentration of 2.5 ppm hydrocarbons. After 48-hour exposure period, the fish were allowed to depurate in fresh, uncontaminated seawater. Depuration time for these fish was 10 days. The taste panelists were able to detect the contaminated fillets very easily while the depurated fish could not be distiguished from the control.

Polysaccharides from Edible Mushroom Hinmogi (Tremella fuciformis) Inhibit Differentiation of 3T3-L1 Adipocytes by Reducing mRNA Expression of $PPAR{\gamma}$, C/$EBP{\alpha}$, and Leptin

  • Water-soluble fraction (WSF) from edible mushroom hinmogi (Tremella fuciformis) were obtained by water extraction, and polysaccharides in the WSF were separated by ethanol precipitation. The inhibitory effects of the polysaccharides on 3T3-L1 adipocyte differentiation were evaluated by the reduction of peroxisome proliferators-activated receptor ${\gamma}$ ($PPAR{\gamma}$) translation, triglyceride accumulation, Oil Red-O staining, and expression levels of $PPAR{\gamma}$, CCAAT/enhancer binding protein a (C/$EBP{\alpha}$), and leptin. The $PPAR{\gamma}$ translation in 3T3-L1 cells was inhibited by the treatment with polysaccharide precipitated by 80% ethanol (P80) which showed highest inhibitory activity among polysaccharides tested. In addition, treatment of P80 to 3T3-L1 cells significantly inhibited the triglyceride accumulation, Oil Red-O staining, and mRNA expression of $PPAR{\gamma}$, C/$EBP{\alpha}$, and leptin in a dose-dependent manner. Based upon these results, P80 from edible mushroom hinmogi shows the inhibitory activity on the differentiation of 3T3-L1 adipocytes. Therefore, it might be employed as a potential anti-obesity material.
